Abstract:Triphenylmethane dyes are widely used in textile dyeing,medicine,biological staining,paper,leather,food and cosmetic industries. Their special stable chemical structures make them difficult to be degradated and decolorized. The biodecolorization research of triphenylmethane dyes will provide the fundamental knowledge for the treatment of dye-containing wastewater and bioremediation of dye-contaminated environment. In this paper,the recent research progress in biodegradation and decolorization of triphenylmethane dyes by microorganisms,such as bacteria,actinomycetes,fungi and alga,as well as the functioning enzymes and their coding genes,were reviewed. The decolorization mechanisms and degradation pathways of triphenylmethane dyes were discussed based on the decolorizing intermediate products of different microbes. The development and application of triphenylmethane dyes biodecolorization and biodegradationwere also prospected.

Abstract:The use of dyes dates to ancient times and has increased due to population and industrial growth, leading to the rise of synthetic dyes. These pollutants are of great environmental impact and azo dyes deserve special attention due their widespread use and challenging degradation. Among the biological solutions developed to mitigate this issue, bacteria are highlighted for being versatile organisms, which can be applied as single organism cultures, microbial consortia, in bioreactors, acting in the detoxification of azo dyes breakage by-products and have the potential to combine biodegradation with the production of products of economic interest. These characteristics go hand in hand with the ability of various strains to act under various chemical and physical parameters, such as a wide range of pH, salinity, and temperature, with good performance under industry, and environmental, relevant conditions. This review encompasses studies with promising results related to the use of bacteria in the bioremediation of environments contaminated with azo dyes in the most diverse techniques and parameters, both in environmental and laboratory samples, also addressing their mechanisms and the legislation involving these dyes around the world, showcasing the importance of bacterial bioremediation, specialty in a scenario in an ever-increasing pursuit for sustainable production.

Abstract:As dye demand continues to rapidly increase in the food, pharmaceutical, cosmetic, paper, textile, and leather industries, an industrialization increase is occurring. Meanwhile, the degradation and removal of azo dyes have raised broad concern regarding the hazards posed by these dyes to the ecological environment and human health. Physicochemical treatments have been applied but are hindered by high energy and economic costs, high sludge production, and chemicals handling. Comparatively, the bioremediation technique is an eco-friendly, removal-efficient, and cost-competitive method to resolve the problem. This paper provides scientific and technical information about recent advances in the biodegradation of azo dyes. It expands the biodegradation efficiency, characteristics, and mechanisms of various microorganisms containing bacteria, fungi, microalgae, and microbial consortia, which have been reported to biodegrade azo dyes. In addition, information about physicochemical factors affecting dye biodegradation has been compiled. Furthermore, this paper also sketches the recent development and characteristics of advanced bioreactors.
Biodegradation of Azo Dyes by Genetically Engineered Azoreductase

Abstract:A azoreductase gene with 537 bp was obtained by PCR amplification from Rhodobacter sphaeroides AS1.1737. The enzyme, with a molecular weight of 18.7 kD, was efficiently expressed in Escherichia coli and its biodegradation characteristics for azo dyes were investigated. Furthermore, the reaction kinetics and mechanism of azo dyes catalyzed by the genetically engineered azoreductase were studied in detail. The presence of a hydrazo-intermediate was identified, which provided a convincing evidence for the assumption that azo dyes were degraded via an incomplete reduction stage.

Abstract:Azo dyes are the most widely applied chemical dyes that have also raised great concerns for environmental contamination and human health issues. There has been a growing interest in discovering bioremediation methods to degrade azo dyes for environmental and economic purposes. Azoreductases are key enzymes evolved in nature capable of degrading azo dyes. The current work reports the identification, expression, and properties of a novel azoreductase (AzoRed2) from Streptomyces sp. S27 which shows an excellent stability against pH change and organic solvents. To overcome the requirements of coenzyme while degrading azo dyes, we introduced a coenzyme regeneration enzyme, Bacillus subtilis glucose 1-dehydrogenase (BsGDH), to construct a recycling system in living cells. The whole-cell biocatalyst containing AzoRed2 and BsGDH was used to degrade a representative azo dye methyl red. The degradation rate of methyl red was up to 99% in 120 min with high substrate concentration (250 μM) and no external coenzyme added. The degradation rate was still 98% in the third batch trial. To sum up, a novel azoreductase with good properties was found, which was applied to construct whole-cell biocatalyst. Both the enzymes and whole-cell biocatalysts are good candidates for the industrial wastewater treatment and environmental restoration.

Abstract:Biodegradation and mineralization of azo dyes using sludge bacteria has become one of the main trends on research and development of treating waste water containing azo dyes.There are two major azorespiration styles of aerobic and anaerobic strains at present.And we identified the differences of the mineralization mechanisms of direct products of azo dye,namely aromatic amine biodegraded by aerobic and anaerobic strains.It is proposed that a sequential anaerobic-aerobic bioreactor treatment of azo dyes-containing waste water can be applied smoothly and extensively.

Abstract:Azo dye has a massive marketplace in various industries, biological laboratories and clinics. A significant drawback is the excessive flush-off of azo dyes and their intermediate products are that they pollute the environment. Several environmental bacteria degrade azo dye under certain specific ecological conditions. Decolourisation by different bacteria occurs under aerobic, anoxic and anaerobic conditions. Bacterial genera such as Pseudomonas, Bacillus and Rhodococcus are used for the rapid decolourisation of azo dyes. Bacterial enzymes play an essential role in the degradation of azo dyes by cleavage of azo bonds (-N=N-). It is degraded by both oxidative and reductive enzymes including azo reductase, laccase, lignin peroxidase, NADH-DCIP reductase, malachite green reductase, polyphenol oxidase (tyrosinase), veratrol alcohol oxidase. This review describes the role and mechanism of these enzymes in the degradation of azo dyes.

Abstract:UV spectrophotometer, HPLC and MS were used to analyze the pathway of degradation of azo dyes. The enzyme activity was defined as the amount of reduced dyes by per mg protein per minute via UV spectrophotometric analysis using Acid Red B as substrate. And the mechanism for azo dyes biodegradation was also analyzed using Methyl Red as substrate. The partially reduced intermediate was found and confirmed.
Structure-Based Long-Term Biodegradation of the Azo Dye: Insights from the Bacterial Community Succession and Efficiency Comparison

Abstract:In this study, microbial community dynamics were explored during biological degradation of azo dyes with different chemical structures. The effect of the different molecular structures of the azo dyes was also assessed against the simultaneous removal of color and the bacterial community. Winogradsky columns were inoculated with dewatered sludge and separately fed with six different azo dyes to conduct the sludge acclimatization process, and nine bacterial decolorizing strains were isolated and identified. The decolorization and biodegradation performances of the acclimated system and isolated strains were also determined. Results showed that the bacterial isolates involved in decolorization and the degradation of the azo dyes were mainly associated with the azo dye structure. After 24 h acclimatization at room temperature without specific illumination, immediate decolorization of methyl red (89%) and methyl orange (78%) was observed, due to their simple structure compared to tartrazine (73%). However, after 8 days of acclimatization, methyl red was easily decolorized up to 99%, and about 87% decolorization was observed for orange G (87%), due to its complex chemical structure. Higher degrees of degradation and decolorization were achieved with Pseudomonas geniculate strain Ka38 (Proteobacteria), Bacillus cereus strain 1FFF (Firmicutes) and Klebsiella variicola strain RVEV3 (Proteobacteria) with continuous shaking at 30 °C. The azo dyes with benzene rings were found to be easier to decolorize and degrade with similar microbial communities. Moreover, it seems that the chemical structures of the azo dyes, in a sense, drove the divergent succession of the bacterial community while reducing the diversity. This study gives a deep insight into the feasible structure-based artificial manipulation of bacterial communities and offers theoretical guidance for decolorizing azo dyes with mixed bacteria cultures.

Abstract:Azo dyes are toxic and carcinogenic and are often present in industrial effluents. In this research, azoreductase and glucose 1-dehydrogenase were coupled for both continuous generation of the cofactor NADH and azo dye removal. The results show that 85% maximum relative activity of azoreductase in an integrated enzyme system was obtained at the conditions: 1 U azoreductase:10 U glucose 1-dehydrogenase, 250 mM glucose, 1.0 mM NAD(+) and 150 mu M methyl red. Sensitivity analysis of the factors in the enzyme system affecting dye removal examined by an artificial neural network model shows that the relative importance of enzyme ratio between azoreductase and glucose 1-dehydrogenase was 22%, followed by dye concentration (27%), NAD(+) concentration (23%) and glucose concentration (22%), indicating none of the variables could be ignored in the enzyme system. Batch results show that the enzyme system has application potential for dye removal. (C) 2012 Elsevier Ltd. All rights reserved.

Abstract:Bio-degradation is the most affordable method of azo dye decontamination, while its drawbacks such as aromatic amines accumulation and low degradation efficiency must be overcome. In this study, a novel mechanism of azo dye degradation by a fungus was discovered. At a concentration of 400 mg/L, the decolorization efficiency of Acid Red 73 (AR73) by Aspergillus tabacinus LZ-M was 90.28%. Metabolite analysis and transcriptome sequencing analysis revealed a self-redox process of AR73 degradation, where the electrons generated in carbon oxidation were transferred to the reduction of -C-N = and –NN. The metabolites, 2-hydroxynaphthalene and N-phenylnitrous amide were mineralized into CO 2 through catechol pathway and a glycolytic process. Furthermore, the mineralization ratio of dye was computed to be 31.8% by the carbon balance and electron balance. By using comparative transcriptome, a novel decoloring enzyme Ord95 was discovered in unknown genes through gene cloning. It hydrolyzed AR73 into 2-hydroxynaphthalene and N-phenylnitrous amide, containing a glutathione S-transferase domain with three arginines as key active sites. Here the new mechanism of azo dye degradation was discovered with identification of a novel enzyme in Aspergillus tabacinus LZ-M.

Abstract:Anthraquinone dyes, which contain anthraquinone chromophore groups, are the second largest class of dyes after azo dyes and are used extensively in textile industries. The majority of these dyes are resistant to degradation because of their complex and stable structures; consequently, a large number of anthraquinone dyes find their way into the environment causing serious pollution. At present, the microbiological approach to treating printing and dyeing wastewater is considered to be an economical and feasible method, and reports regarding the bacterial degradation of anthraquinone dyes are increasing. This paper reviews the classification and structures of anthraquinone dyes, summarizes the types of degradative bacteria, and explores the possible mechanisms and influencing factors of bacterial anthraquinone dye degradation. Present research progress and existing problems are further discussed. Finally, future research directions and key points are presented.

Abstract:Azo dyes are widely used in textile and dyeing industry. Aniline, an important raw material of dyes and auxiliaries, is always the common by-products from the azo dye degradation, which is difficult to be completely removed in dyeing wastewater treatment. The discharge of azo dyes and aniline in textile wastewater arouses many environmental issues, and aniline is listed as the priority pollutant. In recent years, degradation techniques including physical, chemical and biological processes of different dyes and aniline have gained much attention. Their degradation pathways and corresponding mechanisms have been proposed. However, most studies focus on the dye removal or decolorization capacity, the influence factors, and the by-product varieties in certain degradation process. There were very few reports on the behavior of aniline transformation during the azo dye degradation processes, which is crucial for aniline control. This paper aims at comprehensively and critically reviewing the fate of aniline formation and degradation in different degradation technologies. Besides, crucial questions on aniline control are raised and discussed. It is expected that this work will shed new insight and provide valuable information that will aid in avoiding the environmental risk of aniline in textile and dyeing industry.

Abstract:The textile industry effluents are increasingly becoming an environmental problem, and the removal of dyes from aqueous waste streams has great potential for environmental engineering. Among many commonly applied methods, chemical reduction is an emerging prospect in the field of wastewater treatment. This paper reviews a recent 15-year survey on chemical reduction of dyes in the azo family. With respect to the nature of the reducing agents, the scope of the literature was divided into several categories. The available studies aim to reduction of azo dyes using zero-valent iron, other zero-valent metals, amorphous alloys, and finally, a catalytic reduction utilizing different electron donor agents in co-action with a wide range of metal-based catalysts. This paper includes summarization of different configurations of reaction conditions used for the reductive degradation of azo dye, their efficiency, and the influences of various operating parameters of the degradation processes. Several factors affecting the degradation of azo dyes have also been critically analyzed and future perspectives are proposed in this review. From the present study, it can be concluded that a selected reducing agents used for chemical reduction are very effective and environmentally friendly methods for the removal of azo dyes from wastewater.

Abstract:Textile industry consumes a large proportion of available water and releases huge amounts of toxic azo dye effluents, leading to an inevitable situation of acute environmental pollution that has been a significant threat to mankind. Decolorization or detoxification of harmful azo dyes has become a global priority to overcome the disastrous consequences and salvage the ecosystem. Biodegradation of textile azo dyes by endophytes stands to be a lucrative and viable alternative over conventional physico-chemical methods, owing to their eco-friendliness, cost-competitive and non-toxic nature. Especially, plant endophytic microbes exhibit promising biodegradation potential which has wired up the effective removal of textile azo dyes, attributing to their ability to produce dye degrading enzymes, laccases, peroxidases and azoreductases. Although both bacterial and fungal endophytes have been tried for azo dye degradation, endophytic fungi find broader application over bacteria. Despite of the advancements made in microbe-mediated biodegradation, there is still a need to fill the gap in lab to in situ translation of biodegradation research. This review concisely accentuates the xenobiotics of textile azo dyes and microbial mechanisms of biodegradation of textile azo dyes, positing plant endophytic community, especially bacterial and fungal endophytes as the potential dye degraders, highlighting currently reported dye degrading endophytic species.

Abstract:The synthetic azo dyes are widely used in the textile industries for their excellent dyeing properties. They may be classified into many classes based on their structure and application, including direct, reactive, dispersive, acidic, basic, and others. The continuous discharge of wastewater from a large number of textile industries without prior treatment poses detrimental effects on the environment and human health. Azo dyes and their degradation products are extremely poisonous for their carcinogenic, teratogenic and mutagenic nature. Moreover, exposure to synthetic azo dyes can cause genetic changes, skin inflammation, hypersensitivity responses, and skin irritations in persons, which may ultimately result in other profound issues including the deterioration of water quality. This review discusses these dyes in details along with their detrimental effects on aquatic and terrestrial flora and fauna including human beings. Azo dyes degrade the water bodies by increasing biochemical and chemical oxygen demand. Therefore, dye-containing wastewater should be effectively treated using eco-friendly and cost-effective technologies to avoid negative impact on the environment. This article extensively reviews on physical, chemical and biological treatment with their benefits and challenges. Biological-based treatment with higher hydraulic retention time (HRT) is economical, consumes less energy, produces less sludge and environmentally friendly. Whereas the physical and chemical methods with less hydraulic retention time is costly, produces large sludge, requires high dissolved oxygen and ecologically inefficient. Since, biological treatment is more advantageous over physical and chemical methods, researchers are concentrating on bioremediation for eliminating harmful azo dye pollutants from nature. This article provides a thorough analysis of the state-of-the-art biological treatment technologies with their developments and effectiveness in the removal of azo dyes. The mechanism by which genes encoding azoreductase enzymes (azoG, and azoK) enable the natural degradation of azo dyes by bacteria and convert them into less harmful compounds is also extensively examined. Therefore, this review also focuses on the use of genetically modified microorganisms and nano-technological approaches for bioremediation of azo dyes.

Abstract:Azo dyes are significant organic pollutants known for their adverse effects on humans and aquatic life. In this study, anthraquinone-2-sulfonate (AQS) immobilized on biochar (BC) was employed as a novel carrier in up-flow anaerobic fixed-bed reactors to induce specific biofilm formation and promote the biotransformation efficiency of azo dyes. Novel carrier–packed reactor 1 (R1) and BC-packed reactor 2 (R2) were used to treat red reactive 2 (RR2) under continuous operation for 175 days. The decolorization rates of R1 and R2 were 96–83% and 91–73%, respectively. The physicochemical characteristics and extracellular polymeric substances (EPS) of the biofilm revealed a more stable structure in R1. Furthermore, the microbial community in R1 interacted more closely with each other and contained more keystone genera. Overall, this study provides a feasible method for improving the biotransformation of azo dyes, thus providing support for practical applications in wastewater treatment projects.

Abstract:Azo dyes, as the most widely used synthetic dyes, are considered to be one of the culprits of water resources and environmental pollution. Anoxybacillus sp. PDR2 is a thermophilic bacterium with the ability to degrade azo dyes, whose genome contains two genes encoding azoreductases (named AzoPDR2-1 and AzoPDR2-2). In this study, through response surface methodology (RSM), when the initial pH, inoculation volume and Mg 2+ addition amount were 7.18, 10.72% and 0.1 g/L respectively, the decolorization rate of methyl red (MR) (200 mg/L) could reach its maximum (98.8%). The metabolites after biodegradation were detected by UV–Vis spectroscopy, Fourier transform infrared spectroscopy (FTIR), and liquid chromatography mass spectrometry (LC-MS/MS), indicating that MR was successfully decomposed into 4-aminobenzoic acid and other small substrates. In homologous modeling, it was found that both azoreductases were flavin-dependent azoreductases, and belonged to the α/β structure, using the Rossmann fold. In their docking results with the cofactor flavin mononucleotide (FMN), FMN bound to the surface of the protein dimer. Nicotinamide adenine dinucleotide (NADH) was superimposed on the plane of the pyrazine ring between FMN and the activity pocket of protein. Besides, both azoreductase complexes (azoreductase-FMN-NADH) exhibited a substrate preference for MR. Asn104 and Tyr74 played an important role in the combination of the azoreductase AzoPDR2-1 complex and the azoreductase AzoPDR2-2 complex with MR, respectively. This provided assistance for studying the mechanism of azoreductase biodegradation of azo dyes in thermophilic bacteria.

Abstract:Dyes are synthetic organic compounds widely used in various industries such as, textile, leather, plastic, food, pharmaceutical, and paints manufacturing industries. Coloured effluents are highly toxic to the aquatic life and mutagenic to humans. Wastewater containing dyes has become an important issue demanding serious attention. Among the synthetic dyes, azo dyes are the largest and most widely used dyes and account for more than half of the annually produced dyes. The biodegradation of azo dyes is difficult due to their complex structure and synthetic nature. Several treatments have been proposed for efficient azo dye removal, most of them presenting some limitations such as generation of waste sludge, high operational costs, poor efficiency, and incomplete mineralization. Biological treatment is a cost-effective and eco-friendly process for dye degradation. Sequential anaerobic–aerobic biological treatment is considered as one of the most cost-effective methods for the complete mineralization of azo dyes. The anaerobic stage yields decolourization through reductive cleavage of the dye’s azo linkages, resulting in the formation of generally colourless but potentially hazardous aromatic amines. The aerobic stage involves degradation of the aromatic amines. It is the most logical step for removing the azo dyes from the wastewater. Several factors can influence the microbial activity and consequently the efficacy and effectiveness of the complete biodegradation processes. This paper summarizes the results of biological decolourization of azo dyes using various types of reactors, elaborates biochemical mechanisms involved, and discusses influence of various operational parameters on decolourization based on reports published in the last decade.
