Abstract:Extensive investigations have implicated cytokines such as tumour necrosis factor (TNF)- alpha and interleukin (IL)-1, and IL-6 as contributing to the pathology of ischemia-reperfusion (I/R) injury, since an increase in the production of those cytokines was clinically detected after myocardial infarction and cardiopulmonary bypass surgery. Current evidence indicates that these cytokines are autocrine contributors to myocardial dysfunction and cardiomyocyte necrosis in I/R injury, whereas, earlier evidence also suggest that cytokines have controversial roles in cardiovascular pathophysiology. Accordingly, it becomes vital to better define the mechanisms of action of cytokines as important steps towards the development of effective therapeutic strategies to combat their deleterious effects in ischemia-induced myocardial injury. Since TNF-alpha, TGF-beta1, IL-1, IL-6 and IL-8 have been frequently studied in cardiovascular diseases, especially in I/R heart disease, the purpose of this article is to review the cardiodepressant role of these cytokines and their release in I/R injury.

Abstract:Hepatic ischemia/reperfusion injury (IRI) is mainly characterized by high activation of immune inflammatory responses and metabolic responses. Understanding the molecular and metabolic mechanisms underlying development of hepatic IRI is critical for developing effective therapies for hepatic IRI. Recent advances in research have improved our understanding of the pathogenesis of IRI. During IRI, hepatocyte injury and inflammatory responses are mediated by crosstalk between the immune cells and metabolic components. This crosstalk can be targeted to treat or reverse hepatic IRI. Thus, a deep understanding of hepatic microenvironment, especially the immune and metabolic responses, can reveal new therapeutic opportunities for hepatic IRI. In this review, we describe important cells in the liver microenvironment (especially non-parenchymal cells) that regulate immune inflammatory responses. The role of metabolic components in the diagnosis and prevention of hepatic IRI are discussed. Furthermore, recent updated therapeutic strategies based on the hepatic microenvironment, including immune cells and metabolic components, are highlighted.

Abstract:Retinal ischemia reperfusion (RIR) injury is a complicated physiopathologic process, which is mediated by many factors such as free radicals,inflammatory cytokines,leukocytes and so on. There is a close relation between RIR injure and cytokines. On the one hand, RIR may induce the expression of many inflammatory cytokines such as interleukin 1, interleukin 6;on the other hand, many kinds of exogenous neurotrophic factors, such as basic fibroblast growth factor,recombinant human hepatocyte growth factor and brain derived neurotrophic factor, have been shown to protect RIR injury. It is of great value to research the relation between them. So we report the newly progress of foreign researchs about the RIR injury and cytokines.

Abstract:Ischemia-reperfusion injury (IRI) involves a positive amplification feedback loop that stimulates innate immune-driven tissue damage associated with organ procurement from deceased donors and during transplantation surgery. As our appreciation of its basic immune mechanisms has improved in recent years, translating putative biomarkers into therapeutic interventions in clinical transplantation remains challenging. This review presents advances in translational/clinical studies targeting immune responses to reactive oxygen species in IRI-stressed solid organ transplants, especially livers. Here we focus on novel concepts to rejuvenate suboptimal donor organs and improve transplant function using pharmacologic and machine perfusion (MP) strategies. Cellular damage induced by cold ischemia/warm reperfusion and the latest mechanistic insights into the microenvironment's role that leads to reperfusion-induced sterile inflammation is critically discussed. Efforts to improve clinical outcomes and increase the donor organ pool will depend on improving donor management and our better appreciation of the complex mechanisms encompassing organ IRI that govern the innate-adaptive immune interface triggered in the peritransplant period and subsequent allo-Ag challenge. Computational techniques and deep machine learning incorporating the vast cellular and molecular mechanisms will predict which peri-transplant signals and immune interactions are essential for improving access to the long-term function of life-saving transplants.

Abstract:MicroRNAs (miRNAs) are the endogenous non-encoding small RNA molecules that regulate gene expression and participate in important cellular pathways as well as various physiological and pathological processes.Ischemia reperfusion injury (IRI) refers to the blood reperfusion process of tissues and organs after ischemia.The function of the IRI tissues and organs cannot be restored after perfusion which results in dysfunction and structural damage.IRI is a complex systemic process that affects multiple tissues and organs and causes irreversible damage which leads to a cascade of dysfunction of multiple organs.It has been found that a variety of miRNA has obvious expression changes in tissues and organs of IRI,indicating that miRNA can directly or indirectly affect IRI tissues and organs.This review is to provide insights into the regulatory role of miRNA in the IRI of the heart,brain,liver and kidney.MiRNA is not only involved in the pathophysiological process of organ IRI,but also plays a crucial role in clinical diagnosis and intervention treatment as a specific marker of IRI.

Abstract:Ischemia-reperfusion (I/R) injury is a progressive injury that aggravates the pathological state when the organ tissue restores blood supply after a certain period of ischemia, including the myocardial, brain, liver, kidney, and intestinal. With growing evidence that microRNAs (miRNAs) play an important role as posttranscription gene silencing mediators in many I/R injury, in this review, we highlight the microRNAs that are related to I/R injury and their regulatory molecular pathways. In addition, we discussed the potential role of miRNA as a biomarker and its role as a target in I/R injury treatment. Developing miRNAs are not without its challenges, but prudent design combined with existing clinical treatments will result in more effective therapies for I/R injury. This review is aimed at providing new research results obtained in this research field. It is hoped that new research on this topic will not only generate new insights into the pathophysiology of miRNA in I/R injury but also can provide a basis for the clinical application of miRNA in I/R.

Abstract:Ischemia/reperfusion injury (IRI) is caused by a rapid transient reduction in blood flow to a specific organ. IRI is typically accompanied by a strong inflammatory and oxidative stress response to hypoxia and reperfusion, which disrupts organ function. AKI caused by renal IRI contributes to a high morbidity and death rate in a variety of injuries. Although the pathophysiology of IRI is not fully understood, numerous key pathways leading to renal failure have been identified. The production of reactive oxygen species (ROS) during the reperfusion phase of the ischemic kidney and subsequent re-oxygenation begins a cascade of detrimental cellular reactions that lead to inflammation, cell death, and acute kidney failure. Greater knowledge of the cellular pathophysiological mechanisms causing kidney damage may lead to the development of more focused treatments to prevent and treat the damage. We discuss several significant possible mechanisms and treatment methods in renal IRI in this study.

Abstract:Ischemia/reperfusion injury (IRI) permeates a variety of diseases and is a ubiquitous concern in every transplantation proceeding, from whole organs to modest grafts. Given its significance, efforts to evade the damaging effects of both ischemia and reperfusion are abundant in the literature and they consist of several strategies, such as applying pre-ischemic conditioning protocols, improving protection from preservation solutions, thus providing extended cold ischemia time and so on. In this review, we describe many of the latest pharmacological approaches that have been proven effective against IRI, while also revisiting well-established concepts and presenting recent pathophysiological findings in this ever-expanding field. A plethora of promising protocols has emerged in the last few years. They have been showing exciting results regarding protection against IRI by employing drugs that engage several strategies, such as modulating cell-surviving pathways, evading oxidative damage, physically protecting cell membrane integrity, and enhancing cell energetics.

Abstract:Ischemia/reperfusion(I/R) injury is the pathophysiological process of many diseases in clinical .The extent of the damage of organs and tissues and the degree of ischemia and ischemia time length are closely related .The ischemia leads to a significant decrease in ATP production and a decrease in the pH value of the cells ,which can result in the over-load of the calcium in the mitochondria and the cell .During reperfusion,excessive reactive oxygen is produced , and the mitochondria and endoplasmic reticulum in parenchyma cells and endothelial cells are stressed .Subsequently,mitochondrial permeability transition pore is open ,resulting in cell lysis and death .The effects of intestinal flora ,epigenetic changes and the role of particles are all involved in the pathogenesis of I /R.I/R also involves abnormal cell signal transduction ,the acti-vation of the inflammatory response ,the activation of protein kinases ,the participation of humoral factors ,cytokine,comple-ment and abnormal interactions between cells and /or platelet.

Abstract:Acute myocardial ischemia/reperfusion (I/R) injury is a significant, unsolved clinical puzzle. In the disease context of acute myocardial infarction, reperfusion remains the only effective strategy to salvage ischemic myocardium, but it also causes additional damage. Myocardial I/R injury is composed of four types of damage, and these events attenuate the benefits of reperfusion therapy. Thus, inventing new strategies to conquer I/R injury is an unmet clinical need. A variety of pathological processes and mediators, including changes in the pH, generation of reactive oxygen radicals, and intracellular calcium overload, are proposed to be crucial in I/R-related cell injury. Among the intracellular events that occur during I/R, we stress the importance of protein phosphorylation signaling and elaborate its regulation. A variety of protein kinase pathways could be activated in I/R, including reperfusion injury salvage kinase and survivor-activating factor enhancement pathways, which are critical to cardiomyocyte survival. In addition to serine/threonine phosphorylation signaling, protein tyrosine phosphorylation is also critical in multiple cell functions and survival. However, the roles of protein kinases and phosphatases in I/R have not been extensively studied yet. By better understanding the mechanisms of I/R injury, we may have a better chance to develop new strategies for I/R injury and apply them in the clinical patient care.

Abstract:Ischemia/reperfusion (IR) injury contributes to disability and mortality worldwide. Due to the complicated mechanisms and lack of proper therapeutic targets, few interventions are available that specifically target the pathogenesis of IR injury. Regulated cell death (RCD) of endothelial and parenchymal cells is recognized as the promising intervening target. Recent advances in IR injury suggest that small molecules exhibit beneficial effects on various RCD against IR injury, including apoptosis, necroptosis, autophagy, ferroptosis, pyroptosis, and parthanatos. Here, we describe the mechanisms behind these novel promising therapeutic targets and explain the machinery powering the small molecules. These small molecules exert protection by targeting endothelial or parenchymal cells to alleviate IR injury. Therapies of the ideal combination of small molecules targeting multiple cell types have shown potent synergetic therapeutic effects, laying the foundation for novel strategies to attenuate IR injury.

Abstract:Kidney transplantation offers a longer life expectancy and a better quality of life than dialysis to patients with end-stage kidney disease. Ischemia–reperfusion injury (IRI) is thought to be a cornerstone in delayed or reduced graft function and increases the risk of rejection by triggering the immunogenicity of the organ. IRI is an unavoidable event that happens when the blood supply is temporarily reduced and then restored to an organ. IRI is the result of several biological pathways, such as transcriptional reprogramming, apoptosis and necrosis, innate and adaptive immune responses, and endothelial dysfunction. Tubular cells mostly depend on fatty acid (FA) β-oxidation for energy production since more ATP molecules are yielded per substrate molecule than glucose oxidation. Upon ischemia–reperfusion damage, the innate and adaptive immune system activates to achieve tissue clearance and repair. Several cells, cytokines, enzymes, receptors, and ligands are known to take part in these events. The complement cascade might start even before organ procurement in deceased donors. However, additional experimental and clinical data are required to better understand the pathogenic events that take place during this complex process.

Elucidating the Molecular Pathways and Immune System Transcriptome During Ischemia-Reperfusion Injury in Renal Transplantation

Abstract:Ischemia reperfusion injury (IRI) is a major challenge for renal transplantation. This study was performed to explore the mechanisms and potential molecular targets involved in renal IRI. In this study, the gene datasets GSE43974 and GSE126805 from the Gene Expression Omnibus database, which include ischemic and reperfused renal specimens, were analyzed to determine differentially expressed genes (DEGs). Gene ontology annotations, Kyoto Encyclopedia of Genes and Genomes analysis, and gene set enrichment analysis were performed to determine the pathways that are significantly enriched during ischemia and reperfusion. We also determined the microenvironment cell types xCell and performed correlation analyses to reveal the relationship between the molecular pathways and microenvironment cell infiltration. We found 77 DEGs (76 up- and 1 downregulated) and 323 DEGs (312 up- and 11 downregulated) in the GSE43974 and GSE126805 datasets, respectively. Similar signaling pathway enrichment patterns were observed between the two datasets. The combined analyses demonstrate that the NOD-like receptor signaling pathway and its two downstream signaling pathways, MAPK and NF-kβ, are the major significantly enriched pathways. The xCell analysis identified immune cells that are significantly changed after reperfusion, including hematopoietic stem cells, M2 macrophages, monocytes, Treg cells, conventional dendritic cells, and pro B-cells. Enrichment scores of the NOD-like receptor signaling pathway and its downstream pathways during IRI was significantly correlated with the change levels in class-switched memory B-cell and hematopoietic stem cells in both datasets. These data reveal the important role of the NOD-like receptor signaling pathway during IRI, and the close relationship between this pathway and infiltration of specific immune cell types. Our data provide compelling insights into the pathogenesis and potential therapeutic targets for renal IRI.

Abstract:Abstract Ischemia/reperfusion (IR) injury, a main reason of mortality and morbidity worldwide, occurs in many organs and tissues. As a result of IR injury, senescent cells can accumulate in multiple organs. Increasing evidence shows that cellular senescence is the underlying mechanism that transforms an acute organ injury into a chronic one. Several recent studies suggest senescent cells can be targeted for the prevention or elimination of acute and chronic organ injury induced by IR. In this review, we concisely introduce the underlying mechanism and the pivotal role of premature senescence in the transition from acute to chronic IR injuries. Special focus is laid on recent advances in the mechanisms as well as on the basic and clinical research, targeting cellular senescence in multi-organ IR injuries. Besides, the potential directions in this field are discussed in the end. Together, the recent advances reviewed here will act as a comprehensive overview of the roles of cellular senescence in IR injury, which could be of great significance for the design of related studies, or as a guide for potential therapeutic target.

Abstract:Abstract: Hepatic ischemia-reperfusion injury is a complicated systemic process that affects several tissues and organs in addition to being a pathologic process that affects the liver. Several studies have shown that hepatic reperfusion severely impairs liver function. That also produces irreversible damage that causes dysfunction of many organs. The regulation of hepatic ischemia-reperfusion injury (IRI) is influenced by various factors, such as mitochondrial damage, anaerobic metabolism, intracellular Ca2+ overload, oxidative stress and ROS production, and chemokines as well as cytokines generation. Most of the cytokines are generated by helper T cells and neutrophils. A target of Matrix Metalloproteinases (MMPs) in chronic and acute liver injury caused by ischemia can be a significant facilitator of early leukocyte recruitment. Neutrophil gelatinase-associated lipocalin (NGAL) and MMPs could be employed as indicators of the severity of ischemia-reperfusion (I-R) injuries. This review examines the relationship between the components of hepatic IRI, MMPs, and the available pharmaceutical treatments for this condition.

Abstract:Ischemia reperfusion injury (IRI) is associated with a broad array of life-threatening medical conditions including myocardial infarct, cerebral stroke, and organ transplant. Although the pathobiology and clinical manifestations of IRI are well reviewed by previous publications, IRI-related transcriptomic alterations are less studied. This study aimed to reveal a transcriptomic hallmark for IRI by using the RNA-sequencing data provided by several studies on non-human preclinical experimental models. In this regard, we focused on the transcriptional responses of IRI in an acute time-point up to 48 h. We compiled a list of highly reported genes in the current literature that are affected in the context of IRI. We conducted Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses and found many of the up-regulated genes to be involved in cell survival, cell surface signaling, response to oxidative stress, and inflammatory response, while down-regulated genes were predominantly involved in ion transport. Furthermore, by GO analysis, we found that multiple inflammatory and stress response processes were affected after IRI. Tumor necrosis factor alpha (TNF) and nuclear factor kappa-light-chain-enhancer of activated B cells (NF-κB) signaling pathways were also highlighted in the Kyoto Encyclopedia of Genes and Genomes enrichment analysis. In the last section, we discuss the treatment approaches and their efficacy for IRI by comparing RNA sequencing data from therapeutic interventions with the results of our cross-comparison of differentially expressed genes and pathways across IRI.

Abstract:Coronary arterial disease is the most common cardiovascular disease. Myocardial ischemia-reperfusion injury caused by the initial interruption of organ blood flow and subsequent restoration of organ blood flow is an important clinical problem with various cardiac reperfusion strategies after acute myocardial infarction. Even though blood flow recovery is necessary for oxygen and nutrient supply, reperfusion causes pathological sequelae that lead to the aggravation of ischemic injury. At present, although it is known that injury will occur after reperfusion, clinical treatment always focuses on immediate recanalization. Mitochondrial fusion, fission, biogenesis, autophagy, and their intricate interaction constitute an effective mitochondrial quality control system. The mitochondrial quality control system plays an important role in maintaining cell homeostasis and cell survival. The removal of damaged, aging, and dysfunctional mitochondria is mediated by mitochondrial autophagy. With the help of appropriate changes in mitochondrial dynamics, new mitochondria are produced through mitochondrial biogenesis to meet the energy needs of cells. Mitochondrial dysfunction and the resulting oxidative stress have been associated with the pathogenesis of ischemia/reperfusion (I/R) injury, which play a crucial role in the pathophysiological process of myocardial injury. This review aimed at elucidating the mitochondrial quality control system and establishing the possibility of using mitochondria as a potential therapeutic target in the treatment of I/R injuries.
