Abstract:Observations of the temporal and spatial distribution of N,.O in solution are not yet sufficient topermit quantitative assessment of the role of the ocean in the budget of atmospheric N,.O. Consideration of the global nitrogen cycle suggests hat the land should be the primary source of N20. The gas is removed in the atmosphere by photolysis and by reaction with O(•D), and there may be additional sinks in the ocean.

Abstract:The N2O content of waters in the Potomac and Merrimack Rivers was measured on a number of occasions over the period April to July 1977. The concentrations of dissolved N2O exceed those which would apply in equilibrium with air by factors ranging from about 46 in the Potomac to 1.2 in the Merrimack. Highest concentrations of dissolved N2O were associated with sewage discharges from the vicinity of Washington, D.C. and analysis indicates a relatively high yield, 1.3 to 11 percent, for prompt conversion of waste nitrogen to N2O. The yield could be even higher if bubbles originating in sediments should contain as little as 0.3 percent N2O. Measurements of dissolved N2O in fresh water ponds near Boston demonstrate that aquatic systems may provide both strong sources and sinks for atmospheric N2O.

Abstract:Nitrous oxide (N 2 O) is a powerful atmospheric greenhouse gas and cause of ozone layer depletion. Global emissions continue to rise. More than two-thirds of these emissions arise from bacterial and fungal denitrification and nitrification processes in soils, largely as a result of the application of nitrogenous fertilizers. This article summarizes the outcomes of an interdisciplinary meeting, ‘Nitrous oxide (N 2 O) the forgotten greenhouse gas’, held at the Kavli Royal Society International Centre, from 23 to 24 May 2011. It provides an introduction and background to the nature of the problem, and summarizes the conclusions reached regarding the biological sources and sinks of N 2 O in oceans, soils and wastewaters, and discusses the genetic regulation and molecular details of the enzymes responsible. Techniques for providing global and local N 2 O budgets are discussed. The findings of the meeting are drawn together in a review of strategies for mitigating N 2 O emissions, under three headings, namely: (i) managing soil chemistry and microbiology, (ii) engineering crop plants to fix nitrogen, and (iii) sustainable agricultural intensification.

Abstract:Chemolithoautotrophic AOB (ammonia-oxidizing bacteria) form a crucial component in microbial nitrogen cycling in both natural and engineered systems. Under specific conditions, including transitions from anoxic to oxic conditions and/or excessive ammonia loading, and the presence of high nitrite (NO2−) concentrations, these bacteria are also documented to produce nitric oxide (NO) and nitrous oxide (N2O) gases. Essentially, ammonia oxidation in the presence of non-limiting substrate concentrations (ammonia and O2) is associated with N2O production. An exceptional scenario that leads to such conditions is the periodical switch between anoxic and oxic conditions, which is rather common in engineered nitrogen-removal systems. In particular, the recovery from, rather than imposition of, anoxic conditions has been demonstrated to result in N2O production. However, applied engineering perspectives, so far, have largely ignored the contribution of nitrification to N2O emissions in greenhouse gas inventories from wastewater-treatment plants. Recent field-scale measurements have revealed that nitrification-related N2O emissions are generally far higher than emissions assigned to heterotrophic denitrification. In the present paper, the metabolic pathways, which could potentially contribute to NO and N2O production by AOB have been conceptually reconstructed under conditions especially relevant to engineered nitrogen-removal systems. Taken together, the reconstructed pathways, field- and laboratory-scale results suggest that engineering designs that achieve low effluent aqueous nitrogen concentrations also minimize gaseous nitrogen emissions.

Abstract:N2O has been caused more and more attention because of its green-house effect. Microbial activity in biosphere is regarded as an important N2O source. Therefore, to make clear and modify the activity of microorganisms is of fundamental importance to control N2O emission from microorganisms, especially, in Wasetwater treatment situations. In the paper, based on investigation, heterotrophic, nitrate respiring and nitrate reducing microorganisms were emphasized as those being able to produce N2O, besides traditional nitrifiers and denitrifiers. There are different pathways of N2O emission for different organisms under different conditions. The possible mechanisms of N2O yielding were detailed in the paper accompanying with the description of various factors affecting N2O emission. Then, current situation of research on control of N2O emission from wastewater treatment was described and analyzed for making clear the trend of research on N2O emission control in waste water treatment field.
Aerobic denitrification as an N2O source from microbial communities

Abstract:Nitrous oxide (N2O) is a potent greenhouse gas of primarily microbial origin. Oxic and anoxic emissions are commonly ascribed to autotrophic nitrification and heterotrophic denitrification, respectively. Beyond this established dichotomy, we quantitatively show that heterotrophic denitrification can significantly contribute to aerobic nitrogen turnover and N2O emissions in complex microbiomes exposed to frequent oxic/anoxic transitions. Two planktonic, nitrification-inhibited enrichment cultures were established under continuous organic carbon and nitrate feeding, and cyclic oxygen availability. Over a third of the influent organic substrate was respired with nitrate as electron acceptor at high oxygen concentrations (>6.5 mg/L). N2O accounted for up to one-quarter of the nitrate reduced under oxic conditions. The enriched microorganisms maintained a constitutive abundance of denitrifying enzymes due to the oxic/anoxic frequencies exceeding their protein turnover-a common scenario in natural and engineered ecosystems. The aerobic denitrification rates are ascribed primarily to the residual activity of anaerobically synthesised enzymes. From an ecological perspective, the selection of organisms capable of sustaining significant denitrifying activity during aeration shows their competitive advantage over other heterotrophs under varying oxygen availabilities. Ultimately, we propose that the contribution of heterotrophic denitrification to aerobic nitrogen turnover and N2O emissions is currently underestimated in dynamic environments.

Abstract:The rapid expansion of coastal aquaculture has led to an increase in the coverage of aquaculture ponds, where intense feed-derived nitrogen is causing significant emissions of nitrous oxide (N2O). Multiple N2O production pathways and the relative importance of water column vs. sedimentary production in aquaculture ponds remain uncertain. Clarifying these pathways is vital for sustainable aquaculture development. Using 15N-labeled dissolved inorganic nitrogen, the pathways and rates of N2O production in subtropical aquaculture ponds located in south China, cultivating whiteleg shrimp, Japanese seabass, and giant river prawn, were successfully characterized. Total N2O production rates ranged from 6 to 70 µmol-N m-2 d-1, with the shrimp pond exhibiting the highest total N2O production rates, followed by ponds for seabass and prawn. These differences are primarily due to varying feed amounts causing differences in dissolved nutrients in water column and sediment. Particularly, nutrient and organic matter accumulation at the surface sediment stimulated N2O production. The oxygenated sediment on a centimeter scale could produce substantially more N2O compared to the water column above on a meter scale. Partial denitrification, i.e., nitrate and nitrite reduction to N2O, was more important (> 60 %) for N2O production in aquaculture ponds. The availability of nitrite is likely a major factor driving partial denitrification for both sedimentary and water column N2O production.

Abstract:Ammonia-oxidizing archaea (AOA) are key players in the nitrogen cycle and among the most abundant microorganisms in the ocean, thriving even in oxygen-depleted ecosystems. AOA produce the greenhouse gas nitrous oxide (N2O) as a byproduct of ammonia oxidation. Additionally, the recent discovery of a nitric oxide dismutation pathway in the AOA isolate Nitrosopumilus maritimus points toward other N2O production and consumption pathways in AOA. AOA that perform NO dismutation when exposed to oxygen depletion, produce oxygen and dinitrogen as final products. Based on the transient accumulation of N2O coupled with oxygen accumulation, N2O has been proposed as an intermediate in this novel archaeal pathway. In this study, we spiked N2O to oxygen-depleted incubations with pure cultures of two marine AOA isolates that were performing NO dismutation. By using combinations of N compounds with different isotopic signatures (15NO2 - pool +44N2O spike and 14NO2 - pool +46N2O spike), we evaluated the N2O spike effects on the production of oxygen and the isotopic signature of N2 and N2O. The experiments confirmed that N2O is an intermediate in NO dismutation by AOA, distinguishing it from similar pathways in other microbial clades. Furthermore, we showed that AOA rapidly reduce high concentrations of spiked N2O to N2. These findings advance our understanding of microbial N2O production and consumption in oxygen-depleted settings and highlight AOA as potentially important key players in N2O turnover.
Nitrous Oxide Production via Nitroxyl by a Multicopper Oxidase from a Nitrifying Archaeon

Abstract:Ammonia oxidizing archaea (AOA) are among the most abundant microorganisms on earth and are known to be a major source of oceanic nitrous oxide (N2O) emissions, although biochemical origins of this N2O remain unknown. Enzymological details of AOA nitrogen metabolism are broadly unavailable. We report the recombinant expression, purification, and characterization of a multicopper oxidase (MCO), Nmar_1354, from the AOA Nitrosopumilus maritimus. We show that Nmar_1354 selectively produces nitroxyl (HNO) by coupling the oxidation of the obligate nitrification intermediate hydroxylamine (NH2OH) to dioxygen (O2) reduction. This HNO undergoes several downstream reactions, although a significant fraction rapidly dimerizes to yield N2O. These results afford a possible enzymatic origin of AOA-derived N2O and reveal a unique enzymatic reaction for producing HNO.

Abstract:Emission of nitrous oxide (N2O) during biological wastewater treatment is of growing concern since N2O is a major stratospheric ozone-depleting substance and an important greenhouse gas. The emission of N2O from a lab-scale granular sequencing batch reactor (SBR) for partial nitrification (PN) treating synthetic wastewater without organic carbon was therefore determined in this study, because PN process is known to produce more N2O than conventional nitrification processes. The average N2O emission rate from the SBR was 0.32 ± 0.17 mg-N L(-1) h(-1), corresponding to the average emission of N2O of 0.8 ± 0.4% of the incoming nitrogen load (1.5 ± 0.8% of the converted NH4(+)). Analysis of dynamic concentration profiles during one cycle of the SBR operation demonstrated that N2O concentration in off-gas was the highest just after starting aeration whereas N2O concentration in effluent was gradually increased in the initial 40 min of the aeration period and was decreased thereafter. Isotopomer analysis was conducted to identify the main N2O production pathway in the reactor during one cycle. The hydroxylamine (NH2OH) oxidation pathway accounted for 65% of the total N2O production in the initial phase during one cycle, whereas contribution of the NO2(-) reduction pathway to N2O production was comparable with that of the NH2OH oxidation pathway in the latter phase. In addition, spatial distributions of bacteria and their activities in single microbial granules taken from the reactor were determined with microsensors and by in situ hybridization. Partial nitrification occurred mainly in the oxic surface layer of the granules and ammonia-oxidizing bacteria were abundant in this layer. N2O production was also found mainly in the oxic surface layer. Based on these results, although N2O was produced mainly via NH2OH oxidation pathway in the autotrophic partial nitrification reactor, N2O production mechanisms were complex and could involve multiple N2O production pathways.

Abstract:Marine oxygen deficient zones (ODZs) are dynamic areas of microbial nitrogen cycling. Nitrification, the microbial oxidation of ammonia to nitrate, plays multiple roles in the biogeochemistry of these regions, including production of the greenhouse gas nitrous oxide (N 2 O). We present here the results of two oceanographic cruises investigating nitrification, nitrifying microorganisms, and N 2 O production and distribution from the offshore waters of the Eastern Tropical South Pacific (ETSP). On each cruise, high‐resolution measurements of ammonium ([NH 4 + ]), nitrite ([NO 2 ‐ ]), and N 2 O were combined with 15 N tracer‐based determination of ammonia oxidation, nitrite oxidation, nitrate reduction and N 2 O production rates. Depth‐integrated inventories of NH 4 + and NO 2 ‐ were positively correlated with one another, and with depth‐integrated primary production. Depth‐integrated ammonia oxidation rates were correlated with sinking particulate organic nitrogen flux but not with primary production; ammonia oxidation rates were undetectable in trap‐collected sinking particulate material. Nitrite oxidation rates exceeded ammonia oxidation rates at most mesopelagic depths. We found positive correlations between archaeal amoA genes and ammonia oxidation rates and between Nitrospina ‐like 16S rRNA genes and nitrite oxidation rates. N 2 O concentrations in the upper oxycline reached values of >140 nM, even at the western extent of the cruise track, supporting air‐sea fluxes of up to 1.71 μmol m ‐2 d ‐1 . Our results suggest that a source of NO 2 ‐ other than ammonia oxidation may fuel high rates of nitrite oxidation in the offshore ETSP and that air‐sea fluxes of N 2 O from this region may be higher than previously estimated.

Abstract:Nitrous oxide (N 2 O) is a potent greenhouse gas and ozone depletion agent, with a significant natural source from marine oxygen-deficient zones (ODZs). Open questions remain, however, about the microbial processes responsible for this N 2 O production, especially hybrid N 2 O production when ammonia-oxidizing archaea are present. Using 15 N-labeled tracer incubations, we measured the rates of N 2 O production from ammonium (NH 4+ ), nitrite (NO 2- ), and nitrate (NO 3- ) in the eastern tropical North Pacific ODZ and the isotopic labeling of the central ( α ) and terminal ( β ) nitrogen (N) atoms of the N 2 O molecule. We observed production of both doubly and singly labeled N 2 O from each tracer, with the highest rates of labeled N 2 O production at the same depths as the near-surface N 2 O concentration maximum. At most stations and depths, the production of 45 N 2 O α and 45 N 2 O β were statistically indistinguishable, but at a few depths there were significant differences in the labeling of the two nitrogen atoms in the N 2 O molecule. Implementing the rates of labeled N 2 O production in a time-dependent numerical model, we found that N 2 O production from NO 3- dominated at most stations and depths, with rates as high as 1600 ± 200 pM N 2 O d −1 . Hybrid N 2 O production, one of the mechanisms by which ammonia-oxidizing archaea produce N 2 O, had rates as high as 230 ± 80 pM N 2 O d −1 that peaked in both the near-surface and deep N 2 O concentration maxima. Based on the equal production of 45 N 2 O α and 45 N 2 O β in the majority of our experiments, we infer that hybrid N 2 O production likely has a consistent site preference, despite drawing from two distinct substrate pools. We also found that the rates and yields of hybrid N 2 O production were enhanced at low dissolved oxygen concentrations ([O 2 ]), with hybrid N 2 O yields as high as 20 % at depths where [O 2 ] was below detection (880 nM) but nitrification was still active. Finally, we identified a few incubations with [O 2 ] up to 20 μ M where N 2 O production from NO 3- was still active. A relatively high O 2 tolerance for N 2 O production via denitrification has implications for the feedbacks between marine deoxygenation and greenhouse gas cycling.

Abstract:Composting is widely used for organic waste management and is also a major source of nitrous oxide (N2O) emission. New insight into microbial sources and sinks is essential for process regulation to reduce N2O emission from composting. This study used genome-resolved metagenomics to decipher the genomic structures and physiological behaviors of individual bacteria for N2O sources and sinks during composting. Results showed that several nosZ-lacking denitrifiers in feedstocks drove N2O emission at the beginning of the composting. Such emission became negligible at the thermophilic stage, as high temperatures inhibited all denitrifiers for N2O production except for those containing nirK. The nosZ-lacking denitrifiers were notably enriched to increase N2O production at the cooling stage. Nevertheless, organic biodegradation limited energy availability for chemotaxis and flagellar assembly to restrain nirKS-containing denitrifiers for nitrate reduction toward N2O sources but insignificantly interrupt norBC- and nosZ-containing bacteria (particularly nosZ-containing nondenitrifiers) for N2O sinks by capturing N2O and nitric oxide (NO) for energy production, thereby reducing N2O emission at the mature stage. Furthermore, nosZII-type bacteria included all nosZ-containing nondenitrifiers and dominated N2O sinks. Thus, targeted strategies can be developed to restrict the physiological behaviors of nirKS-containing denitrifiers and expand the taxonomic distribution of nosZ for effective N2O mitigation in composting.

Abstract:Nitrous oxide (N2O) production pathways in a single stage, continuously fed partial nitritation-anammox reactor were investigated using online isotopic analysis of offgas N2O with quantum cascade laser absorption spectroscopy (QCLAS). N2O emissions increased when reactor operating conditions were not optimal, for example, high dissolved oxygen concentration. SP measurements indicated that the increase in N2O was due to enhanced nitrifier denitrification, generally related to nitrite build-up in the reactor. The results of this study confirm that process control via online N2O monitoring is an ideal method to detect imbalances in reactor operation and regulate aeration, to ensure optimal reactor conditions and minimise N2O emissions. Under normal operating conditions, the N2O isotopic site preference (SP) was much higher than expected - up to 40‰ - which could not be explained within the current understanding of N2O production pathways. Various targeted experiments were conducted to investigate the characteristics of N2O formation in the reactor. The high SP measurements during both normal operating and experimental conditions could potentially be explained by a number of hypotheses: i) unexpectedly strong heterotrophic N2O reduction, ii) unknown inorganic or anammox-associated N2O production pathway, iii) previous underestimation of SP fractionation during N2O production from NH2OH, or strong variations in SP from this pathway depending on reactor conditions. The second hypothesis - an unknown or incompletely characterised production pathway - was most consistent with results, however the other possibilities cannot be discounted. Further experiments are needed to distinguish between these hypotheses and fully resolve N2O production pathways in PN-anammox systems.

Abstract:Nitrous oxide (N 2 O) is a potent greenhouse gas and ozone depleting substance, with the ocean accounting for about one third of global emissions. In marine environments, a significant amount of N 2 O is produced by biological processes in Oxygen Deficient Zones (ODZs). While recent technological advances are making surface N 2 O concentration more available, high temporal and spatial resolution water-column N 2 O concentration data are relatively scarce, limiting global N 2 O ocean models' predictive capability. We present a N 2 O concentration, stable isotopic composition and isotopomer dataset of unprecedently large spatial coverage and depth resolution in the broader Pacific, crossing both the eastern tropical South and North Pacific Ocean ODZs collected as part of the GO-SHIP P18 repeat hydrography program in 2016/2017. We complement these data with dissolved gases (nitrogen, oxygen, argon) and nitrate isotope data to investigate the pathways controlling N 2 O production in relation to apparent oxygen utilization and fixed nitrogen loss. N 2 O yield significantly increased under low oxygen conditions near the ODZs. Keeling plot analysis revealed different N 2 O sources above the ODZs under different oxygen regimes. Our stable isotopic data and relationships between the N 2 O added by microbial processes (ΔN 2 O) and dissolved inorganic nitrogen (DIN) deficit confirm increased N 2 O production by denitrification under low oxygen conditions near the oxycline where the largest N 2 O accumulations were observed. The slope for δ 18 O-N 2 O versus site preference (SP, the difference between the central (α) and outer (β) N atoms in the linear N 2 O molecule) in the eastern tropical North Pacific ODZ was lower than expected for pure N 2 O reduction, likely because of the observed decrease in δ 15 N β . This trend is consistent with prior ODZ studies and attributed to concurrent production of N 2 O from nitrite with a low δ 15 N or denitrification with a SP >0‰. We estimated apparent isotope effects for N 2 O consumption in the ETNP ODZ of 3.6‰ for 15 N bulk , 9.4‰ for 15 N α , -2.3‰ for 15 N β , 12.0‰ for 18 O, and 11.7‰ for SP. These values were generally within ranges previously reported for previous laboratory and field experiments.

Abstract:Significance Nitrous oxide (N 2 O) is a potent ozone-depleting greenhouse gas. This work identifies a means by which N 2 O is generated during nitrification, or biological ammonia oxidation. Fertilizer use in agriculture stimulates nitrification, thus increasing the volume of N 2 O emissions worldwide. The results presented herein will inform models and strategies toward optimized, sustainable agriculture. Moreover, these results highlight a rare example of biological N–N bond formation.

Abstract:Abstract The ocean is estimated to contribute up to ~20% of global fluxes of atmospheric nitrous oxide (N 2 O), an important greenhouse gas and ozone depletion agent. Marine oxygen minimum zones (OMZs) contribute disproportionately to this flux. To further understand the partition of nitrification and denitrification and their environmental controls on marine N 2 O fluxes, we report new relationships between oxygen concentration and rates of N 2 O production from nitrification and denitrification directly measured with 15 N tracers in the Eastern Tropical Pacific. Highest N 2 O production rates occurred near the oxic‐anoxic interface, where there is strong potential for N 2 O efflux to the atmosphere. The dominant N 2 O source in OMZs was nitrate reduction, the rates of which were one to two orders of magnitude higher than those of ammonium oxidation. The presence of oxygen significantly inhibited the production of N 2 O from both nitrification and denitrification. These experimental data provide new constraints to a multi‐component global ocean biogeochemical model, which yielded annual oceanic N 2 O efflux of 1.7 – 4.4 Tg‐N (median 2.8 Tg‐N, 1 Tg = 10 12 g), with denitrification contributing 20% to the oceanic flux. Thus, denitrification should be viewed as a net N 2 O production pathway in the marine environment.

Abstract:Nitrous oxide (N2O) is an important greenhouse gas (GHG) with substantial global warming potential and also contributes to ozone depletion through photochemical nitric oxide (NO) production in the stratosphere. The negative effects of N2O on climate and stratospheric ozone make N2O mitigation an international challenge. More than 60% of global N2O emissions are emitted from agricultural soils mainly due to the application of synthetic nitrogen-containing fertilizers. Thus, mitigation strategies must be developed which increase (or at least do not negatively impact) on agricultural efficiency whilst decrease the levels of N2O released. This aim is particularly important in the context of the ever expanding population and subsequent increased burden on the food chain. More than two-thirds of N2O emissions from soils can be attributed to bacterial and fungal denitrification and nitrification processes. In ammonia-oxidizing bacteria, N2O is formed through the oxidation of hydroxylamine to nitrite. In denitrifiers, nitrate is reduced to N2 via nitrite, NO and N2O production. In addition to denitrification, respiratory nitrate ammonification (also termed dissimilatory nitrate reduction to ammonium) is another important nitrate-reducing mechanism in soil, responsible for the loss of nitrate and production of N2O from reduction of NO that is formed as a by-product of the reduction process. This review will synthesize our current understanding of the environmental, regulatory and biochemical control of N2O emissions by nitrate-reducing bacteria and point to new solutions for agricultural GHG mitigation.
