Calcium Channels and SNARE Proteins

Abstract:The interactions between the presynaptic Ca 2+ channel synprint site and soluble N -ethylmaleimide-sensitive factor attachment protein receptor (SNARE) proteins provide a molecular basis for a physical and functional coupling between Ca 2+ influx and subsequent exocytosis. SNARE proteins also retrogradely modulate the voltage dependence of steady-state inactivation on Ca v 2 channels, thus providing a feedback mechanism for fine-tuning Ca 2+ influx and, hence, synaptic transmission. Further regulation is imposed by the interplay between the synprint site interactions and the modulation of calcium channels through phosphorylation and G-proteins, suggesting that the mammalian brain has devised numerous pathways for the precise control of sequential events resulting in efficient neurotransmitter release.
