Effect of Different Dose Insulin Combined with Growth Hormone on the Secretion of Cytokine in Endotoxin-Stimulated Monocytes

Abstract:Objective:To investigate effect of insulin combined with growth hormone on the secretion of cytokine in endotoxin-stimulated monocytes in vitro.Methods: Peripheral blood monocytes were extracted from 8 healthy volunteers and then stimulated by endotoxin.According to the concentration of insulin and GH,monocytes were divided into five groups: A(without insulin and GH);B(100 ng/ml GH,no insulin);C-E(100 ng/ml GH,500,1 000 or 2 000 μU/ml insulin).After stimulation of lipopolysaccharide and incubation for 24 hours,tumor necrosis factor(TNF-α),interleukin-1β(IL-1β),interleukin-6(IL-6),interleukin-4(IL-4) and interleukin-10(IL-10) were tested by ELISA.Results: Concentrations of all pro-inflammatory and anti-inflammatory cytokine were significantly higher than that of group A after adding GH into culture media(P0.05).However,the concentrations of TNF-α in group D and E were lower than that of group B(P0.01) and the secretions of IL-1β and IL-6 in group C,D and E were also lower than that of group B.Compared with group B,insulin could notably increase the secretion of IL-4 in group D and E(P0.01).IL-10 upgraded in group E obviously(P0.01).Conclusions: GH has an effect on endotoxin-stimulated monocytes to secrete pro-inflammatory and anti-inflammatory cytokine and different doses of insulin combined with GH has the opposite effect.
