Abstract:The design and fabrication of a microfluidic cytometer system and its application for reticulocyte detection are described. This chip can count the target cells, which are focused at the detection window without sheath flow. This cytometer system based on optimized epifluoresence has a competitive advantage in the signal-to-noise ratio. Induced fluorescence from the reticulocyte binded with antibody is detected by the optical module and then transformed into the electronic signal by a photo multiplier tube. After signal processing, the results are automatically read out by a digital module and displayed on the system. To evaluate this microfluidic cytometer system, experiments employing polystyrene (PS) micro beads and induced reticulocyte of mice anemia are carried out, respectively, and the results illustrate that the microfluidic cytometer system is effective in detecting the reticulocyte.

Abstract:Summary Elucidation of the pathophysiological basis of platelet disorders in murine models requires a reliable method for the frequent determinations of platelet counts in individual mice. Here, we present a rapid, reproducible and accurate flow cytometric method for enumeration of platelets that involves fluorescent staining of platelets in whole blood with specific antibody and the addition of known numbers of fluorescent beads for standardization of the sample volume. Analysis of platelets obtained by tail bleeding indicated that this sampling procedure did not activate platelets, and that only five microliters of blood were required for platelet counting. Using this method, we followed platelet counts in mice infected with the relapsing fever spirochete Borrelia turicatae for 26 days, and found that this bacterium induces thrombocytopenia, a common manifestation of human relapsing fever. Therefore, this method can be used to follow the number and the activation state of circulating platelets from individual mice over extended periods of time and is applicable to a wide range of murine models of platelet disorders.

Abstract:The immature platelet fraction (IPF) is a measure of newly released platelets, which has been used as a marker of platelet production in multiple human studies but is not widely available in multispecies analyzers. We developed gates to measure the IPF in diluted and undiluted murine blood samples on the Sysmex XN-1000V multispecies hematology analyzer. IPF gates were created using undiluted and diluted (1/10) blood samples obtained from adult and newborn (postnatal day 10, P10) C57BL/6J wild-type (WT) mice, and from 3 murine models of thrombocytopenia: c-MPL-/- mice, which lack the thrombopoietin receptor (hyporegenerative); antibody-mediated thrombocytopenia; and acute inflammation-induced thrombocytopenia. P10 mice were chosen because, at their size, we could consistently obtain (by terminal phlebotomy) the blood volume needed to run an undiluted sample. The undiluted blood IPF gate successfully differentiated between mechanisms of thrombocytopenia in both adult and P10 mice. For diluted samples, 2 IPF gates were generated: a thrombocytopenic (T) gate, which performed well in samples with platelet counts (PCs) <800 × 109/L in adult mice and <500 × 109/L in newborn mice, and a non-thrombocytopenic (NT) gate, which performed well in samples with PCs above these thresholds. PCs and IPFs measured in diluted blood using these gates agreed well with those measured in undiluted blood and had good reproducibility. These diluted gates allow for the accurate measurement of PCs and IPFs in small (10 µL) blood volumes, which can be obtained easily from adult and newborn mice as small as P1 to assess platelet production serially.
Microfluidic assessment of functional culture-derived platelets in human thrombi under flow

Abstract:Despite their clinical significance, human platelets are not amenable to genetic manipulation, thus forcing a reliance on mouse models. Culture-derived platelets (CDPs) from human peripheral blood CD34(+) cells can be genetically altered and may eventually be used for transfusions. By use of microfluidics, the time-dependent incorporation of CD41(+)CD42(+) CDPs into clots was measured using only 54,000 CDPs doped into 27 mu L, of human whole blood perfused over collagen at a wall shear rate of 100 sec(-1). With the use of fluorescence-labeled human platelets (instead of CDPs) doped between 0.25% and 2% of total platelets, incorporation was highly quantitative and allowed monitoring of the anti-alpha IIb beta 3 antagonism that occurred after collagen adhesion. CDPs were only 15% as efficient as human platelets in their incorporation into human thrombi under flow, although both cell types were equally antagonized by alpha(IIb)beta(3) inhibition. Transient transfection allowed the monitoring of GFP(+) human CDP incorporation into clots. This assay quantifies genetically altered CDP function under flow. Abstract:Megakaryopoiesis and platelet production are complex biological processes that require tight regulation of successive lineage commitment steps and are ultimately responsible for maintaining and renewing the pool of circulating platelets in the blood. Despite major advancements in the understanding of megakaryocytic biology, the detailed mechanisms driving megakaryocytic differentiation have yet to be elucidated. Here we show that automated image analysis algorithms applied to two-photon excited fluorescence (TPEF) images can non-invasively monitor structural and metabolic megakaryocyte behavior changes occurring during differentiation and platelet formation in vitro. Our results demonstrate that high-contrast, label-free two photon imaging holds great potential in studying the underlying physiological processes controlling the intricate process of platelet production.

Abstract:Objective To establish an in vitro model for observing the real- time interaction between platelets and vascular vessel.Methods Thin glass tubes coated with von Willebrand faeter(vWF) were used to form the flow chambers.Siphonage system,peristaltic pump and a constant flow syringe pump were adopted to supply different kinds of flow and well defined shear rates.With inverted microscope and charge- coupled device(CCD),a series of reactions of platelets and chinese hamster ovary(CHO) cells transfected with glycoprotein Ib- Ⅸ under flow conditions were recorded simultaneously.Results Platelets and CHO cells transfected with glycoprotein Ib- Ⅸ rolled and steadily adhered on the tube wall through binding to vWF under different kinds of flow and well defined shear rates.Conclusion This flow chamber allows real- time observation and record of the interaction between platelets and vWF under well- defined shear rate.This approach can be applied to platelet adhesion research.

Abstract:An accurate and comprehensive assessment of platelet function is essential for managing patients who receive antiplatelet therapies or require platelet transfusion either for treating active bleeding or for prophylaxis. Platelets contribute to clotting by undergoing a series of highly regulated functional responses including adhesion, spreading, granular secretion, aggregation, and cytoskeletal contraction. However, current platelet function assays evaluate only partial aspects of this intricate process and often under non-physiological testing conditions. Herein, we describe the development of a new approach to measure multiple key platelet function-related parameters, in a more physiologically relevant ex vivo semi-rigid microenvironment using a membrane capacitance sensor (MCS). MCS response to clotting provided three sensing parameters with sensitivities towards platelet counts, stimulation strengths, and activation pathways. Live confocal fluorescent imaging of stimulated platelets on MCS suggests that the presented system can readily and accurately convert the dynamics of cytoskeletal reorganization into analyzable electrical signals. Together, this new completely electrical sensing platform can be a promising diagnostic venue to recognize the impairment of primary hemostatic functions, evaluate the efficacy of therapeutic interventions, and gain further insights into the mechanisms of platelets in hemostasis and thrombosis.
Establishment and identification of humanized SCID mouse model

Abstract:Objective:To establish and identify humanized-SCID mouse model(hu-SCID).Methods:SCID mouse was treated by CTX to inhibit the hemocytopoiesis. With successive 4-day injection, human peripheral blood mononuclear cells(PBMC) were engrafted into SCID mouse through intraperitoneal injection. After 4, 8 and 12 weeks of engraftment, peripheral blood, spleen and liver tissues of engrafted SCID mouse were harvested. Human CD3~+, CD19~+ cells in peripheral blood were analyzed by inflorescence microscopy and FCM, human CD3~+, CD19~+ cells in spleen and liver tissues were observed by immune histochemistry, and human IgG level in SCID mouse serum was measured by ELISA.Results:After engraftment of 4, 8 and 12 weeks, human CD3~+, CD19~+ cells in SCID peripheral blood were identified by inflorescence microscopy and the percents were 31% and 10% respectively by FCM analysis. And these cells could be evidenced after 12 weeks later. Through immune histochemistry human CD3~+、CD19~+ cells were detected in mouse spleen but not in liver tissue. Furthermore the titer of human IgG in mouse serum was 390,1 100 and 1 040 μg/ml at each time point respectively.Conclusion:Our experimental results demonstrated that a bona fide humanized SCID model was established.
Development of a novel flow cytometry‐based approach for reticulocytes micronucleus test in rat peripheral blood

Abstract:The micronucleus test (MNT) is the most widely applied short‐term assay to detect clastogens or spindle disruptors. The use of flow cytometry (FCM) has been reported for micronucleated erythrocytes scoring in peripheral blood. The aim of this study was to develop a novel and practical protocol for MNT in rat peripheral blood by FCM, with the method validation. CD71‐fluorescein isothiocyanate and DRAQ5 were adopted for the fluorescent staining of proteins and DNA, respectively, to detect micronuclei. To validate the method, groups of male Sprague–Dawley rats (five per group) received two oral gavage doses at 0 and 24 h of six chemicals (four positive mutagens: ethyl methanesulphonate [EMS], cyclophosphamide [CP], colchicine [COL], and ethyl nitrosourea [ENU]; two nongenotoxic chemicals: sodium saccharin and eugenol). Blood samples were collected from the tail vein before and on the five continuous days after treatments; all of which were analyzed for micronuclei presence by both the manual (Giemsa staining) and FCM methods. The FCM‐based method consistently demonstrated highly sensitive responses for micronucleus detection at all concentrations and all time points for EMS, CP, COL, and ENU. Sodium saccharin and eugenol could be identified as negative in this protocol. Results obtained with the FCM‐based method correlated well with the micronucleus frequencies (r = 0.659–0.952), and the proportion of immature erythrocytes (r = 0.915–0.981) tested by Giemsa staining. The method reported here, with easy operation, low background, and requirement for a regular FCM, could be an efficient system for micronucleus scoring.

Abstract:An in vivo flow cytometer is developed that allows the real-time detection and quantification of circulating fluorescently labeled cells in live animals. A signal from a cell population of interest is recorded as the cells pass through a slit of light focused across a blood vessel. Confocal detection of the excited fluorescence allows continuous monitoring of labeled cells in the upper layers of scattering tissue, such as the skin. The device is used to characterize the in vivo kinetics of red and white blood cells circulating in the vasculatúre of the mouse ear. Potential applications in biology and medicine are discussed.

Abstract:Objective To establish a flow-cytometric method to detect microvesicles (MVs) in rat peripheral blood, and to detect platelets-derived MVs (PMVs) and endothelial cell-derived MVs (EMVs) in blood from ischemic precondition-ing (IPC) treated rats. Methods Blood was withdrawn from rat abdominal aorta and anticoagulated with sodium citrate. Platelets-free plasma (PFP) was isolated through two centrifugations at room temperature. PFP was incubated with FITC-conjugated mouse anti-rat CD61 or PE-conjugated mouse anti-rat CD144. Standard beads in diameter of 1 and 2μm were used for calibration and absolute counting, respectively. Analysis was performed on flow cytometer. Results When 3.5%so-dium citrate was mixed with blood at volume ratio of 1∶4, clear supernatant was collected after centrifugation. Signals of parti-cles smaller than 1μm accounted for more than 99%of overall signals. PMVs and EMVs were CD61 positive and CD144 positive, respectively. Their diameters were both smaller than 1 μm. The concentration of PMVs and EMVs in peripheral blood from IPC treated rats was (4 053±1 987)/μL and (4 870±825)/μL, respectively. Conclusion The method for MVs de-tection by flow cytometry was successfully established and optimized, and verified through detecting PMVs and EMVs in pe-ripheral blood from IPC treated rats.

Abstract:Background: As cancer metastasis is the deadliest aspect of cancer, causing 90% of human deaths, evaluating the molecular mechanisms underlying this process is the major interest to those in the drug development field. Both therapeutic target identification and proof-of-concept experimentation in anti-cancer drug development require appropriate animal models, such as xenograft tumor transplantation in transgenic and knockout mice. In the progression of cancer metastasis, circulating tumor cells (CTCs) are the most critical factor in determining the prognosis of cancer patients. Several studies have demonstrated that measuring CTC-specific markers in a clinical setting (e.g., flow cytometry) can provide a current status of cancer development in patients. However, this useful technique has rarely been applied in the real-time monitoring of CTCs in preclinical animal models. Methods: In this study, we designed a rapid and reliable detection method by combining a bioluminescent in vivo imaging system (IVIS) and quantitative polymerase chain reaction (QPCR)-based analysis to measure CTCs in animal blood. Using the IVIS Spectrum CT System with 3D-imaging on orthotropic-developed breast-tumor-bearing mice. Results: In this manuscript, we established a quick and reliable method for measuring CTCs in a preclinical animal mode. The key to this technique is the use of specific human and mouse GUS primers on DNA/RNA of mouse peripheral blood under an absolute qPCR system. First, the high sensitivity of cancer cell detection on IVIS was presented by measuring the luciferase carried MDA-MB-231 cells from 5 to 5x1011 cell numbers with great correlation (R2 = 0.999). Next, the MDA-MB-231 cell numbers injected by tail vein and their IVIS radiance signals were strongly corrected with qPCR-calculated copy numbers (R2 > 0.99). Furthermore, by applying an orthotropic implantation animal model, we successfully distinguished xenograft tumor-bearing mice and control mice with a significant difference (p < 0.001), whereas IVIS Spectrum-CT 3D-visualization showed that blood of mice with lung metastasis contained more than twice the CTC numbers than ordinary tumor-bearing mice. We demonstrated a positive correlation between lung metastasis status and CTC numbers in peripheral mouse blood. Conclusion: Collectively, the techniques developed for this study resulted in the integration of CTC assessments into preclinical models both in vivo and ex vivo, which will facilitate translational targeted therapy in clinical practice.

Abstract:Platelets are anucleate cytoplasmic fragments derived from megakaryocytes (MKs) that play crucial roles in blood clotting system and innate immune response. The clinical needs of platelet transfusion are extensive and increasing, while current donor-derived platelet supply confronts challenges including limited donors, short shelf life of platelet products, the risk of bacterial and viral contamination and transfusion refractoriness. The construction of a non-donor-dependent system for efficient platelet generation to provide a consistent supply of platelets could address these concerns. Therefore, in vitro differentiation of human pluripotent stem cells (PSCs) have been studied to serve as a potential source of MKs and platelets to meet the demands of clinical transfusion. The immortalized megakaryocyte cell lines (imMKCLs) were previously established as cryopreservable master cells that can be robustly expanded and then produce platelets upon maturation, but the yield of platelets was far too short from that postulated in vivo (Nakamura, 2014). To overcome this issue, we sought to establish a reporter system that can monitor imMKCL maturation in vitro in high throughput manner to identify compounds that improve their maturation and platelet release.

Abstract:The occurrence of vaso-occlusive crisis greatly depends on the competition between the sickling delay time and the transit time of individual sickle cells, i.e., red blood cells from sickle cell disease (SCD) patients, while they are traversing the circulatory system. Many drugs for treating SCD work by inhibiting the polymerization of sickle hemoglobin (HbS), effectively delaying the sickling process in sickle cells (SS RBCs). Most previous studies on screening anti-sickling drugs, such as voxelotor, rely on in vitro testing of sickling characteristics, often conducted under prolonged deoxygenation for up to 1 hour. However, since the microcirculation of RBCs typically takes less than 1 minute, the results of these studies may be less accurate and less relevant for in vitro-in vivo correlation. In our current study, we introduce a computer vision-enhanced microfluidic framework designed to automatically capture the transient sickling kinetics of SS RBCs within a 1-min timeframe. Our study has successfully detected differences in the transient sickling kinetics between vehicle control and voxelotor-treated SS RBCs. This approach has the potential for broader applications in screening anti-sickling therapies.

A Microfluidic Digital Single-Cell Assay for the Evaluation of Anticancer Drugs.

Abstract:Digital single-cell assays hold high potentials for the analysis of cell apoptosis and the evaluation of chemotherapeutic reagents for cancer therapy. In this paper, a microfluidic hydrodynamic trapping system was developed for digital single-cell assays with the capability of monitoring cellular dynamics over time. The microfluidic chip was designed with arrays of bypass structures for trapping individual cells without the need for surface modification, external electric force, or robotic equipment. After optimization of the bypass structure by both numerical simulations and experiments, a single-cell trapping efficiency of ∼90 % was achieved. We demonstrated the method as a digital single-cell assay for the evaluation of five clinically established chemotherapeutic reagents. As a result, the half maximal inhibitory concentration (IC50) values of these compounds could be conveniently determined. We further modeled the gradual decrease of active drugs over time which was often observed in vivo after an injection to investigate cell apoptosis against chemotherapeutic reagents. The developed method provided a valuable means for cell apoptotic analysis and evaluation of anticancer drugs.

Abstract:As a label-free and high-throughput single cell analysis platform, impedance flow cytometry (IFC) suffers from clogging caused by a narrow microchannel as mechanical constriction (MC). Current sheath constriction (SC) solutions lack systematic evaluation of the performance and proper guidelines for the sheath fluid. Herein, we hypothesize that the viscosity of the non-conductive liquid is the key to the performance of SC, and propose to employ non-conductive viscous sheath flow in SC to unlock the tradeoff between sensitivity and throughput, while ensuring measurement accuracy. By placing MC and SC in series in the same microfluidic chip, we established an evaluation platform to prove the hypothesis. Through modeling analysis and experiments, we confirmed the accuracy (error < 1.60% ± 4.71%) of SC w.r.t. MC, and demonstrated that viscous non-conductive PEG solution achieved an improved sensitivity (7.92×) and signal-to-noise ratio (1.42×) in impedance measurement, with the accuracy maintained and free of clogging. Viscous SC IFC also shows satisfactory ability to distinguish different types of cancer cells and different subtypes of human breast cancer cells. It is envisioned that viscous SC IFC paves the way for IFC to be really usable in practice with clogging-free, accurate, and sensitive performance.
Establishment of Cherimic Hupbl-Scid-hcc Model in SCID Mice by Intraperitoneal Injection of Human Peripheral Blood Monocytes

Abstract:Objective To explore the feasibility of constructing cherimic huPBL SCID HCC model in SCID mice by intraperitoneal injection of human peripheral blood monocytes. Methods Human PBLs were isolated from peripheral blood of healthy donors and intraperitoneally injected into SCID mice at a dose of 2×10 7 cells/mouse. Simultaneously, 5×10 5 HepG2 cells in 0 2 ml were intracutaneously injected into the right flank of the mice. Two and 4 weeks later, CD3 expressin in PBMCs isolated from the mice were analyzed by FACS and human IgG in mouse blood was determined by ELISA. In week 4, splenic cells of the mice were prepared to determine cytotoxicity against HepG2 cells and detect human lymphocytes immunohisochemically in cryostat section. Results The tumorigenecity rate was 100% with latent period of 12 to 18 days. In the second and fourth week after injection, the content of human IgG in the mouse blood reached 69 8 μg/ml and 125 9 μg/ml and the ratio of CD3 + cells in the mouse blood was 10 5% and 3 5%, respectively. In the fourth week after the injection, cytotoxicity of the mouse spleen cells was 20 3% and existence of CD3 + cells in the mouse spleen was demonstrated immunohisochemically. Pathological examination showed tumor growth. Conclusions The huPBL SCID HCC model can be successfully established by combination of intraperitoneal injection of human peripheral blood monocytes and subcutaneous injection of liver cancer cells in SCID mice.
A New Assay Method for Late CFU-S Formation and Long-Term Reconstituting Activity Using a Small Number of Pluripotent Hemopoietic Stem Cells.

Abstract:We have previously reported that Lin(-)/CD71(-)/MHC class I-high/c-kit(<low) bone marrow cells (c-kit(<low) cells) are pluripotent hemopoietic stem cells (P-HSCs), since they have the capacity to self-renew for at least 2 years in mice and differentiate into all hemopoietic lineage cells over the long term when serial bone marrow transplantation is carried out using 500 c-kit(<low) cells. In addition, we have found that the c-kit(<low) cells do not form colony-forming units-spleen (CFU-S) on days 8 to 14 but form late CFU-S (after 16 days). In the present study, to confirm that c-kit(<low) cells are truly P-HSCs, we examine whether a few (less than or equal to50) c-kit(<low) cells can form late CFU-S and reconstitute lethally irradiated recipients. We have established a new method to rescue lethally, irradiated mice by transplantation of a few cells so that they survive for more than 16 days: 0.2 ml of 20 Gy-irradiated peripheral blood (PB) was injected into the recipients every 3 days. All the mice that had been transplanted with 25 or 50 c-kit(<low), cells alone died within 12 days, and no CFU-S were detected in their spleens. However, when 25 or 50 c-kit(<low) cells were injected and 0.2 ml of 20 Gy-irradiated PB was injected every 3 days, the recipients survived, and a small number of CFU-S were detected after 16 days. About 40% of the recipients injected with 50 c-kit(<low) cells and about 15% of those injected with 25 c-kit(<low) cells survived for more than 6 months. Moreover, donor-derived multilineage cells were detected in all the hematolymphoid organs of the recipient mice. This new assay method using a small number of cells would be of great advantage for clarifying which cells are truly P-HSCs.
A Microfluidic Approach for Cryoprotectant Screening: Preliminary Validation with Human Red Blood Cells.

Abstract:BACKGROUND:The application of microfluidics has overcome a series of long-term existing bottlenecks in the field of cryobiology. However, there is still no investigation regarding the on-chip rapid screening of cryoprotectant concentration optimization for the integration of the entire freeze, thaw and cell counting process.OBJECTIVE:To establish an on-chip approach for rapid cryoprotectant screening and concentration optimization.MATERIALS AND METHODS:Rapid freezing of red blood cells with glycerol and dimethyl sulfoxide was used to validate the proposed on-chip method.RESULTS:The optimal cryoprotectant and its concentration can be determined with the described method, without the need of additional samplings and assays. The optimal concentration for glycerol and dimethyl sulfoxide used in the validation experiments was 10% (w/v).CONCLUSION:The microfluidic approach can be used for rapid cryoprotectant screening and concentration optimization.
An Integrated Microfluidic Blood Sampler for Determination of Blood Input Function in Quantitative Mouse Micropet Studies

Abstract:In this study, we developed and constructed a blood sampling system on an integrated microfluidic platform to withdraw sub-microliter blood samples from mice up to a rate of one to two samples per second. On one microfluidic chip, eighteen blood samples can be collected on designated wells and can be retrieved later for further analysis. The chip is flushed with Heparin lock flush between sample collections. The device is remotely controlled by a user-friendly interface and can be programmed to take samples at specific time interval. Preliminary results show that both saline solution and FDG blood samples (/spl sim/220 nano-liter/sample) can be taken with a consistent volume (variation < 2.2% s.d.) at a rate of one sample per second. Although the volumes are small, the FDG activities in blood samples are detectable with low variability (<1.2%). Our study demonstrates the feasibility of deriving input function from mice using a microfluidic blood sampling device. The blood loss (<100 /spl mu/l) and the impact on physiological change is expected to be minimized.
