Abstract:The pollution of heavy metals and harmful elements in traditional Chinese medicine is one of the main problems that hinder the internationalization of traditional Chinese medicine. At present, major developed countries have incorporated elemental residues into drug risk assessment to ensure drug quality and safety, while China's drug risk assessment system is still in its infancy. In recent years, China has made efforts to improve the risk assessment system of traditional Chinese medicine. Researchers have reported the risk assessment of heavy metals and harmful elements in traditional Chinese medicine, aiming to evaluate the quality and safety of traditional Chinese medicine and promote the integration of traditional Chinese medicine with international standards. This study reviews the research reports in recent years, compares the pharmacopoeia standards of major developed countries, pinpoints the similarities and differences between different drugs in terms of hazard identification, hazard characteristics, and assessment methods, and discusses the possible problems and development directions of the risk assessment system of traditional Chinese medicine. This review is expected to improve the limit standards of traditional Chinese medicine elements and the risk assessment system of traditional Chinese medicine, thus providing support for enhancing the international competitiveness of traditional Chinese medicine products.

Abstract:The accumulation of heavy metal in circulating TCMs has attracted widespread attention because the security and therapeutic efficacy are inevitably imperiled by the survival ecological environment and human production activities. How to reduce the pollution level and improve the toxicity damage becomes an urgent issue. This article provides a comprehensive overview of the current status of heavy metal contamination over a thousand types of single herbal (botanical, animal and mineral medicines) and TCM preparations published over nearly two decades. The survey revealed that growth ecosystems (soil, water sources), anthropogenic factors (harvesting, processing, storage), specific varieties and medicinal parts utilized as well as the inherent resistance capacity are the key factors that affect the accumulation of heavy metals in TCMs. And Pb, Cu and Cr are the major cumulative elements for botanicals, while mineral and animal medicines are dominated by As and Cu elements, respectively. Ongoing efforts aimed at mitigating the level and translocation rate of heavy metals by optimized cultivation processes, appropriate processing methodologies and advanced adsorption techniques are effective removal strategies. And the prospects of TCMs as a detoxifying agent for heavy metal toxicity damage posed development potential. Besides, the correlation between the speciation of arsenic (As) and chromium (Cr) and their toxicity should also be elaborated in order to provide effective references for standardizing drug dosage and cycle. And the imperative from the perspective of improving limitations standards of HMs for animal medicines, external preparations and folk medicines as well as exploring the interaction mechanisms between heavy metals and active ingredients of TCMs provides the direction for the follow-up study.

Abstract:The products of traditional Chinese medicine play an important role in keeping healthy of people.Limited quantity of heavy metals in traditional Chinese medicine is a pledge in the international market of herbal medicine.In this paper the factors affecting the amount of heavy metals in traditional Chinese medicine including soils,characters of medicinal plants,the discharge of industrial waste,the pollution of pesticides and fertilizers and pollution in process of store were firstly analyzed and some principal measures of controlling the pollution of heavy metals in traditional Chinese medicine were raised.Meanwhile,the study state of the pollution of heavy metals in traditional Chinese medicine and the main problems in it were also expounded.At last,in order to offer some beneficial basis to the development of traditional Chinese medicine industry the some suggestions were raised and some prospects in the pollution of heavy metals in traditional Chinese medicine were put forward.

Abstract:A total of 33 pesticides have been banned from Chinese medicinal materials and decoction pieces(plants) according to Chinese Pharmacopoeia(2020 edition). According to the chemical structures, they are mainly divided into seven categories: organophosphorus compounds, organochlorines, carbamates, amidines, sulfonylureas, phenylpyrazoles, and ethers. These banned pesticides exhibit neurotoxicity, reproductive toxicity, immune system toxicity, teratogenicity, carcinogenesis, and mutagenesis, seriously damaging human and animal health. They affect not only the quality and safety of traditional Chinese medicines and resulting products, but also their competitiveness in the international market. Due to the numerous varieties of traditional Chinese medicines and their complex substrates, it is necessary to establish a universal and highly sensitive method for pesticide residue detection. This review systematically summarized the residual status, toxicity, and analytical methods of banned pesticides in traditional Chinese medicines, and forecasted the prospects of different analytical techniques, so as to provide reference for further safety and risk assessment of banned pesticide residues in traditional Chinese medicines, thus ensuring the safe production of traditional Chinese medicines.

Current situation analysis and countermeasures on contamination of heavy metal in traditional Chinese medicinal materials in China

Abstract:Recent years, with the rapid development of industrialization in China, the contamination of heavy metal was worsening, and traditional Chinese medicinal materials(TCMM) were also suffered. The data were collected in the literatures in the past five years, and Green Trade Standard of Importing Exporting Medicinal Plants Preparations was used as the standard to evaluate the pollution conditions of lead(Pb), cadmium(Cd), mercury(Hg), arsenic(As), and copper(Cu). The five heavy metals exceeded the limits of the standard and the percentages were 9.66%, 26.35%, 13.0%, 9.32%, and 16.09%, respectively. Moreover, statistics on the residues of the heavy metals in different categories of TCMM from various origins were also carried out in this paper. The results showed that the contamination of heavy metals in TCMM differently depended on the origins and categories. Studies on the rapid detection methods and the limit standards of heavy metal should be established immediately. At the same time, the regulations should be formulated timely and supervision also should be intensified. To solve the problems of excessive amounts of heavy metals in TCMM, necessary measures should be taken to control the contamination of TCMM.

Abstract:With the increasing trade of traditional Chinese medicine(TCM),the issue of excessive levels of toxic heavy metals metals in TCM has raised worldwide concerns. Some countries and re?gions have set strict limits on the heavy in TCM. There are currently no uniform limits on heavy metals, partly for lack of a good understanding of heavy metals in TCM. The toxicity of heavy metals varies with their chemical forms so that it is not rational to evaluate safety in terms of total contents. Cinnabar and realgar are widely used TCM containing heavy metals in clinical practice. Because of the potential toxicity of heavy metals such as mercury or arsenic,and the related adverse reactions reported occa?sionally,the safety of cinnabar,realgar and their preparations has also attracted public attention. In the present paper,the progress in toxicological studies on cinnabar,realgar as well as commonly used preparations was reviewed while priorities of future research were proposed.

Abstract:Traditional Chinese medicine (TCM) has unique effects in the prevention and treatment of diseases, and it makes great contributions to the development of society and the progress of medical civilization. However, the problem of excessive heavy metals in TCM has seriously hindered its development. This paper presents a brief and systemic review for the determination of heavy metals by the recently developed atomic techniques, including atomic absorption spectrometry (AAS), atomic fluorescence spectrometry (AFS), inductively coupled plasma-optical emission spectrometry (ICP-OES) and inductively coupled plasma-mass spectrometry (ICP-MS). Abstract:Marine traditional Chinese medicines (MTCMs) hold a significant place in the rich cultural heritage in China. It plays an irreplaceable role in addressing human diseases and serves as a crucial pillar for the development of China's marine economy. However, the rapid pace of industrialization has raised concerns about the safety of MTCM, particularly in relation to heavy metal pollution. Heavy metal pollution poses a significant threat to the development of MTCM and human health, necessitating the need for detection analysis and risk assessment of heavy metals in MTCM. In this paper, the current research status, pollution situation, detection and analysis technology, removal technology and risk assessment of heavy metals in MTCM are discussed, and the establishment of a pollution detection database and a comprehensive quality and safety supervision system for MTCM is proposed. These measures aim to enhance understanding of heavy metals and harmful elements in MTCM. It is expected to provide a valuable reference for the control of heavy metals and harmful elements in MTCM, as well as the sustainable development and application of MTCM.

Abstract:A comprehensive evaluation of the chemical forms of heavy metals, rather than their total amount of elements, is the basis of the scientific and objective evaluation of safety in heavy metals. As mercury, the most toxic heavy metal in the environment (such as in water, soil, and air), accumulates in medicinal plants, chemical forms of mercury in medicinal plants and their preparation need to be clearly understood. This study aims to summarize the chemical status of mercury in bulk Chinese medicinal herbs and traditional Chinese medicine preparations and to discuss research methods for their analysis. Further, widespread high-performance liquid chromatography–inductively coupled plasma mass spectrometry, advanced synchrotron X-ray absorption fine structure spectroscopy, and X-ray fluorescence were used for the in situ analysis of the chemical state of mercury. The results were then analyzed to improve the analytical methods and evaluation standards for the chemical state of mercury in Chinese herbal medicines.

[Safety evaluation of heavy metals contaminated Xiaochaihu Tang using health risk assessment model].

Abstract:In order to further improve the quality and safety evaluation standards of traditional Chinese medicine compound preparation,Xiaochaihu Tang compound preparations were selected to analyze the pollution level of heavy metals deeply,and the potential health risks were evaluated under taking such kind of compound preparations. In this study,the contents of copper( Cu),arsenic( As),cadmium( Cd),mercury( Hg),and lead( Pb) in different Xiaochaihu Tang compound preparations were determinated by the method of inductively coupled plasma mass spectrometry( ICP-MS). Moreover,combined with target hazard coefficient method and in vitro artificial system,the bioaccessibility and health risk level was investigated in three main consumption ways including powder,decoction and granule. The result was showed that,under the maximum residual limit set by International Standard Organization,only one batch of raw herb was eight times exceeded the Hg MRL,however,in water decoctions and granules,the heavy metal residue rate was reduced to 2. 02%( Hg in granules)-42. 85%( Cd in granules). So,the heavy metal pollutions and health risks can be reduced to safe region in spite of the serious pollution in raw herbs. Besides,the THQ and CR values of the three consumption methods were lower than the standard values of non-carcinogenic and carcinogenic risks of each heavy metal. It can be seen that even if the heavy metals in the raw herbs exceed the standard,the use of Xiaochaihu Tang decoction and granules can reduce the harm of heavy metals to the human body. Above all,the establishment of this health risk assessment model can be provided experimental basis and reference value for improving the safety evaluation standard of other heavy metals contained traditional Chinese medicine( TCM) compound preparations,and further improving the quality control methods of other different toxic compounds in clinical use.

Abstract:Traditional Chinese medicine has become an important pillar of "Healthy China" and the national medical system in recent years. Due to the wide range of raw materials in traditional Chinese medicinal materials (TCMM), the issue of metals has attracted more and more attention. In this paper, a comprehensive review of public reports on metals in TCMM in recent decades was conducted. From a total of 1969 reported articles, a total of 296 research reports on metals in TCMM were screened. The 296 reports involved 255 species in 85 families, with a total of 274 medicinal materials. These TCMM were divided into taproot-type, leaf-type, flower and fruit-type, herba-type, stem-type and bark-type medicinal materials according to the medicinal parts. The content of five metals lead (Pb), cadmium (Cd), mercury (Hg), arsenic (As), chromium (Cr) in these TCMM was noted, and the distribution rules for metals were analyzed. The results showed that: (1) For the distribution of metals in different medicinal parts, Pb was mainly distributed in leaves; Cd was mainly distributed in flowers and fruits, stems and leaves; Hg was mainly distributed in barks; As was mainly distributed in stems; Cr was mainly distributed in stems, flowers and fruits. (2) The areas with the highest risk of metal residues were the Qinghai-Tibet Plateau, south China, and southwest China. (3) Among all types of TCMM, herba-type medicinal materials had the highest risk of metal content. (4) Combined with the pharmacopoeia metal limit standards implemented in 2019, the exceeding rate of Pb in TCMM was the highest, with a maximum value of 37.67%; among the six major types of TCMM, the medicinal materials with the highest exceeding rate were herba-type medicinal materials, among which Hg had the highest exceeding rate of 23.08%; in terms of medicinal parts, the highest exceeding rate of metals was in leaf-type medicinal materials, among which Pb had the highest exceeding rate of 37.67%. On the whole, the situation in regard to metal residues in TCMM was acceptable, but it cannot be ignored. It needed to be paid attention to in the industrialization and management of TCMM.

Abstract:Scientific risk assessment of exogenous and endogenous toxic substances in traditional Chinese medicine (TCM) is of great significance. The present review comprises a comprehensive summary of progress in the health risk assessment of harmful exogenous substances in TCMs. Such substances include heavy metals, pesticide residues, biotoxins, and endogenous toxic components involving pyrrolizidine alkaloids. The review also discusses the strengths and weaknesses of various bioaccessibility and bioavailability models, and their applications in risk assessment. Future avenues of risk assessment research are highlighted, including further exploration of risk assessment parameters, innovation of bioaccessibility and bioavailability techniques, enhancement of probabilistic risk assessment combined with bioavailability, improvement of cumulative risk assessment strategies, and formulation of strategies for reducing relative bioavailability (RBA) values in TCMs. Such efforts represent an attempt to develop a risk assessment system that is capable of evaluating the exogenous and endogenous toxic substances in TCMs to ensure its safe use in clinics, and to promote the sustainable development of the TCM industry.

Abstract:Traditional Chinese medicines (TCMs) are widely used to treat various diseases in China and some countries, and TCM products are becoming increasingly available and popular worldwide. But TCMs are facing the challenge of heavy metal pollution. In this work, we examined the total contents and fractionations of Pb, Cd, Hg, and Cu in six TCMs (Angelicae Sinensis Radix (ASR), Chuanxiong Rhizoma (CR), Polygonati Rhizoma (PR), Astragali Radix (AR), Carthami Flos (CF), and Paeoniae Radix Rubra (PRR)) and evaluated the health risk of four heavy metals in these TCMs. The results showed that Cd, Pb, and Cu contents were considerably high and the amount of Cd in six TCMs, Pb in CR, ASR, AR, and CF, and Hg in ASR, PR, and PRR exceeded the limit values. The predominant fractions of Pb, Cd, and Cu were exchangeable and carbonate fractions in six TCMs; Hg mainly existed in organic and residual fractions. The average daily intake dose (ADD) and target hazard quotient (THQ) of Pb based on total content and total THQ of four heavy metals based on bioaccessible fractions in AR and PRR exceeded the safety guideline. These results indicated that the potential health risk could occur by taking these TCMs.

Abstract:Herbal medicines that are widely used worldwide are easily contaminated by pesticides and heavy metals, threatening human health. In this study, a modified QuEChERS pre-treatment method combined with HPLC/GC-MS/MS was established for the determination of 24 pesticide residues in Dendrobium candidum. The average recoveries of 24 pesticides in D. candidum were 76.9-110.0% with the relative standard deviation (RSD) of 0.28-11.40%, and their limits of detection (LOD) and limits of quantitation (LOQ) were 0.005-10 and 0.011-22 mu g kg(-1), respectively. The results showed that 83.33% of all samples had detected pesticide residues with the concentrations of 0.06-312.83 mu g kg(-1). Meanwhile, microwave digestion combined with ICP-MS was used to detect the residues of 8 heavy metals in D. candidum. The average recoveries of 8 heavy metals were 82.7-108.1% with an RSD of 1.4-8.0%, and their LOD and LOQ were 0.0001-0.05 mg kg(-1) and 0.0003-0.2 mg kg(-1), respectively. The results indicated that 8 heavy metals were all detected in all samples, and the highest concentration of Zn was 11.97 mg kg(-1). Furthermore, the health risk assessment showed that the risk of the detected pesticides and heavy metals in samples to humans, specifically to the general population including adults and children, was acceptable.

Abstract:<h3 class="a-plus-plus">Abstract</h3> Little is known about the extent of heavy metal accumulation in traditional Chinese medicines (TCMs). In this study, the levels of lead (Pb), cadmium (Cd), arsenic (As), and mercury (Hg) in traditional animal medicines were monitored using inductively coupled plasma mass spectroscopy (ICP-MS). Additionally, for the first time, a heavy metal risk assessment strategy was used to evaluate the potential risks of traditional animal medicines by calculating estimated daily intake (EDI), target hazard quotient (THQ), and cancer risk (CR). To obtain a refined risk assessment, the frequency of exposure to traditional animal medicines was determined from questionnaire data, and the safe factor for TCM was applied. Based on the standard levels for leech, it was found that earthworm, hive, scorpion, and leech accumulated high levels of heavy metals. The combined THQ (cTHQ) values indicated that ingestion of most traditional animal medicines would not pose a risk to the health of either male or female human beings. However, it was indicated that attention should be paid to the potential risk associated with cicada slough, earthworm, scorpion, turtle shells, and hive. Among heavy metals, As and Hg contributed to a major extent to the risk to human health. The CR assessment for Pb and As indicated that, with the exception of earthworm, the cancer risk was less than the acceptable lifetime risk for both males and females. Owing to the higher body weight, both THQ and CR were generally lower for males than for females.
