The Biochemical Changes of Inferior Colliculus in Rats Before and after Auditory Deprivation:A Preliminary in Vivo 1H-Magnetic Resonance Spectroscopy Study

Abstract:Objective To employ in vivo proton magnetic resonance spectroscopy(7.0 T MRI)study the biochemical changes of inferior colliculus in rats after auditory deprivation and to explore the possible molecular mechanisms of plasticity in the central auditory system.Methods Proton magnetic resonance spectroscopy (1H-MRS) was carried out in 8 male adult rats.Each rat underwent two MRS scan:one scan assessed before auditory deprivation and the other scan assessed 4 days after auditory deprivation(bilateral cochlear ablation).The region of interest(ROI)was placed on the left inferior colliculus.The changes of the ratios of NAA /Cr and Cho/Cr were detected and analyzed.Results The ratio of Cho/Cr(0.637±0.044) in left inferior colliculus of auditory deprivation rats was significantly lower than that in normal rats(0.764±0.062,t=4.800,P0.01).The ratios of NAA/Cr had no significant difference between before and after auditory deprivation(1.192±0.057 vs 1.235±0.082,t=1.265,P0.05).Conclusion 1H-MRS is a potential tool for studying the metabolic changes in central auditory system in vivo.The ratio of Cho/Cr decrease in the inferior colliculus may be an earlier indicator of the damage to neuron by auditory deprivation.
