Intravenous Transplantation of Bone Marrow Mesenchymal Stem Cells Reduces Neointima Formation after Carotid Artery Injury in Rats

Abstract:To study the effects of bone marrow mesenchymal stem cells(BM-MSCs) intravenous transplantation on reendothelialization and neointima formation after carotid artery injury in rats,38 carotid deendothelialization rats were randomized into two groups: BM-MSCs group receiving intravenous transplantation of 1×10~6 cultured BM-MSCs,and control group receiving normal saline.7 days and 28 days after transplantation,animals were killed and the samples of carotid artery were harvested respectively.The samples were studied with scanning electron microscopy(SEM) for endothelialization,morphometrically analizing I/M ratio,immunohistochemistry staining for nuclear antigen(PCNA) and CD34 to evaluate whether intravenous transplantation of BM-MSCs affects proliferation of vessel intima and reendothelialization.SEM and CD34 immunohistochemistry staining showed that reendothelialization degree in BM-MSCs group was significantly higher compared with those in control group at both 7d and 28d(P<0.05).Meanwhile,I/M ratio and PCNA positive cells in BM-MSCs group were reduced significantly compared with those in control group at both 7d and 28d(P<0.05).Intravenous transplantation of bone marrow mesenchymal stem cells has a beneficial effect on the reendothelialization and may reduce neointima thickeness induced by the ballon injury in rats.
