Abstract:Aims To investigate novel biomarker for diagnosis of cervical cancer, we analyzed the datasets in Gene Expression Omnibus (GEO) and confirmed the candidate biomarker in patient sample. Materials and methods We collected major datasets of cervical cancer in GEO, and analyzed the differential expression of normal and cancer samples online with GEO2R and tested the differences, then focus on the GSE63514 to screen the target genes in different histological grades by using the R-Bioconductor package and R-heatmap. Then human specimens from the cervix in different histological grades were used to confirm the top 8 genes expression by immunohistochemical staining using Ki67 as a standard control. Results We identified genes differentially expressed in normal and cervical cancer, 274 upregulated genes and 206 downregulated genes. After intersection with GSE63514, we found the obvious tendency in different histological grades. Then we screened the top 24 genes, and confirmed the top 8 genes in human cervix tissues. Immunohistochemical (IHC) results confirmed that keratin 17 (KRT17) was not expressed in normal cervical tissues and was over-expressed in cervical cancer. Cysteine-rich secretory protein-2 (CRISP2) was less expressed in high-grade squamous intraepithelial lesions (HSILs) than in other histological grades. Conclusion For the good repeatability and consistency of KRT17 and CRISP2, they may be good candidate biomarkers. Combined analysis of KRT17, CRISP2 expression at both genetic and protein levels can determine different histological grades of cervical squamous cell carcinoma. Such combined analysis is capable of improving diagnostic accuracy of cervical cancer.
Analysis of Molecular Pathological Factors of Unfavorable Prognosis for Young Cervical Cancer Patients

Abstract:The aim of this study was to explore the molecular pathological characteristics of cervical cancer in women aged below 35, disclose the factors that indicate the unfavorable prognosis, and provide some useful targets for improving therapeutic outcome. Patients and Methods: 64 cases of pathological data originating from cervical cancer patients aged below 35 served as the study group to perform a retrospective research, and pathological data from 90 cases of cervical cancer patients beyond age 35, who underwent treatment during the same time, were selected at random as the reference group. Immunohistochemistry and quantified image analysis were utilized to detect synchronously the protein expression difference of survivin and p27 between the 2 groups. Results: The total 5-year survival rate of the study group was 65.6% compared to 84.4% in the reference group, while the lymph node metastasis rate was 25% compared to 12.2% in the reference group. Both differences were statistically significant (p < 0.05). Survivin expression is much greater in the study group than in the reference group (p < 0.05); there is no statistical difference of the p27 expression between the 2 groups (p > 0.05), but p27 expression is related to lymph node metastasis. Conclusion: Younger female patients have a worse prognosis than older ones, and survivin expression is associated with poor prognosis. Targeting these molecular factors may lead to a hopeful biomedical therapy to improve the prognosis of young female cervical cancer patients.

Abstract:Objective:To evaluate multiple biopsies and cervical conization in the clinical diagnosis and treatment of cervical cancer.Methods: Two thousand and five hundred females aged 30~49 years in a high risk area for cervical cancer underwent liquid based pap smear,high risk human papilloma virus(HPV) testing(13 sub-types),visual inspection of the cervix using acetic acid(VIA) and visual inspection of the cervix using lugol's iodine(VILI).Those with positive findings in any of these four screening tests underwent digital colposcopic exam,and some had biopsies taken from 4 regions of the cervix as well as endocervical curretage.Seventy-two patients with pathological findings of cervical intraepithelial neoplasia(CIN)2 or greater(including CIN2,CIN3,Carcinoma in situ(CIS) and microinvasive cancer) underwent cervical conization(the conventional first-line treatment),with final treatment(regularly scheduled follow-up or more extensive therapy) based on surgical pathology.Results: Pathological findings of the cone biopsies were correlated with that from colposcopy-directed biopsy in 39 patients(55%).In 16 patients,the diagnosis pointed to cancer;five patients increased from CIN2 to CIN3,and one increased from CIS to microinvasive cancer.Eleven patients(15%) had less findings on surgical pathology,for a total discrepancy rate of 45%.Conclusion:(1)The use of multiple biopsies should be promoted in screening for cervical cancer.(2)Cases of CIN3 should be actively treated.(3)Cervical conization should be the first line method used to diagnose and treat cases of pathology-confirmed high grade cervical changes(CIN2 and greater).
Analysis of 174 Cases with Cervical Cancer in Women under 35 Years Old

Abstract:OBJECTIVE:To analyse the prevalence, etiology, presentation, preserving ovary and prognosis of cervical cancer in women under 35 years old. METHODS:The clinical information of 174 patients were retrospectively analysed. RESULTS:The percentage of new cervical cancer cases from 1991 to 2001 was 1.2%, 1.2%, 4.3%, 4.2%, 4.6%, 4.5%, 7.3%, 9.0%, 10.7%, 9.4%, 10.8%, respectively (P < 0.01). Contact bleeding was the main symptom that occurred in 101 cases (101/174, 58.0%). Cervicitis was incorrectly diagnosed in 45 cases (45/174, 25.9%). Sex irregularities occurred in 51 cases (51/174, 29.3%). The number of cases with carcinoma in situ, stage I, stage II, stage III was 22, 40, 94, 18, respectively. There are 29 cases with lymph vascular involvement, 60 with 1/2 or more penetration of the cervical stroma. The incidence of HPV16/18 infection was 34%. The ovarian metastasis was 0.8%. The 5 year survival rate of stage I - III was 71.6%, 60.4% and 13.3%, respectively (P < 0.01). COX regression analysis indicated that stage, lymph-vascular involvement and the depth of tumor infiltration were independently prognostic factors. For patients with one risk factor after radical surgery, the survival of patients with adjuvant radiation therapy and chemotherapy or not was 100%, 88%, respectively (P > 0.05). For patients with two or more risk factors, it was 68%, 1/7, respectively (P < 0.05). CONCLUSIONS:There is an increasing prevalence of cervical cancer in women under 35 years old, which is possibly related to the sexual transition. The preserving ovary in young cervical cancer patients is safe and effective. Patients with late stage, lymph-vascular involvement and deep infiltration of cervical stroma have poor prognosis. Adjuvant treatment may be helpful for patients with 2 or more risk factors after radical surgery.

Abstract:Objective To evaluate the related risk factors that affect the prognosis of cervical cancer.Methods The clinical data of 505cases of cervical cancer patients who treated in the Department of Gynecologic Oncology of Peking University Shenzhen Hospital in the past 10years were analyzed retrospectively.The clinic pathologic clinicopathologic features and risk factors that related to the prognosis of cervical cancer were analyzed by statistical software SPSS 19.0.A statistically significant difference was indicated by P<0.05.Results ① The peak age of onset focus on 35~45years old which showed a single-peak,the 3-year and 5-year survival rates of all patients were 94% and 93%.② The multivariate regression analysis indicated that clinical stage,histology differentiated grades,platinum-based chemoradiotherapy were independent prognosis factors for cervical cancer patients(P<0.05).Conclusions The peak age of cervical cancer patients in this study focus on 35~45years old which showed a single-peak.The 3-year and 5-year survivals were higher than that reported in the literature.Histology differentiated grades,platinum-based chemoradiotherapy were independent prognosis factors for cervical cancer patients.
Demographics, Risk Factors and Survival Analysis of Lung Cancer in Young Adults (&lt;30 Years) in China-A Retrospective Cohort Study

Abstract:Abstract Background Lung cancer in very young adults in China aged <30 is on the rise and the clinicopathological characteristics of this unique population are incomplete and are yet to be elucidated. Our study aims to explore the characteristics, trends and distinctive features of lung cancer this age group to provide more information that can aid in developing optimal treatment regimens and improve prognosis for this exclusive age group. Method A retrospective review of 141 lung cancer patients admitted at our hospital from August 2011 to October 2021 was performed. The focused age group in our study was 18-30 years old. The data collected included demographics, tumor pathology, gene mutations, and treatment. Overall survival and prognosis were systematically analyzed. Results From a total of 141 of young lung cancer patients, 139 were included in final analysis. The gender distribution in our series favored female(65.5%), and only 7.2% of the study cohort had a history of smoking. Adenocarcinoma was the most common histological type in our study, presenting 93.5% of all patients. The most common stages at diagnosis was stage I(61.2%) and stage IV(30.2%). Genetic testing was performed in only 24.5% (34/139) of the patients. From the 23/34 patients in our study who tested positive for a genetic mutation, 19 (82.3%) were stage IV. The most common gene mutation was Anaplastic Lymphoma kinase(ALK) rearrangement observed in 13/23 (56.5%) gene mutation positive patients. Among stage IV NSCLC, OS was longer in patients who received targeted therapy, compared to the patients not receiving target therapy: 88 months vs 24 months (95% CI: 0.15-1.0, p=0.0282). Conclusion Lung cancer in the young adult population has particular characteristics (Common in female, non-smoker and adenocarcinoma patients). Higher stages of disease were observed in those with a gene mutation (notably ALK rearrangement). Although the sample size was small, the current data suggests a higher mutation rate in the younger population. These findings have fundamental implications for practice and we propose that all patients under 30 diagnosed with lung cancer should undergo genetic testing so they can receive personalized treatment and therefore achieve better outcomes.

Abstract:Introduction: Cervical cancer remains a significant healthcare problem, notably in low- to middle-income countries. While a negative test for hrHPV has a predictive value of more than 99.5%, its positive predictive value is less than 10% for CIN2+ stages. This makes the use of a so-called triage test indispensable for population-based screening to avoid referring women, that are ultimately at low risk of developing cervical cancer, to a gynecologist. This review will give an overview of tests that are based on epigenetic marker panels and protein markers. Areas covered: There is a medical need for molecular markers with a better predictive value to discriminate hrHPV-positive women that are at risk of developing cervical cancer from those that are not. Areas covered are epigenetic and protein markers as well as health economic considerations in view of the fact that most cases of cervical cancer arise in low-to-middle-income countries. Expert opinion: While there are biomarker assays based on changes at the nucleic acid (DNA methylation patterns, miRNAs) and at the protein level, they are not widely used in population screening. Combining nucleic acid-based and protein-based tests could improve the overall specificity for discriminating CIN2+ lesions that carry a low risk of progressing to cervical cancer within the screening interval from those that carry an elevated risk. The challenge is to reduce unnecessary referrals without an undesired increase in false-negative diagnoses resulting in cases of cervical cancer that could have been prevented. A further challenge is to develop tests for low-and middle-income countries, which is critical to reduce the worldwide burden of cervical cancer.
Clinical features of 153 female patients with advanced breast cancer under 35 years of age and their prognosis analysis

Abstract:Objective: To explore the molecular type, metastasis characteristics and the prognostic factors of advanced breast cancer in women under 35 years of age. Methods: Retrospective analysis on clinical data of 153 patients from January 2011 to January 2016 treated in Department of Breast Oncology, Peking University Cancer Hospital was carried out in this study. Univariate and multivariate analyses were conducted using Kaplan-Meier method and COX hazard model, respectively. Results: The median age of this cohort was 31.7 years, with a median disease-free survival (DFS) of 28.4 months. Of the 153 patients, 24 patients (15.7%) had the immediate family history of tumor, and 11 patients (7.2%) were diagnosed with initial TNM stage Ⅳ. Bone metastasis was observed in 63 patients (41.2%), and was most commonly seen in first metastasis. By the end of April 2016, the median survival time was 31.3 months, and the 5-year survival rate was 18.1%. Multivariate analysis revealed that the immediate family history of tumor, estrogen receptor (ER)-negative, progesterone receptor (PR)-negative, and larger tumor volume were the independent adverse prognostic factors (all P u003c 0.05). Conclusion: The prognosis of advanced breast cancer in young women is significantly affected by the age at the diagnosis, and is worse than other subtypes of breast cancer. Abstract:Cervical cancer is the most prevalent gynecologic malignancy, especially in women of low- and middle-income countries (LMICs). With a better understanding of the etiology and pathogenesis of cervical cancer, it has been well accepted that this type of cancer can be prevented and treated via early screening. Due to its higher sensitivity than cytology to identify precursor lesions of cervical cancer, detection of high-risk human papillomavirus (HR-HPV) DNA has been implemented as the primary screening approach. However, a high referral rate for colposcopy after HR-HPV DNA detection due to its low specificity in HR-HPV screening often leads to overtreatment and thus increases the healthcare burden. Emerging evidence has demonstrated that detection of host cell gene and/or HPV DNA methylation represents a promising approach for the early triage of cervical cancer in HR-HPV-positive women owing to its convenience and comparable performance to cytology, particularly in LMICs with limited healthcare resources. While numerous potential markers involving DNA methylation of host cell genes and the HPV genome have been identified thus far, it is crucial to define which genes or panels involving host and/or HPV are feasible and appropriate for large-scale screening and triage. An ideal approach for screening and triage of CIN/ICC requires high sensitivity and adequate specificity and is suitable for self-sampling and inexpensive to allow population-based screening, particularly in LMICs. In this review, we summarize the markers of host cell gene/HR-HPV DNA methylation and discuss their triage performance and feasibility for high-grade precancerous cervical intraepithelial neoplasia or worse (CIN2+ and CIN3+) in HR-HPV-positive women.

Deciphering age-specific molecular features in cervical cancer and constructing an angio-immune prognostic model

Abstract:Cancer incidence is increasingly seen in younger individuals. Molecular distinctions between young and elderly patients at onset are understudied. This study used public databases to explore genomic, transcriptomic, and immune-related features across age groups in cervical cancer. Additionally, it aims to create a prognostic model applicable across diverse age cohorts, enabling precise patient stratification, and personalized therapies. Gene mutations, expression data, and clinicopathological information were obtained from 317 cervical cancer patients. These patients were divided into a young group and an old group based on the median age of onset. The characteristics of differential gene mutation, gene expression, and immune cells analysis were analyzed by R software. Finally, the prognostic model was constructed by univariate Cox, least absolute shrinkage and selection operator, and multivariate Cox regression analyses of angiogenic and immune gene sets. Its validity was further confirmed using an additional 300 cervical squamous cell carcinoma and endocervical adenocarcinoma tissues. Cervical cancer patients at elderly onset age exhibit a significantly higher frequency of NOTCH1 and TP53 driver mutations compared to young patients, along with a notably higher tumor mutational burden. However, there were no significant differences between the 2 groups in terms of genomic instability and age-related mutational signatures. Differential gene expression analysis revealed that the young group significantly upregulated interferon-alpha and gamma responses and exhibited significantly higher activity in multiple metabolic pathways. Immune microenvironment analysis indicated enrichment of dendritic cells and natural killer cells in the young group, while transforming growth factor-β signature was enriched in the elderly group, indicating a higher degree of immune exclusion. A multigene prognostic model based on angiogenesis and T cell immune gene sets showed excellent prognostic performance independent of clinical factors such as age. High-risk groups identified by the model exhibit significant activation of tumor-promoting processes, such as metastasis and angiogenesis. Our study reveals distinct patterns in cancer-driving mechanisms, biological processes, and immune system status between young and elderly patients at onset with cervical cancer. These findings shed light on the age-specific underlying mechanisms of carcinogenesis. Furthermore, an independent molecular prognostic model is constructed to provide valuable references for patient stratification and the development of potential drug targets.

Abstract:Cervical cancer is one of the most common types of cancer and the fourth leading cause of cancer‑related deaths in women. The occurrence and development of cervical cancer is a multifactorial and multilevel process, which usually occurs alongside a continuous high‑risk human papillomavirus infection. With further developments in molecular biology and the advancement of sequencing technology, the role of biomarkers in cervical diseases has been gradually recognized. Therefore, it remains a priority to identify key molecular markers that can be used for the screening and triaging of the lesions. In recent years, numerous studies have been conducted in order to identify important markers for cervical diseases. The present review aimed to summarize the molecular alterations and clinical relevance of chromosomal alterations, DNA polymorphisms, the DNA methylation status, histone modifications, and alterations in microRNA and protein expression levels. Accumulating evidence suggests that molecular alterations may reflect the degree and the prognosis of the disease. Although significant progress has been made in the field of cervical cancer research, further samples and experiments are still required to identify crucial molecules.
A novel methylation-detection panel for HPV associated high-grade squamous intraepithelial lesion and cervical cancer screening

Abstract:Cervical cancer (CC) was considered to be the most common gynaecological cancer, with an estimated 342,000 deaths worldwide each year, as the majority of patients were diagnosed at an advanced stage of the disease. The purpose of this study was to evaluate the predictive value of multi-locus methylation assay for the early detection of CC. The cervical exfoliated cell samples from 492 HPV-positive females with cervical lesions were collected and subjected to methylation detection of gene FAM19A4, EPB41L3 and PAX1 after bisulfite conversion. The levels of gene methylation in patients with different severity of cervical lesions were evaluated and compared. The receiver-operating characteristic (ROC) curve was established and efficacy indexes such as sensitivity, specificity and area under the curve (AUC) were calculated to assess the diagnostic value of DNA methylation detection at multiple gene loci for CC. The methylation levels of FAM19A4, EPB41L3 and PAX1 were significantly increased with the grade of cervical squamous intraepithelial lesions. The sensitivities of FAM19A4, EPB41L3 and PAX1 alone for high-grade squamous intraepithelial lesion (HSIL) and CC diagnosis were 84.6%, 86.3% and 88.0%, respectively; when three markers were combined by a logistic regression model, the sensitivity was 88.0%, with a high specificity of 97.7% and AUC of 0.957 (95% CI 0.937–0.977). Methylation status of FAM19A4, EPB41L3 and PAX1 were highly specific and effective for monitoring the progression of cervical lesions and the tri-gene methylation assay could be used as a triage tool for CC early screening.

Abstract:Sensitive and specific biomarkers for the early detection of cervical cancer are urgently required to reduce the high morbidity and mortality of this disease. We previously demonstrated that circulating microRNAs (miRNAs) are correlated with certain types of human cancer. The aim of this study was to investigate the altered profile of serum miRNAs in cervical cancer patients in order to predict cervical cancer at a relative early stage. Serum samples were collected from 213 cervical cancer patients and 158 age-and ethnicity-matched controls. An initial screening of miRNA expression was performed by Solexa sequencing. Differential expression was validated using the stem-loop miRNA quantitative polymerase chain reaction (qPCR) assay in individual samples and the samples were arranged by two-phase selection and validation. The Solexa sequencing results revealed 12 markedly upregulated serum miRNAs in cervical cancer patients compared with controls. The reverse transcription-qPCR analysis identified a profile of 5 serum miRNAs (miR-21, -29a, -25, -200a and -486-5p) as a cervical cancer biomarker. The receiver operating characteristic curves indicated that a panel of 5 miRNAs constitutes a more sensitive and specific diagnostic test compared with any single miRNA-based assay, the squamous cell carcinoma antigen or the carbohydrate antigen 125. More importantly, miR-29a and miR-200a may indicate tumor histological grade and progression stage. Therefore, a 5-miRNA signature identified from genome-wide serum miRNA expression profiling may serve as a fingerprint for cervical cancer diagnosis.
The Rule of Mortality Risk in Young Women (≤35 Years) with Primary Breast Cancer

Abstract:21098 Background: The prognosis of young women (=35 years) is poorer than that of women over 35 years, which may indicate that they bear different biologic characteristics. However, such data is still not available at home and abroad as the rule of mortality risk in young women and its distinction from that in women over 35 years. We sought to investigate the clinical characteristics, prognosis and the rule of mortality risk in young women (=35 years) with primary breast cancer. Methods: We performed a retrospective study of 2331 female unilateral breast cancer patients undergoing surgery in our hospital from January 1990 to October 2005.They were divided into two groups with the cut-off of 35 years. Selection criteria included complete data on the following: age, tumor size, axillary nodal status, estrogen receptor (ER) status, menopausal status.Overall survival curve was estimated by Kaplan-Meier method and annual mortality hazard by hazard function. Their clinicopathologic indices, overall survival curve and annual mortality hazard were analyzed. Results: There was statistical significance between =35 years and >35 years group in terms of tumor size( P =0.0231), axillary nodal status( P <0.001) and estrogen receptor status( P =0.064). 10-year overall survival was 72% and 79% ( P = 0.0081), respectively, for =35 years and >35 years group. Although annual mortality hazard curve for both groups showed double-peaked pattern, there also exist some differences. Mortality surges for =35 years patients appeared earlier than those for >35 years counterparts. Moreover, =35 years group showed a minor increase between double peaks, which cannot be found in >35 years group. Conclusions: Between the two groups, different tumor biology leads to the discrepancy of overall survival and mortality pattern, which might indicate distinct principles and measures in the follow-up and treatment for young patients to provide the possibility of improving the therapeutic effect and reducing the mortality hazard. Identification of Circulating MicroRNAs as a Promising Diagnostic Biomarker for Cervical Intraepithelial Neoplasia and Early Cancer: A Meta-Analysis

Abstract:Background . Cervical cancer (CC) is one of the most common female malignant tumors. And cervical intraepithelial neoplasia (CIN) is the precancerous lesion of CC, which can progress to invasive CC. MicroRNAs (miRNAs) have been found to be potential diagnostic biomarkers for CIN or CC. However, recently, the lack of sufficient studies about the diagnostic value of miRNAs for CIN made it challenging to separately investigate the diagnostic efficacy of miRNAs for CIN. Likewise, the conclusions among those studies were discordant. Therefore, we conducted this meta-analysis, aimed at evaluating the diagnostic efficacy of miRNAs for CIN and CC patients. Methods . Literature search was performed in PubMed, Embase, and Web of Science databases. Pooled sensitivity, specificity, and other diagnostic parameters were calculated through Stata 14.0 software. Furthermore, subgroup analyses and metaregression analysis were conducted to explore the main sources of heterogeneity. Results . Ten articles covering 50 studies were eligible, which included 5,908 patients and 4,819 healthy individuals. The pooled sensitivity, specificity, positive likelihood ratio (PLR), negative likelihood ratio (NLR), diagnostic odds ratio (DOR), and area under the curve (AUC) were 0.81 (95% CI, 0.77-0.85), 0.86 (95% CI, 0.83-0.89), 5.9 (95% CI, 4.5-7.7), 0.22 (95% CI, 0.17-0.28), 27 (95% CI, 17-44), and 0.91 (95% CI, 0.88-0.93), respectively. Additionally, the ethnicity and internal reference were the main sources of heterogeneity. Conclusions . Circulating miRNAs can be a promising noninvasive diagnostic biomarker for CIN and early CC, especially miR-9 and miR-205, which need to be verified by large-scale studies.

Abstract:Purpose: The early detection of breast cancer in women under the age of 40 has posed significant challenges. This can be attributed in part to the limited research conducted on the breast cancer in this age group, particularly with regards to large sample sizes. We aimed to address this gap by analyzing and comparing the ultrasound imaging and pathological characteristics of breast cancer in women aged under 40 and those aged 40 and above. Methods: A retrospective assessment was conducted to examine the ultrasound imaging and clinicopathologic characteristics of 555 women with surgically confirmed breast cancers. The patient cohort consisted of 160 individuals below the age of 40 and 395 individuals aged 40 years and above. Results: Our study identified the breast cancer in patients under 40 years was more likely to show regular shape (p = 0.043) compared with tumors in patients who were 40 years and over. Furthermore, in young female patients (<40 years), irregular shape was correlated with the HER2-enriched type (p = 0.02), circumscribed margin (p = 0.001), and a lack of calcifications (p = 0.02) were associated with the triple-negative type. In another group (>= 40 years), only a lack of calcifications (p = 0.003) were associated with the triple-negative type. Conclusion: Breast cancer in women under the age of 40 exhibits distinct ultrasonographic characteristics patterns that vary across different immunophenotypes, which may provide certain predictive information for physicians.
Establishment of a molecular risk model for the prognosis of cervical cancer based on microRNA expression

Abstract:Background: Globally, the incidence of cervical cancer (CC) is highest among all tumors of the female reproductive system. Numerous studies have shown that the expression level of microRNA (miRNA) is highly correlated with cancer. This study aimed to establish a molecular prognostic model of CC based on miRNAs in order to provide more individualized treatment to CC patients.Methods: Human tissues were selected from the Cancer Hospital (Chinese Academy of Medical Sciences and Peking Union Medical College) for <i>miRNA</i> gene sequencing. CC transcriptome expression and clinical data were downloaded from The Cancer Genome Atlas (TCGA). We distinguished between common differentially expressed miRNAs of CC miRNA-seq and TCGA-CC. To obtain a molecular prognostic model, R package was used to perform univariate Cox proportional hazard regression and least absolute shrinkage and selection operator (LASSO) Cox regression for common differentially-expressed miRNAs. Next, the model performance was evaluated by survival analysis, receiver operating characteristic (ROC) curve analysis, as well as univariate and multivariate analyses in the TCGA-CC dataset. Quantitative Real-time polymerase chain reaction (qPCR) detection was to verify the expression changes of miRNA. Transwell was used to verify the role of molecules in CC cell migration and invasion.Results: Thirty-nine miRNAs were distinguished in TCGA-CC and CC miRNA-seq, LASSO regression analysis to obtain the risk model (risk score =-0.310× expression of hsa-miR-142-3p +0.439× expression of hsa-miR-100-3p). The survival analysis, ROC curve analysis, patient risk assessment, and univariate and multivariate analyses showed that the risk score model had good predictive ability in assessing patient survival (P<0.01), risk of death, and independent prognosis (P<0.01). qPCR detection of clinical samples and cells showed that the expression of hsa-miR-142-3p and hsa-miR-100-3p was consistent with the results of the database analysis. The Transwell results indicated that miR-142-3p is an inhibiting factor and miR-100-3p serves as a promoting factor in CC cell migration and invasion.Conclusions: Twelve miRNA-seq and TCGA-CC tissues were used to build a prognostic model for CC. We have obtained a two-miRNA risk score model. Our results provide a new strategy for the individualized diagnosis and treatment of CC.

Clinical Analysis of 301 Patients with Cervical Carcinoma

Abstract:Objective:To compare the clinical symptoms and pathological features of cervical cancer in different age types.Methods: A total of 301 patients with cervical cancer were reviewed.The patients were divided into two groups: young group(age,≤35 years) and old group(age,>35 years).Results: The average age was 45.07±0.52 years and the peak age was 35-46 years(54.2%).The proportion of patients with constant pregnancy(G≥3) and constant giving-birth(P≥2) were 56.5% and 33.9%,respectively.The proportion of stage Ia in the young group(44.2%) was higher than that in the old group(25.6%).The rate of adenocarcinoma was 16.3% in the young group but 6.7% in the old group(P<0.05).The rate of lymph node metastases was lower in the young group than that in the old group.The depths of vascular invasion,parametrium invasion and stromal invasion in the young group were less than those in the old group.Conclusions: In the young group,the clinical stage of cervical cancer is earlier than that in the old group,and the rate of adenocarcinoma is higher than that in the old group.
Clinical Pathological Analysis of 201 Young Women with Breast Cancer

Abstract:Objective To explore the clinicopathological characteristics in young women with breast cancer.Methods The clinicopathological data of 201 cases of breast cancer with age below 35 years from Apr.1997 to Apr.2010 in Anyang Tumor Hospital were retrospectively analyzed and compared with those of 251 cases of breast cancer patients selected randomly with age more than 60 years during the same period.All these cases had undergone surgical treatment and been confirmed by pathologist.Results In the young group,the percentage of patients with the course of disease less than six months was higher than that of the old-aged group [82.09 ％ (165/201),72.11％ (181/251)] (x2 =6.19,P ＜ 0.05).The incidence of the tumor size ≤ 2 cm in the young group was less than that of the old-aged group [19.90 ％ (40/201),30.28 ％ (76/ 251)] (x2 =6.302,P ＜ 0.05).The incidence rate of metastasis of lymph nodes in the young group was less than that of the old-aged group [63.68 ％ (128/201),54.19 ％ (136/251)] (x2 =4.145,P ＜ 0.05).The incidence rate of stage 0～ Ⅰ in the young group was more than that of the old-aged group [11.94 ％ (24/201),17.53 ％ (44/251)] (x2 =2.729,P ＞ 0.05).Conclusion The tumor size in the young group is larger than that in the old-aged group,pTNM classification in the young group is later than that in the latter group and the incidence rate of metastasis of lymph nodes in the young group is higher than that in the latter group.In young patients with breast cancer the percentage of tumors with advanced tumor stage and poor prognosis are more than that in elder breast cancer patients.In addition,these young patients have lower awareness to mammary adenocarcinoma.Young ladies should examine breast cancer by themselves and go to hospital for regular inspections in order to discovere,diagnose and treat cancer earlier.

Abstract:Objective To explore the clinical and pathological characteristics of breast cancer in young women(<35 years old)and to analyze their association with treatment and prognosis. Methods The records of 110 breast cancer in young women treated from January 2008 to December 2014 were reviewed. Statistical methods were used for analyzing the correlation between clinical and pathological characteristics such as T-stage, N-stage, IHC (Immunohistochemistry) and the morbidity as well as prognosis. The clinical and pathological characteristics of breast cancer in young women were compared with patients ≥35years using the χ2 test. Kaplan-Meier curves were reported for OS (overall survival) and PFS (progression-free survival), and the log-rank test was used to compare the difference in groups. Cox proportional models were fitted for multivariate analysis. Results All of the patients were women and the median age was 32 (21 to 34)-year old, accounted for 5.71%(110/2189) of the breast cancer treated at the same period. The morbidity of Luminal A-like and Luminal B-like(HER2 negative) in <35-year group were higher than ≥35-year group(P<0.001). Patients<35 years chose breast-conserving or mastectomy combined reconstruction surgery were much more than older group(P<0.001). The median follow-up duration of breast cancer in young women was 31.5 months, the 5-year OS rate was 93.3%and the 5-year PFS rate was 89.3%. It is concluded that the N-stage(P<0.001)and M-stage(P<0.001)were the influencing factors of the PFS by multivariate Cox regression analysis. The N-stage(P<0.001)was the influencing factor of OS. Conclusion Breast cancer in young women had special clinical and pathological characteristics. The endocrine therapy should be strengthened as well as protecting fertility. Patients<35 years demanded high quality of life and appearance, and the option of surgical treatment should be personalized.
