The Hepatoprotective Effect of Diltiazem on Acute Hepatitis Induced by Cocaine in Mice

Abstract:Objective To study the hepatoprotective effect of Diltiazem on acute experimental hepatitis induced by cocaine in mice and its possible mechanism.Methods The animals were randomly divided into five groups: three doses of Diltiazem groups(15 mg/kg,30 mg/kg and 60 mg/kg),a control group and a cocaine model group.The three groups were administration with Diltiazem for four days.At the fourth day,the three groups and the cocaine model group were given cocaine(80 mg/kg,ip);30 minutes after the Diltiazem were given.And all animals were killed after 24 hours.Serum alanine aminotransferase(ALT),lactate dehydrogenase(LDH),aspartateminotransferase(AST) were examined.Pathological changes of liver were observed.Results Comparing with the cocaine model group,the serum ALT,LDH,AST activities in Diltiazem groups were significantly decreased in response to cocaine(P0.01).Histopathological studies indicated that the liver pathological changes were lightened.Conclusion Diltiazem has some certain protective effect on acute liver injury caused by cocaine.The mechanism is probably concerned about blocking the hepatic microsomal cytochrome P450-mediated cocaine bioactivation and free radical scavenging effects.
