Study of effect of Yishenjuanshi decoction on extracelluar matrix in cultured glomerular mesangial cell

Abstract:Objective It is to observe the inhibiting effect of Yishenjuanshi decoction on the secretion of extracelluar matrix in cultured glomerular mesangial cells(GMCs).Methods GMC were cultured in cultured solutions contained low,medium and high dose of Yishenjuanshi decoction respectively with 10% serum-DMEM for 24?h,48?h and 72?h.After that,cell supernatant was collected to determine the concentrations of fibronectin(FN),laminin(LN) and collagen Ⅳ(Col Ⅳ) in it with Elisa method.Results Drug serum contained Yishenjuanshi could significantly inhibit GMC cultured in vitro to secrete FN,LN and ColⅣ.There was a dose-effect relationship between the inhibiting degree and drug concentration.The inhibition ratios increased accordingly when the dose of the decoction increased.Conclusion Yishenjuanshi has an obvious inhibiting function on secretion of FN,LN and ColⅣ of GMC cultured in vitro.It indicates that inhibiting GMC to secret extracelluar matrix is one of the important mechanisms of this decoction treating for chronic nephritis.
