Abstract:Paclitaxel, the first of a new class of microtubule-stabilizing anti-tumor agents, has been hailed as the most significant chemotherapy advance in 15 years. Early clinical results showed response rates of 24%-30% in platinum refractory ovarian cancer at doses ranging from 110-250 mg/m(2) repeated every three to four weeks. The dose limiting toxicity is neutropenia which is characteristically profound, but brief. Severe hypersensitivity reactions which occurred in the phase I studies can be prevented by the use of anti-allergic pre-medication. From a slow start constrained by drug availability, the clinical development of paclitaxel has accelerated dramatically as the issue of product supply has been solved. Substantial activity has been reported in advanced breast cancer (56% RR in pre-treated patients; 62% RR in patients untreated for advanced disease) and non-small cell lung cancer (NSCLC) (20%-30% RR in advanced stage IIIb/IV patients). Significant activity has been reported also in melanoma, small cell lung cancer (SCLC), bladder cancer, squamous cell carcinoma of the head and neck, and of the esophagus, cervical and endometrial cancer, as well as lymphomas. Combination chemotherapy studies have shown high activity in a variety of tumor types. Superiority over standard therapy has been demonstrated in ovarian and lung cancer in phase III randomized studies. Paclitaxel's novel mechanism of action and relative lack of clinical cross-resistance with other agents suggest a role in the combination therapy of early stage disease.

Abstract:Paclitaxel, a natural secondary metabolite isolated and purified from the bark of the Taxus tree, is considered one of the most successful natural anticancer drugs due to its low toxicity, high potency and broad-spectrum anticancer activity. Taxus trees are scarce and slow-growing, and with extremely low paclitaxel content, the contradiction between supply and demand in the market is becoming more and more intense. Therefore, researchers have tried to obtain paclitaxel by various methods such as chemical synthesis, artificial culture, microbial fermentation and tissue cell culture to meet the clinical demand for this drug. This paper provides a comprehensive overview of paclitaxel extraction, combination therapy, total synthesis, semi-synthesis and biosynthesis in recent years and provides an outlook, aiming to provide a theoretical basis and reference for further research on the production and application of paclitaxel in the future.

Taxol was originally isolated from the Pacific yew tree in 1967 and triggered an intense scientific and engineering venture to provide the compound reliably to cancer patients. The choices available for production include synthetic and biosynthetic routes (and combinations thereof). This chapter focuses on the currently utilized and emerging biosynthetic options for Taxol production. A particular emphasis is placed on the biosynthetic production hosts including macroscopic and unicellular plant species and more recent attempts to elucidate, transfer, and reconstitute the Taxol pathway within technically advanced microbial hosts. In so doing, we provide the reader with relevant background related to Taxol and more general information related to producing valuable, but structurally complex, natural products through biosynthetic strategies.

Abstract:Paclitaxel is a tricyclic diterpenoid first isolated from the bark of Taxus brevifolia. It is notably one of the most effective natural anticancer drugs discovered so far, and has been widely used in the treatment of breast, ovarian, lung cancer, as well as other diseases. However, the content of paclitaxel in the bark of yew trees is extremely low, thus alternative sources need to be developed urgently to meet the clinical demands. Using methods of synthetic biology to realize the heterologous expression of paclitaxel-related genes in microbial hosts could be a good choice. This paper reviews the advances in the researches on the biosynthesis of paclitaxel to provide new research strategies and insights.

Abstract:Paclitaxel, a rare diterpene extracted from the bark of Chinese yew (Taxus chinensis), is renowned for its anti-cancer activity and serves as a primary drug for treating cancers. Due to the exceptionally low content of paclitaxel in the bark, a semi-synthetic method that depletes Chinese yew resources is used in the production of paclitaxel, which, however, fails to meet the escalating clinical demand. In recent years, researchers have achieved significant progress in heterologous biosynthesis and metabolic engineering for the production of paclitaxel. This article comprehensively reviews the advancements in paclitaxel production, encompassing chemical synthesis, heterologous biosynthesis, and cell engineering. It provides an in-depth introduction to the biosynthetic pathway and transcriptional regulation mechanisms of paclitaxel, aiming to provide a valuable reference for further research on paclitaxel biosynthesis.

Abstract:The diterpenoid paclitaxel (Taxol ® ) is a blockbuster anticancer agent that was originally isolated from the Pacific yew ( Taxus brevifolia ) five decades ago. Despite the wealth of information gained over the years on Taxol research, there still remains supply issues to meet increasing clinical demand. Although alternative Taxol production methods have been developed, they still face several drawbacks that cause supply shortages and high production costs. It is highly desired to develop biotechnological production platforms for Taxol, however, there are still gaps in our understanding of the biosynthetic pathway, catalytic enzymes, regulatory and control mechanisms that hamper production of this critical drug by synthetic biology approaches. Over the past 5 years, significant advances were made in metabolic engineering and optimization of the Taxol pathway in different hosts, leading to accumulation of taxane intermediates. Computational and experimental approaches were leveraged to gain mechanistic insights into the catalytic cycle of pathway enzymes and guide rational protein engineering efforts to improve catalytic fitness and substrate/product specificity, especially of the cytochrome P450s (CYP450s). Notable breakthroughs were also realized in engineering the pathway in plant hosts that are more promising in addressing the challenging CYP450 chemistry. Here, we review these recent advances and in addition, we summarize recent transcriptomic data sets of Taxus species and elicited culture cells, and give a bird’s-eye view of the information that can be gleaned from these publicly available resources. Recent mining of transcriptome data sets led to discovery of two putative pathway enzymes, provided many lead candidates for the missing steps and provided new insights on the regulatory mechanisms governing Taxol biosynthesis. All these inferences are relevant to future biotechnological production of Taxol.

Abstract:During a screening performed by the National Cancer Institute in the 1960s, the terpenoid paclitaxel was discovered. Paclitaxel expanded the treatment options for breast, lung, prostate and ovarian cancer. Paclitaxel is only present in minute amounts in the bark of Taxia brevifolia. A sustainable supply was ensured with a culture developed from Taxus chinensis, or with semi-synthesis from other taxanes. Paclitaxel is marketed under the name Taxol. An intermediate from the semi-synthesis docetaxel is also used as a drug and marketed as Taxotere. O-Methylated docetaxel is used for treatment of some paclitaxel-resistant cancer forms as cabazitaxel. The solubility problems of paclitaxel have been overcome by formulation of a nanoparticle albumin-bound paclitaxel (NAB-paclitaxel, Abraxane). The mechanism of action is affinity towards microtubules, which prevents proliferation and consequently the drug would be expected primarily to be active towards cancer cells proliferating faster than benign cells. The activity against slowly growing tumors such as solid tumors suggests that other effects such as oncogenic signaling or cellular trafficking are involved. In addition to terpenoids, recently discovered microtubule-targeting polyketide macrolides and non-ribosomal peptides have been discovered and marketed as drugs. The revolutionary improvements for treatment of cancer diseases targeting microtubules have led to an intensive search for other compounds with the same target. Several polyketide macrolides, terpenoids and non-ribosomal peptides have been investigated and a few marketed.

Abstract:Breast cancer, as the most common malignancy in women worldwide, compels enormous efforts in therapeutic research. Paclitaxel, a microtubule stabiliser, has shown its versatility and efficacy in treating various breast cancer subtypes. However, its use is often associated with dose-dependent toxicity and tumour cell resistance. Recent advances in nanotechnology have opened new avenues for delivering paclitaxel, offering potential solutions to these problems. This study aims to summarise paclitaxel's latest applications and prospects in breast cancer treatment, focusing on nanoformulations. The research methods include reviewing and analyzing the latest data from preclinical testing of nano-paclitaxel formulations and their evaluation in clinical practice. In addition, paclitaxel's mechanism of action, clinical application, main side effects, and development of its nano-formulations are also extensively discussed. This paper thoroughly summarises the present condition and prospects of using paclitaxel for treating breast cancer, especially nanoformulations. It also highlights the need for continued research and development to improve the efficacy of paclitaxel and reduce its consequences to serve breast cancer patients better.

Abstract:Introduction: Paclitaxel and docetaxel were two epoch-making anticancer drugs and have been successfully used in chemotherapy for a variety of cancer types. In the year 2010, a new taxane, cabazitaxel, was approved by FDA for use in combination with prednisone for the treatment of metastatic hormone-refractory prostate cancer. Albumin-bound paclitaxel (nab-paclitaxel; abraxane) nanodroplet formulation was another notable invention (FDA approval 2005 for refractory, metastatic, or relapsed breast cancer). Abraxane in combination with gemcitabine for the treatment of pancreatic cancer was approved by FDA in 2013. Accordingly, there have been a huge number of patent applications dealing with taxane anticancer agents in the last 5years. Thus, it is a good time to review the progress in this area and find the next wave for new developments. Area covered: This review covers the patent literature from the year 2010 to early 2015 on various aspects of taxane-based chemotherapies and drug developments. Expert opinion: Three FDA-approved taxane anticancer drugs will continue to expand their therapeutic applications, especially through drug combinations and new formulations. Inspired by the success of abraxane, new nano-formulations are emerging. Highly potent new-generation taxanes will play a key role in the development of efficacious tumor-targeted drug delivery systems.

Abstract:Over the past decade, Taxol and its closely related structural analogue Taxotere have emerged as very important antitumor agents. Their widespread use in the treatment of a variety of cancer types, their likely approval for the treatment of additional forms of cancer, and their use at earlier stages of intervention will lead to increased demand for these drugs in the future. Because of yield considerations, Taxol and Taxotere are currently derived via semisynthesis from the advanced taxoid 10-deacetylbaccatin III, which must be isolated from yew (Taxus) trees. Thus, efforts are underway to produce Taxol (and other advanced taxoids for use in semisynthesis) by alternate, biotechnological means. This article provides a current overview of research on taxoid biosynthesis and an assessment of bioengineering applications for taxoid production in yew cell culture.

Abstract:Paclitaxel and its semi-synthetic analog docetaxel are currently considered two of the most effective drugs in cancer chemotherapy.However,their clinical utility is limited due to a number of undesirable difficulties,such as the dose-limiting toxicity related to paclitaxel′s side effects,low solubility and inactivity against drug resistant tumors.Various prodrugs of paclitaxel have been studied and developed to improve its bioavailability and tumor specificity and to keep or increase its activity against drug-resistant human cancer,which significantly increased the level of its treatment.In this article,the progress on the prodrugs of paclitaxel in recent years were reviewed.

Abstract:OBJECTIVE: Recently, many oversea and domestic researchers have carried out the various researches to study the anticancer mechanisms of taxinol extracted from plants, and reveal that taxinol has significant therapeutic effects on many cancer cells, which may conduce to the better application of taxinol in clinical works. DATA SOURCES: We indexed the pertinent literatures in Medline database from January 1979 to February 2006 on compute by the key words of "chemotherapy, tumor, taxinol", and the language was limited to English. STUDY SELECTION: All the related articles about the mechanism and clinical application of taxinol were selected. Inclusion criteria:①random control study,②basic or clinical research including parallel control group. Exclusion criteria: reproducible papers or reviews. DATA EXTRACTION: After the first trail, 58 papers referred to taxinol and anticancer mechanism were collected. Twenty papers were random control studies, basic or clinical research including parallel control group, which accorded to the inclusion criteria; Thirty-eight papers, which were reproducibility papers or reviews, were expelled. DATA SYNTHESIS: ①Taxinol, as a broad-spectrum anticancer drug, has satisfactory therapeutic effects for some advanced cancer which are resistant to ordinary anticancer drugs, and has significant curative effects for many clinical malignancies. Recent study results show that taxinol has a significant cytotoxic effect to Carpse sarcoma, high effects to P388 and P1534 leukemia, and inhibition effect to the growth of W256 sarcoma, S180 and lung cancer.②Taxinol may suppress tumor through regulating microtubule stabilization, inducing apoptosis and adjusting immunologic mechanism.③It is generally thought that the main target of taxinol is microtubule protein/microtubule system. Taxinol can promote the polymerization of microtubule and inhibit their degradation, through which taxinol can block cell division in G2/M stage and induce apoptosis of tumor cells. However, numerous researches have indicated that the effect of taxinol to microtubule and the blockage of cell division in G2/M stage are not the only mechanisms for the apoptosis induced by taxinol.④The novelty of taxinol's pharmacological action is that it can induce apoptosis through inhibiting the formation of microtubule and regulating the dynamic balance of microtubule protein dimer. CONCLUSION: ①Taxinol is thought as a broad-spectrum anticancer drug, which can further promote the therapeutic effect if combined with other ordinary anticancer drugs. Therefore taxinol is the other important anticancer drug after cyclophosphamide and adriamycin.②There are many signal conduction paths and mechanisms that may participate in the apoptosis reduced by taxinol.

Abstract:Paclitaxel and docetaxel are among the most widely used chemotherapeutic drugs against various types of cancer. However, these drugs cause undesirable side effects as well as drug resistance. Therefore, it is essential to develop next-generation taxoid anticancer agents with better pharmacological properties and improved activity especially against drug-resistant and metastatic cancers. The SAR studies by the authors have led to the development of numerous highly potent novel second- and third-generation taxoids with systematic modifications at the C-2, C-10, and C-3' positions. The third-generation taxoids showed virtually no difference in potency against drug-resistant and drug-sensitive cell lines. Some of the next-generation taxoids also exhibited excellent potency against cancer stem cells. This account summarizes concisely investigations into taxoids over 25 years based on a strong quest for the discovery and development of efficacious next-generation taxoids. Discussed herein are SAR studies on different types of taxoids, a common pharmacophore proposal for microtubule-stabilizing anticancer agents and its interesting history, the identification of the paclitaxel binding site and its bioactive conformation, characteristics of the next-generation taxoids in cancer cell biology, including new aspects of their mechanism of action, and the highly efficacious tumor-targeted drug delivery of potent next-generation taxoids.
