Abstract:Carotenoids serve diverse functions in vastly different organisms that both produce and consume them. Enhanced carotenoid accumulation is of great importance in the visual and functional properties of fruits and vegetables. Significant progress has been achieved in recent years in our understanding of carotenoid biosynthesis in tomato (Solanum lycopersicum) using biochemical and genetics approaches. The carotenoid metabolic network is temporally and spatially controlled, and plants have evolved strategic tactics to regulate carotenoid metabolism in response to various developmental and environmental factors. In this review, we summarize the current status of studies on transcription factors and phytohormones that regulate carotenoid biosynthesis, catabolism, and storage capacity in plastids, as well as the responses of carotenoid metabolism to environmental cues in tomato fruits. Transcription factors function either in cooperation with or independently of phytohormone signaling to regulate carotenoid metabolism, providing novel approaches for metabolic engineering of carotenoid composition and content in tomato.

Abstract:Carotenoids is a family of nutrient and bioactive components in vegetable and fruits.Recently considerable epidemical studies found that the vegetable and fruits containing carotenoids could reduce the risk of some chronic diseases.The bioactivities of carotenoids except for the sources of VA are focused on again,especially the common antioxidant activity of all the carotenoids,the preventing of prostate cancer of lycopene,the protecting of eyes and skin and the preventing of osteoporosis.The paper briefly introduced the structure and occurrence in food and human body,emphasized especially the new researches of bioactivities,and discussed the problems and possible researching fields.

Abstract:Abstract Carotenoids are isoprenoid metabolites synthesized de novo in all photosynthetic organisms. Carotenoids are essential for plants with diverse functions in photosynthesis, photoprotection, pigmentation, phytohormone synthesis, and signaling. They are also critically important for humans as precursors of vitamin A synthesis and as dietary antioxidants. The vital roles of carotenoids to plants and humans have prompted significant progress toward our understanding of carotenoid metabolism and regulation. New regulators and novel roles of carotenoid metabolites are continuously revealed. This review focuses on current status of carotenoid metabolism and highlights recent advances in comprehension of the intrinsic and multi-dimensional regulation of carotenoid accumulation. We also discuss the functional evolution of carotenoids, the agricultural and horticultural application, and some key areas for future research.

Abstract:Carotenoids have a range of diverse biological functions and actions, especially playing an important role in human health with provitamin A activity, anti-cancer activity, enhancing immune ability and so on. Human body can't synthesis carotenoids by itself and must absorb them from outside. However, carotenoid contents in many plant are very low, and many kinds of carotenoid are difficult to produce by chemical ways. With the elucidation of carotenoid biosynthetic pathway and cloning genes of relative enzymes from microorganisms and higher plants, it is possible to regulate carotenoid biosynthesis via genetic engineering. This article reviews gene cloning of carotenoid biosynthetic enzymes in microorganisms and higher plants, and advances in the studies of carotenoid production in heterologous microorganisms and crop plants using gene-manipulated carotenoid biosynthesis.

Abstract:Carotenoids are indispensable to plants and play a critical role in human nutrition and health. Significant progress has been made in our understanding of carotenoid metabolism in plants. The biosynthetic pathway has been extensively studied. Nearly all the genes encoding the biosynthetic enzymes have been isolated and characterized from various organisms. In recent years, there is an increasing body of work on the signaling pathways and plastid development, which might provide global control of carotenoid biosynthesis and accumulation. Herein, we will highlight recent progress on the biosynthesis, regulation, and metabolic engineering of carotenoids in plants, as well as the future research towards elucidating the regulatory mechanisms and metabolic network that control carotenoid metabolism.

Abstract:Carotenoids are one of the most important groups of natural food colorants beneficial for human health. Carotenoids exist widely in higher plants, alga, a few microorganisms and some animals, while biosynthetic pathways and compositions of carotenoids accumulated vary greatly. Through optimization of culture conditions, application of genetic transformation and hydrolases-assisted extraction, production of carotenoids was greatly improved and the cost reduced, facilitating the wider consumption of natural carotenoids as healthy food colorants.

Abstract:Carotenoids are naturally occurring pigments that are abundant in the natural world. Due to their excellent antioxidant attributes, carotenoids are widely utilized in various industries, including the food, pharmaceutical, cosmetic industries, and others. Plants, algae, and microorganisms are presently the main sources for acquiring natural carotenoids. However, due to the swift progress in metabolic engineering and synthetic biology, along with the continuous and thorough investigation of carotenoid biosynthetic pathways, recombinant strains have emerged as promising candidates to produce carotenoids. The identification and manipulation of gene targets that influence the accumulation of the desired products is a crucial challenge in the construction and metabolic regulation of recombinant strains. In this review, we provide an overview of the carotenoid biosynthetic pathway, followed by a summary of the methodologies employed in the discovery of gene targets associated with carotenoid production. Furthermore, we focus on discussing the gene targets that have shown potential to enhance carotenoid production. To facilitate future research, we categorize these gene targets based on their capacity to attain elevated levels of carotenoid production.

Abstract:Carotenoids are well-known isoprenoid pigments naturally produced by plants, algae, photosynthetic bacteria as well as by several heterotrophic microorganisms. In plants, they are synthesized in plastids where they play essential roles in light-harvesting and in protecting the photosynthetic apparatus from reactive oxygen species (ROS). Carotenoids are also precursors of bioactive metabolites called apocarotenoids, including vitamin A and the phytohormones abscisic acid (ABA) and strigolactones (SLs). Genetic engineering of carotenogenesis made possible the enhancement of the nutritional value of many crops. New metabolic engineering approaches have recently been developed to modulate carotenoid content, including the employment of CRISPR technologies for single-base editing and the integration of exogenous genes into specific “safe harbors” in the genome. In addition, recent studies revealed the option of synthetic conversion of leaf chloroplasts into chromoplasts, thus increasing carotenoid storage capacity and boosting the nutritional value of green plant tissues. Moreover, transient gene expression through viral vectors allowed the accumulation of carotenoids outside the plastid. Furthermore, the utilization of engineered microorganisms allowed efficient mass production of carotenoids, making it convenient for industrial practices. Interestingly, manipulation of carotenoid biosynthesis can also influence plant architecture, and positively impact growth and yield, making it an important target for crop improvements beyond biofortification. Here, we briefly describe carotenoid biosynthesis and highlight the latest advances and discoveries related to synthetic carotenoid metabolism in plants and microorganisms.

Abstract:Carotenoids are mostly C40 terpenoids, a class of hydrocarbons that participate in various biological processes in plants, such as photosynthesis, photomorphogenesis, photoprotection, and development. Carotenoids also serve as precursors for two plant hormones and a diverse set of apocarotenoids. They are colorants and critical components of the human diet as antioxidants and provitamin A. In this review, we summarize current knowledge of the genes and enzymes involved in carotenoid metabolism and describe recent progress in understanding the regulatory mechanisms underlying carotenoid accumulation. The importance of the specific location of carotenoid enzyme metabolons and plastid types as well as of carotenoid-derived signals is discussed.

Abstract:Carotenoids in plant foods are sources of pro-vitamin A and nutrients with several health benefits, including antioxidant and anticancer activities. However, humans cannot synthesize carotenoids de novo and must obtain them from the diet, typically via plant foods. We review the chemical changes of carotenoids in plant foods from farm to table and nutrition, including nutrient release and degradation during processing and metabolism in vivo. We also describe the influencing factors and proposals corresponding to enhancing the release, retention and utilization of carotenoids, thus benefiting human health. Processing methods influence the release and degradation of carotenoids, and nonthermal processing may optimize processing effects. The carotenoid profile, food matrix, and body status influence the digestion, absorption, and biotransformation of carotenoids in vivo; food design (diet and carotenoid delivery systems) can increase the bioavailability levels of carotenoids in the human body. In this review, the dynamic fate of carotenoids in plant foods is summarized systematically and deeply, focusing on changes in their chemical structure; identifying critical control points and influencing factors to facilitate carotenoid regulation; and suggesting multi-dimensional strategies based on the current state of food processing industries to achieve health benefits for consumers.

Abstract:Flavonol has a wide range of biological activities and studies on biosynthesis and regulation of flavonol in plant attract more attention.Here,research progress in key enzymes (FLS,F3H,F3′H,F3′5′H,UGT) and regulatory factors (e.g.MYB transcription factors) in flavonol biosynthetic pathway were summarized from biochemical and molecular biological points of view.Important evidence illustrating key structure genes or regulatory factors playing critical roles in flavonol biosynthesis are emphasized,which may result in efficient regulation of target molecules and accumulation of flavonol compounds with important biological functions via genetic engineering.

Abstract:Carotenoids are a group of widely distributed natural pigments. They give many horticultural plants the bright red, orange, and yellow colors, as well as the aroma and flavor. Carotenoids enhance the health value and represent an essential quality trait of horticultural products. Significant efforts have been made to correlate specific carotenoid production with pathway gene expression. Some transcription factors that directly regulate transcription of the pathway genes have been identified. Horticultural crops have evolved with complicated and multifaceted regulatory mechanisms to generate the enormous diversity in carotenoid content and composition. However, the diverse and complex control of carotenoid accumulation is still not well understood. In this review, we depict carotenoid accumulation pathways and highlight the recent progress in the regulatory control of carotenoid accumulation in horticultural plants. Because of the critical roles of chromoplasts for carotenoid hyperproduction, we evaluate chromoplast ultrastructures and carotenoid sequestrations. A perspective on carotenoid research in horticultural crops is provided.

Abstract:Pepper(Capsicum spp.)belongs to the genus Capsicum in the Solanaceae family and is widely cultivated around the world as a vegetable and spice crop.In addition to its application as a cooking ingredient and spice,pepper also has a wide range of uses in the pharmaceutical and cosmetic fields.Carotenoids are a class of natural pigments involved in many important metabolic processes in plants,such as photosynthesis,photoprotection,photomorphogenesis and growth and development.Carotenoid is one of the main nutrients of pepper fruit,which has various biological activities.To cultivate varieties with higher carotenoid content,it is necessary to understand its biosynthesis and molecular mechanism of regulation.The development of molecular biology and biotechnology has promoted the identification of carotenoid biosynthesis genes,providing opportunities for breeding new varieties of pepper with higher carotenoid content.In this review,the physiological role of carotenoid,carotenoid and pepper fruit color,carotenoid biosynthesis pathway,structural genes and regulatory factors of carotenoid biosynthesis pathway,molecular genetics of pepper fruit color and QTL sites related to pepper fruit color are described.GGPS,PSY,PDS,ZDS,LCY,CCS are port enzymes and rate-limiting enzymes in the carotenoid biosynthesis pathway of pepper,and the expression abundance and activity of them directly regulate the carotenoid content in pepper.Transcription factors such as MYB306,DIVARICATA1 and BBX20 are involved in the regulation of carotenoid biosynthesis in pepper.The diversity of carotenoid biosynthesis and regulatory genes determines the amount and content of carotenoid monomer and affects the color of pepper fruit.Based on the current research progress,future research on carotenoid metabolism of pepper is prospected in order to provide new ideas for breeding new varieties of biofortified pepper rich in carotenoids in the future.

Abstract:Carotenoids, natural tetraterpenoids found abundantly in plants, contribute to the diverse colors of plant non-photosynthetic tissues and provide fragrance through their cleavage products, which also play crucial roles in plant growth and development. Understanding the synthesis, degradation, and storage pathways of carotenoids and identifying regulatory factors represents a significant strategy for enhancing plant quality. Chromoplasts serve as the primary plastids responsible for carotenoid accumulation, and their differentiation is linked to the levels of carotenoids, rendering them a subject of substantial research interest. The differentiation of chromoplasts involves alterations in plastid structure and protein import machinery. Additionally, this process is influenced by factors such as the ORANGE (OR) gene, Clp proteases, xanthophyll esterification, and environmental factors. This review shows the relationship between chromoplast and carotenoid accumulation by presenting recent advances in chromoplast structure, the differentiation process, and key regulatory factors, which can also provide a reference for rational exploitation of chromoplasts to enhance plant quality.

Abstract:Carotenoids, which are inherent pigments occurring in plants and microorganisms, manifest a diverse array of vivid hues. Owing to their multifarious health advantages, carotenoids have engendered substantial interest among scholars and consumers alike. Presently, carotenoids are extensively employed in the realms of food, nutrition and health commodities, pharmaceuticals, and cosmetics, rendering them an indispensable constituent of our quotidian existence. Therefore, the objective of this review is to present a succinct and methodical examination of the sources, constituents, and factors influencing formation of carotenoids. Particular attention will be given to encapsulation strategies that maintain intrinsic characteristics, as the growing desire for carotenoids is propelled by individuals' escalating standards of living. Moreover, the applications of natural carotenoids in multiple fields, including pharmaceutical, food and feed, as well as cosmetics, are discussed in detail. Finally, this article explores the main challenges hindering the future advancement of carotenoids, aiming at facilitating their effective integration into the circular economy.
