Abstract:Human defensin is a family of cationic antimicrobial peptides in human being. During the last two decades a series of endogenous alpha-and beta-human defensins have been discovered. They are important components of the first barrier in human's body against the invasion of various microorganisms, and they are thought to play an important role in linking the innate and adaptive defense system of human being. The recent advances in the research of human defensins were reviewed, including their discovery, molecular and genetic properties, expression regulation, and mechanisms of antimicrobial activity. The possibility to produce human defensins via genetic engineering was also discussed. And the application outlook of human defensins in medicine and curing patients infected with antibiotics-resistant microbials was presented.

Abstract:Human defensins are a family of cationic antimicrobial peptides with molecular weights of 4–5kDa, containing a conserved six disulphide-linked cysteine motif. During the last two decades a series of endogenous α- and β-human defensins were discovered. They exhibit a broad range of antimicrobial properties and are thought to be ideal therapeutic agents because of their potential ability to circumvent the problems of acquired resistance often observed with other antimicrobial therapies. Because of their appealing medical and pharmaceutical potential there has been an emphasis on human defensins in medical and molecular biology research in recent years. This paper aims to present a comprehensive review of recent advances in the study of human defensins including their discovery, classification, molecular properties, expression, mechanisms of action and potential medical applications. In addition, the advances in producing human defensins via genetic engineered cells are summarized from research works in our group (besides host cells including E. coli, B. subtilis and yeast systems, the cell-free protein synthesis system was also employed to express human β-defensin-2) along with other related published works. The present challenges and prospects for the potential application of human defensins are also discussed.

Abstract:Antimicrobial peptides have important effects on the innate resistance system of human and livestock due to their activity against bacteria,viruses and fungi.They also are expected to be used as preventive and therapeutic medicine.Therefore they are called 'new generation antibiotics'.Antimicrobial peptides are cationic peptides,and the most numerous group of them is a family of defensins.Among them the most common are peptides with a beta-sheet structure containing three intra-molecular disulphide bonds,called defensins,comprising three classes: alpha-,beta-and theta-defensin.The family of beta-defensins is the largest one,and their transcripts have been found in many tissues of human and livestock.The aim of this paper is to present the current knowledge about antimicrobial peptides from the defensins family in farm animals,their expression,polymorphism,as well as the potential of their use as genetic markers of health and production traits.

Abstract:Defensin is a kind of cysteine-rich small peptide, which has a broad spectrum of resistance to bacteria with a special resistance mechanism. So far, a large number of studies on defensins have been reported, and the different types of defensins have been isolated from various organisms. A broad prospect of application on defensins has been displayed both in genetic engineering and medicine field. This article reviewed the classification and the biological characteristics of defensins, including mammalian α-, β-, θ-defensins, insect defensins, and plant defensins. The molecular structures, antibacterial activities, and antibacterial mechanisms of these definsins were summarized. The two mechanisms of de-fensin, including independent membrane mechanism and targeting of intracellular compounds by defensins, are ex-pounded. This paper also summarized the researches on isolation and expression of defensin genes and disease resistance genetic engineering of mammal and plant defensins. A prospect of the future applications of defensin both in biophar-maceutical sciences and plant disease resistance genetic engineering was discussed.

Abstract:Defensins are relatively small molecules and cationic antimicrobial peptides,less than 100 amino acids, which are active against the microorganisms including Gram-positive and Gram-negative bacteria, fungi, spirochaeta and some enveloped viruses. So research on defensins has become a hot spot in the world. Thereinto,β-defensins exist in poultry only. Different types of avian antimicrobial peptides were classified in terms of its source, homologous residues of amino acids and molecular framework. Avian defensins were abroad distributed in epithelium of blood, enteron, urine and procreative tracts of chicken. Evolutionary relationship between the avian β-defensins and the other vertebrates β-defensins indicated that there was a common ancestor of β-defensin genes. The peptides have shown potentially economical value. Therefore, this article focused on avian antimicrobial peptides.The classification and tissue distribution, evolution, bioactivities, gene engineering, and application of avian defensins were briefly reviewed.

Abstract:β-Defensins are cationic antimicrobial peptides (AMPs) that play an important role in the innate immune defense of bovines. They are constitutively expressed in mammary glands and induced differently in response to pathogens. Their expression is influenced by various factors, including hormones, plant-derived compounds, and dietary energy imbalance. The toll-like receptors (TLRs)/nuclear factor-kappa B (NF-κB) pathway plays a crucial role in β-defensin induction, while alternative pathways such as mitogen-activated protein kinase (MAPK) and epigenetic regulation also make substantial contributions. β-Defensins exhibit bactericidal activity against a wide range of pathogens, including two major mastitis pathogens, Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus), primarily through membrane disruption. β-Defensins have low cytotoxicity to host cells and demonstrate immunomodulatory properties, and pathogens also display minimal resistance to these AMPs. Given the increasing concern in antimicrobial resistance, the potential of β-defensins as natural antimicrobials has garnered considerable attention. This article provides an overview of the characteristics of bovine β-defensins, their expression pathways, their mode of action, and factors influencing their expression in the mammary glands of cattle. Additionally, it identifies the current gaps in research within this field and suggests areas that require further investigation. Understanding the regulation and function of β-defensins offers valuable insights to develop effective strategies for strengthening the immune system of mammary glands, reducing the reliance on synthetic antimicrobials, and explore novel natural antimicrobial alternatives.

Abstract:The arbitrary abuse of chemical additives and antibiotics has become one of the factors that restricting the development of animal husbandry,the result is that it produces the drug resistance and impacting the ruminants and human health.Plant and plant extracts posses the characteristics of antibacterial,growth-promoting,immunity-improving and anti-oxidation.For ruminant animals,it plays an important role,including ruminal fermentation,nutrition metabolism,products,growth and reproduction.As an efficient,safe,stable and controllable plant extracts and Chinese herbal medicine feed additives have increasingly become the substitutes of antibiotics and become the natural feed additives.This paper will discuss the application of the Chinese herbal medicine and plant extracts in the production of ruminants.

Abstract:Defensins are endogenous antimicrobial peptides (AMPs) produced by professional phago- cytes, Paneth cells, and epithelial cells at mucosal surfaces, which mediate innate immunity through their potent antimicrobial activity in the intestinal tract. In addition, defensins also regulate the function of diverse host immune cells, thereby play an important role in both innate and adaptive immune responses against pathogenic microbes. Abundant evidences have proved that attenuated changes in defensins expression are observed in inflammatory bowel disease (IBD). Further studies have discovered the concentration of different defensins subgroups has the relationship with various clinical characteristics of IBD and that mucosal surface destruction causes defensins deficiency as a result of in- flammatory damage. This article is to review new current approaches on defensins expression in the intestinal mucosa, particularly in IBD and their potential roles in immune responses in the gut mucosa.

Abstract:We report the biochemical and functional properties of a novel bovine beta-defensin (bBD-1). Cloned from bovine mammary papillary duct epithelia, the bBD-1 cDNA predicts a 69 amino acid propeptide that is much more similar to human beta-defensin-1 (hBD-1) than to other bovine defensins. The bBD-1 gene contains two exons and one 8.5 kb intron. Using RT-PCR, we detected the bBD-1 transcript in the teat mucosa, kidney, vagina, ovary, oviduct, and colon. A synthetic bBD-1 peptide demonstrates potent antibacterial activity against Escherichia coli. The widespread expression of bBD-1 mRNA indicates that bBD-1 may play an important role in the bovine host defense against infections.

Abstract:Insect defensin, a member of the family of small cationic antibiotic peptides, was found in the hemolymph of several orders. They contribute to natural and acquired immune against Gram-negative microorganisms, and they work with a novel mechanism without the help of antibody, complement system, and phagocyte. The molecular characteristics, gene and expression, biological effects, and mechanisms of insect defensins are completely reviewed, and also the recent advances in insect defensins are mentioned briefly.

Abstract:β-Defensins are small antimicrobial proteins expressed in various organisms and have great potential for improving animal health and selective breeding programs. Giant pandas have a distinctive lineage in Carnivora, and it is unclear whether β-defensin genes have experienced different selective pressures during giant panda evolution. We therefore characterized the giant panda (Ailuropoda melanoleuca) β-defensin gene family through gap filling, TBLASTN, and HMM searches. Among 36 β-defensins identified, gastrointestinal disease may induce the expression of the DEFB1 and DEFB139 genes in the digestive system. Moreover, for DEFB139, a significant positive selection different from that of its homologs was revealed through branch model comparisons. A Pro-to-Arg mutation in the giant panda DEFB139 mature peptide may have enhanced the peptide's antimicrobial potency by increasing its stability, isoelectric point, surface charge and surface hydrophobicity, and by stabilizing its second β-sheet. Broth microdilution tests showed that the increase in net charge caused by the Pro-to-Arg mutation has enhanced the peptide's potency against Staphylococcus aureus, although the increase was minor. We expect that additional gene function and expression studies of the giant panda DEFB139 gene could improve the existing conservation strategies for the giant panda.

Abstract:Vertebrate β-defensins comprise an important family of antimicrobial peptides that protect organisms from a diverse spectrum of bacteria, viruses, fungi, and protozoan parasites. Previous studies have shown a marked variation in the number of β-defensins among species, but the underlying reason is unclear. To address this question, we performed comprehensive computational searches to study the intact β-defensin genes from 29 vertebrates. Phylogenetic analysis of the β-defensin genes in vertebrates identified frequent changes in the number of β-defensin genes and multiple species-specific gene gains and losses that have been occurring throughout the evolution of vertebrates. The number of intact β-defensin genes varied from 1 in the western clawed frog to 20 in cattle, with numerous expansions and contractions of the gene family throughout vertebrates, especially among tetrapods. The β-defensin gene number in a species is relevant to the ever-changing microbial challenges from the environment that they inhabit. Selection pressure analysis shows there exist three amino acid sites under significant positive selection. Protein structural characteristics analysis suggests that structural diversity determines the diverse functions of β-defensins. Our study provides a new perspective on the relationships among vertebrate β-defensin gene repertoires and different survival circumstances, which helps explain how β-defensins have evolved.
