Abstract:A novel glutathione peroxidase (GPx) mimic has been prepared by incorporation of a selenium-based catalytic unit into the focal point of a fully-branched hyperbranched polymer. First, an AB2 monomer consisting of isatin and an electron rich aromatic moiety was polycondensed in the presence of 5-nitroisatin as a core reagent, resulting in a polymer with 100% degree of branching. The latter was coupled to the catalytically active moiety, Br(CH2)5SeSe(CH2)5Br, by nucleophilic substitution of the bromides by the residual amide groups of the incorporated nitroisatin core. The obtained polymer has demonstrated prominent GPx activity as desired, which could be attributed to the hydrophobic, densely branched and core-shell structure of the polymer surrounding the catalytic center.

Abstract:As one of the antioxidative enzymes exerting important biological roles in living bodies, the selenoenzyme glutathione peroxidase has received increased attentions. Over the past decades, much effort has been paid to mimic the catalytic behavior of glutathione peroxidase for exploring the enzymatic catalytic mechanisms and for developing potential antioxidative medicines. A great number of artificial GPxs, ranging from small molecular GPx mimics to macromolecular ones, and to nanoenzymes have been designed and prepared based on chemical, biological and supramolecular strategies. In this article, we specify the ideas and strategies for developing artificial GPxs based on macromolecules as scaffolds. The artificial system contains synthesized macromolecule-based selenoenzymes, self-assembled selenoenzymes, biomacromolecule-based selenoenzymes and so on.
A Novel Glutathione Peroxidase Mimic with Antioxidant Activity.

Abstract:Many diseases are associated with the overproduction of hydroperoxides that inflict cell damage. A novel cyclodextrin derivative, 6A,6B-diseleninic acid-6A',6B'-selenium bridged beta-cyclodextrin (6-diSeCD), was synthesized to be a functional mimic of glutathione peroxidase (GPX) that normally removes these hydroperoxides. The mimic had high catalytic GPX activity of 13.5 U/micromol, which is 13.6-fold higher than ebselen (PZ51), and was chemically and biologically stable in vitro. Antioxidant activity was studied by ferrous sulfate/ascorbate-induced mitochondria damage model system. These data show that the mimic has great antioxidant activity. Such mimics may result in better clinical therapies for diseases mediated by hydroperoxides.
Selenium-containing 15-Mer Peptides with High Glutathione Peroxidase-like Activity

Abstract:Glutathione peroxidase (GPX) is one of the most crucial antioxidant enzymes in a variety of organisms. Here we described a new strategy for generating a novel GPX mimic by combination of a phage-displayed random 15-mer peptide library followed by computer-aided rational design and chemical mutation. The novel GPX mimic is a homodimer consisting of a 15-mer selenopeptide with an appropriate catalytic center, a specific binding site for substrates, and high catalytic efficiency. Its steady state kinetics was also studied, and the values of k(cat)/K(m)(GSH) and k(cat)/ K(mH(2)O(2)) were found to be similar to that of native GPX and the highest among the existing GPX mimics. Moreover, the novel GPX mimic was confirmed to have a strong antioxidant ability to inhibit lipid peroxidation by measuring the content of malondialdehyde, cell viability, and lactate dehydrogenase activity. Importantly, the novel GPX mimic can penetrate into the cell membrane because of its small molecular size. These characteristics endue the novel mimic with potential perspective for pharmaceutical applications.

Abstract:Glutathione peroxidase (GPX) is a well-known selenoenzyme that functions as an antioxidant and catalyzes the reduction of harmful peroxide by glutathione and protects cells against oxidative damage. Because many diseases are related to oxidative stress, GPX is an ancient foe of many diseases. Antioxidants are very useful for biological bodies, and considerable effort has been spent to find compounds that could imitate the properties of GPX. This paper reviews GPX mimics developed so far and describes a new, more effective strategy for fabricating them. Although many GPX mimics have been made, they possess serious disadvantages: low activity, low solubility in water, and, in some cases, toxicity. In order to overcome these drawbacks, we have proposed a new strategy of imitating GPX. First, a receptor with a substrate binding site is generated. Next, a catalytic group is incorporated into the receptor near the substrate binding site, allowing the catalytic group access to the functional group of the substrate. Finally, a highly efficient enzyme mimic is obtained. Using this strategy, we successfully fabricated GPX mimics that use antibodies, cyclodextrins, some enzymes and proteins as receptors and chemical modification to incorporate the catalytic group, selenocysteine (Sec). The general principle of combining a functional group involved in catalysis with a specific binding site for the substrate is an approach that could be applied to the generation of other efficient semisynthetic biocatalysts. We describe the antioxidant activities of these GPX mimics and the reasons of their being promising candidates for medicinal applications. Abstract:Selenium (Se), known as an essential trace elements, is closely related to human health and diseases. This essential trace element exists in at least 25 selenoproteins in the form of selenocysteine residue, which performs a variety of biological functions. Among these selenoproteins,glutathione peroxidase (GPx) is a well-known antioxidase,since it catalyzes the reduction of harmful peroxide by glutathione and protects cells against oxidative damage. Herein,some strategies and methods are presented to construct GPx mimics based on protein scaffolds. Through these studies, we expect to gain better understanding of the biological properties of selenoprotein,and even develop more effective approaches to create new selenoprotein mimics.
Gene Manipulation Based Selenium-Containing Peptide Exhibiting Synergism of Sod and Gpx

Abstract:In this paper, we constructed a novel bifunctional superoxide dismutase(SOD)/glutathione peroxidase(GPx) mimic, a selenium-, copper-containing 35-mer peptide conjugate(Se-Cu-35P) in which a three-amino acid linker(Gly-Asn-Gly) connects the C-terminus of 17-mer polypeptide SOD mimic with the N-terminus of 15-mer polypeptide GPx mimic. The SOD and GPx activities of Se-Cu-35P are two orders of magnitude lower than those of natural SOD and GPx, respectively. It provides a GPx activity 56-fold higher than Ebselen(a well-known GPx mimic). The glutathione(GSH) binding constant is 5.6×102 L·mol−1. Se-Cu-35P synergistically resists against the inactivation by H2O2 and protects the mitochondria from oxidative damage in a dose dependent manner. These results highlight the challenge of generating an efficient SOD/GPx synergism mimic. It could facilitate the studies of the cooperation of GPx and SOD and could be a potential therapeutic agent for the treatment of ROS-mediated diseases.

Abstract:The development of artificial enzyme mimics has been rapidly growing in recent years, and it is attracting increasing attention owing to their remarkable advantages over natural enzymes. Herein, we developed a general and facile method to fabricate efficient glutathione peroxidase (GPx) mimics by grafting selenium-containing molecules (phenylselenylbromide, PhSeBr) to a Zr(IV)-based UiO-66-NH2 framework. In the presence of glutathione (GSH) serving as substrate, the fabricated UiO-66-Se catalysts can catalyze the reduction of hydroperoxides. The as-prepared UiO-66-Se systems show good catalytic activity over three cycles. These high-efficiency GPx mimic metal-organic frameworks (MOFs) are endowed with excellent thermal and structural stability, providing a promising avenue for the development of artificial enzyme mimics.
Construction Of The Active Site Of Glutathione Peroxidase On Polymer-Based Nanoparticles

Abstract:A new nanoenzyme model with glutathione peroxidase-like active site was constructed on polystyrene nanoparticle (PN1) via microemulsion polymerization. In this model system, two functional monomers were designed: one is a tellurium-containing compound that was introduced on the surface of the nanoparticle and acts as a catalytic center, and the other one is an arginine-containing compound designed as a binding site for the complexation of the carboxyl group of substrate 3-carboxy-4-nitrobenzenethiol (ArSH, 1). As a new glutathione peroxidase (GPx) mimic, it demonstrated excellent catalytic activity and substrate specificity. In ArSH assay system, it was at least 316000-fold more efficient than PhSeSePh for the reduction of cumene hydroperoxide (CUOOH) by ArSH. In contrast to model PN2, which lacks of substrate binding site, PN1 exhibits an obvious enhancement in catalytic activity. To further promote the catalytic efficiency, a substrate ArSH surface-imprinted nanoenzyme model (I-PN) was developed. By correctly incorporating and positioning the catalytic center tellurium and functional binding factor guanidinium, a continuative activity enhancement of 596000-fold for the reduction of CUOOH by catalyst I-PN compared with diphenyl diselenide (PhSeSePh) was observed. The results clearly show that polymeric nanoparticle can be developed as an excellent model for combining most of catalytic factors of enzyme into one scaffold.

Abstract:In this communication, we report the synthesis of the three generations of Fréchet-type poly(aryl ether) dendrimers with a diselenide core that demonstrate generation-dependent glutathione peroxidase (GPx) activity with initial reduction rates as high as 2431.20 muM min-1 for the third-generation product, around 1400 times faster than Ebselen. It represents a successful example of using a dendrimer as a model for a GPx mimic.

Abstract:Glutathione peroxidase (GPx, EC 1.11.1.9) is a key enzyme involved in scavenging of reactive oxygen species in biological system. For developing an efficient GPx-like antioxidant, catalytically necessary amino acid derivatives which located near the GPx active center were prepared as functional monomers. Via predetermined imprinting with substrate glutathione (GSH), a polymer-based GPx mimic with a similar structure of catalytic center of natural GPx was developed, and it demonstrated high-catalytic efficiency and substrate specificity. The imprinting polymer (I-PEM) exhibits GPx-like activity about three times higher than that of 2-SeCD, a cyclodextrin-based GPx mimic. The detailed studies on kinetics revealed that not only the substrate binding but also positional arrangement of reacting groups contribute significantly to the catalytic efficiency of the peroxidase model.

Abstract:Superoxide dismutase (SOD), glutathione peroxidase (GPX), and catalase (CAT) play crucial roles in balancing the production and decomposition of reactive oxygen species (ROS) in living organisms. These enzymes act cooperatively and synergistically to scavenge ROS. In order to imitate the synergism of these enzymes, we designed and synthesized a novel 32-mer peptide (32P) on the basis of the previous 15-mer peptide with GPX activity and a 17-mer peptide with SOD activity. Upon the selenation and chelation of copper, the 32-mer peptide was converted to a new Se- and Cu-containing 32-mer peptide (Se-Cu-32P) that displayed both SOD and GPX activities, and its kinetics was studied. Moreover, the novel peptide was demonstrated to be able to better protect vero cells from the injury induced by the xanthine oxidase (XOD)/xanthine/Fe2+ damage system than its parents. Thus, this bifunctional enzyme imitated the synergism of SOD and GPX and could be a better candidate of therapeutic medicine.

Abstract:6-Selenium bridged cyclodextrin was synthesized by regioselective tosylation of β-cyclodextrin and NaHSe displacement on the basis of the active site structure of glutathione peroxidase(GPX). The compounds displayed a higher activity of GPX than Ebselen(PZ51), its GPX activity was 4.3 times more than that of PZ51. The structure of the mimic was characterized by elemental analysis, IR and 1H NMR. The Se content and its valence in the mimic was determined by X-ray photoelectron spectrum.

Abstract:A novel method to functionalize graphene oxide (GO) by hyperbranched polysiloxane via polyhydrosilylation is reported, taking advantage of hyperbranched polysiloxane that possesses good properties such as low viscosity, good rheology, good solubility and high reactivity. The changes in GO surface morphology, chemistry and physical conditions at different stages are characterized by Fourier-transform infrared spectra (FT-IR), X-ray photoelectron spectroscopy (XPS) and Transmission electron microscopy (TEM). XPS analysis shows that the oxygen content of GO is 29.90% and the silicon content of hyperbranched polysiloxane grafted graphene oxide (HBPGO) is 18.66%. The results indicate that hyperbranched polysiloxane is successfully grafted onto the surface of GO and this novel nanostructure may have potential applications in composites.
A Novel Dicyclodextrinyl Diselenide Compound with Glutathione Peroxidase Activity

Abstract:A 6A,6A′‐dicyclohexylamine‐6B,6B′‐diselenide‐bis‐β‐cyclodextrin (6‐CySeCD) was designed and synthesized to imitate the antioxidant enzyme glutathione peroxidase (GPX). In this novel GPX model, β‐cyclodextrin provided a hydrophobic environment for substrate binding within its cavity, and a cyclohexylamine group was incorporated into cyclodextrin in proximity to the catalytic selenium in order to increase the stability of the nucleophilic intermediate selenolate. 6‐CySeCD exhibits better GPX activity than 6,6′‐diselenide‐bis‐cyclodextrin (6‐SeCD) and 2‐phenyl‐1,2‐benzoisoselenazol‐3(2H)‐one (Ebselen) in the reduction of H2O2, tert‐butyl hydroperoxide and cumenyl hydroperoxide by glutathione, respectively. A ping‐pong mechanism was observed in steady‐state kinetic studies on 6‐CySeCD‐catalyzed reactions. The enzymatic properties showed that there are two major factors for improving the catalytic efficiency of GPX mimics. First, the substrate‐binding site should match the size and shape of the substrate and second, incorporation of an imido‐group increases the stability of selenolate in the catalytic cycle. More efficient antioxidant ability compared with 6‐SeCD and Ebselen was also seen in the ferrous sulfate/ascorbate‐induced mitochondria damage system, and this implies its prospective therapeutic application.

Abstract:We report on the use of block copolymer micelles of polystyrene-b-poly( acrylic acid) ( PS-b-PAA) as matrixes for incorporating dibenzyl diselenide. We found that the water-insoluble diselenide, after being incorporated into the micelles, demonstrates glutathione peroxidase ( GPx) activity in water. Surprisingly, the mimicking system can be adjusted to show higher GPx activity by increasing the ionic strength of the solution simply upon addition of NaCl. Moreover, dibenzyl diselenide incorporated into the micelles is quite stable and maintains its GPx activity even after exposure to the atmosphere.

Abstract:Amphiphilic block copolymers poly(ethylene glycol)-block-poly(N-3-(methylthio)propyl glycine) (PEG-b-PMeSPG) were synthesized via ring-opening polymerization of N-3-(methylthio)propyl glycine N-thiocarboxyanhydride (MeSPG-NTA) initiated by amino-terminated PEG. The self-assemblies of three PEG-b-PMeSPG copolymers with different PMeSPG block lengths were first prepared by nanoprecipitation method using THE and DMF, respectively, as the organic solvent, and their morphologies were studied by Cryo-EM and DLS. To prepare polymersomes loaded with glucose oxidase (GOx), double emulsion method followed by extrusion treatment was employed. The oxidation-responsive disruption of polymersomes was achieved upon the introduction of glucose because of the oxidants generated insitu by GOx/glucose. Abstract:Synthetic reactive oxygen species (ROS)-responsive biomaterials have emerged as a useful platform for regulating critical aspects of ROS-induced pathologies and can improve such hostile microenvironments. Here, we report a series of new hyperbranched poly(β-hydrazide ester) macromers (HB-PBHEs) with disulfide moieties synthesized via an "A2 + B4" Michael addition approach. The three-dimensional structure of HB-PBHEs with multiacrylate end groups endows the macromers with rapid gelation capabilities to form (1) injectable hydrogels via cross-linking with thiolated hyaluronic acid and (2) robust UV-cross-linked hydrogels. The disulfide-containing macromers and hydrogels exhibit H2O2-responsive degradation compared with the counterparts synthesized by a dihydrazide monomer without disulfide moieties. The cell viability under a high ROS environment can be well-maintained under the protection of the disulfide containing hydrogels.

Abstract:To construct an efficient GPx mimic, a novel method for preparing polymer-based vesicles carrying GPx-active sites was developed. A series of block copolymers loaded with recognition and catalytic sites were synthesized based on polystyrene-block-poly[tri(ethylene glycol) methyl ether acrylate]s (PS-PMEO3MAs). By altering the molar ratio of the functional copolymers, vesicles with GPx activity were obtained by self-assembly of these functional copolymers through blending. The optimum GPx mimic constructed by the blending process exhibited high catalytic activity and acted as a real catalyst with typical saturation kinetics behavior. The method may be of benefit for designing other enzyme mimics and may cast a light on constructing other biologically related functional nanoparticles.
