Abstract:Objective To study the immune response against hepatitis B virus induced by HBsAg recombinant adenovirus-transduced dendritic cells (DC) in vivo and in vitro. Methods DC were isolated from bone marrow cells of BALB/c mice, transfected with HBsAg recombinant adenovirus Ad-S (DC/Ad-S) or pulsed with HBsAg (DC/HBsAg) and used for analysis of the phenotypic markers by flow cytometry and the stimulatory capacity in mixed leukocyte reaction (MLR) . HBsAg-specific activity of splenic cytotoxic T lymphocyte (CTL) of BALB/c mice was measured by LDH release assay and the anti-HBs titers in sera was examined by RIA after being immunized with DC/Ad-S (12), DC/HBsAg (12) or pcDNA3.1(+)-S plsmid(8). Intracellular cytokines of proliferative T cells in autologous MLR or of splenic T cells after immunization was detected by flow cytometry. Results Either the Ad-S- or Ad-lacZ-transduced DC or HBsAg-pulsed DC or untreated DC expressed similar levels of phenotypic markers, and showed similar stimulatory capacity in either allogeneic or autologous MLR. DC/Ad-S and DC/HBsAg induced more TH or Tc secreting IFN-γrather than IL-4 in autologous MLR. DC/Ad-S more efficiently stimulated splenic TH to secrete IFN-γthan pcDNA3.1( + )-S (P0.05) and splenic Tc to product IFN-γthan DC/HBsAg (P0.01) and plasmid DNA (P 0.01), and induced stronger splenic HBsAg-specific CTL responses than that induced by DC/HBsAg. However, DC/Ad-S and DC/HBsAg primed splenic Tc to secret IFN-γand splenic HBsAg-specific CTL, more reductively than plasmid DNA at 4 weeks after immunization. Anti-HBs response elicited by DC vaccination was much weaker than that induced by pcDNA3.1( + )-S at either 2 or 4 weeks after immunization. Conclusion HBsAg recombinant adenovirus-transduced DC can induce more enhanced TH1/ Tcl (typeⅠ) cell-mediated immunity and HBsAg-specific CTL than HBsAg-pulsed DC or plasmid DNA, and may be an efficient inducer in priming HBV-specific T cell response.

Abstract:The elimination of hepatitis B virus (HBV) infection is partially facilitated by the prophylactic HB vaccine. As the loss of seroprotection over time remains a conundrum for long-lasting protection, a comprehensive dynamic analysis of immunogenic targets of the HB vaccine will provide novel insights into the improvement and design of potential targets. In this study, 36 healthy subjects without prior history of hepatitis B infection and negative for hepatitis B surface antibody (anti-HBs) were enrolled. Participants were given a series of three doses of HB vaccine on a 0-, 1-, and 6-month schedule and longitudinally followed up. We systematically mapped 55 overlapping 15-mer peptides covering the small S protein of hepatitis B virus (SHBs) of vaccinees' serum samples at seven time points by performing an ELISA assay. Additionally, the frequencies and function dynamics of adaptive immune response were assessed by flow cytometry. We found that the SHBs peptide coverage presented an overall upward trend along with the vaccination progress, and the individual subpartition recognition was strongly correlated with the anti-HBs titers. Moreover, we identified one dominant epitope (S29) located on "a determinant region" associated with effective vaccine response. Besides, significant correlations between the proportion of plasmablasts and proliferating B cells and levels of anti-HBs were ascertained. Taken together, our data characterized the dynamics of HB vaccine-induced neutralizing antibodies against B-cell linear epitopes on SHBs and adaptive immune response, which will be constructive to develop the next-generation vaccine.

Abstract:ABSTRACT The hepatitis B virus core protein (HBcAg) is a uniquely immunogenic particulate antigen and as such has been used as a vaccine carrier platform. The use of other hepadnavirus core proteins as vaccine carriers has not been explored. To determine whether the rodent hepadnavirus core proteins derived from the woodchuck (WHcAg), ground squirrel (GScAg), and arctic squirrel (AScAg) viruses possess immunogen characteristics similar to those of HBcAg, comparative antigenicity and immunogenicity studies were performed. The results indicate that (i) the rodent core proteins are equal in immunogenicity to or more immunogenic than HBcAg at the B-cell and T-cell levels; (ii) major histocompatibility complex (MHC) genes influence the immune response to the rodent core proteins (however, nonresponder haplotypes were not identified); (iii) WHcAg can behave as a T-cell-independent antigen in athymic mice; (iv) the rodent core proteins are not significantly cross-reactive with the HBcAg at the antibody level (however, the nonparticulate “eAgs” do appear to be cross-reactive); (v) the rodent core proteins are only partially cross-reactive with HBcAg at the CD4+ T-cell level, depending on MHC haplotype; and (vi) the rodent core proteins are competent to function as vaccine carrier platforms for heterologous, B-cell epitopes. These results have implications for the selection of an optimal hepadnavirus core protein for vaccine design, especially in view of the “preexisting” immunity problem that is inherent in the use of HBcAg for human vaccine development.

Abstract:Objective To investigate the immune response against Hepatitis B virus (HBV) in- duced by HBsAg recombinant adenovirus (Ad-S)-transduced dendritic cells (DCs) in HBV transgenic mice (HBV-Tg),and evaluate its potential in treating chronic HBV infection.Methods DCs were generated from bone marrow cells from the HBV Tg mice.and transfected with Ad-S (DC/Ad-S) or pulsed with HBsAg protein (DC/HBsAg).HBV-Tg mice were randomly assigned to receive either DC/Ad-S (n=10) or DC/HBsAg(n=10) or pcDNA3.1(+) S plasmid (n=6),and the same im munization schedule was repeated twice at 3 week intervals.Intracellular cytokines of splenic T cells, HBsAg-specific activity of splenic cytotoxic T lymphocyte (CTL),and HBsAg and anti-HBs and HBV DNA titers in sera were detected after immunization by flow eytometry,LDH release assay, ELISA and quantitative PCR,respectively.Hepatic injury was monitored at various time points after immunization by measuring serum alanine aminotransferase (ALT) activity.Tissue samples of the liver were stained with hematoxylin and eosin for histological analysis,and the expression of hepatitis B core Ag(HBcAg) and HBsAg was assessed by immunohistochemical analysis.Results DC/Ad-S stimulated splenic Tc more efficiently to secrete IFNγthan DC/HBsAg or pcDNA3. 1 (+)-S (P＜0.05 or P＜0.01),and induced stronger splenic HBsAg-specific CTL responses than that induced by DC/HBsAg (P＜0.05 or P＜0.01 at 10:1,20:1,40:1 of E:T ratio),which were stronger than that induced by DNA vaccination (all P＜0.01 at 10:1,20:1,40:1 of E:T ratio),at 1 or 2 weeks after immunization, respectively. The CTL response elicited by DC/Ad-S was transient, rising to strongest at 1 to 2 weeks after immunization and waning gradually thereafter,but reduced more slowly than that elicited by DC/HBsAg.The suppression of serum HBsAg and HBV DNA and the reduction of expression of HBcAg and HBsAg in liver from HBV-Tg mice induced by DC/Ad-S were the strongest in all groups of immunization,and the second was that induced by DC/HBsAg from 1 to 4 weeks after immunization.The serum anti-HBs titer and ALT activity were slightly elevated,and histological analysis of the liver displayed only a very slight focal inflammatory infiltration up to 4 weeks after immunization,and the inter-group differences were not significant.Conclusions HBsAg recombinant adenovirus-transduced DCs can induce Tcl and CTL response,reduce the titer of serum HBsAg and HBV DNA,and inhibite the expression of HBcAg and HBsAg in liver in HBV-Tg mice, stronger than that induced by HBsAg-pulsed DCs or plasmid DNA.

Abstract:The hepatitis B core antigen (HBcAg) has been proposed as a useful particulate carrier platform for poorly immunogenic peptidic and carbohydrate B cell epitopes. However, biochemical and immunologic impediments have plagued this technology. Specifically, the “assembly” problem characterized by the low yield of unstable hybrid particles resulting from the insertion of foreign sequences and the “pre-existing immunity” problem due to the fact that the HBcAg is derived from a human pathogen have limited the development of this carrier technology. As a means of addressing the “pre-existing immunity” problem we have used the core proteins from the rodent hepdnaviruses. A number of advantages to the use of the rodent hepadnaviral core proteins as opposed to the HBcAg for vaccine design were defined including: equal or superior immunogenicity at the T and B cell levels; the use of the rodent core proteins does not compromise the anti-HBc diagnostic assay; the efficacy of the rodent core proteins as vaccine carriers will not be limited by pre-existing anti-HBc antibodies that are present in previously and currently HBV-infected persons; and the HBcAg-specific tolerance present in HBV chronic carriers can be circumvented by the use of the rodent core proteins.

Hepatitis B core-related antigen: A novel and promising surrogate biomarker to guide anti-hepatitis B virus therapy

Abstract:The current requirement for biomarkers to detect hepatitis B virus (HBV) infection is polarized. One is a fully-automated and highly sensitive measurement system; the other is a simple system for point-of-care testing (POCT) in resource-limited areas. Hepatitis B core-related antigen (HBcrAg) reflects intrahepatic covalently closed circular DNA and serum HBV DNA. Even in patients with undetectable serum HBV DNA or HBsAg loss, HBcrAg may remain detectable. Decreased HBcrAg levels are associated with reduction of the occurrence of hepatocellular carcinoma (HCC) in chronic hepatitis B. Recently, a fully-automated, novel high-sensitivity HBcrAg assay (iTACT-HBcrAg, cut-off value: 2.1 logIU/mL) has been developed. This attractive assay has been released in Japan very recently. iTACT-HBcrAg can be useful for monitoring HBV reactivation and prediction of HCC occurrence, as an alternative to HBV DNA. Moreover, monitoring HBcrAg may be suitable for determining the therapeutic effectiveness of approved drugs and novel drugs under development. Presently, international guidelines recommend anti-HBV prophylaxis for pregnant women with high viral loads to prevent mother-to-child transmission of HBV. However, >95% of HBV-infected individuals live in countries where HBV DNA quantification is not available. Worldwide elimination of HBV needs the scaling-up of examination and medication services in resource-limited areas. Based on this situation, a rapid and easy HBcrAg assay as a POCT is valuable. This review provides the latest information regarding the clinical use of a new surrogate marker, HBcrAg, in HBV management, based on iTACT-HBcrAg or POCT, and introduces novel agents targeting HBV RNA/protein.

Abstract:Chronic hepatitis B (CHB) is one of the major public health challenges in the world. Due to a strong interplay between specific T-cell immunity and elimination of hepatitis B virus (HBV), efforts to develop novel immunotherapeutics are gaining attention. TG1050, a novel immunotherapy, has shown efficacy in an animal study. To support the clinical development of TG1050 in China, specific immunity to the fusion antigens of TG1050 was assessed in Chinese patients. One hundred and thirty subjects were divided into three groups as CHB patients, HBV spontaneous resolvers, and CHB patients with HBsAg loss after antiviral treatment. HBV-specific T-cell responses to pools of HBV Core or Polymerase genotype D peptides included in TG1050 were evaluated. HBV Core- or Polymerase-specific cells were detected in peripheral blood mononuclear cells (PBMCs) from the different cohorts. The frequencies and intensities of HBV Core-specific immune responses were significantly lower in CHB patients than in HBsAg loss subjects. In CHB patients, a dominant pool derived from Polymerase (Pol1) was the most immunogenic. CHB patients with low viral loads (<10(6) IU/mL) were more likely to have a positive response specific to the Core peptide pool. Overall, genotype D-derived peptides included in TG1050 could raise broad and functional T-cell responses in PBMCs from Chinese CHB patients infected with genotype B/C isolates. Core-specific immunogenic domains appeared as hot spots with the capacity to differentiate between CHB vs HBsAg loss subjects. These observations support the extended application and associated immune monitoring of TG1050 in China.

Abstract:<span>Background: Hepatitis B core-related antigen (HBcrAg) is a promising disease-monitoring marker for chronic hepatitis B (CHB). Abstract:Hepatitis B surface antibody (HBsAb) plays a critical role in protecting against infection of hepatitis B virus (HBV) and were extensively studied in literature. At the same time, the status of hepatitis B surface antigen (HBs)-specific B cells in both vaccinated and HBV infected people received limited attention. In the current study, we adopted a highly specific B-cell Enzyme Linked ImmunoSpot (ELISpot) assay to analyze HBs-specific B cells in various clinical settings: healthy individuals with the history of HBV vaccination before and after receiving an extra HBV vaccine boost, people chronically infected with HBV (CHB) in various clinical stages, with or without a particular type anti-viral treatment, or whether receiving a dose of HBV vaccine. In all of these cases, B-cell ELISpot assay was used effectively in enumerating the frequency of HBs-specific B cells. While the focus of the current report was to establish the utility of this assay for HBV research, a number of interesting observations were made in this pilot study based on the profiles and dynamics of HBs-specific B cells in various conditions. Such information is useful to guide the future work in designing novel therapeutic strategies against CHB.

Abstract:Hepatitis B virus (HBV) infection is a global public health problem that is closely related to liver cirrhosis and hepatocellular carcinoma (HCC). The prevalence of acute and chronic HBV infection, liver cirrhosis, and HCC has significantly decreased as a result of the introduction of universal HBV vaccination programs. The first hepatitis B vaccine approved was developed by purifying the hepatitis B surface antigen (HBsAg) from the plasma of asymptomatic HBsAg carriers. Subsequently, recombinant DNA technology led to the development of the recombinant hepatitis B vaccine. Although there are already several licensed vaccines available for HBV infection, continuous research is essential to develop even more effective vaccines. Prophylactic hepatitis B vaccination has been important in the prevention of hepatitis B because it has effectively produced protective immunity against hepatitis B viral infection. Prophylactic vaccines only need to provoke neutralizing antibodies directed against the HBV envelop proteins, whereas therapeutic vaccines are most likely needed to induce a comprehensive T cell response and thus, should include other HBV antigens, such as HBV core and polymerase. The existing vaccines have proven to be highly effective in preventing HBV infection, but ongoing research aims to improve their efficacy, duration of protection, and accessibility. The routine administration of the HBV vaccine is safe and well-tolerated worldwide. The purpose of this type of immunization is to trigger an immunological response in the host, which will halt HBV replication. The clinical efficacy and safety of the HBV vaccine are affected by a number of immunological and clinical factors. However, this success is now in jeopardy due to the breakthrough infections caused by HBV variants with mutations in the S gene, high viral loads, and virus-induced immunosuppression. In this review, we describe various types of available HBV vaccines, along with the recent progress in the ongoing battle to develop new vaccines against HBV.

Abstract:Hepatitis B core-related antigen (HBcrAg) reflects the activity of intrahepatic covalently closed circular DNA. HBcrAg can be detected even in chronic hepatitis B patients in whom serum HBV DNA or hepatitis B surface antigen is undetectable. The HBcrAg measurement system was developed based on two concepts. One is a fully-automated and highly-sensitive HBcrAg assay (iTACT-HBcrAg) and the other is a point-of-care testing (POCT) that can be used in in resource-limited areas. iTACT-HBcrAg is an alternative to HBV DNA for monitoring HBV reactivation and predicting the development of hepatocellular carcinoma. This validated biomarker is available in routine clinical practice in Japan. Abstract:Chronic hepatitis caused by hepatitis B virus infection pose a major public health problem worldwide,and incur substantial economic burden. Current treatment mostly relies on nucleoside/nucleotide analogue and interferon. Because of high rates of resistance,poor tolerability and possible adverse reactions,new therapeutic options are warranted. Due to the promising ability,studies on therapeutic vaccine became the hot point. Therapeutic hepatitis B vaccines under developing include protein-based,peptide-based,DNA-based and autologous dendritic cell-based vaccines. Presently,three therapeutic vaccines are being evaluated in chronic patients in China.
Humoral and cellular immunogenecity of nucleic acid vaccine of hepatitis B core antigen in rhesus monkeys

Abstract:Objective To observe humoral and cellular immunogenecity of nucleic acid vaccine of hepatitis B core antigen (HBcAg) in rhesus monkeys. Methods Rhesus monkeys of the experimental group and the control group received intramuscular injections of a HBcAg nucleic acid vaccine(pJW4303/HBc) and a control plasmid (pJW4303), respectively. Anti HBc titers, isotypes of anti HBc IgG in sera of the rhesus monkeys pre and post vaccinations, and IFN γ as well as IL 4 levels in the culture supernatant of PBMC isolated from the monkeys were detected by an enzyme linked immunoabsorbent assay. HBcAg specific proliferation activities of PBMC in the monkeys were measured by 3H TdR incorporation assay. Results It was observed in rhesus monkeys of experimental group an obvious anti HBc response after immunization with HBcAg nucleic acid vaccine. The major isotypes of anti HBc IgG was IgG2 and IFN γ was predominant compared with IL 4 in the culture supernatant of rhesus monkeys' PBMC, both indicating Th1 type of immune responses. HBcAg specific proliferation activities of PBMC in the experimental group were significantly stronger than those in the control group. Conclusions The nucleic acid vaccine based on HBcAg shows a good humoral and cellular immunogenecity in rhesus monkeys.

Abstract:Chronic hepatitis B (CHB) poses a serious threat to the health of the Chinese people. Viral markers are of great significance during antiviral therapy and research and development of innovative drugs. However, traditional serovirological markers such as HBV DNA and hepatitis B surface antigen (HBsAg) have certain limitations in assessing the efficacy of antiviral treatment and accurately reflecting the transcriptional activity of covalently closed circular DNA (cccDNA). In recent years, the role of new viral markers such as HBV RNA, hepatitis B core-related antigen (HBcrAg) and quantitative anti-hepatitis B core protein (qAnti-HBc) have attracted more and more attention because of accurately reflecting the transcription activity of cccDNA, determining the therapeutic effect and predicting the risk of recurrence after drug discontinuation. This article briefly introduces the application of new and old markers in various stages of antiviral therapy in CHB patients, and combines the application of new viral markers in the evaluation of drug efficacy, and expounds its possible role in the research and development of new antiviral therapy.

Abstract:OBJECTIVESTo observe immunogenicity of new DNA vaccine encoding for hepatitis B virus core antigen (HBcAg).METHODSA new DNA vaccine (pSW3891/HBc) encoding for hepatitis B virus core antigen was constructed using plasmid pSW3891 which can be used in human. Control and experiment groups of Balb/c mice were immunized with pSW3891 or pSW3891/HBc by gene gun. Anti-HBc in sera of mice was tested by ELISA (enzyme linked immune sorbent assay). Specific cytotoxicity of cytotoxic T lymphocyte (CTL) of mice was detected by LDH release assay.RESULTSpSW3891/HBc can express in 293T cell line effectively. Mice immunized with pSW3891/HBc showed strong anti-HBc response and specific high cytotoxicity of CTL.CONCLUSIONpSW3891/HBc induced significantly humoral and cellular immune responses in Balb/c mice.

Abstract:OBJECTIVE:To evaluate the immunization effects of HBV core antigen and surface antigen fusion protein.METHODS:The DNA fragments encoding HBsAg 100-162 aa; HBcAg 1-78 aa and HBcAg 83-144 aa were PCR-amplified, and then cloned into pcDNA3 plasmid. The chimeric gene was subcloned into the prokaryotic vector, pRSET-B. The E.coli expressed recombinant protein purified. BALB/c mice were immunized with recombinant protein or eukaryotic expression plasmid, humoral response and cellular response were examined.RESULTS:The plasmid containing the chimeric gene of HBsAg and HBcAg induced effective anti-HBs antibody response and strong HBcAg specific lymphocyte proliferative response, but could not induce anti-HBc antibody response. Fusion protein induced strong anti-HBs and anti-HBc antibody response, and it also caused significant HBcAg specific lymphocyte proliferation. Compared to the recombinant fusion protein, the plasmid containing the chimeric gene of HBsAg and HBcAg can induce more effective cellular response but weaker humoral response.CONCLUSION:Compared to the recombinant fusion protein, the plasmid containing the chimeric gene of HBsAg and HBcAg is a more effective vaccine.

Abstract:Hepatitis B virus core antibodies are specific antibodies produced after viral infection that appear early and last for a long time, and its levels in serum are measured by the double-antigen sandwich chemiluminescent microparticle immunoassay method, which has higher sensitivity and specificity, providing new clinical indicators for hepatitis B patients diagnosis, treatment, and drug withdrawal management. This article reviews the clinical significance and research progress of quantitative hepatitis B core antibody measurement and expounds on its research applications and prospects in clinical practice.

Abstract:Hepatitis B core antigen (HBcAg) was prepared from human liver tissue and used in an immunoelectro-osmophoresis screening test to detect antibody to HBcAg (anti-HBc) in patients with evidence of liver disease and in blood donors. With the exception of two immunosuppressed HBsAg carriers, anti-HBc was found in all cases of hepatitis B infection even when HBsAg was detectable only by radioimmunoassay. Non-specific reactions were observed in 'non-B' hepatitis but, in spite of this problem, antib-HBc screening was considered a useful addition to routine tests in the clinical hepatitis laboratory.
