Abstract:The focus of this work was on the synthesis of two bidesmosidic ursolic acid saponins bearing a 2,3-branched trisaccharide residue. Therefore, 3-O-{[beta-D-glucopyranosyl-(1-->2)]-[alpha-L-arabinopyranosyl-(1-->3)]-alpha-L-arabinopyranosyl}ursolic acid-28-O-[beta-D-glucopyranosyl] ester 1 was concisely synthesized by two strategies in 22% and 41% overall yield, respectively, and another congener 3-O-{[beta-D-xylopyranosyl-(1-->2)]-[beta-D-glucopyranosyl-(1-->3)]-alpha-L-arabinopyranosyl}ursolic acid-28-O-[beta-D-glucopyranosyl] ester 2 was also efficiently prepared in 81% overall yield. The (1)H NMR and (13)C NMR signals of saponin 2 are all consistent with those reported for the natural product.

Abstract:An efficient synthetic strategy for three natural seco-type cholestane alkaloids isolated from the Veratrum plants, based on commercially available naturally occurring and abundant (-)-diosgenin (1), as exemplified in the concise asymmetric synthesis of (-)-verazine (4), (-)-veramiline (5) (proposed structure), and its 22-epimer, (-)-oblonginine (6), is presented. This work highlights the application of a cascade ring-switching process of (-)-diosgenin to achieve the E-ring opening and construction of chiral six-membered lactone challenges in seco-type cholestane alkaloid synthesis. This approach enables the synthesis of related natural and nature-like novel cholestane alkaloids, opening up opportunities for more extensive exploration of cholestane alkaloid biology.

Abstract:A bioinspired synthesis of spirochensilide A from commercially available lanosterol is reported. The synthesis features a directed C-H oxidation, a Wagner-Meerwein-type double methyl migration, a Meinwald rearrangement, and a double-bond isomerization/spiroketal formation cascade. The proposed biosynthetic speculation was modified by this synthetic sequence, which also served as a platform for the synthesis of other lanostanes with migrating methyl groups.

Abstract:Two new spirostanol saponins, tuberosides D and E, have been isolated from the seeds of Allium tuberosum. On the basis of spectral data and chemical reactions, their structures were established as (25S)-5 alpha-spirostane-2 alpha,3 beta-diol 3-O-alpha-L-rhamnopyranosyl-( 1 --> 2)-O-[alpha-L-rhamnopyranosyl-( 1 --> 4)]-O-beta-D-glucopyranoside and (25S)-5 alpha-spirostan-2 alpha,3 beta-diol 3-O-beta-D-glucopyranosyl-(1 --> 2)O-[alpha-L-rhamnopyranosyl-(1 --> 4)]-O-beta-D-glucopyranoside, respectively.

Abstract:Main conclusionThis work reviews recent advances in the pathways and key enzymes of steroidal saponins biosynthesis and sets the foundation for the biotechnological production of these useful compounds through transformation of microorganisms.Steroidal saponins, due to their specific chemical structures and active effects, have long been important natural products and that are irreplaceable in hormone production and other pharmaceutical industries. This article comprehensively reviewed the previous and current research progress and summarized the biosynthesis pathways and key biosynthetic enzymes of steroidal saponins that have been discovered in plants and microoganisms. On the basis of the general biosynthetic pathway in plants, it was found that the starting components, intermediates and catalysing enzymes were diverse between plants and microorganisms; however, the functions of their related enzymes tended to be similar. The biosynthesis pathways of steroidal saponins in microorganisms and marine organisms have not been revealed as clearly as those in plants and need further investigation. The elucidation of biosynthetic pathways and key enzymes is essential for understanding the synthetic mechanisms of these compounds and provides researchers with important information to further develop and implement the massive production of steroidal saponins by biotechnological approaches and methodologies.

Abstract:3-O-[ss-D-Glucopyranosyl-(1 -> 3)-alpha-L-arabinopyranosyl]-oleanolic acid-28-O-[ss-D-glucopyranosyl] ester 1 was synthesized concisely by a convergent strategy. Using stepwise fashion for the synthesis of saponin 2, 3-O-{[ss-D-glucopyranosyl-(1 -> 2)]-[alpha-L-arabinopyranosyl-(1 -> 3)]-alpha-L-arabinopyranosyl}-oleanolic acid-28-O-(ss-D-glucopyranosyl) ester, an abnormal phenomenon, that the terminal arabinosyl residue took the C-1(4) conformation instead of typical C-4(1) form, was observed. Deprotection or heating could not resume the normal conformation, which resulted in the product of 2 ' not 2.

Abstract:Comprehensive Summary Sepositoside A (1) is a prototypical cyclic steroid glycoside bearing a hybrid 16‐membered ring composed of the steroid skeleton and a 1,2‐trans‐linked trisaccharide. Herein, we report an expedient access toward two simplified analogues, in which the strained 16‐membered ring is constructed via Au(I)‐catalyzed intramolecular addition of alcohol to epoxide. A similar macroetherification in relevant steroid trisaccharides has been intensively examined, however, failed to furnish the macrocyclic skeleton of Sepositoside A. Abstract:Leveraging on Schmidt glycosylation method, a highly efficient approach to obtain oleanane-type triterpene saponins was fixed, whereby oleanyl mono-, disaccharide (guaianin N), trisaccharide (elatoside E), as well as tetrasaccharide (elatoside F) were obtained efficiently. The synthetic investigation has resulted in the discovery of the effect of branch-sugar incorporation sequence on the overall synthetic efficiency. Moreover, through bioactivity investigation, the cytotoxic activity of the obtained triterpenoid saponins was evaluated, and the preliminary structure-activity relationship was deduced.

Abstract:A mechanistically interesting and practical method for the synthesis of functionalized spiro[4.5]decanes is developed, featuring oxidative dearomatization-induced ring expansion of cyclobutanes as the key element. The new method enables facile access to a variety of spiro[4.5]cyclohexadienones with good efficiency and generality. Further elaboration of the resulting products into other valuable scaffolds is also explored, leading to the discovery of an interesting compound that displays a promising biological profile. Moreover, we have also conducted a comprehensive computational study that provides a deep insight into the mechanism of the reaction.

Abstract:Chemical synthetic approach toward acyl group‐free escin derivatives has been successfully established, which would speed up the process of pharmaceutical applications of these valuable natural D/E‐ring simultaneously functionalized oleanane‐type saponins. Comprehensive Summary With protoescigenin as starting material and through orchestrated application of Yu and Schmidt glycosylation protocols, the synthesis of acyl group‐free escin derivatives was achieved for the first time. As the undesired non‐specific toxicity, originating from the existence of acyl groups on aglycone, prohibits the wide application of escins, the established strategies toward non‐acylated protoescigenin‐type saponins would dramatically ease the access to escin derivatives dispense of acyl groups, thereby speeding up the pace of pharmaceutical use of these valuable compounds.

Abstract:Star quality: Goniopectenoside B, a minor asterosaponin from starfish Goniopecten demonstrans with antifouling activity, has been synthesized in a convergent 21 steps and in 4.3 % overall yield starting from adrenosterone. This represents the first synthesis of a complex asterosaponin, which are ubiquitous and characteristic in starfish as defense chemicals (see figure).
Naturally Occurring Polyhydroxylated Spirostanol Saponins, A Review of the Classification, Sources, Biosynthesis, Biological Activities, and Toxicity

Abstract:Polyhydroxylated spirostanol saponins, characterized by three or more hydroxy substitutions in the aglycone, have various interesting biological activities. In the present study, "steroids", "saponins", "polyhydroxylated", "spirostanol saponins", and "steroidal saponins" were used as search terms to screen the literature. Cited references were collected between 1950 and 2023 from the Web of Science, SciFinder, and China National Knowledge Internet (CNKI). A total of 407 polyhydroxylated spirostanol saponins were included in this review. These saponins were classified into three types, α, β, and γ. Polyhydroxylated spirostanol saponins have potential benefits, primarily anti‐inflammatory, antimicrobial, cytotoxic, and cAMP phosphodiesterase inhibitory activities. These compounds were found in 11 plant families and 36 genera. The top three families containing the most saponins were Asparagaceae, Melanthiaceae, and Amaryllidaceae, and the top five genera were Trillium, Helleborus, Allium, Dracaena, and Paris. The top five plants were Trillium tschonoskii Maxim., Ypsilandra thibetica Franch., Paris polyphylla var. yunnanensis (Franch.) Hand.‐Mazz., Helleborus thibetanus Franch., and Helleborus foetidus L. On the basis of their diverse biological activities, these saponins and related plant resources are worthy of further development and utilization.

Abstract:The first asymmetric total synthesis of bioactive diterpenoid hypoestin A with an unprecedented [5-8-5-3] tetracyclic skeleton is accomplished in 15 steps from commercially available (R)-limonene. Furthermore, the second asymmetric total syntheses of sesterterpenoids albolic acid and ceroplastol II in 21 steps are also reported. The synthetically challenging and highly functionalized [X-8-5] (X = 5 or 7) tricarbocyclic ring systems found in hypoestin A, albolic acid, ceroplastol II, and schindilactone A, as well as other natural products, are efficiently and directly constructed via a unique intramolecular Pauson-Khand reaction of an allene-yne. This work represents the first reported use of the Pauson-Khand reaction to access synthetically challenging eight-membered-ring systems in natural product synthesis.

Abstract:Saponins are amphiphilic molecules consisting of carbohydrate and either triterpenoid or steroid aglycone moieties and are noted for their multiple biological activities—Fungicidal, antimicrobial, antiviral, anti-inflammatory, anticancer, antioxidant and immunomodulatory effects have all been observed. Saponins from natural sources have long been used in herbal and traditional medicines; however, the isolation of complexed saponins from nature is difficult and laborious, due to the scarce amount and structure heterogeneity. Chemical synthesis is considered a powerful tool to expand the structural diversity of saponin, leading to the discovery of promising compounds. This review focuses on recent developments in the structure optimization and biological evaluation of synthetic triterpenoid and steroid saponin derivatives. By summarizing the structure–activity relationship (SAR) results, we hope to provide the direction for future development of saponin-based bioactive compounds.

Abstract:Saponin as a natural surfactant has been used for the synthesis of spiro[indoline‐3,4′‐pyran]‐2‐ones through one‐pot three‐component reaction of isatins, malononitrile, and cyclic 1,3‐dicarbonyl compounds in water at room temperature. Natural surfactants are biodegradable and therefore environmentally benign. To avoid the detrimental effects of synthetic surfactants on the environment, it is highly desirable to replace them with natural surfactants as alternatives. In this context, we are reporting herein the use of saponins derived from Indian soapberry (Sapindus mukorossi) fruit pericarp as a natural surfactant for the synthesis of spiro[indoline‐3,4′‐pyran]‐2‐one derivatives through one‐pot three‐component reaction of isatins, malononitrile, and cyclic 1,3‐dicarbonyl compounds. Wide substrate scope has been established with the synthesis of as many as 18 compounds in yields ranging from 72% to 94%.

Abstract:The first synthesis of four natural triterpene saponins, which exhibit significant antitumor activities, was concisely achieved by adopting a stepwise glycosylation. The key intermediate 13 was afforded via Bu(2)SnO-mediated regioseletive benzoylation. During the preparation of the target compounds, it was found that the alpha-L-arabinopyranosyl unit in intermediates 17 and 20 existed in the unusual (1)C(4) conformation, and after removing the benzoyl groups, the alpha-L-arabinopyranosyl unit was normal in a typical (4)C(1) form.

Abstract:A series of 18-Ar-spirotryprostatin A analogs were synthesized stereoselectively from isatin via a five-step reaction sequence involving Wittig reaction, 1,3-dipolar cycloaddition, amidated ring closure reaction, hydrolyze and decarboxylation. The two one-pot reactions employed in the synthetic route resulted in high yields. The 1,3-dipolar cycloaddition reaction exhibited a diastereomeric ratio (dr) exceeding 20:1 and an enantiomeric ratio (er) reaching 90:10. Additionally, the amidated ring closure reaction achieved yields of up to 93%.
