Abstract:Wild Rosa resources in Yunnan are very abundant and hold many important and useful traits for improving modern rose. However, many of them have no cytological data and few were karyologically analysed. In this article, karyological study of 24 wild Rosa taxa belonging to different sections was carried out by means of traditional squashing and pressing. The chromosome number of 8 taxa and karyotypes of most studied ones were reported for the first time. 22 resources were diploids with 14 chromosomes (2n = 2x = 14), while Rosa moyesii Hemsl. et Wils. and Rosa macrophylla Lindl. from Sect. Cinnamomeae DC . were hexaploids with 42 chromosomes (2n = 6x = 42). Only type 1A and type 2A been found, karyotypes of genus Rosa in Yunnan were symmetric. Also, the studied taxa differed much from each other in chromosome length, position of centromere, satellite position and satellite number, etc. The results implied that wild Rosa germplasm in Yunnan were rich in karyologcal diversity, and polyploidization might have played and were still playing very important roles in the evolution and speciation in genus Rosa , mainly in Sect. Cinnamomeae in alpine areas of northwestern Yunnan. The cytological data would supply chromosomal proofs not only for the taxonomy, phylogeny and evolution of genus Rosa in this special geological area, but also for the exploitation of useful traits of wild Rosa into the breeding of new cultivars.

Abstract:The results demonstrated that the same species of different geographical populations have the same amount of chromosome and karyotypic formulae. Rana limmocharis boie and Rana j. Japonica have 2n=26, NF=52 and 5+8 karyotypic formulae. Microhyla ornata has 2n=24, NF=48 and 6+6 karyotypic formulae. But some dissimilarities were found among them. First, the number and sequence of submetacentric chromosome were different among them, and then the secondary contriction (SC) or satellite (Sat) were also different. It was showed that the chromosomes of the same species of different geographical population have diversities.

Abstract:The karyological analysis on 4 varieties of R.odorata from 11 localities was carried out by conventional pressed slice method.The results showed that:(1)R.odorata var.odorata was diploidy with a karyotype of 1A and karyotype fomula of 2n=2x=14=12m+2sm.(2)Chromosomal characters of R.odorata var.gigantea from 4 natural populations were almost similar.All the 4 populations were diploidy with the same karyotype of 1A and karyotype fomula of 2n=2x=14=12m+2sm.The main difference of chromosome morphology among populations lay in the constitution of chromosomal relative length of the genome.(3)R.odorata var.pseudoindica was diploidy with a karyotype of 1A with a karyotype fomula of 2n=2x=14=10m+4sm.(4)Chromosomal number and morphology of R.odorata var.erubescens from different places differed greatly.The plants from Mujiaqiao and Yongchun were both triploid with the karyotype fomula of 2n=3x=21=18m+3sm and 2n=3x=21=21m,respectively.Plants from Hongqiao,Fumin and Xiaohe were diploid with 2x=14=12m+2sm,2n=2x=14=10m+4sm and 2n=2x=14=12m+2sm,respectively.Karyotypes of all the R.odorata var.erubescens were 1A,except those plants which from Fumin being 1B.The karyomophological characteristics,cyto-genetic divergence mode,rich variation and their possible utilization in rose breeding of R.odorata were discussed.

Abstract:The karyological study on 11 cultivars of Chinese old garden roses such as Rosa 'Slater's crim-son China' was carried out by conventional pressed slice method. The results showed that: (1) The ploidy varied among culitvars. R. 'Viridiflora' , R. 'Yulinglong' , R. 'Slater's crimson China' and R. 'Parsons Pink China' were diploid with 14 chromosomes (2n=2x=14). R. 'Chunshui Lu bo' and R. ' Huzhongyue' were triploid with 21 chromosomes (2n=3x=21). R. 'Dafugui' , R. 'Jinfenlian' , R. 'Juhongchao' ,R. ' Yingri Hehua' and R. ' Sichun' were tetraploid with 28 chromosomes (2n=4x=28). (2) Among the cultivars, the absolute length of chromosomes ranged from 1 pan to 3 μm and the asymmetry index were between 56.560% and 60.376%. Except for R. ' Sichun' and R. ' Yingri Hehua', no chromosome satellite was observed in the cells of other cultivars. (3) The karyotypes of R. ' Chunshui Lubo' and R. ' Huzhongyue' were both 1A with the same karyotype formula of 2n=3x=21=3sm + 18m. Those of R. ' Jinfenlian' and R.' Juhongchao' were both 1A and their karyotype formulas were 2n=4x=28=4sm + 24m. With a karyotype formula of 2n=4x=28=2sm + 26m, the karyotype of R. ' Dafugui' was also 1 A, while that of R. 'Viridiflora' was 1B with 2n=2x=14=2sm + 12m being its karyotype formula. With the karyotypeformula being 2n=4x=28=4sm + 22m + 2m (SAT), the karyotypes of R. 'Sichun' and R. ' Yingri Hehua' were both 1A. R. 'Yulinglong' had the karytype of 1A and karyotype formula of 2n=2x=14=2sm + 12m. As far as R. ' Parson's Pink China' and R. 'Slater's crimson China' were concerned, their karyotypes were both 2A with a formula of 2n=2x=4sm + 10m.

Abstract:[Objective] The study was to provide the cytological basis for development and utilization and genetic breeding of Kengyilia thoroldiana(Oliv.)J.L.Yang,Yen et Baum.[Method] The chromosome karyotype of root tip cells of K.thoroldiana was analyzed with squash method.[Result] The results of chromosome counting showed that the chromosome number of root tip cells of K.thoroldiana was 2n=42,among them there were 16 pairs of metacentric chromosome,4 pairs of submetacentric chromosome and 1 pair of acrocentric chromosome,and their karyotype formula was 2n=6x=32m+8sm(4SAT)+2st.The total length of genome of K.thoroldiana was 178.26 μm with average length of 8.48 μm.The variation range of chromosome length was 2.93%-8.40%.According to the karyotype classification standard of stebbins,the karyotype of K.thoroldiana was 2B type.[Conclusion] K.thoroldiana was hexaploid with basic chromosome number of x=7.

Abstract:The karyotype of Rheum officinale was analyzed in the present study. The chromosome features were conducted by conventional root-tip squashing method and photomicrography technique. Results were as follows: The karyotype formula of R. officinale from Luanchuan in Henan was 2n = 2x = 22 = 18m + 4M; karyotype was 1A type; karyotype asymmetry index (As.K.%) was 54.81%. Chromosome ploidy was diploid, and no polyploidy was observed. The chromosome number and the morphology of R. officinale are stable and clear, which can provide the foundation for further research.

Abstract:In order to explore the karyotype and evolutionary trend of Clematis and provide taxonomically useful data, the chromosome number, the number and position of satellites, the karyotype formulae, the karyotype, arm ratio, relative length centromeric index, and the index of the karyotype asymmetry of 11 species of the genus Clematis were studied. All the analyzed species showed the same stable chromosome number (2n = 2x = 16) and basic chromosome number 8. Clematis karyotypes are composed by metacentric, submetacentric, and terminal centromeric chromosomes. Clematis lanuginosa, C. otophora, C. lasiandra, C. shenlungchiaensis, C. florida var. plena, C. fusca var. violacea, C. crispa, and C. viticella belong to Type 2A, a very symmetric and plesiomorphic type. Clematis henryi, C. integrifolia, and C. japonaca belong to Type 2B, which is apomorphic relative to the other group. The 11 species of Clematis are different in karyotype parameters providing data for the taxonomy of the genus.

Abstract:Karyotype was studied via squash method in seventeen chrysanthemum cuhivars with small in- florescences which differ in plant architecture and purpose.Karyotypic findings showed that all cultivars were mixoploid with chromosome number ranging from 49 to 57,and 54 was the most frequent number observed.In addition,diversity of chromosome morphology was observed.Metacentric,submetaeentric and acrocentric chromosomes coexisted in most cultivars.The ratio of chromosome length,i.e.,ratio of the length of the lon- gest chromosome to that of the shortest one,varied from 1.62 to 2.39.The percentage of chromosome with arm ratio beyond 2 was between 11.11% and 29.62%.Asymmetry index (As.K%) ranged from 59.74% to 63.37%.The karyotypes of Dendranthema×grandiflorum‘Aoyunchengguang’,‘Jinlingbaifeng’(2n=53) and three ground cover cultivars were characterized as"2B"type,whereas,those of the remaining cuhivars were characterized as"2A"type.Karyotype diversity not solely resulted from the mutation of chromosome,re- combination of chromosome also likely contributed to the diversity.

Abstract:In this study, both Clausena lansium and C. anisum-olens were shown to be diploids, 2 n=2 x=18. The karyotype was 2 n=18=16 m+2 sm for C. lansium and 2 n=18=14 m+4 sm for C. anisum-olens, both being of the 1 A type and relatively promitive. A brief discussion is given about the phylogenetic relationship between the two species.

Abstract:In order to investigate the systematic relationship and to complete the karyological data of rose, for the purpose of exploring desirable traits for genetic improvement of modern cultivars, nuclear DNA content of 204 accessions representing almost all sections of genus Rosa were estimated by flow cytometry. The 1Cx-value of each taxon was calculated according to the estimated DNA content. The results were as follows: (1) nuclear DNA content of the studied taxa varied from 0.73 pg in diploid R. zhongdianensis (Section Pimpinellifoliae) to 5.07 pg in decaploid R. praelucens (Section Microphyllae), and nuclear DNA content of rose species generally increased with, but not simply proportional to, ploidy level; (2) 1Cx-value varied from 0.37 pg in R. zhongdianensis to 0.89 pg in R. brunonii (Section Synstylae), and the studied taxa were significantly different from each other in 1Cx-value; (3) there exist significant differences of 1Cx-values among sections, varying from 0.46 pg in Section Pimpinellifoliae to 0.77 pg in Section Synstylae; (4) ploidy level varied within most sections except in Section Banksianae, Section Bracteatae, Section Laevigatae and Section Synstylae, while the variation of 1Cx-value with ploidy level differed greatly among sections. The results suggested that taxonomic section should be taken into consideration when assigning ploidy of rose species from rapid estimates of DNA content by flow cytometry. Also, although it does not reflect the taxonomy and biosystematics sufficiently, nuclear DNA content and 1Cx-value can be used as reference points in the biogeographic, systematic and evolutionary study of genus Rosa.

Abstract:In this paper, we reported the karyotypes of 30 Chinese large flowered pot chrysanthemum cultivars, differing with respect to flower type, petal type, and flower colour. The interphase nuclei and prophase chromosomes of all the cultivars are, respectively, of the complex chromocentre and the interstitial type. Somatic chromosome number varies from 49 to 62, mostly falling in the range 51-56 or 58. Most of the cultivars are chromosomal mosaics, with three showing 2n = 6x = 54, and two 2n +1 = 6x + 1 = 55. At mitotic metaphase, most of the chromosomes are of the metacentric or submetacentric type, with a small number of acrocentrics and telocentrics. B chromosome (s) are present in about 22% of the entireties. The asymmetry index of the chromosomes ranges between 61 and 66%. The karyotypes can be categorized as reversely symmetrical types "2A" or "2B".

Abstract:The article introduced a new method of chromosomes preparation by callus of Cortex Magnoliae Officinalis and then analysed the karyotype of Cortex Magnoliae Officinalis. The results showed that the chromosome amounted to 38. The karyotype formula was K_((2n))=38=32m+4sm(2SAT)+2st, among which there were 16 pairs of metacentric chromosomes(m), 2 pairs of submetacentric chromosomes(sm) and 1 pair of acrocentric chromosome(st).

Abstract:[Objective] Rosa ‘ Yunfen'was a plant breeders' right owned new cut rose cultivar.The rose hip and achene setting rates were very low and differred significantly among crossing combinations when R.‘ Yunfen'was used as the paternal parents in hybridization breeding.[Method] To find the reason,pollen mother cell meiosis of R.‘ Yunfen'was observed and the meiotic phases in different periods of the division were recorded in this study.[Result] The optimal time for observation of meiosis was when the bud was about 1.03-1.07 cm wide and 2.9-3.4 cm long.At that stage,the anther colour turned from light green to yellowish green.The meiotic behaviors of pollen mother cells were non-synchronic and several meiosis phases could be observed in the same anther.Abnormal phenomina of meiosis such as univalent,trivalent and chromosome-bridge could be observed in the first division of meiosis.Then,triad and dyad were produced in the second division of meiosis of R.‘ Yunfen',by which 2n pollens were formed.[Conclusion] The abnormal division of PMCs of R.‘ Yunfen'might be a reason for the low hip and achene setting rate when used as paternal parent in breeding.

Abstract:Chinese narcissus(Narcissus tazetta var.chinese Roem.)only blooms without seeds and it propagates using its bulbs.There were some different reports for its karyotype.Our result of karyotype analysis supports the viewpoint that Chinese narcissus belongs to triploid plant,and the karyotype leans to a heterogeneous triploid.During the meiosis of microspore mother cells(MMC),the most cells display abnormal chromosomes:some chromosomes moved out of the spindle of the cell at metaphase;some chromosome bridges and lagging ones were formed at anaphase;and many microspores displayed the micronucleus at the telophase.The abnormality of chromosomes during MMC meiosis may induce microspore abortion during its development.

Abstract:A hybrid between Triticum aestivum and Psathyrostachys huashanica(H9802-6),and a hybrid between Triticum aestivum and Haynaldia villosa(V9910-15-4) were used to carry out cytology observation in the meiophase of their pollen.The results proved that there were some alien substitution lines and alien addition lines in the hybridization of H9802-6,which may not be steady.In the hybridization of V9910-15-4,the chromosome number and chromosome configuration were diverse and complicate.There were unusual situations in the meiophase.A lot of ring-like synaptic chromosome,tip synaptic chromosome and some un-synaptic univalent had been found.It reflected that the hybridization of Tritcium aestivum and Haynaldia villosa was not easy to stay steady in application.

Abstract:Fifty‐two populations representing thirty‐two taxa of Lilium L. from China are karyologically analyzed. The results showed that all populations have the same basic chromosome number x=12, and all species are diploid except Lilium tigrinum, which is triploid with 2n=36. In addition, one population of L. sulphureum is aneuploid with 2n=23. The karyotype evolution in the genus Lilium is mainly in terms of alterations of the fine structure of the chromosomes, not in ploidy or basic chromosome number variation. When combining previous studies and our results, we consider the major driving forces for evolution of Lilium to be an increase in karyotypic asymmetry, and unequal translocation and formation of secondary constrictions. The existence of intercalary satellites on the two largest pairs of chromosomes is correlated with the geographic distribution pattern of Lilium across the world. The Hengduan Mountains is a modern diversity and differentiation center of Lilium and it also could be the center of origin for this genus. In addition, some taxonomic conclusions were verified on the subgenus level. Among the species investigated, the chromosome number and karyotypes of 13 taxa were documented for the first time.

Abstract:Two wild populations of L. houdyshelii and L. straminea were first discovered in Anhui province and cytologically studied. The results showed that two kinds of cytology were found in L. houdyshelii, with one having the karyotype 2n = 31 = 3m + 13st + 10t + 5T, and the other having the karyotype 2n = 30 = 3m + 21st (SAT) + 3t (SAT) + 3T. The karyotypes and secondary constriction are reported for the first time. The karyotype of L. straminea is formulated as 2n = 19 = 3m + 7st + 4t + 5T, which is similar to some previous reports and confirmed here. The karyotypes of these two taxa are both of 3B type according to the degree of asymmetry and Stebbins' criteria. According to the gross floral morphology, phenology and karyotype of these two taxa, it is suggested that L. houdyshelii is probably a hybrid between a taxon with 2n = 6M + 10T, such as L. longtuba or L. chinensis and L. radiata var. pumila, and L. straminea putatively originates in hybridization between L. radiata var. pumila and L. chinensis.
