Dose-time-toxicity relationship study on hepatic toxicity caused by single dose total Bupleurum saponin extracts to mice

Abstract:Objective: To study on the time-toxicity and dose-toxicity relationships caused by single dose total Bupleurum saponin extracts to mice.Methods: Mice are grouped according to different time or dose points,to observe the death condition and toxicity of mice,detect ALT 、AST in serum,calculate liver,spleen,thymus index,and observe the morphological changes of hepatic tissue under light microscope.Results: ALT,AST in serum are peaked after 4 hours' administration,and restored near normality after 48 hours.Compared with the normal group,31.25 g/kg dose group of total Bupleurum saponin extracts produced significant hepatic injury,and ALT,AST in serum increased significantly with the dose increasing.Under light microscope,the hepatic tissue of different dose mice all had different degree's edema and fatty degeneration to hepatic cells,and the high dose group even appeared to be necrosis and lobular structure unclear.Conclusion: The total Bupleurum saponins extracts gavaged certain dose one time to mice could cause acute hepa-injury and even death,and there were certain time-toxicity and dose-toxicity relationships.
