Abstract:Objective To investigate the potential correlation between pathogenesis of primary Sjgren′s syndrome(pSS) and the function of monocyte-derived dendritic cells.Methods Monocytes from 11 pSS patients and 11 healthy controls were isolated and induced to dendritic cells in the presence of IL-4 and GM-CSF.Polyinosinic polycytidylic acid(polyI∶ C,50 μg/mL) was used to simulate the monocyte-derived dendritic cells.Serial cytokines and TLR3 mRNA were assessed by ELISA and RT-PCR,respectively,at the 6th,12th and 14th hour after polyI∶ C treatment.Results Significantly more IFN-α,IFN-β and TNF-α were produced in monocyte-derived dentritic cells from pSS patients when challenged with polyI∶ C.Although TLR3 mRNA expression was down-regulated by polyI∶ C stimulation,it was impaired in pSS patients.Conclusion The Hypersensitivity to polyI∶ C,which may associated with TLR3 abnormality,of the monocyte-derived dendritic cells from pSS patients was identified.The abnormality of monocyte-derived dendritic cells from pSS patients may involved in the pathogenesis of pSS.
