Abstract:A novel asymmetric hydrogenation of vinylthioethers was developed using a ruthenium(II) NHC complex. This method provides an efficient approach to optically active 1,5-benzothiazepines featuring stereocenters with C-S bonds. Excellent enantioselectivities (up to 95%ee) and high yields (up to 99%) were obtained for a variety of substrates bearing a range of useful functional groups. Moreover, this methodology could be directly applied to the synthesis of the antidepressant drug R-(-)-thiazesim.
Regioselective Synthesis of Acyclic Cis-Enediynes Via an Acid-Catalyzed Rearrangement of 1,2-Dialkynylallyl Alcohols. Syntheses, Computational Calculations, and Mechanism

Abstract:A novel synthesis of acyclic cis-enediynes 2 has been established by an acid-catalyzed rearrangement of 1,2-diyn-2-propen-1-ols 1 possessing a C-3-aryl group in the presence of water, alcohols, or thiols. Reactivity of allyl alcohols and regio- and cis/trans diastereoselectivity of the allylic migration were examined. In the presence of(+/-)-10-camphorsulfonic acid (CSA), the parent allyl alcohol 5 and the C-3-methyl-substituted 9 failed to give enediynes, whereas the C-3-aryl-substituted 12 and 29 underwent the allylic rearrangement to provide predominantly cis-enediynes 16 and 31 at room temperature or below. Under similar acidic conditions, enediyne alcohol 13 produced 16b and 16d with the same regio and cis/trans diastereoselectivity observed for 12. Allyl alcohol 30, an isomer of 29, also provided enediynes 31c and 32c after a prolonged reaction (90 h) at room temperature in the presence of CSA and EI;OH. These results suggested that the same allylic cations were obtained from allyl alcohols 12 and 13 or 29 and 30 even though the ease of ionization differed for each substrate. Involvement of allylic cations in the product-forming step was confirmed by the finding that chiral allyl alcohols (-)-12 and (-)-18c furnished racemic products. In general, the p-MeOPh-substituted allyl alcohol 29 gave a better regioselectivity than the Ph-substituted 12. In the reactions with alcohols, the regioisomeric ratios were 100:0 (31:33) for 29 and ca. 96:4 (16:17) for 12; the ratios decreased to ca. 90:10 (31:33) for 29 and ca. 10:30 (16:17) for 12 when thiols were used. The cis/trans diastereoselectivity is higher for allyl alcohol 12 (100% for 16 at 20 degrees C) compared to that for 29 (31:32 = 80:20-94:6 at 0 degrees C). Computational calculations at the RHF/3-21G level, carried out on the model compounds and allylic cations, indicated that nucleophilic trapping takes place preferentially at the Cg carbon to form the thermodynamically much more stable enediynes. Under the best reaction conditions (1 equiv of CSA and 2 equiv of EtOH in CH2Cl2, 20 degrees C), a number of acyclic cis-enediynes can be synthesized in three steps from the commercially available alpha-bromocinnamaldehyde (10).

These compounds 3a-3p were synthesized in good yield by refluxing alpha-aminoacetophenones and hydrazines in glacial acetic acid. The reaction proceeds under mild conditions and is compatible with various functional groups.

Abstract:A highly efficient asymmetric hydrogenation of azepine/oxazepine-type seven-member cyclic imine hydrochlorides was successfully developed using Rh/bisphosphine-thiourea ligand ZhaoPhos, affording various chiral seven-member cyclic amines with full conversions, high yields, and excellent enantioselectivities (up to 96% yield, >99% ee). Additionally, this asymmetric hydrogenation can proceed well on gram scale with excellent ee value. Moreover, control experimental results displayed that the anion-bonding interaction between the chloride ion of the substrate and thiourea motif of the ZhaoPhos played an important role to obtain excellent enantioselectivity.

Abstract:Chiral aryl glycines are biologically relevant compounds that received a great deal of attention over the last years. They can be prepared by aminohydroxylation (K. L. Reddy, K. B. Sharpless J. Am. Chem. Soc. 1998, 120, 1207-1217), amination of enolates (D. A. Evans, S. G. Nelson J. Am. Chem. Soc. 1997, 119, 6452-6453) or reductive amination (R. Kadyrov, T. H. Riermeier, U. Dingerdissen, V. Tararov, A. Börner J. Org. Chem. 2003, 68, 4067-4070) but in some cases low regioselectivity, high catalyst loading or poor reactivity is observed. Abstract:Transition-metal-catalyzed reductive coupling reactions have emerged as powerful protocols to construct C-C bonds. However, the development of enantioselective C(sp3)-C(sp3) reductive coupling remains challenging. Herein, we report a highly regio-, diastereo-, and enantioselective reductive homocoupling of allylic acetates through cooperative palladium and photoredox catalysis using diisopropylethylamine or Hantzsch ester as a homogeneous organic reductant. This straightforward protocol enables the stereoselective construction of C(sp3)-C(sp3) bonds under mild reaction conditions. A series of C2-symmetrical chiral 1,5-dienes were easily prepared with excellent enantioselectivities (up to >99% ee), diastereoselectivities (up to >95:5 dr), and regioselectivities (up to >95:5 rr). The resultant chiral 1,5-dienes can be directly used as chiral ligands in asymmetric synthesis, and they can be also transformed into other valuable chiral ligands.

Abstract:A chemoselecive cyclodehydration of α‐acyl‐β‐arylaminoacrylamides via an electrophilic activation strategy has been described. A series of substituted 4‐aminopyridines are chemoselectively prepared in 73‐92% yields from α‐acyl‐β‐arylaminoacrylamides activated by hexaphenyloxodiphos phonium triflate (Hendrickson reagent), whereas substituted diaza[n]phenacenes (n = 4‐6) are obtained in 72‐93% yields directly from α‐acyl‐β‐arylaminoacrylamides using Hendrickson reagent in combination with triflic anhydride (Tf2O). The synthetic protocols feature with readily available starting materials, mild reaction conditions, simple execution, and wide range of synthetic potential of products.

Abstract:A palladium-catalyzed enantioselective decarboxylative allylic alkylation of α-benzyl cyanoacetates with methylene cyclic carbamates has been successfully developed. An array of enantioenriched products bearing nitrile-containing acyclic quaternary carbon stereocenters were obtained in high yields with good enantioselectivities (up to 99% yield and 93% ee). Abstract:An efficient method for asymmetric synthesis of acyclic α-tertiary amine derivatives has been achieved through enantioselective aminations of α-branched ynones with azodicarboxylates enabled by chiral phosphoric acid catalysis. Moreover, kinetic resolution of racemic starting material was realized under these conditions, which gave access to valuable enantioenriched α-substituted ketones. Abstract:We have developed an efficient method for the preparation of chiral fluorinated compounds by sequential organocatalyzed desymmetrization of 3-substituted glutaric anhydrides and photoredox-catalyzed decarboxylic fluorination. Chiral fluorides can be prepared in yields of up to 58% and with excellent enantioselectivities of up to 98% ee.

Abstract:Chiral cyclopentadienyl-rhodium(III) CpxRh(III) catalysis has been demonstrated to be competent for catalyzing highly enantioselective aziridination of challenging unactivated terminal alkenes and nitrene sources. The chiral CpxRh(III) catalysis system exhibited outstanding catalytic performance and wide functional group tolerance, yielding synthetically important and highly valuable chiral aziridines with good to excellent yields and enantioselectivities (up to 99% yield, 93% ee). This protocol presents a novel and effective strategy for synthesizing enantioenriched aziridines from simple alkenes. Various transformations were performed on the aziridine products, illustrating the versatility and synthetic potential of this protocol for constructing highly functionalized compounds.

Abstract:A dual nickel/photoredox-catalyzed enantioselective reductive cross-coupling of aryl halides with CF<sub>3</sub>-substituted racemic alkyl electrophiles has been established. The approach accommodates a broad palette of aryl iodides and alkyl bromides to access a variety of chiral CF<sub>3</sub>-containing compounds. Abstract:We report the development of a modular, one pot, sequential chemoenzymatic system for the formal enantioselective construction of the C-C bond in 2-aryl 1,4-dicarbonyl compounds. This sequence comprises a rhodium catalyzed diazocoupling that provides >9:1 selectivity for heterocoupling of two diazoesters and a reduction mediated by an ene-reductase (ER), which occurs in up to 99% enantiomeric excess (ee). The high yield and enantioselectivity of this system result from the preferential generation of an (E)-alkene from the diazo coupling reaction and selective reduction of the (E)-alkene in a mixture of (E) and (Z) isomers by the ER. Screening of a panel of ERs revealed that OPR1 from Lycopersicum esculentum catalyzes the reduction of bulky tert-butyl or benzyl esters to afford chiral diesters that are poised for orthogonal reactions at the two distinct ester units of the product. Overall, this work demonstrates the benefit of combining organometallic and enzymatic catalysis to create unusual overall transformations that do not require the isolation and purification of intermediates.

Abstract:An efficient and concise asymmetric hydrogenation of acrylate esters promoted by the cationic ruthenium monohydride complex [Ru(H)(eta(6)-cot)SYNPHOS]+BF4- is reported. A full investigation of the effects of catalyst precursors, solvents, temperature, hydrogen pressure, substrates as well as steric and electronic properties of ligands was carried out. The corresponding valuable Roche ester derivatives were obtained in good to excellent isolated yields and high enantioselectivities under mild conditions. The robustness and practicability of this highly enantioselective hydrogenation was demonstrated by the synthesis of the 3-hydroxy-2-methylpropanoic acid tert-butyl ester on a multigram scale, resulting III excellent yield and ee up to 94%.
