[An Experimental Study of Endothelin Receptor A Expression in Brain Tissue after Subarachnoid Hemorrhage in Rabbits].

Abstract:Objective To study the expression and the significance of calcitonin gene-related peptide (CGRP) and endothelin receptor A (ETRA) in the brain tissue after subarachnoid hemorrhage (SAH). Methods Forty-five Japanese rabbits were randomly divided into four groups, including SAH-group (n = 20), saline-group (n = 15), puncture-group (n = 5) and blank-group (n = 5). The SAH animal model was established by autologus arterial blood injection into the cisterna magna twice. Immunohistochemical staining for GCRP and ETRA were used for observing their expression. Results Immuno-labeled granules of CGRP and ETRA were expressed in all 20 cases of SAH-group. The CGRP was expressed positively in few cells of SAH-group at 1 hour, strongly from 3 d to 7 d, especially at 5 d, and then the positive cells decreased at 10 d, but never came back to the normal. ETRA was expressed mainly in the cytoplasm of neurons and neuroglial cells in SAH-group. It was lightly stained at 1 hour after injecting the blood into cisterna magna at the first time, and then turned into dark brown at 3 d; it was stained lighter at 5 d than 3 d, and then become lighter gradually. Conclusion The expressions of CGRP and ETRA are obvious in the brain tissue after SAH, with a significant dynamic change, but their trends are not synchronous. CGRP and ETRA may play an important role in the development and progression of delayed neurological deficit.
