Abstract:The aim of this study is to investigate how firing patterns of neuron in hippocampus change with coupling effect between soma and dendrite via simulation of single neuron firings by a two-compartment (soma-dendrite) model. The results will help to understand how firing patterns relative to synaptic plasticity in further study. The neuron model used in this study consisted two compartments: the soma compartment and the dendrite compartment, which can describe synaptic input functions. Simulations of neural firings were performed with fourth order Runge-Kutta algorithm with different range of coupling conductance. Firing patterns of neuron were simulated under different current applied to soma (I s ). Abstract:What is the difference between the efferent spike train of a neuron with a large soma versus that of a neuron with a small soma? We propose an analytical method called the decoupling approach to tackle the problem. Two limiting cases-the soma is much smaller than the dendrite or vica versa-are theoretically investigated. For both the two-compartment integrate-and-fire model and Pinsky-Rinzel model, we show, both theoretically and numerically, that the smaller the soma is, the faster and the more irregularly the neuron fires. We further conclude, in terms of numerical simulations, that cells falling in between the two limiting cases form a continuum with respect to their firing properties (mean firing time and coefficient of variation of inter-spike intervals).

Abstract:The capacity of short-term memory has been studied using an integrate-and-fire neuronal network model. It is found that the storage of events depend on the manner of the correlation between the events, and the capacity is dominated by the value of after-depolarization potential. There is a monotonic increasing relationship between the value of after-depolarization potential and the memory numbers. The biophysics relevance of the network model is discussed and different kinds of the information processes are studied too.

Abstract:Here a two-compartment model is used to investigate how spike threshold depends on the rate of membrane depolarization leading to the action potential, i.e., dV(S)/dt. The model is comprised of a soma and a passive dendrite, and incorporates a morphological parameter that describes the area proportion occupied by soma. The threshold potential of somatic chamber is determined within a range of dV(S)/dt for different values of morphological parameter and internal coupling conductance. By analyzing the interactions of inward and outward membrane currents prior to spike initiation, the biophysical basis associated with each threshold dynamic has been identified. Based on the simulations, we conceptualize the threshold dynamics of neuron as the outward level of somatic net current at the perithreshold potentials. These results provide a detailed description about how the morphology and biophysics of neurons participate in their threshold dynamics, which could facilitate to interpret the modulatory mechanism of subthreshold electromagnetic fields as well as the biophysical basis of neural coding. Abstract:We have developed two-, three- and four-compartment models of a mammalian olfactory bulb mitral cell as a reduction of a complex 286-compartment model [1]. A minimum of three compartments, representing soma, secondary (basal) dendrites and the glomerular tuft of the primary dendrite, is required to adequately reproduce the behaviour of the full model over a broad range of firing rates. Adding a fourth compartment to represent the shaft of the primary dendrite gives a substantial improvement. The reduced models exhibit behaviours in common with the full model which were not used in fitting the model parameters. The reduced models run 75 or more times faster than the full model, making their use in large, realistic network models of the olfactory bulb practical.

Abstract:Mounting experimental evidence suggests that brain-state-specific neural mechanisms, supported by connectomic architectures, play a crucial role in integrating past and contextual knowledge with the current, incoming flow of evidence (e.g., from sensory systems). These mechanisms operate across multiple spatial and temporal scales, necessitating dedicated support at the levels of individual neurons and synapses. A notable feature within the neocortex is the structure of large, deep pyramidal neurons, which exhibit a distinctive separation between an apical dendritic compartment and a basal dendritic/perisomatic compartment. This separation is characterized by distinct patterns of incoming connections and brain-state-specific activation mechanisms, namely, apical amplification, isolation, and drive, which are associated with wakefulness, deeper NREM sleep stages, and REM sleep, respectively. The cognitive roles of apical mechanisms have been demonstrated in behaving animals. In contrast, classical models of learning in spiking networks are based on single-compartment neurons, lacking the ability to describe the integration of apical and basal/somatic information. This work aims to provide the computational community with a two-compartment spiking neuron model that incorporates features essential for supporting brain-state-specific learning. This model includes a piece-wise linear transfer function (ThetaPlanes) at the highest abstraction level, making it suitable for use in large-scale bio-inspired artificial intelligence systems. A machine learning evolutionary algorithm, guided by a set of fitness functions, selected the parameters that define neurons expressing the desired apical mechanisms.

Abstract:For single neuron models, reproducing characteristics of neuronal activity such as the firing rate, amplitude of spikes, and threshold potentials as functions of both synaptic current and conductance is a challenging task. In the present work, we measure these characteristics of regular spiking cortical neurons using the dynamic patch-clamp technique, compare the data with predictions from the standard Hodgkin-Huxley and Izhikevich models, and propose a relatively simple five-dimensional dynamical system model, based on threshold criteria. The model contains a single sodium channel with slow inactivation, fast activation and moderate deactivation, as well as, two fast repolarizing and slow shunting potassium channels. The model quantitatively reproduces characteristics of steady-state activity that are typical for a cortical pyramidal neuron, namely firing rate not exceeding 30 Hz; critical values of the stimulating current and conductance which induce the depolarization block not exceeding 80 mV and 3, respectively (both values are scaled by the resting input conductance); extremum of hyperpolarization close to the midpoint between spikes. The analysis of the model reveals that the spiking regime appears through a saddle-node-on-invariant-circle bifurcation, and the depolarization block is reached through a saddle-node bifurcation of cycles. The model can be used for realistic network simulations, and it can also be implemented within the so-called mean-field, refractory density framework.
Extending Integrate-and-Fire Model Neurons to Account for the Effects of Weak Electric Fields and Input Filtering Mediated by the Dendrite

Abstract:Transcranial brain stimulation and evidence of ephaptic coupling have recently sparked strong interests in understanding the effects of weak electric fields on the dynamics of brain networks and of coupled populations of neurons. The collective dynamics of large neuronal populations can be efficiently studied using single-compartment (point) model neurons of the integrate-and-fire (IF) type as their elements. These models, however, lack the dendritic morphology required to biophysically describe the effect of an extracellular electric field on the neuronal membrane voltage. Here, we extend the IF point neuron models to accurately reflect morphology dependent electric field effects extracted from a canonical spatial "ball-and-stick" (BS) neuron model. Even in the absence of an extracellular field, neuronal morphology by itself strongly affects the cellular response properties. We, therefore, derive additional components for leaky and nonlinear IF neuron models to reproduce the subthreshold voltage and spiking dynamics of the BS model exposed to both fluctuating somatic and dendritic inputs and an extracellular electric field. We show that an oscillatory electric field causes spike rate resonance, or equivalently, pronounced spike to field coherence. Its resonance frequency depends on the location of the synaptic background inputs. For somatic inputs the resonance appears in the beta and gamma frequency range, whereas for distal dendritic inputs it is shifted to even higher frequencies. Irrespective of an external electric field, the presence of a dendritic cable attenuates the subthreshold response at the soma to slowly-varying somatic inputs while implementing a low-pass filter for distal dendritic inputs. Our point neuron model extension is straightforward to implement and is computationally much more efficient compared to the original BS model. It is well suited for studying the dynamics of large populations of neurons with heterogeneous dendritic morphology with (and without) the influence of weak external electric fields.

Firing Patterns of the CA1 Pyramidal Neuron with Geometric Singular Perturbation: a Model Study

Abstract:An investigation of CA1 pyramidal model is an important issue for applications, which is intimately related to the composition of ions in the extracellular environment and external stimulation. In this paper, it is demonstrated that the effects of different electrophysiological parameters such as muscarinic-sensitive potassium current activation variable and sustained sodium current inactivation variable on the firing sequence of model by numerical simulations. Furthermore, the paper also discusses that the temperature affects the firing of the CA1 model from direct current (DC) and alternating current (AC) stimuli. It is found that the model exhibits excellent spiking and bursting patterns, even chaotic patterns occur. Meanwhile, generalized mixed oscillations emerge in the model. Additionally, the firing modes are depicted by providing the response curve (RC), inter-spike interval curve (ISI), phase diagram curve (PDC) and the number of spikes per burst curve (NC). Mathematically, the paper elaborates the results which are presented to obtain two lower dimensional subsystems, which govern the fast and slow dynamics for giving insight into the dynamic behaviors of the full 5D system based on the geometric singular perturbation theory (GSPT). Particularly, we analyse the phase diagrams of the CA1 model to understand the properties better. The present results may contribute to further understand the information processing of the CA1 pyramidal neurons.
Compartmental models of mammalian motoneurons of types S, FR and FF and their computer simulation

Abstract:Mathematical models of motoneurons (MNs) of types S, FR and FF were developed based on cat MN data. Each of the three models has an initial segment, a soma and a dendritic tree. The initial segment and the soma include models of several types of ionic currents, including a calcium-dependent slow potassium current. The dendritic tree is modeled as a series association of several electrically passive cylinders. Afterhyperpolarization parameters, current to frequency relation and the responses to input current steps, ramps and sinusoids were used for model validation. The effects of sinusoidally varying synaptic inputs at different levels of the dendritic tree were studied by computer simulation. The corresponding frequency response functions resulted of lowpass type with cutoff frequencies from 10 to 40 Hz, for synapses occurring more distally or more proximally, respectively. The nonlinear effects caused by two sinusoidally varying synaptic conductances (at 7 and 11 Hz), acting at different dendritic segments, were quantified by spectral analysis of the current reaching the soma. The simulations pointed to two main nonlinear effects: (i) harmonics of the two input frequencies (e.g., 14 Hz) and (ii) intermodulation terms (e.g., 4 Hz). When the two synaptic inputs occurred on more distal dendritic compartments the nonlinear effects were more pronounced.
Metabolic Energy Consumption and Information Transmission of a Two-Compartment Neuron Model and Its Cortical Network

Abstract:The physiological structure of the brain and the morphology of its neurons result from selective pressure, the nervous system must be energy efficient. In this paper, a two compartment model consisting of dendritic and axon compartments is proposed to simulate the effects of morphology and temperature on neuronal kinetics. Based on the equivalent circuit method, we calculated the metabolic energy consumption of neurons with morphological and temperature variations. It is shown that higher temperatures increase the separation of Na+ and K+ currents in the axon, resulting in higher energy efficiency, while neuron with moderate morphological parameter is more likely to fire, but accompanied by a higher energy cost. We integrate this two-compartment model into a feedforward network to simulate changes in network energy consumption affected by neuron morphology and temperature during information propagation. It is found that moderate temperature and morphology allow stable propagation of synchronization spikes in the feedforward network and maximize the energy utilization for information transmission. In addition, we present a simple statistical method to detect the robustness of network information transmission. This paper may provide some insights for further studies on energy consumption in the cerebral cortex.

Abstract:Modulating the spike patterns of single neuron is an important content of closed-loop cellular electrophysiology. This paper investigates the spike pattern control of a physiologically accurate two-compartment Pinsky-Rinzel model with a fast and local optimized generalized predictive control algorithm. In order to simulate the electrophysiological experiments more realistically, uncertainties including model parameter variations, noises with arbitrary intensities and changes of inner states are introduced in the model. Simulation results demonstrate the robustness of the proposed algorithm on different uncertainties. What is more, the control performances rely on the value of the geometric parameter that characterizes the ratio between the soma area and the dendrite area while has little to do with the internal coupling conductance. The derived theoretical simulations represent a new direction towards the design of online optimal stimulus waveforms in real cellular electrophysiological experiments, which can effectively cope with the physiological uncertainties.

Abstract:Medium spiny neurons are the principal neurons in striatum, which are closely related to many diseases of the central nervous system. Based on medium spiny neuron model of biological systems, the authors obtained various firing patterns by comparing the responses of the medium spiny neuron under direct current, alternating current and square wave firstly, and the results showed that the change of alternating current had a huge influence on the firing rhythm of this neuron. Then, this study researched the impact of three different ions conductance or permeability(sodium, potassium, calcium) on the firing properties of medium spiny neuron. Finally, by removing part of dendrites which simulated the death and degradation of them, the difference between the firing pattern of the healthy neuron and the incomplete neuron had been gained. According to the simulated results above, both the endogenic factor and extrinsic factor have effect on the firing rhythm of the medium spiny neuron.

Abstract:A complete single-neuron model must correctly reproduce the firing of spikes and bursts. We present a study of a simplified model of deep pyramidal cells of the cortex with active dendrites. We hypothesized that we can model the soma and its apical tuft with only two compartments, without significant loss in the accuracy of spike-timing predictions. The model is based on experimentally measurable impulse-response functions, which transfer the effect of current injected in one compartment to current reaching the other. Each compartment was modeled with a pair of non-linear differential equations and a small number of parameters that approximate the Hodgkin-and-Huxley equations. The predictive power of this model was tested on electrophysiological experiments where noisy current was injected in both the soma and the apical dendrite simultaneously. We conclude that a simple two-compartment model can predict spike times of pyramidal cells stimulated in the soma and dendrites simultaneously. Our results support that regenerating activity in the dendritic tuft is required to properly account for the dynamics of layer 5 pyramidal cells under in-vivo-like conditions.

Abstract:We assess, both numerically and theoretically, how positively correlated Poisson inputs affect the output of the integrate-and-fire and Hodgkin-Huxley models. For the integrate-and-fire model the variability of efferent spike trains is an increasing function of input correlation, and of the ratio between inhibitory and excitatory inputs. Interestingly for the Hodgkin-Huxley model the variability of efferent spike trains is a decreasing function of input correlation, and for fixed input correlation it is almost independent of the ratio between inhibitory and excitatory inputs. In terms of the signal to noise ratio of efferent spike trains the integrate-and-fire model works better in an environment of asynchronous inputs, but the Hodgkin-Huxley model has an advantage for more synchronous (correlated) inputs. In conclusion the integrate-and-fire and Hodgkin-Huxley models respond to correlated inputs in totally opposite ways.

Abstract:Pre-Bötzinger complex (PBC) neurons located in mammalian brain are the necessary conditions to produce respiratory rhythm, which has been widely verified experimentally and numerically. At present, one of the two different types of bursting mechanisms found in PBC mainly depends on the calcium-activated of non-specific cation current ( I CaN ). In order to study the influence of I CaN and stimulus current I exc in PBC inspiratory neurons, a single compartment model was simplified, and firing patterns of the model was discussed by using stability theory, bifurcation analysis, fast, and slow decomposition technology combined with numerical simulation. Under the stimulation of different somatic applied currents, the firing behavior of neurons are studied and exhibit multiple mix bursting patterns, which is helpful to further understand the mechanism of respiratory rhythms of PBC neurons.

Abstract:Leakage current is a critical factor which can balance ions flow across channels in neuron. In this study, a simple case is considered about the influence of leakage conductance gL on neuronal firing activities in a two compartment model, the detailed analysis includes two aspects: (1). the values of gL in the two compartments are equal, (2). the values of gL in the two compartments are not equal. With computer simulation, it is found that numerical results obtained from the variation of gL in somatic compartment and dendritic compartment show much difference, simulation results obtained from variation of gL in dendritic compartment have much similarity with results got in case (1) when gL varies, while the variation of gL in soma gives some difference with the case in (1). In the final the changing of membrane capacitance to the potential spike is also discussed here. All these results may give us a general rule of neuronal firing properties.

Abstract:Many central neurons, and in particular certain brainstem aminergic neurons exhibit spontaneous and fairly regular spiking with frequencies of order a few Hz. A large number of ion channel types contribute to such spiking so that accurate modeling of spike generation leads to the requirement of solving very large systems of differential equations, ordinary in the first instance. Since analysis of spiking behavior when many synaptic inputs are active adds further to the number of components, it is useful to have simplified mathematical models of spiking in such neurons so that, for example, stochastic features of inputs and output spike trains can be incorporated. In this article we investigate two simple two-component models which mimic features of spiking in serotonergic neurons of the dorsal raphe nucleus and noradrenergic neurons of the locus coeruleus. The first model is of the Fitzhugh-Nagumo type and the second is a reduced Hodgkin-Huxley model. For each model solutions are computed with two representative sets of parameters. Frequency versus input currents are found and reveal Hodgkin type 2 behavior. For the first model a bifurcation and phase plane analysis supports these findings. The spike trajectories in the second model are very similar to those in DRN SE pacemaker activity but there are more parameters than in the Fitzhugh-Nagumo type model. The article concludes with a brief review of previous modeling of these types of neurons and its relevance to studies of serotonergic involvement in spatial working memory and obsessive-compulsive disorder.
