Cytogenetic analysis of Hemibarbus labeo Pallas

Abstract:Metaphase chromosome specimens of Hemibarbus labeo injected with PHA and colchicin,were prepared from short-term culture of kidney cells with air-drying technique.Its diploid chromosome number was 2n=50.The karyotype is composed of 16m,16sm and 18st+t chromosomes,NF=82.Diploid nucleus DNA content was measured from the somatic cells of Hemibarbus labeo pallas using flow cytometer(Beckman cellL ab Quant,made in USA) and the DNA content(2.3 pg·N-1) oferythrocytes of chick(Gallus sp) as standard.The Hemibarbus labeo Pallas DNA content was 2.34 pg·N-1,which was correspond with the result measured chromosome number as ploidy.According to the present results and date reported,Hemibarbus labeo Pallas was possibly a diploid(2n).
