Effects of Perfluorooctane Sulphonate Subchronic Exposure on Learning and Memory Abilities and Hippocampal Neurons in Mice

Abstract:Objective To explore learning and memory abilities and changes of hippocampal neurons in mice after subchronic exposure to perfluorooctane sulphonate(PFOS).Methods ICR mice were randomly divided into four groups,eight in each and intraperitoneally exposed to PFOS for six consecutive weeks at the doses of 0,5,20 and 40 mg/kg body weight respectively.Learning and memory ability of mice were assayed by Morris water maze.Neuropathology by HE staining and statistical analysis for morphology in hippocampus were carried out.Results In Morris water maze test,the escape latency in 20 mg/kg PFOS group was significantly increased compared with the control group in the first training day(P0.05).While,in the first,second and fifth day,the escape latency all significantly delayed after subchronic exposure to PFOS in the dose of 40 mg/kg compared with the control(P0.05).Hippocampal neurons in CA1 region showed smaller cell body with dark stained nucleus in the higher dose exposure group.In comparison with the control group,neuron density declined both in CA1 and CA3 region,while darkness degree of cell nuclear significantly increased in CA1 of the higher dose exposure group.Conclusion Subchronic PFOS exposure may reduce learning and memory ability,which is relative to neural damage and loss of hippocampal neurons in mice.
