Apoptosis and Mitochondrial Membrane Potential in Circulating Neutrophils in Children Underwent Surgery

Abstract:Objective To investigate the perioperative changes of apoptosis and mitochondrial membrane potential in children's circulating neutrophils after operation.Methods Sixteen children underwent selective surgery under general anesthesia were studied.Blood samples were taken for assessment of neutrophils apoptosis and mitochondrial membrane potential on the day before surgery(T_0) and at 24 and 72 hours after operation(T_1 and T_2), respectively.After isolated,the neutrophils were cultured for 15 hours.Then,the cultured cells were stained with 7-amino-actinomycin D(7-AAD) to quantify the apoptosis and the cationic lipophilic fluorochrome 3,3′-dihexyloxacarbocy-anine iodide [DioC_6(3)] to analyze mitochondrial membrane potential by flow cytometer.Results The apoptotic rates of the cells at T_1 and T_2 were 7.55±4.70% and 5.47±2.67% respectively,which were significantly lower than those at T0 (13.27±7.11%,P0.05).The cationic lipophilic fluorochrome DioC_6(3) at T_1 and T_2 was 30.72±15.84 and 29.63±17.39 respectively,which were significantly less than that at T_0(30.72±15.84)(P0.05).Conclusions Surgical trauma is associated with a significant decrease in neutrophils apoptosis with lower levels of DioC_6(3),indicating that mitochondrial membrane potential of less cells is reduced.This delay of neutrophils apoptosis may be one of the causes involved in systemic inflammatory response syndrome.
