Abstract:【Objective】 To construct the recombinant adenovirus with pAdxsi-eGFP-rNIS and identify the characterization.【Methods】Recombinant shuttle vector of pShuttle-eGFP-CMV and primitive plasmid contained rNIS gene were cut down with enzyme.And vector fragment and inserted fragment were regained after electrophoresis.The two fragments were linked with ligase and translated into DH5a strain which smeared to the solid medium with kanamycin assistance.Monoclonal colony was inoculated into the fluid medium with kanamycin assistance.The recombinant shuttle plasmid of pShuttle-eGFP-CMV-rNIS was abstracted from the bacterium and was identified.pShuttle-eGFP-CMV-rNIS was transferred into the recombinant adenovirus frame vector of pAdxsi with the similar methods.The recombinant adenovirus plasmid of pAdxsi-eGFP-rNIS was extracted after amplification in the bacterium and then was identified.A549 cells were observed under fluorescence microscope after tansfection by Ad-eGFP-rNIS.Then the cells were incubated with 125I solution and measured by γ counter.【Results】RNIS gene was constructed into the recombinant shuttle vector of pShuttle-eGFP-CMV.And pShuttle-eGFP-CMV-rNIS was transferred successfully into the recombinant adenovirus frame vector of pAdxsi.The genes of rNIS and eGFP were expressed exactly.A549 cells infected by Ad-eGFP-rNIS could be visualized and those without infection could not be seen under fluorescence microscope.The radioactivity counter in the infected cells was 292501441 cpm.That in the cells without infection was 49127 cpm.There was significant difference between them(t=15.2,P0.001).【Conclusion】The recombinant adenovirus vector containing rNIS gene can be constructed successfully.rNIS is charac-terized after being infected into tumor cells.It will play an important role in the process of tumor target therapy and molecular imaging mediated by rNIS.
