The Role of Methionine-Enkephalin and Antibody Against Opioid Receptor on the Proliferation of Mouse NS-1 Cells

Abstract:Objective: To investigate the Effects and mechanisms of Methionine-enkephalin on Signal Transduction System in Mouse NS-1 Cells. Methods: The mouse NS-1 cells were treated with different concentrations of methionine-enkephalin (MENK) and monoclonal antibody against delta opioid receptor. The activity of protein kinase A (PKA) and protein kinase C (PKC) in the NS-1 cells were determined by phosphoration of histone, and the level of inositol trisphosphate (IP3) was measured with the method of radioimmunoassay (RIA). Results: MENK could enhance the activity of PKA in both of cytoplasm and membrane, which was antagonized by the antibody. A diphase effects of MENK on PKC was noted. 0.1 μmol/L of MENK could increase the activity of PKC in cytoplasm and lower the activity of PKC in membrane. However,an opposite effect of MENK on PKC could be observed at the concentration of 10 μmol/L. The activity of PKC in both of cytoplasm and membrane could obviously be down-regulated by MENK at the concentration of 1 μmol/L and the antibody could reverse the effects. MENK could reduce the level of IP3 in cells and the effect could be antagonized by the antibody. Conclusion: It might be appraised that MENK could modulate the function of cells via various signal transduction systems and therefore result in different biological effects.
