Abstract:A parallel process for the new synthesis of 2,7-dimethyl-2,4,6-octatriene-1,8-dial was investigated.3-(5,5-Dimethyl-1,3-dioxan-2-yl)-2-butenal was obtained by a process involving aldehyde protection,ester exchange and catalytic oxidation from 4-acetoxy-2-methyl-2-butenal.A new catalytic oxidation process was proposed to get 3-(5,5-dimethyl-1,3-dioxan-2-yl)-2-butenal by oxidating 3-(5,5-dimethyl-1,3-dioxan-2-yl)-2-buten-1-ol with copper(Ⅰ) chloride、2,2,6,6-tetramethyl-1-piperidinyloxy and oxygen.3-(5,5-Dimethyl-1,3-dioxan-2-yl)-2-buten-1-ol was chlorizated with SOCl_2 in DMF to get 2-(3-chloro-1-methyl-1-propenyl)-5,5-dimethyl-1,3-dioxane.2,7-Dimethyl-2,4,6-octatriene-1,8-dial was acquired in an overall yield of 48% by one-pot synthesis method from 3-(5,5-dimethyl-1,3-dioxan-2-yl)-2-butenal,2-(3-chloro-1-methyl-1-propenyl)-5,5-dimethyl-1,3-dioxane and triethylphosphite.

Abstract:Derivatives of podocarpane-type diterpenoids, including cassane-type, abietane-type and totarane-type diterpenoids, are either widely distributed natural products or common intermediates in synthetic and medicinal chemistry. Although unmasked ortho-benzoquinone has been used in [4+2] cycloadditions in sparse cases due to its multiple reactivity as diene, dienophile, heterodiene and heterodienophile, applications of this motif in cycloadditions of complex molecules are very rare. We report herein that this [4+2] process can be successfully extended to various polycyclic substrates, followed by a photo-decarbonylation, to generate versatile derivatives of podocarpane-type diterpenoids. Racemic synthesis of isolophanthins A & B and abietatrien-3β-ol was accomplished on the basis of this methodology.

Abstract:ConspectusTetracyclic diterpenoids (C20) mainly refer to the plant terpenoids bearing biogenetically related carbon skeletons derived from copalyl diphosphates (ent-CPP and syn-CPP). This large family contains over 1600 known members that can be categorized into 11 major structural types. Among them, more than three-quarters share a bridged bicyclo[3.2.1]octane subunit, which is also an important branching point in biosynthesis en route to the other types of bicyclic scaffolds, such as bicyclo[2.2.2]-, bicyclo[3.3.0]-, and tricyclo[3.2.1.0]octanes. Combined with the significance of its stereochemical importance in biological activity, the assembly of the bicyclo[3.2.1]octane skeletons is critical to the success of the whole synthesis blueprint toward tetracyclic diterpenoids. Although a number of inspiring methodologies have been disclosed, general approaches by the incorporation of innovative cascade reactions permitting access to diverse structural types of tetracyclic diterpenoids remain limited and in urgent demand.Because of the long-standing interest in the synthesis of bridged diterpenoids, we have recently developed two complementary types of oxidative dearomatization induced (ODI) cascade approaches to the rapid and efficient construction of bicyclo[3.2.1]octane skeletons. In this Account, we summarize our original synthesis design, methodology development, and the application of these two strategies in tetracyclic diterpenoid synthesis during the past few years in our laboratory.First, we detail our preliminary investigation of the ODI-[5 + 2] cycloaddition/pinacol rearrangement cascade reaction, which showed a wide scope of vinylphenol substrates and led to cyclopentane and cyclohexane-fused bicyclo[3.2.1]octanes in good yields with excellent dr values. Next, we describe the utilization of this ODI-[5 + 2] cascade reaction which resulted in the asymmetric total syntheses of four highly oxygenated ent-kauranoids. The strategy concerning accurate stereochemical control in the ODI-[5 + 2] cycloaddition was then successfully transplanted to the total syntheses of three stemaranoids, thus providing a straightforward and diastereoselective route to C9-ethano-bridged tetracyclic diterpenoids. To access more complex diterpenoid rhodomollanol A, we exploited two additional biomimetic rearrangements, namely, the retro-Dieckmann fragmentation/vinylogous Dieckmann cyclization cascade and the photo-Nazarov cyclization/intramolecular cycloetherification cascade. Taken together with the ODI-[5 + 2] cascade, the asymmetric total synthesis of the target molecule was realized, which shed light on the biogenetic pathway of the unprecedented rhodomollane-type carbon framework. Finally, we describe an ODI-Diels–Alder/Beckwith–Dowd cascade approach as a valuable supplement to the ODI-[5 + 2] cascade for the fabrication of cycloheptane-fused bicyclo[3.2.1]octane skeletons. Its versatility was also demonstrated by the total syntheses of two challenging grayanane diterpenoids. In view of the high functional-group compatibility and scalability, we anticipate that the two novel cascade approaches will find further use in the field of complex natural product synthesis.

Abstract:A new Ti-III-mediated reductive epoxide-opening/ Beckwith-Dowd rearrangement process efficiently assembles the bicyclo[3.2.1]octane framework of highly oxidized grayanane diterpenoids. By incorporation of a Cu(tbs)(2)-catalyzed (tbs=N-tert-butylsalicylaldiminato) intramolecular cyclopropanation, a diastereoselective oxidative dearomatization-induced Diels-Alder cycloaddition and a MeReO3-catalyzed Rubottom oxidation, this approach has enabled the first total syntheses of rhodomolleins XX and XXII in 23 and 22 steps, respectively.

Abstract:An ethanol extract of the powdered twigs of Podocarpus imbricatus afforded 14 new diterpenoids (1–14), which all share an aromatized C ring. These isolates belong to five diterpenoid types that include abietanes (1–3), semperviranes (4–9), totaranes (10–12), a C-17 norabietane (13), and an icetexane (14). Their structures were assigned mainly by analysis of the spectroscopic data, and the absolute configuration of 1 was determined by X-ray crystallography. A biosynthetic pathway for five of the biogenetically related types of diterpenoids was proposed. Abstract:Efficient formal syntheses of (±)-Asterisca-3(15),6-diene, a natural product with a bicyclo[6.3.0]undecane skeleton, and (±)-Pentalenene, a natural product with a tricyclo[6.3.0.04,8]undecane skeleton, have been achieved by using Rh(I)-catalyzed [(5+2)+1] cycloaddition. The [(5+2)+1] reaction provides an expeditious approach to reach the bicyclic cyclooctenone 4, which was quickly transformed (via hydroboration then oxidation) to diketone 14, a key advanced intermediate for the total synthesis of (±)-Asterisca-3(15),6-diene. Through further transformations, 14 was converted to diene 18, an advanced intermediate for the total synthesis of (±)-Pentalenene.

Abstract:A synthesis of chiral dimethylcyclopropane derivatives, useful for the synthesis of casbane, lathyrane, and ingenane-type diterpenoids, from easily available (+)-3-carene is described. The silyl enol ether (8) was derived from (-)-cis-4-caranone (7b), which was obtained from (+)-3-carene (6). Ozonolysis of 8 gave 9a and 10. The right half segment (11b) for a synthesis of crotonitenone (3) was formed from 9a in three steps. The epoxide (12) was isomerized to a mixture of the allylic alcohols (13a) and (14a), which was transformed to the ketone (20) in five steps. Methylation of 20 followed by phosphorylation and reduction gave (+)-cis-4-caranone (23) in about 40% overall yield from 12. The methylester (+)-(32b) and its enantiomer (-)-(34b) were synthesized from (+)-6. Ozonolysis of a mixture of the silyl enol ether (30) and (31) followed by methylation with diazomethane gave 32a, which was hydrolyzed to give the desired (+)-32b. The enantiomer (-)-34b was derived from 32a in five steps.

A highly stereoselective intramolecular Diels–Alder addition was designed as the key step to construct the ring D, and the absolute configurations of natural 16-deacetoxy-12-epi-scalarafuran acetate was supported by the X-ray diffraction analysis of single crystal of corresponding 16-deacetoxy-12-epi-scalarafuran.

In this work, the first total syntheses of two aromatic ent-pimaranes were achieved via a C-ABC construction sequence enabled by chiral auxiliary controlled asymmetric radical polyene cyclization, and the subsequent substrate-controlled stereo-/regio-specific hydroboration of alkene allowed for access to both natural products with C19 oxidation modifications.
