A new method to culturing the human craniopharyngioma cell line

Abstract:Objective To establish cell line of craniopharyngiomas(CP) and to investigate the characteristics of the tumor cell for providing a useful research tool and basing rationale of research on cell level.Methods Human craniopharyngiomas cells were primarily cultured with a special medium in vitro,and the biological characteristics of tumor cells were investigated by cell count and MTT experiment.Results We established the cell line of primary craniopharyngiomas in vitro by the way of digestion culture successfully.Now we got the purified cells and serial subcultivation in cell line.We can see the cells digested by enzyme at the same time if we culture cells by the way of digestion culture,all of them have the morphology of round shape,they became little shuttle shape in the second day.The CP cells have the same shapes approximately,slabstone-like shapes and polygonal shapes.The cell line we found was identified as CP cell line by a series of tests.The growth curve showed that the cell can grow stably,and the doubling time was about 3 days.Conclusion Craniopharyngioma cell line can be cultured with a special medium by enzymatic digestion through the use of fresh tumor tissue in the CP patients.
