Abstract:This is a study to determine 5 main compounds of rhubarb in Rhubarb and Coptidis Xiexin decoctions,using an assay method,in an attempt to compare the concentration of anthraquinones in different compatibility infusions.We developed a HPLC method,using a phenyl column(250mm×4.6mm,5μm)under an ambient temperature,and using gradient elution with mobile phase of methanol-0.01% phosphoric acid and detected at 254 nm,to determine the con- centration of 5 compounds in Rhubarb.The calibration curve of aloe-emodin,rhein,emodin,and chrysophanol indi- cating a good linearity was obtained within the range of 3.15～64.00μg/ml,and 1.70～33.92μg/ml of physcion.The RSD of 5 compounds were less than 2%.The average extraction recovery of the 5 main compounds was 102.1%,with an RSD less than 5%.The compatibility of rhubarb and coptis(scutellaris)has a significant effect on the concentration of anthraquinones.It is concluded that this method has been proved to be accurate and reliable for determining the concentration of anthraquinones in prescriptions containing Radix et rhizoma rhei.The assay results show that an in- creased or reduced scutellaris concentration in Rhubarb and Coptis Xiexin Decoction has a significant effect on the concentration of anthraquinones.

Abstract:In order to study the compatibility of Chinese herbal drugs, a method was proposed by investigating chemical reactions with combination of chemical constituents or major groups of chemical compounds of Chinese herbal drugs. The content variation of chemical constituents during combination of rhizoma coptidis alkaloids with fructus evodiae flavonoids was analyzed by high performance liquid chromatography (HPLC). The experimental results showed that only small changes of the relative peak areas of different chemical constituents were observed and no new peaks were detected. No chemical reactions were observed during the compatibility test of rhizoma coptidis alkaloids with fructus evodiae flavonoids.
Determination of Multiple Active Constituents in Da-Huang-Xiao-Shi Decoction Using HPLC-LTQ-Orbitrap Mass Spectrometry: Application in Comparing the Differences in the Formula and Its Constituent Herbs.

Abstract:Da-Huang-Xiao-Shi decoction (DHXSD) is a traditional Chinese medicine formula and is used to treat cholestasis. In this study, we developed a reliable and comprehensive HPLC coupled with a linear ion trap-Orbitrap mass spectrometry method for the separation and determination of 21 components including six alkaloids, five anthraquinones, three tannins, three terpenes, two iridoid glycosides, one organic acid and one flavonoid in DHXSD. A C18 column was eluted using a gradient mobile phase at a flow rate of 1 ml/min. Detection was operated with an electrospray ionization source in positive and negative ion modes using selective ion monitoring. The calibration curves for all analytes showed good linearity (r > 0.9901), and the inter- and intra-day precision did not exceed 4.98%. The recovery, repeatability and stability were also within the acceptable limits. The method was successfully applied to determine multiple active constituents in DHXSD and its constituent herbs. Compared with Da Huang, the total contents of the five anthraquinones were significantly higher in DHXSD. However, the changes in the components from Zhi Zi/Huang Bo were complicated in DHXSD. The study could serve as a fundamental reference for establishing comprehensive DHXSD quality control measures and be helpful to understand some compatibility laws of DHXSD.
Simultaneous Chemical Fingerprinting and Quantitative Analysis of Crude and Processed Radix Scrophulariae from Different Locations in China by HPLC.

Abstract:A validated liquid chromatography method was first developed to evaluate the quality of crude and processed Radix Scrophulariae extracts through establishing chromatographic fingerprint and simultaneous determination of five bioactive compounds, namely 5-hydroxymethylfurfural (5-HMF), acteoside, angroside C, harpagoside and cinnamic acid. The chromatographic were separated on an Agilent Zorbax Extend C(18) column (250 mm × 4.6 mm, 5 μm) and detected by diode array detector (DAD). Mobile phase was composed of (A) aqueous phosphoric acid (0.03%, v/v) and (B) acetonitrile using a gradient elution. Analytes were performed at 30 °C with a flow rate of 1.0 mL/min and UV detection at 280 nm. All calibration curves showed good linear regression (r(2) ≥0.9996) within the tested ranges, and the recovery of the method was in the range of 98.12-103.38%, with RSD values ranging from 0.6 to 2.8%. In addition, the contents of those five bioactive compounds in crude and processed Radix Scrophulariae prepared by different locations of China were determined to establish the effectiveness of the method. The results demonstrate that the developed method is accurate and reproducible and could be readily utilized as a suitable quality control method for the quantification of Radix Scrophulariae.
Pharmacokinetics of Anthraquinones in Xiexin Decoction and in Different Combinations of Its Constituent Herbs

Twenty rats were randomly divided into four groups and were administered one of the four extracts: DH, DH-HQ, DH-HL and XXD (DH-HQ-HL) via intragastric gavage. Anthraquinone concentrations in plasma were determined by an HPLC technique. Pharmacokinetic parameters were calculated from the plasma concentration-time data. Compared with DH alone, the DH-HL combination decreased C-max of all five anthraquinones and A UC of four anthraquinones (except physcion), and the DH-HQ combination decreased AUC of aloe-emodin and C-max of rhein. Finally, XXD increased AUC of all five anthraquinones compared with DH-HL combination. These results showed that the oral bioavailabilities of five anthraquinones were decreased significantly by combining DH with HL, whereas HQ weakened the effect of HL that inhibited the absorption of anthraquinones. Compared with DH alone, the DH-HL combination showed maximum plasma concentration decreased (Cmax) and area under curve (AUC) for the values anthraquinones, and a prolonged eliminatun half life (T1/2) for rhein, while the DH-HQ combination showed a decrease Cmax for rein and a prolonged T1/2 for aloe-emodin and physcion. Finally, XXD (DH-HQ-HL) administration resulted in an increased AUC for all five anthraquinones compared to DH-HL, and increased the total of AUC for rhein, emodin, chrysophanol and physcion compared to DH alone. These results showed that the oral bioavailability of the five anthraquinones was significantly decreased by combining DH with HL, whereas HQ increased the amounts of absorbed anthraquinones (except for aloe-emodin), and weakened the effect of HL which inhibited the absorption of anthraquinones.
Identification and Determination of Active Anthraquinones in Chinese Teas by Micellar Electrokinetic Capillary Chromatography

Abstract:The simultaneous determination of emodin, aloe-emodin and rhein, in Semen Cassiae, Qinghai Wild Dahuang tea, Ningxia Juemingzi tea, Lanzhou Juemingzi tea and Surong Juerningzi tea, has been investigated by micellar electrokinetic capillary chromatography for the first time. With an electrolyte containing 15 mm borax, 30 mm SDS, 10% (v/v) ethanol, at pH 9.60 and 20 kV applied voltage, the three analytes were completely separated within 12 min. The effects of the concentration of borax, electrolyte pH, the concentrations of SDS and organic modifier and the applied voltage on electropheoretic behavior and separation were studied. The linear calibration range was 4-120 mug/mL (r = 0.9921) for emodin, 10-200 mug/mL (r = 0.9970) for aloe-emodin and 2-100 mug/mL (r = 0.9971) for rhein, respectively. Under the optimum conditions, the relative standard deviation (RSD) values (n = 6) of the migration time and the peak area of each peak was 0.59-0.80% and 1.30-3.22%, respectively. The contents of the analytes in Rheum, Qinghai Wild Dahuang tea, Semen Cassiae and three other kinds of teas were easily determined with recoveries ranging from 95.3 to 104.4%. Abstract:Xiexin decoction (XXD) is composed of Rhei Rhizoma (DH), Radix Scutellaria (HQ), and Coptis Chinensis (HL). Free anthraquinones in DH are the basic effective constituents in XXD. Reportedly, HL decreases the bioavailability of the anthraquinones, while HQ antagonizes the effect of HL. In this study, we aimed to determine the underlying mechanisms. The metabolisms of anthraquinones by intestinal flora were studied using rat fecal suspension (RFS); the metabolisms of rhein (a typical anthraquinone in DH) by rat intestine and liver were studied using rat intestine microsomes (RIMs) and rat liver microsomes (RLMs), respectively; the intestinal transport of rhein was studied using everted gut sacs. The results showed that HL decreased the amount of the free anthraquinones after incubation in RFS and inhibited the intestinal transport of rhein, but HQ antagonized the effect of HL. On the other hand, HQ strongly inhibited the glucuronidation of rhein in both RIMs and RLMs. The results suggested that HL decreased the oral bioavailability of the anthraquinones due to inhibiting the conversion of conjugated anthraquinones to free anthraquinones by intestinal flora and decreasing the intestinal transport of the anthraquinones; HQ confronted the effect of HL by inhibiting the glucuronidation of the anthraquinones in intestine and weakening the inhibitory effects of HL.

Abstract:A simple, reliable method for simultaneous determination of the ten components in Danhong injection (DHI) in combination of chromatographic fingerprint analysis was developed by high performance liquid chromatography coupled with diode-array-detector (HPLC-DAD). The separation was performed on an Agilent Zorbax SB-C18 column with a linear gradient elution of acetonitrile and 0.5% formic acid. The method was validated by linearity, precision, stability and recovery. The developed method was subsequently applied to evaluate 15 batches of DHI and testified to be suitable for its quality control.
[Simultaneous Determination of 4 Anthraquinones in Kuhuang Injection by RP-HPLC].

Abstract:OBJECTIVE:To develop a new high-performance liquid chromatographic method for simultaneous quantitative determination of 4 anthraquinones in Kuhuang injection.METHOD:The chromatographic separation was performed on a Lichrospher C18 column (4.6 mm x 250 mm, 5 microm,), A linear gradient elution of A (CH3COOH: H2O = 1:100) and B (CH3COOH: H2O: CH3CN = 1:20:80) was used. The flow rate was 0.8 mL x min(-1) and column temperature was set at 35 degrees C. The UV detection wavelength was set at 254 nm.RESULT:The recoveries of rhein, emodin, chrysophanol and physcion were 98.9%, 100.5%, 102.5% and 99.0%, respectively. Their linear ranges were 0.0875-1.75 microg for rhein, 0.0825-1.65 microg for emodin, 0.159-3.17 microg for chrysophanol and 0.0525-1.05 microg for physcion, respectively.CONCLUSION:The method was accurate, repeatable and suitable to determine the contents of 4 anthraquinones in Kuhuang injection.

Abstract:A sensitive and reliable ultra high performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry method was established to separate and identify the chemical constituents of Zhi-Zi-Da-Huang decoction, a classic traditional Chinese medicine formula. The chromatographic separation was achieved on a Shim-pack XR-ODS C18 column (75 × 3.0 mm, 2.2 μm) using a gradient elution program. The detection was performed on a Waters Xevo G2 Q-TOF mass spectrometer equipped with electrospray ionization source in both positive and negative modes. With the optimized conditions, a total of 82 compounds were identified or tentatively characterized. Of the 82 compounds, 21 compounds were identified by comparing the retention time and MS data with reference standards, the rest were characterized by analyzing MS data and retrieving the reference literature. In addition, 31 compounds were identified from Gardenia jasminoides Ellis, ten compounds were identified from Rheum palmatum L., 33 compounds were identified from Citrus aurantium L., and eight compounds were identified from Sojae Semen Praeparatum. Results indicated that iridoids, anthraquinones, flavonoids, isoflavonoids, coumarins, glycosides of crocetin, monoterpenoids, and organic acids were major constituents in Zhi-Zi-Da-Huang decoction. It is concluded that the developed ultra high performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry method with high sensitivity and resolution is suitable for identifying and characterizing the chemical constituents of Zhi-Zi-Da-Huang decoction, and the analysis provides a helpful chemical basis for further research on Zhi-Zi-Da-Huang decoction.

DECOCTION PROCESS OPTIMIZATION AND QUALITY EVALUATION OF YI-HUANG DECOCTION BY HPLC FINGERPRINT ANALYSIS.

Abstract:Yi Huang decoction (YHD) has been used as one of famous traditional formula because of its unique effectiveness against gynecological diseases. YHD is composed of five herbs, including the rootstock of Dioscoma opposita Thunb. (Dioscoreaceae), the kernel of Etayale ferx Salisb. (Nymphaeaceae), the bark of Phellodendron chinense Schneid. (Rutaceae), the seed of Plantago asiatica L. (Plantaginaceae), and the seed of Ginkgo biloba L. (Ginkgoaceae). To effectively control the quality, the processing method for YHD was optimized by means of single factor test as well as orthogonal test in this study. A completely validated method based on HPLC coupled with diode array detector was performed on a Kromasil C(18) column at 30° with mobile phase of 0.1% aqueous phosphoric acid and acetonitrile. As a result, HPLC fingerprint on the basis of the chromatographic data from 32 batches of samples was obtained, which contained 44 common peaks. Among these common peaks, 6 peaks were identified as geniposidic acid, berberine hydrochloride, palmatine hydrochloride, phellodendrine chloride, magnoflorine, and verbascoside, respectively, based on their retention time relative to the standards. Meanwhile, the contents of these 6 compounds were also simultaneously examined. In sum, this study offered valuable information for the proper processing and quality control for YHD.
Establishment of Method for Analysis of Decoction Huanglianjiedutang by HPLC

Abstract:The decoction Huanglianjiedutang consists of four medicinal plants, i.e. Coptis japonica Makino, Scutellaria baricaleins Georgi, Phellodendron amuranse Ruprecht and Gardenia jasminonoides Ellis and its constituents mainly include three groups of flavonoids, alkaloid and iridoid glycosides with great difference of chemico-physical properties, which assume different mobile phases when analyzed by HPLC respectively. This article describes via comparison the influence of formi acid-triethylamine mobile phase additive with different concentrations on the analysis of the aqueous extract of the decoction Huanglianjiedutang by HPLC, thereby having developed a stable and repetitive method of HPLC analysis, in which the analysis is composed of a chromatographic column Hypersil ODS2(250 × 4.6mm i.d, 5 um particle size), a mobile phase consisting of the aqueous solution of 0.3%(v/v) formic acid-0.3%(v/v) triethylamine and the acetonitrile solution of 0.3%(v/v) formic acid-0.3(v/v) triethylamine, and an ultraviolet detector with the wavelength 260nm. This method can be used to analyze alkaloids, flavonoids and iridoid glycosides of TCM compound drugs at the same time, thas providing a refernce for the analysis of TCM extracts with complex constituents.
[Compatibility chemistry of acid-alkaline pair medicine of Dahuang and Huangbai in Dahuang Xiaoshi decoction]

Abstract:Objective: To determine the change pattern of alkaline and acid components of Dahuang and Huangbai amoung different combinations in Dahuang Xiaoshi decotion. Method: The contents of anthraquinones and berberine were determined by HPLC in samples of Dahuang extracts, Huangbai extracts, Dahuang and Huangbai pair medicine extracts, and extracts of whole recipe. Result: The contents of anthraquinones and berberine were decreased with the changes of combinations: from component medicinal material alone to pair medicines, and to whole recipe. the change pattern was more apparent in water decoctions than in ethanol extracts. Conclusion: The contents of alkaline and acid components are changed with different extract methods and different combinations.

Abstract:The main active ingredients, ferulic acid, vanillin, and three senkyunolides in Chinese traditional herb medicine, Chuanxiong, were separated and identified by HPLC, HPLC-MS, and EI-MS. Tetramethylpyrazine (TMP), which was reported as the major active ingredient in Chuanxiong, could not be detected by HPLC in the present paper.Although TMP is the major active ingredient of Chuanxiong, its content was below 1.2x10(-7)g/g, which is less than the lower detection limit by HPLC.
Quantitative Analysis of Six Phenolic Acids in Artemisia capillaris (Yinchen) by HPLC-DAD and Their Transformation Pathways in Decoction Preparation Process

Abstract:We aimed to establish a quantitative analysis method of six constituents (5-caffeoylquinic acid, 3-caffeoylquinic acid, 4-caffeoylquinic acid, 1,3-dicaffeoylquinic acid, 3,4-dicaffeoylquinic acid, and 4,5-dicaffeoylquinic acid) in Artemisia capillaris (Yinchen) and its decoction by using HPLC coupled with DAD. Besides, the transformation paths of the six constituents were analyzed in decoction preparation processing. The analytical method was fully validated in terms of linearity, sensitivity, precision, repeatability, and recovery and applied to assess the transformation trend and quantitative analysis of the six constituents in Yinchen decoction. The contents of six constituents varied greatly in Yinchen herb and Yinchen decoction, and there were inextricable internal relationships between them. Presumably 3-caffeoylquinic acid was isomerized to generate 5-caffeoylquinic acid and 4-caffeoylquinic acid. Similarly, 1,3-dicaffeoylquinic acid and 3,4-dicaffeoylquinic acid were produced by isomerization of 4,5-dicaffeoylquinic acid. In conclusion, this study provides a chemical basis for quality control of Yinchen decoction, and the changes of selected markers in decoction could give us some novel perspectives to study the relationship between substances and drug efficacy.

Abstract:An HPLC/DAD/ESI/MS method was established for the qualitative analysis of the major constituents in Shi-Quan-Da-Bu decoction, a traditional Chinese medicinal formula. Based on the chromatographic separation of most of the constituents of Shi-Quan-Da-Bu decoction on a Zorbax SB-C18 column with water-acetonitrile as the mobile phase, more than 150 components were detected by LC-MS. Among them, 36 major components, including flavonoids, triterpenoid saponins, iridoid glycosides, phathalides and phenylpropionic acids were either identified or tentatively characterized. Twenty one compounds were identified by ESI-MS and comparison of their retention times with those of reference standards. The other 15 compounds were deduced according to their MS and UV spectra and retention behaviors by comparing the results with those reported in the literature. Most of these compounds were derived mainly from Paeonia lactiflora, Glycyrrhiza uralensis, Panax ginseng, Ligusticum chuanxiong, Angelica sinensis and Astragalus membranaceus.

Abstract:Da-Huang-Xiao-Shi decoction (DHXSD), a traditional Chinese medicinal formula, has been used mainly to treat jaundice for more than 1700 years in China. In this study, we developed a rapid, sensitive, and accurate LC-MS/MS method to simultaneously determine multiple, potentially bioactive compounds of DHXSD, including five alkaloids (berberine, phellodendrine, palmatine, jatrorrhizine, and magnoflorine), five anthraquinones (rhein, aloe-emodin, emodin, chrysophanol, and physcion), two iridoid glycosides (geniposide and genipin 1-gentiobioside), and one iridoid aglycone (genipin) in rat plasma. Plasma samples collected from rats were treated immediately with 5% acetic acid to avoid the degradation of genipin. After protein precipitation with acetonitrile containing 5% acetic acid, the compounds were reconstituted in acetonitrile-water (50:50, v/v) solution containing 6.5% formic acid and separated on the ACQUIT (TM) UPLC BEH C-18 column (2.1 x 100 mm; 1.7 mu m) using a mobile phase composed of 2 mM ammonium formate in water (solvent A) and acetonitrile (solvent B) at a flow rate of 0.3 mL/min. Quantitation was performed on a Triple Quand 5500 tandem mass spectrometer coupled with an electrospray ionization (ESI) source. Multiple reaction monitoring (MRM) was used to quantify compounds in positive and negative ion modes. The method validation results showed that the specificity, linearity, precision and accuracy, recovery, matrix effect, and stability of the 13 compounds met the requirements for their quantitation in biological samples. This newly established method was successfully used in a pharmacokinetic study on rats orally treated with DHXSD. Besides, glucuronide and sulfate metabolites were also determined in rat plasma after hydrolysis. This is the first method developed for the simultaneous quantification of multiple compounds of DHXSD in vivo. Our study provides relevant information on the pharmacokinetics of DHXSD and the relationship between the compounds of DHXSD and their therapeutic effects. (C) 2019 Elsevier B.V. All rights reserved.

Abstract:Objective:To establish an HPLC method for the determination of xanthotoxol,bergapten,imperatorin and isoimperatorin in Angelicae Dahuricae Radix from different sources.Method:A Zorbax SB-C_(18)(150 mm×4.6 mm,5μm) column was used with the mobile phase consisted of acetonitrile-water with gradient elution.The detection wavelength was 300 nm and the flow rate was 1.0 ml·min~(-1).Result:The calibration curve of xanthotoxol,bergapten,imperatorin and isoimperatorin were in good linearity within certain ranges and the average recoveries were also good.Conclusion:The method is simple,accurate and repeatable and can be used for the determination of xanthotoxol,bergapten,imperatorin and isoimperatorin in Angelicae Dahuricae.
A comparison of the chemical compounds in single- and co-boiled Zhi-Zi-Da-Huang decoctions with UPLC-qTOF-MS/MS and UPLC-DAD

Abstract:Abstract Background Zhi-Zi-Da-Huang decoction (ZZDHD) is a famous Traditional Chinese Medicine decoction due to its therapeutic effects on clinical hepatobiliary disorders. ZZDHD is composed of Gardeniae Fructus, Rhei Radix et Rhizoma, Fructus Aurantii Immaturus, and Sojae Semen Praeparatum. With the development of current technology, dispensing granules have been widely used for convenience. However, limited research has been conducted to determine differences in the chemical compounds between dispensing granules and traditional decoction. Methods A strategy based on UPLC-qTOF-MS/MS and UPLC-DAD was established to quantitatively and qualitatively analyze the chemical compounds present in single- and co-boiled ZZDHD. First, we utilized UPLC-qTOF-MS/MS to identify the compounds in single- and co-boiled ZZDHD. Then, 15 compounds were quantitatively analyzed in ZZDHD by UPLC-DAD. Finally, fingerprint and chemometric analyses were adopted to evaluate the difference between single- and co-boiled ZZDHD. Results One hundred and forty-seven compounds were authenticated or determined according to their retention behaviors, mass mensuration, and characteristic fragment ions. In addition, 15 analytes of single- and co-boiled ZZDHD indicated a significant difference in the content of many compounds, including geniposide (G2), emodin, chrysophanol, daidzin, and crocin-I. The similarities of single and co-boiled ZZDHD were higher than 0.99, as evaluated by fingerprinting; 30 common peaks were evaluated by the chemometric analysis, which found 13 important variable values (VIP > 1) including C3, A3, crocetin, daidzin, C5, physcion, G2, genipin-1-O- β -D-gentiobioside (G1), chrysophanol, emodin, A2, A5, and C6. Conclusions This study provided an experimental basis for explaining the rationality of single-and co-boiled ZZDHD, and a study model for comparing the chemical substances between traditional decoctions and dispensing granules.
