Abstract:Fish is one of eight foods that can cause severe allergic reactions. The growing consumption of fish and fish derivatives contributes to an increase in the incidence of fish allergies. Distinct regional variations in fish allergies are evident with higher prevalence rates observed in coastal countries, which may be related to the high consumption of local fish in these regions. The diagnostic methods of fish allergies include in vivo and in vitro detection, and the skin prick test is one of the most common screening methods. To date, several fish allergens have been identified, including parvalbumin, enolase, aldolase, collagen, and other allergens. Despite the homologous structure of parvalbumin from different fish species, variations in the IgE epitope may be one of the reasons leading to the complexity of fish allergies. To further understand the mechanisms of fish allergies, this review summarizes the recent literature and focuses on the clinical characteristics, mechanism, allergens, epitopes, and cross-reactivity.

Abstract:This paper is a brief review of major seafood allergens and their properties. According to the analysis of current situation, we bring forward some major problems and difficulties of seafood allergenicity. In the end, developmental ways of hypoallergenic seafood products were given and wish to offer some suggestions to seafood processing.
Detection and analysis of the binding activities of Ig E from allergic patients to sarcoplasmic protein of different kinds of fishes

Abstract:Sera from 80 fish- allergic and only shrimp and crab- allergic patients were collected in Zhejiang region,and the binding activities of Ig E in patients sera to sarcoplasmic proteins from 7 species of fish were examined by enzyme- linked immunosorbent assay( ELISA). The protein composition and allergen distribution of fish were analyzed by SDS- PAGE and Western blot. Results demonstrated that there was no significant difference in specific Ig E positive rate between fish allergic and only shrimp and crab- allergic patients. But allergic patients’ sera had different positive rate in reaction with different kinds of fish proteins. The proteins of Oncorhynchus mykiss,Scophthalmus maximus and Micropterus salmoides were more allergenic with the positive rate of 70. 67%,46. 67% and 26. 39%,respectively. In constrast,Aristichthys nobilis,Cyprinus carpio,Ctenopharyngodon idellus and Acipenser sinensis had relative-ly low positive rate. SDS- PAGE analysis showed that the composition of protein was different in 7 fish species. Western blot results showed that the components of fish species had various reaction with positive sera. Thereinto,three species of fish( Oncorhynchus mykiss,Scophthalmus maximus and Micropterus salmoides),which was higher positive rate,were detected about 12 k Da specific protein bands. In summary,the study was detected and analyzed the specific Ig E binding capacity in sera from fish- allergic and shrimp and crab- allergic patients to sarcoplasmic protein of varieties of fish. These provided some references for the diseases prevention of aquatic products and the development of hypoallergenic aquatic food.

Abstract:China is a large country both in area and population size, with diverse landscapes, climates and environments, and its domestic economy, international trade and tourism is rapidly growing. Under such conditions and in line with the above growths, the occurrence of allergy problems is unexpectedly increasing. A scarcity of public, governmental and food-industry knowledge and awareness of allergies and allergenic sources in China may cause a serious burden to the national population as well as to foreign consumers. Numerous allergenic sources have been reported in China, most of which are however only available in Chinese literature and have not yet been systematically reviewed and summarized in English. Here, we present an overview of the most relevant allergen types as well as their sources and the most prominent examples in China.

Abstract:People eat many varieties of food to satiate their hunger. Among them, a few numbers of food cause overreaction of the body's immune system, and fish holds a permanent position on that list. Processing methods, including one treatment or a combination, can have different effects on the allergenic potential of food proteins. An important point to note, however, is that not all of these methods can eliminate the potential for protein allergy. Thus, it is essential to understand the risk involved with the consumption of processed fish and its derivatives. Fish could be prepared in various ways before come to the dining plate. It has shown some of these methods can effectively manipulate the allergenicity owing to the alterations occurred in the protein conformation. This article provides an overview of the impact of fish processing methods (thermal and non-thermal) on the allergenic potential of fish along with possible causative structural modification provokes allergen stability. The article begins with current trends related to fish consumption, proceeds with the prevalence and underlying mechanism of fish allergy. Properties of clinically relevant fish proteins, projected IgE epitopes of PV, cross-reactivity of fish allergens are also addressed in this context to understand and compare the behavioral patterns of PV profiles of different species on processing methods.

Abstract:Qingdao is a coastal city of China where the problem of fish allergy is increasingly prominent.There have been no data providing a comprehensive picture of the major fish allergens to local people so far.Based on the above situation,the aim of this research was to study on the cross-reactivity of different fish allergens,to provide primary basis of safe eating guide for local fish allergic individuals.To determine the allergens of 7 commonly edible fishes:Alaska pollock(Theragra chatcogramma),red snapper(Pagrosomus major),salmon(Salmo salar),spanish mackerel(Scomberomrus niphonius),turbot(Scophthatmus maximus),large yellow croaker(Larimichthys crocea) and carp(Cyprinus carpio),SDS-PAGE was applied to observe the protein components of fish extracts.Sera from 18 fish allergic patients were used in Western-blot.Cross-reactivity was investigated by IgG Western-blot,IgG ELISA and ELISA inhibition used rabbit anti-spanish mackerel parvalbumin polyclonal antibody.Results showed that the major fish allergens were 48-57,33-41 ku,28 ku and 17 ku proteins to Qingdao residents,which was distinct from the common consensus that parvalbumin was the major allergen in fish.Meanwhile,the study on the cross-reactivity of fish parvalbumin also indicated there was a quite strong cross-reactivity between different species of fish proteins.Therefore,despite the allergens might be different from each other,it was suggested that the subjects not eat other 6 fishes if allergic to one of them.

Abstract:The prevalence of allergic diseases has increased dramatically in recent years in China, affecting the quality of life in 40% of the population. The identification of allergens is the key to the diagnosis of allergic diseases. Presently, several methods of allergy diagnostics are available in China, but they have not been standardized. Additionally, cross-sensitization and co-sensitization make allergy diagnostics even more complicated. Based on 4 aspects of allergic disease (mechanism, diagnosis procedures, allergen detection in vivo and in vitro as well as the distribution map of the most important airborne allergens in China) and by referring to the consensus of the European Society of Allergy and Clinical Immunology, the World Allergy Organization, and the important literature on allergy diagnostics in China in recent years, we drafted this consensus of allergy diagnostics with Chinese characteristics. It aims to standardize the diagnostic methods of allergens and provides a reference for health care givers. The current document was prepared by a panel of experts from the main stream of professional allergy associations in China.

Abstract:High hydrostatic pressure (HHP), a novel non-thermal processing technology, can inactivate microorganisms in food with ultra-high pressure over 100 MPa. In recent years, it has shown unique potential in alleviating seafood allergenicity. Seafood, as a primary high-quality protein source, is one of popular food products in many human populations, while seafood allergy remains an obstacle to the consumption of seafood and calls for processing raw materials to reduce their allergenicity. Heating and fermentation as conventional methods, along with HHP as a rising novel technology, have been applied in seafood processing, such as shrimp and squid. This review provides a brief introduction of current key publications and limitations of researches on seafood allergy. In addition, characteristics and principles, processing parameters and effects of HHP treatment on seafood of current researches were detailed. Our main goal was to support readers to keep abreast with knowledge on seafood allergy and provide new insights of using HHP for seafood processing to achieve lower allergenicity.

Abstract:<span>The increasing number of people with seafood allergy has caused a series of problems for practitioners and consumers in the seafood industry year by year. Thereby, development of efficient, convenient and low-cost allergen detection methods is urgently needed. This review introduces three important existing seafood allergen detection methods associated with DNA-based, protein-based and aptamer-based. Their principles and biological characteristics are firstly presented. The core of these three methods are DNA amplification techniques, specific binding of antigens and antibodies, and specific binding of aptamers and ligands, respectively. Among them, DNA-based detection method is an indirect analysis, which takes the gene of allergen as the detection object and is characterized by good stability and high sensitivity. Protein-based and aptamer-based, methods employ indirect analysis for allergen detection. The difference is that the latter uses an easily synthesized and highly efficient aptamer as the detection probe, showing great promising potentials. The advantages and disadvantages of the three mentioned detection methods are also discussed. In the future, as more efficient and reliable detection methods for seafood allergens come into practice, the possibility of seafood allergy patients eating seafood products by mistake will be greatly reduced, which will ensure the food safety and the health of allergy patients.</span>

Food-Pollen Cross-Reactivity and its Molecular Diagnosis in China

Abstract:Purpose of review: Plant-derived foods are one of the most common causative sources of food allergy in China, with a significant relationship to pollinosis. This review aims to provide a comprehensive overview of this food-pollen allergy syndrome and its molecular allergen diagnosis to better understand the cross-reactive basis. Recent findings: Food-pollen cross-reactivity has been mainly reported in Northern China, Artemisia pollen is the major related inhalant source, followed by tree pollen (Betula), while grass pollen plays a minor role. Pollen allergy is relatively low in Southern China, with allergies to grass pollen being more important than weed and tree pollens. Rosaceae fruits and legume seeds stand out as major related allergenic foods. Non-specific lipid transfer protein (nsLTP) has been found to be the most clinically relevant cross-reacting allergenic component, able to induce severe reactions. PR-10, profilin, defensin, chitinase, and gibberellin-regulated proteins are other important cross-reactive allergen molecules. Artemisia pollen can induce allergenic cross-reactions with a wide range of plant-derived foods in China, and spring tree pollens (Betula) are also important. nsLTP found in both pollen and plant-derived food is considered the most significant allergen in food pollen cross-reactivity. Component-resolved diagnosis with potential allergenic proteins is recommended to improve diagnostic accuracy and predict the potential risk of causing allergic symptoms.

Abstract:Introduction: Adverse reactions to food (ARF) are a major worldwide public health and food safety problem. Among the various causes of ARF, food allergies (FA) are particularly serious as the immune response that is triggered can be fatal even at very low doses. However, the prevalence of ARF and FA in the general population in China is presently unclear. This study aims to determine the epidemiological characteristics and risk factors for ARF and FA, which can be a basis for estimating thresholds for major food allergens. Methods: This is a multicenter, cross-sectional, epidemiologic survey with a case-control study nested among a selected population in China. Random individuals were recruited using stratified cluster random sampling; ARF and FA were comprehensively assessed using modified EuroPrevall FA Project questionnaires as well as structured interviews, sensitization testing, and double-blind placebocontrolled food challenges (DBPCFC). Results: This method of epidemiologic study on ARF and FA was a pilot application in Jiangxi Province from January 2020; among the total 21,273 children and adults that completed the questionnaire, 5.8% reported ARF and 4.3% reported FA. ARF were determined to be associated with age, gender, and region. Animal-derived foods were the dominant offending foods, especially shrimp, and skin symptoms were the most commonly reported ARF. Discussion: This is the first multi-center, largescale, epidemiologic study on ARF and FA using standardized methods, including DBPCFC, in the Chinese general population. This study presents an important approach to assessing ARF and FA, provides significant insights about the prevalence of ARF and FA, and facilitates support for updating the list of allergenic food labels: which will be essential for improving ARF prevention and management in China.

Abstract:Aquatic product allergy is an important food safety problem. As a new green food processing method, non-thermal processing technology is gradually applied to the research field of reducing the allergenicity of aquatic products. Therefore, this paper briefly introduces food allergy, and summarizes the non-thermal processing technologies such as irradiation, ultrasound, ultra-high pressure, low-temperature plasma, glycosylation treatment and enzymatic hydrolysis to reduce the allergenicity of aquatic products and the allergenicity evaluation methods of aquatic products such as serology, simulated digestion, cell experiment and animal experiment, so as to provide reference and guidance for the development of aquatic products with low allergenicity.

Abstract:Background: Food allergy (FA) is an immune system reaction that occurs soon after eating certain food, which severely damages the health of allergic patients. Since medical approaches for FA may trigger severe adverse effects, the natural substances with anti-allergy functions have received more and more attention.Scope and approach: This paper reviewed the natural bioactive substances with anti-allergic properties in food allergy, which mainly include polyphenols, polysaccharides, oligosaccharides, traditional Chinese medicine (TCM) and probiotics. Moreover, this paper analyzed the limitations of current investigations and provided future outlook for the development of anti-allergic products.Key findings and conclusions: This paper extensively reviews the natural substances with anti-allergy functions. As the results, the polyphenols could inhibit the formation of allergen-IgE complex, influence the allergen-IgE complex bind to the receptor (Fc epsilon RI) on the surface of effector cells (e.g. mast cells and basophils). The poly-saccharides and oligosaccharides could inhibit the secretion of allergic mediators and cytokines, suppressing Th2 responses and promoting Th1/Treg responses. The TCM could reduce allergen presentation to dendritic cell, weaken the secretion of allergen-specific IgE and the degranulation of mast cell, upregulate Th1 cytokines (e.g. IL-12, IFN-gamma) and downregulate Th2 cytokines (e.g. IL-4, IL-13). The probiotics could help maintain the integrity of intestinal epithelial barriers by promoting mucus production via goblet cells, which could reduce the intestinal permeability and improved mucus generation to decrease the risk of food allergen into immune systems. This review could provide a better understanding and future outlook on the natural anti-allergy substances and supply a novel insight into their potential application for FA prevention and treatment.

Abstract:Seafoods are fashionable delicacies with high nutritional values and culinary properties, while seafood belongs to worldwide common food allergens. In recent years, many seafood allergens have been identified, while the diversity of various seafood species give a great challenge in identifying and characterizing seafood allergens, mapping IgE-binding epitopes and allergen immunotherapy development, which are critical for allergy diagnostics and immunotherapy treatments. This paper reviewed the recent progress on seafood (fish, crustacean, and mollusk) allergens, IgE-binding epitopes and allergen immunotherapy for seafood allergy. In recent years, many newly identified seafood allergens were reported, this work concluded the current situation of seafood allergen identification and designation by the World Health Organization (WHO)/International Union of Immunological Societies (IUIS) Allergen Nomenclature Sub-Committee. Moreover, this review represented the recent advances in identifying the IgE-binding epitopes of seafood allergens, which were helpful to the diagnosis, prevention and treatment for seafood allergy. Furthermore, the allergen immunotherapy could alleviate seafood allergy and provide promising approaches for seafood allergy treatment. This review represents the recent advances and future outlook on seafood allergen identification, IgE-binding epitope mapping and allergen immunotherapy strategies for seafood allergy prevention and treatment.

Abstract:This review highlights significant regional variations in food allergy prevalence across China, with higher rates in economically developed coastal areas. Despite this growing burden, challenges such as limited diagnostic tools, a shortage of allergists and inadequate access to epinephrine autoinjectors hinder effective management of food allergies nationwide. The prevalence of food allergies in China seems to be increasing, but there are limited studies describing the pattern of food allergies across the country. This review highlights regional variations observed across China, with data indicating a higher prevalence in the more economically developed eastern and southern coastal regions compared to inland areas. Egg and milk are the most common allergies among children under 3 years old; for children above 3 years old, specific food allergens also show regional differences, with shellfish allergies being more common in southern and eastern coastal areas, while wheat and fruit allergies are more prevalent in northern regions. Emerging peanut and tree nut allergies have also been observed in China's megacities, although the prevalence remains relatively low compared to Western countries. These geographic and environmental influences highlight the complexity of the food allergy landscape in China and the need for a more nuanced understanding of the underlying drivers. Despite the growing burden of food allergies, significant gaps exist in effectively managing these conditions in China. Lack of standardised diagnostic tools, limited access to oral food challenges and a shortage of trained allergists pose major challenges. Another critical gap is the limited availability and affordability of epinephrine autoinjectors, essential for managing life‐threatening anaphylactic reactions. Addressing these systemic deficiencies in China's food allergy management infrastructure will require concerted efforts from policymakers, healthcare systems and pharmaceutical companies. Investing in the development of standardised diagnostics, expanding the allergy speciality workforce and ensuring equitable access to emergency care and treatment options are crucial steps towards improving health outcomes for the millions of individuals affected by food allergies in China.

Abstract:Food provides energy and various nutrients and is the most important substance for the survival of living beings. However, for allergic people, certain foods cause strong reactions, and sometimes even cause shock or death. Food allergy has been recognized by the World Health Organization (WHO) as a major global food safety issue which affect the quality of life of nearly 5% of adults and 8% of children, and the incidence continues to rise but there is no effective cure. Drug alleviation methods for food allergies often have shortcomings such as side effects, poor safety, and high cost. At present, domestic and foreign scientists have turned to research and develop various new, safe and efficient natural sources of hypoallergenic or anti-allergic drugs or foods. There are many kinds of anti-allergic substances obtained from the plants and animals have been reported. Besides, probiotics and bifidobacteria also have certain anti-allergic effects. Of all the sources of anti-allergic substances, the ocean is rich in effective active substances due to its remarkable biodiversity and extremely complex living environment, and plays a huge role in the field of anti-food allergy. In this paper, the anti-food allergic bioactive substances isolated from marine organisms encompassing marine microbial, plant, animal sources and their mechanism were reviewed and the possible targets of anti-allergic substances exerting effects are illustrated by drawing. In addition, the development prospects of marine anti-allergic market are discussed and forecasted, which can provide reference for future research on anti-allergic substances.

Abstract:The consumption of seafood is widespread in China.The increase in the rate of seafood allergies,as well as their persistence and severity,become an important food safety issue.To improve seafood allergy patients' life quality,it is important to find a kind of seafood which has little or even none allergens.Different kinds of seafood live in different circumstances,it may have different allergens.This study compared the difference among varieties of shrimp.In the experiments,three kinds of shrimp were selected and the protein profiles were analyzed using SDS-PAGE.Then Immunoblotting and EAST inhibition were used to determine the allergenicity of them.The results show that main allergens of the three kinds of shrimp have molecules-36 kD,similar allergenicity,and critical cross-reactivity.
