Abstract:Efficient post-triazole regioselective N-2 arylation was developed from C-4, C-5 disubstituted-1,2,3-NH-triazoles. Three different approaches had been investigated, including S(N)Ar, Cu(I) catalyzed aryl amidation and Cu(II) mediated boronic acid coupling. The N-2-aryl triazoles were successfully synthesized with excellent yields. The structures were characterized by X-ray crystallography and some N-2-triazole products gave strong fluorescence with various emission controlled by the C-5 groups.

Abstract:A new class of bicyclic silaheterocycles, 6,6-dimethyl-2-aryl-6,7-dihydro- 5H-[1,2,4] triazolo[5,1-b][1,3,5] thiazasilines, which could be developed as potential sila-drugs. The reactions of several 3-aryl- 5-mercapto-1,2,4-triazoles with bis(chloromethyl) dimethylsilane under basic conditions gave high yields of bicyclic products. Reaction, product workup and isolation were streamlined without requiring an anhydrous solvent, inert gas or chromatographic purification. The products were structurally characterized by single-crystal X-ray diffraction.

Abstract:The 1,3-dipolar cycloaddition of azides and active internal alkynes has been well studied, but is rarely utilized as a tool for polymer preparation. In this work, an efficient polymerization route is developed. Polycycloaddition of diazide (4) and bis(benzoylethynyl)-benzenes and -butane (3) at elevated temperature has produced the first examples of soluble 1,4,5-trisubstituted polytriazoles PI with satisfactory molecular weights (Mw up to 16 400) in excellent yields (up to 98.6%). All the obtained polymers are thermally stable, losing merely 5% of their weights at temperatures higher than 367 degrees C. They exhibit higher refractive indices than some commercial plastics and can be crosslinked upon UV irradiation to generate a 3D photopattern with high resolution. The metal-free feature of such a methodology offers a facile tool to prepare functional materials free from the contamination of metal species.

Abstract:A series of novel heterocyclic polymers with fluorescent brightening properties are synthesized via Click polymerization. Fast synthesis of poly-1,2,3-triazoles (M n ≥ 9.31 kDa) is described herein, with a high yield of up to 95%. The Click polymerization approach has a number of advantages, including facile operation and outstanding isolation yield. The resultant polymers have a high thermal stability, excellent UV resistance, as well as acid and light fastness. On embedding with optical brightening agents, the polymers display strong fluorescent brightening properties in the tetrahydrofuran solution. Moreover, these products have a strong solution emission intensity and extraordinary photostability under UV light.

Abstract:2-Azido-3,5-dichloropyridine and 2-azido-5-chloro-3-fluoropyridine were given by reaction of sodium azide with 2,3,5-trichloropyridine, 3,5-dichloro-2-fluoropyridine or 5-chloro-2,3-difluoropyridine in ionic liquids. 1,3-Dipolar cycloaddition of 2-azido-3,5-dichloropyridine or 2-azido-5-chloro-3-fluoropyridine to alkynes in ionic liquids afforded the corresponding 1,4,5-trisubstituted [1,2,3]-triazoles in good yields and regioselectivities.

Abstract:Rapid parallel liquid-phase synthesis of pyrazoles has first been developed. The 1,3-dipolar cycloaddition between nitrilimines generated in situ and soluble polymer-supported alkynyl or alkenyl dipolarophiles in parallel one-pot fashion gave the corresponding PEG-supported regioisomeric pyrazoles or regiospecific pyrazolines. The latter was assuredly oxidated by DDQ to PEG-supported regiospecific pyrazoles. Cleavage from the support under mild conditions afforded pyrazoles in good yields and high purity.

Abstract:A series of unsymmetrical triazine-triazole conjugates were prepared by the copper-catalyzed cycloaddition reactions of 2-azide-4,6-dimethoxy-1,3,5-triazine with terminal alkynes under mild reaction conditions. Abstract:Visible light-enhanced [3 + 2] cycloaddition of N,N-disubstituted hydrazines with N-cyano-N-aryl-p-toluenesulfonamides is an efficient reaction pathway to polysubstituted 1,2,4-triazol-3-amines. The reaction is performed under mild conditions without the addition of any transition metals. This strategy involves a C(sp3)-H bond activation, a cyano cycloaddition, and the formation of two new C═N bonds. The protocol shows the advantages of good functional group tolerance and broad substrate scope. The late-stage modification experiments provide practical applications in the field of organic synthesis and medicinal chemistry.

Abstract:The solid-phase synthesis of 1,3-disubstituted and 1,3,5-trisubstituted 1,3,5-triazine-2,4,6-triones from MBHA and Wang resin is described. Reaction of resin-bound amino acids with isocyanates yield resin-bound ureas, which further react with chlorocarbonyl isocyanate in toluene at 65 degreesC to selectively afford the resin-bound 1,3-disubstituted 1,3,5-triazine-2,4,6-triones. Selective alkylation at the N-5 position of the resin-bound 1,3-disubstituted 1.3,5-triazine-2,4,6-triones was accomplished by treatment with alkyl halides in the presence of 1,8-diazabicyclo[5.4.0]undec-7-ene (DBU). The desired products were cleaved from their solid support and obtained in good yield and purity. The method can be employed in production of toltrazuril analogue libraries for identification of new anticoccidial agents.

Abstract:A general and regioselective synthesis of 3-trifluoromethyl 1,2,4-triazoles has been achieved through photocycloaddition of sydnone with trifluoroacetonitrile. This method employed trifluoroacetaldehyde O-(aryl)oxime as the CF3CN precursor and tolerated various functional groups to furnish 3-trifluoromethyl 1,2,4-triazole products in moderate to good yields. Mechanistic experiments revealed an energy transfer from photocatalyst 4-CzIPN to the sydnone substrates.

Abstract:We report a Cu(II)-catalyzed C-H amidation/cyclization of azomethine imines with dioxazolones as acyl nitrene transfer reagents under additive- and ligand-free conditions. An array of 1,2,4-triazolo[1,5-a]pyridine derivatives were afforded in moderate to good yields with excellent functional group tolerance. In addition, scale-up reaction and photoluminescence properties were discussed.

Abstract:In this work, a series of 4-aryl-3-(trifluoromethyl)-1 H -1,2,4-triazol-5(4 H )-one derivatives is synthesized through nucleophilic intramolecular cyclization reactions of ethyl 2-(2,2,2-trifluoro-1-(arylimino)ethyl)hydrazine-1-carboxylate intermediates, which are themselves prepared in high yields via reactions of ethyl chloroformate and N -aryl-2,2,2-trifluoroacetimidoyl chloride derivatives. Some of the merits of the reported procedure are an operationally simple and concise method, the ready availability of starting materials, excellent product yields, no formation of harmful by-products and easy purification of the products. Abstract:An efficient and rapid parallel liquid-phase synthesis of pyrazolines has been developed. The one-pot three-components reaction of polyethylene glycol (PEG)-supported acrylate 1, aldehyde 2 and aryl hydrazine 3 in the presence of chloramine-T in methanol gave the corresponding PEG-supported pyrazolines 5. Cleavage from the support under mild conditions afforded pyrazolines 6 in good yields (69-91%) and high purities (91-100%).

Abstract:Iodo-1,2,3-triazoles are of considerable interest for chemical and biomedical applications. However, current synthetic methods of iodo-1,2,3-triazoles can be hardly applied into direct modifications of bioactive molecules in water solvent. In this work, through novel combination of water-compatible oxidative iodination and CuAAC reaction, we developed a novel copper-catalyzed aqueous multicomponent synthetic method of 5-iodo-1,2,3-triazoles. The method shows to be highly effective and selective for substrates including biologically relevant compounds bearing nucleoside, sugar and amino acid moieties. Dramatically, based on this aqueous tandem reaction, a direct single-step multi-component dual-modification of peptide is developed from readily available starting materials. Furthermore, the method could also be applied into concise and fast multi-component radioactive I 125 labelings from commercial sodium [ 125 I]iodide aqueous solution as starting materials.
