Abstract:Rice noodles, a traditional Chinese food, are made using natural fermentation. The fermentation process plays an important role for the texture and flavour, which are due to the metabolism of the microorganisms involved, especially lactic acid bacteria (LAB). However, the fermentation of rice noodles is a non-controlled and spontaneous process; the quality of products is not stable. In order to improve and stabilize the quality of the production, the LAB functions on rice noodles during fermentation need to be elucidated. In this study, the community composition and dynamics of LAB associated with the fermented rice were investigated by using a culture-dependent approach in combination with a culture-independent approach, polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE). Sixty-four strains were isolated by culture-dependent analysis in different rice noodles factories and mainly 10 species in total were found. In addition, PCR-DGGE detected 17 species (including uncultured species) in total. Seven Lactobacillus species were detected by both methods, namely Lb. delbrueckii, Lb. salivarius, Lb. helveticus, Lb. reuteri, Lb. fermentum, Lb. amylovorus and Lb. oris, and the rest were detected by different methods separately. The potential role of these predominant LAB in the fermentation process is discussed. In general, the two techniques complement each other to give a better understanding on the LAB composition, which is necessary for the isolation of functional strains and improvement of the quality and consistency of rice noodles.

Abstract:This paper focused on isolation and identification species of psychrophilic microorganisms from raw milk provided by the pastures in Hangzhou district.Most of psychrophilic microorganisms in raw milk of Hangzhou district were Gram-negative psychrophilic microorganisms.Pseudomonas fluorescens、Acinetobacter lwoffii and Enterobacter sakazakii were dominant microorganisms.All of the resalts were the basis of set up a new rapid methods for dairy industry to detect the contamination of psychrophilic microorganisms in raw milk.

Abstract:The evolution of microbial floras in producing natural fermented rice noodles was investigated in this study. The microbial varieties were isolated and screened on the basis of morphological properties of colonies grew on the selective media and their fermentation effects on the rheological properties as well as eating qualities. The results revealed that the microorganisms mainly come from the rice granules with initial bacterial count up to 5 log cfu/mL. The natural fermentation was an anaerobic process, and the anaerobic plate count reached its maximal value of 10.21 log cfu/mL after 72 It of fermentation. LAB was the dominant microorganism, up to as high as 10.63 log cfu/mL. The second dominant microorganisms were yeasts, about 4 - 5 log cfu/mL. By further isolation and screening, five bacteria, two LAB and three yeasts were obtained. Rheometer measurements indicated that the larger quantity of acid yielded by the microorganisms produced more significant effects on the improvement of rheological properties. Therefore, natural fermentation process for producing the rice noodles is actually a lactic acid fermentation one. LAB plays, a key role in improving the physical properties and eating qualities of rice noodles.

Abstract:16 strains of Bacillus natto were screened from 24 kinds of natto powder, fresh natto and fermented soybean, 14 strains of Saccharomyces cerevisiae were screened from 20 species of distiller's yeast, and 14 strains of Lactobacillus plantarum were screened from 20 kinds of pickled Chinese cabbage. The colony characteristics, bacteria morphology and physiological-biochemical characteristic were observed and identified. The experiment was in order to provide experimental basis to the production and application of composite probiotic fermented feed.

Abstract:In this study, microbes were isolated from the rice slurry of a fermented rice cake to obtain lactic acid bacteria and yeast species. These species were identified using microbial physiology and gene sequence analyses. As the growth of the lactic acid bacterial strain R-2b and the yeast J-3a strains were found to be the best, a composite starter comprising these microbes was used for the preparation of fermented rice cakes. Based on single factor and orthogonal experiments, when the proportion of Lactobacillus plantarum, Saccharomyces cerevisiae, and Candida humilis was 1:3:6, the optimal fermentation conditions were addition of sugar and starter amounts of 20% and 6%, respectively, a fermentation temperature of 32 C, and fermentation time of 8 h. The fermented rice cake with this optimum ratio had the most abundant volatile components and qualified physicochemical and microbial indexes. Additionally, the overall quality was better than that of commercially available products.

Abstract:Lactic acid bacteria are commonly used in food industry,mainly in the fermentation process of meat products,prickles and diary products. They can add special flavor and color to the products,depress the growth of pathogens,and reduce nitrite concentration in the products,which makes the products safer to the customers.12 strains of lactic acid bacteria were isolated from the pickled vegetables.After amplification and sequencing of 16S rDNA gene,the isolated strains were identified by sequence homology as follows: 9 strains of Lactobacillus plantarum,2 strains of Lactobacillus sakei and 1 strain of Pediococcus pentosaceus.Some of their biochemical characteristics were studied and compared,as well as their abilities on the tolerance and degradation of nitrite.A L.plantarum strain,named J-10,was approved to degrade nitrite quickly with good biochemical characteristics. When grown in 0.25 mg/ml nitrite sodium added medium,strain J-10 could degrade 99.2% nitrite sodium at 37℃ and pH 6.2 for 24 h.

Abstract:Understanding bacteria and yeasts can reduce unpredictable changes in fresh fermented rice noodles (FFRN). The effects of selected strains (Limosilactobacillus fermentum, Lactoplantibacillus plantarum, Lactococcus lactis and Saccharomyces cerevisiae) on the edible quality, microbial community, and volatile component of FFRN were studied. The results indicated that the fermentation time could be shortened to 12 h when Limosilactobacillus fermentum, Lactoplantibacillus plantarum, and Lactococcus lactis were added, whereas it still required approximately 42 h after adding Saccharomyces cerevisiae. Only a steady bacterial composition was provided by adding Limosilactobacillus fermentum, Lactoplantibacillus plantarum, and Lactococcus lactis, and only a steady fungal composition was provided by adding Saccharomyces cerevisiae. Therefore, these microbial results indicated that the selected single strains cannot improve the safety of FFRN. However, the cooking loss was decreased from 3.11 ± 0.11 to 2.66 ± 0.13 and the hardness of FFRN was increased from 1186 ± 178 to 1980 ± 207 when it was fermented with single strains. Finally, a total of 42 volatile components were determined by Gas chromatography-ion Mobility Spectrometry and 8 aldehydes, 2 ketones, and 1 alcohol were added during the entire fermentation process. The main volatile components were different during fermentation depending on the added strain, and there was the greatest variety of volatiles in the group with added Saccharomyces cerevisiae.

Abstract:Lactic acid bacteria (LAB) are a prevalent bacterial group in rice wine maturation that contributes to flavor, texture, and nutritive value. To better understand LAB diversity in rice wines, 6 rice wine varieties from different regions in China were investigated using denaturing gradient gel electrophoresis (DGGE). Lactobacillus plantarum, L. namurensis, and Pediococcus acidilactici were identified using DGGE. Forty nine isolates were screened using a culture-dependent method. Prominent taxa were identified as Enterococcus sp., Lactobacillus delbrueckii, L. rhamnosus, L. plantarum, and Pediococcus acidilactici. Isolates were grouped and used for fermentation of rice wines. Greater numbers of species involved in fermentation lead to better sensory attributes of wine. DGGE analysis combined with a culture-dependent method can be a tool for investigation of the bacterial compositions of fermented rice wines.

Abstract:Six strains of lactic acid bacteria were obtained from the fermentation broth of Chinese rice wine.By using methods of physiological-biochemical and 16S rDNA tests,strains R1,R4,R5,R8 and R20 were identified as Lactobacillus rossiae,and strain R2 was identified as Lactobacillus casei.The ability of producing biogenic amines by these strains was evaluated by MRS plates and showed that only strain R1 could produce biogenic amines.HPLC confirmed that strain R1 could produce 598.61mg/L putrescine,and the others could not produce biogenic amines.The consistency of the results indicated that MRS plates analysis could be used as an effective,simple and economical method to evaluate the lactic acid bacteria's ability of producing biogenic amines.

Abstract:In this study,the bacterial diversities in three Jiangshui (a traditional Chinese fermented vegetable product) samples collected from three different locations were analyzed in order to isolate lactic acid bacteria for massive production of Jiangshui.The dominant lactic acid bacteria with a significant bacteriostatic effect against four common foodborne pathogens were isolated and identified from the Jiangshui samples.Total bacterial DNA was extracted from Jiangshui for sequencing of the 16S rRNA V3-V4 region using the MiSeq sequencing technique to understand the bacterial diversity.The results showed that Lactobacillus was dominant in all three Jiangshui samples,showing differences in their bacterial diversity.The compositions of bacterial communities in Jiangshui samples from Chang'an district of Xi'an and Taian county of Tianshui were similar,but significantly different from that of the sample from Yanta district of Xi'an.Both spoilage bacteria and potential pathogens (or opportunistic pathogens) were detected in each sample.A total of 35 stains able to form calcium dissolution circle,were isolated using a modified MRS medium.Ten strains were selected for their outstanding inhibitory activity against Staphylococcus aureus,Escherichia coil,Salmonella enteritidis and Shigella dysenteriae,which were identified through 16S rDNA sequencing as 8 strains of Lactobacillus plantarum and 2 strains of Lactobacillus rhamnosus,indicating that Lactobacillus plantarum and Lactobacillus rhamnosus are the dominant bacteria in Jiangshui.This study can provide some theoretical and practical bases for the production of Jiangshui by applying both lactic acid bacteria or one of them in the future.

Abstract:[Objective]To isolate microorganisms in specific ecological environment( rotten straw),and study the degrading abilities of these microorganisms on rice stalk. [Method]By using microcrystalline cellulose powder as carbon source,we isolated and identificated microbes that degrading rice stalks in certain natural ecological environment. [Result] From straw stalks,7 strains were isolated. Morphological observation and biochemical identification showed that 2 isolates belong to penicillium,2 belong to Aspergillus furnigatus,1 belongs to Rhizopus microsporus,1 Gongronella butleri and 1 Mucor racemosus. Furthermore,the ability of these isolates to degrade rice stalks was studied. Among which,the abilities of degrading straw cellulose of 1 Aspergillus furnigatus and 1 Gongronella butleri are strongger,the abilities of degrading straw hemicellulose of another Aspergillus furnigatus and 1 Gongronella butleri are strongger. [Conclusion] The study can provide reference for microbial strains degrading rice stalk.

Abstract:Pathogenic microorganisms have been detected in fermented food. Combining the enormous class of the pathogens and their continuously appearing mutants or novel species, it is important to select suitable and safe antibacterial agents for fermented food safety. Lactic acid bacteria (LAB) which produce diverse imperative antimicrobial metabolites have an immense number of applications in the food industry. Here, the human-derived strain YT was isolated due to its cell-free supernatant (CFS-YT) and cells (Cs-YT), respectively performed obvious inhibitory ring to Gram-positive and -negative spoilage bacteria. Strain YT was identified as Lacticaseibacillus rhamnosus by the 16s rDNA sequence and morphology. The antibacterial activity of CFS-YT was demonstrated to be growth-dependent, pHs-sensitive, broadly thermostable and enzyme-insensitive. Cs-YT displayed a broad antibacterial spectrum with the action mode of bacteriostasis. The antibacterial activity of Cs-YT was due to substances located at the cell surface which were sensitive to heat, stable at broad pH gradients and sensitive to specific enzymes. These data suggested that L. rhamnosus YT could be used as an alternative antimicrobial agent in fermented food biopreservation.
Links between microbial compositions and volatile profiles of rice noodle fermentation liquid evaluated by 16S rRNA sequencing and GC-MS

Abstract:The study investigated the correlations between microbial compositions and volatile profiles of six mixed culture rice noodle fermentation liquid (RNFL) samples labelled by YI, XX, HL, XL, KJ, and JJ, respectively. Through 16S rRNA high-throughput sequencing technology (HTS), the main microbial genera identified in RNFL samples were Lactobacillus, Burkholderia, Gluconacetobacter, Lactococcus, and Leuconostoc (≥5% of operational taxonomic units). A total of one hundred and ten volatile compounds were detected by using SPME/GC-MS. E-nose data showed that HL, XX, and JJ shared similarities in flavor characteristics. Correlation analysis between bacterial genus and volatile compounds indicated that Lactobacillus and Gluconacetobacter were strongly correlated with most flavor compounds detected. Moreover, YI and XX had higher fragrance scores in sensory evaluation, indicating thicker fermented rice fragrance than other RNFL samples.

Abstract:Rice wine, a traditional fermented alcoholic beverage in China, is produced with grains such as rice, which are fermented with saccharifying starter-koji. Its flavor quality is closely associated to the starter culture-koji, which is made by mixing botanical materials with high-class glutinous rice in certain ecological context. However, there are few reports on the microbial community structure of rice wine koji. In this paper, bacterial community structures of rice wine koji were analyzed using 16S rRNA gene sequencing based on Illumina MiSeq high-throughput technology in 20 samples collected from Xiaogan area, Hubei province and Dazhu area, Sichuan province (10 from each area). We found rice wine koji flora mainly consisted of Weissella, Lactobacillus, Lactococcus, Bacillus, Enterococcus, and Cronobacter, with relative abundances of 29.49%, 10.93%, 8.85%, 4.75%, 1.16% and 1.15%, respectively, as well as an accumulative average relative abundance of 58.71%. They all belonged to Firmicutes and Proteobacteria—the two known dominant genus. Genus-level PCA (Principal component analysis) and OTU-level PCoA (Principal coordinates analysis) based on unweighted UniFrac distances showed that the bacterial community structure differed significantly between the samples from the 2 areas. 7 OTUs were detected in all samples, accounting for 4.4% of the total qualified assembly. Among the 7 OTUs, 3 OTUs were identified as Enterococcus, 2 OTUs were identified as Cronobacter, 1 OTU was identified as Bacillus and 1 OTU was identified as Alkaliphilus. Fifty-eight lactic acid bacteria (LAB) strains were isolated from the 20 koji samples with traditional microbial methods. Among them, Enterococcus faecium and Pediococcus pentosaceus were the dominant LAB isolates, with relative abundances of 51.72% and 31.03%. Despite the differences, a large number of shared bacteria were detected in samples from the two areas.

Abstract:In order to study lactic acid bacteria species which plays a major role in the fermentation process of Italian ryegrass(Lolium multiflorum)silage,6lactic acid bacteria strains(L-1,L-3,L-4,L-5,L-8and L-9)were isolated from previously fermented juice of Changjiang No.2Italian ryegrass.The results of traditional biochemical identification,experiment of acid-production rate,acid tolerance test,salt tolerance test,and the 16SrRNA sequence analysis indicated that L-1,L-3,L-4,L-5,L-8and L-8are Lactobacillus plantarum,Lactococcus lactis,Lactococcus garvieae,Leuconostoc lactis,Weissella confuse and Weissella cibaria,respectively,and all of them have good acid productivity,strong acid and salt tolerance.Further studies on using these strains as feed additives can be made.
