Calcium silicate cements prepared by hydrothermal synthesis for bone repair

Abstract:Calcium silicate materials have attracted a great deal of interest because of their osteogenesis. The purpose of this study was to use the hydrothermal method without the use of ethanol and an acidic medium to synthesize calcium silicate powder. After mixing the powder with water, the in vitro bioactivity and osteogenesis of calcium silicate cements were examined in comparison with the cement prepared by the sol-gel route. The morphology, phase composition, weight loss, and diametral tensile strength of the cement after soaking in a simulated body fluid (SBF) for different time periods were assayed. Human fetal osteoblasts (hFOBs) cultured on the cement surfaces were used to evaluate the in vitro osteogenesis. The results indicated that after 1 day of soaking in SBF, a precipitated apatite layer was found on the two cement surfaces. Notably, although the two cements were derived from the different powder technologies, there were no significant differences (P> 0.05) in bioactivity including phase evolution, morphology change, strength variation, and degradation between cements during soaking in SBF. The hydrothermally-derived calcium silicate cement showed similar cell attachment, proliferation, differentiation, and mineralization expressions compared to the sol-gel-derived cement, but being higher than those obtained in the tissue culture plate. The present results suggest that the calcium silicate cements prepared by a hydrothermal method had an excellent bioactivity and osteogenesis and may be suitable for bone repair. In Vitro Bioactivity of Calcium Silicate Prepared by Sol-gel Process

Abstract:The high-purity calcium silicate ceramic was sintered at 1300℃ under the atmospheric pressure with the calcium silicate powder synthesized by the sol-gel method. The calcium silicate ceramic was characterized by X-ray diffraction (XRD) and scanning electron microscopy (SEM) and the bioactivity、cytotoxicity and cell adhension were also studied. The results showed that the sample was mainly constituted by the β-calcium silicate(β-CS). The bone-like HAp was formed on the surface of the calcium silicate ceramic after 14 d soaking in simulated body fluid (SBF), and more bone-like HAp formed after 28 d soaking. The result of cell experiment indicates that the calcium silicate possesses good bioactivity and can promote the osteoblast growth.

Abstract:ObjectivesIn tissue regeneration research, the term “bioactivity” was initially used to describe the resistance to removal of a biomaterial from host tissues after intraosseous implantation. Hydraulic calcium silicate cements (HCSCs) are putatively accepted as bioactive materials, as exemplified by the increasing number of publications reporting that these cements produce an apatite-rich surface layer after they contact simulated body fluids.MethodsIn this review, the same definitions employed for establishing in vitro and in vivo bioactivity in glass–ceramics, and the proposed mechanisms involved in these phenomena are used as blueprints for investigating whether HCSCs are bioactive.ResultsThe literature abounds with evidence that HCSCs exhibit in vitro bioactivity; however, there is a general lack of stringent methodologies for characterizing the calcium phosphate phases precipitated on HCSCs. Although in vivo bioactivity has been demonstrated for some HCSCs, a fibrous connective tissue layer is frequently identified along the bone–cement interface that is reminiscent of the responses observed in bioinert materials, without accompanying clarifications to account for such observations.ConclusionsAs bone-bonding is not predictably achieved, there is insufficient scientific evidence to substantiate that HCSCs are indeed bioactive. Objective appraisal criteria should be developed for more accurately defining the bioactivity profiles of HCSCs designed for clinical use.
Advances in the use of calcium silicate-based materials in bone tissue engineering

Abstract:As the main means of treating bone defects, bone grafts are in high demand. Recently, the development of suitable bone graft replacement materials has received significant research attention because of the drawbacks of autologous and allogeneic bone grafts. The applications of ceramic bone graft materials, such as calcium phosphate, calcium sulfate, and hydroxyapatite, are limited by various factors. Calcium silicate,a new bone graft material, exhibits high biocompatibility and osteoinductivity. This paper summarizes the main methods for synthesizing calcium silicate and compares their advantages and disadvantages in detail. The mechanical and antimicrobial properties of calcium silicate can be improved to fulfil the biological and mechanical requirements of bone graft materials by adding materials such as B 2 O 3 , SiO 2 , Ag, Zn, Ti, and chitosan. Recent research progress in calcium silicate ceramics in preliminary clinical applications as well as 3D printing applications is also reviewed, which provide the impetus for the clinical application of calcium silicate ceramics. Finally, the article suggests future directions for the development of calcium silicate-based materials and offer prospects for their application as bone repair and replacement materials.

Abstract:BACKGROUND:Calcium sulfate is mixed with other biomaterials in order to overcome its weaknesses or failings. OBJECTIVE:To investigate the application of calcium sulfate as a mixture with other materials acting as drug release system and scaffold material for tissue engineering. METHODS:Using“calcium sulfate, bone repair material, drug release, biomaterials, compound materials”in Chinese and“calcium sulfate, mixture, bone defect, drug release”in English as the key words, a computer-based online search of China Academic Journal Ful-text database and PubMed database (1980-01/2012-03) was performed for articles about the application of calcium sulfate as a mixture with other materials. RESULTS AND CONCLUSION:Calcium sulfate has more advantages and peculiarities than other biomaterials and make a wel development in clinic. However, some disadvantages such as too rapid resorption or too short drug release period hinder its widespread use in clinic. Therefore, many studies have been focused on the application of calcium sulfate as a mixture with other materials recently. Combining calcium sulfate with other material to obtain an ideal material may be a promising and sane choice. This can make the best use of calcium sulfate advantages and bypass its disadvantages. Future research is required to enhance and improve the resorption property, drug release profile, handling characteristics and physical properties of calcium sulfate through mixing with other materials while preserving its biocompatibility and osteoconduction.

Abstract:Calcium (Ca) and silica (Si) ions have attracted intense interest in biomedical applications. The two ions are directly involved in many biological processes; for instance, Ca plays a key role in regulating cellular responses to bioceramics, promoting cell growth and differentiation into osteoblasts. Si plays a significant role in bone calcification and is helpful for bone density improvement and inhibiting osteoporosis. Calcium silicate ceramics including a large group of trace metal containing calcium silicate‐based compounds are involved in biomedical applications such as repairing hard tissue texture, bone scaffolds, bone cements or implant coatings. The aim of the paper is to provide a comprehensive overview of developments in research on calcium silicate‐based ceramics, such as wollastonite (CaSiO3), diopside (CaMgSi2O6), akermanite (Ca2MgSi2O7), bredigite (Ca7Mg(SiO4)4), merwinite (Ca3MgSi2O8), monticellite (CaMgSiO4), hardystonite (Ca2Zn(Si2O7) and baghdadite (Ca3ZrSi2O9) including degradation, apatite mineralization and mechanical properties. Finally, the biological in vitro and in vivo presentation for bone tissue repair are summarized, which show promise with regard to application of calcium silicate‐based ceramics as bone repair and replacement materials.

Abstract:Calcium silicate is a common implant material with excellent mechanical strength and good biological activity. In recent years, the addition of strengthening materials to calcium silicate has been proven to promote bone tissue regeneration, but its degradation properties require further improvements. In this paper, calcium silicate was used as the matrix, and 10 wt% hydroxyapatite and 10 wt% strontium phosphate were added to im prove the biological activity of the scaffold. The effect of adding different amounts of calcium sulfate dihydrate (CaSO4·2H2O) on the degradation of the scaffold was explored. A porous ceramic scaffold was prepared by digital light processing (DLP) technology, and its performance was evaluated. Cell experiments showed that the addition of calcium sulfate improved cell proliferation and differentiation. Simulated body fluid (SBF) immersion tests showed that small amounts of apatite deposits appeared on the fourth day, larger deposits appeared on the 14th day, and degradation occurred on the surface after 28 days of immersion. Mechanical tests showed that the addition of 5 wt% CaSO4·2H2O improved the compressibility of the composite. After soaking in SBF for 14 days, it retained its compressive strength (11.8 MPa), which meets the requirements of cancellous bone, demonstrating its potential application value for bone repair.

Abstract:The poor bioactivity and mechanical properties have restricted its biomedical application, although poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV) had good biocompatibility and biodegradability. In this study, calcium silicate (CS) was incorporated into PHBV for improving its bioactivity and mechanical properties, and the porous PHBV/CS composite scaffolds were fabricated via selective laser sintering (SLS). Simulated body fluid (SBF) immersion tests indicated the composite scaffolds had good apatite-forming ability, which could be mainly attributed to the electrostatic attraction of negatively charged silanol groups derived from CS degradation to positively charged calcium ions in SBF. Moreover, the compressive properties of the composite scaffolds increased at first, and then decreased with increasing the CS content, which was ascribed to the fact that CS of a proper content could homogeneously disperse in PHBV matrix, while excessive CS would form continuous phase. The compressive strength and modulus of composite scaffolds with optimal CS content of 10 wt % were 3.55 MPa and 36.54 MPa, respectively, which were increased by 41.43% and 28.61%, respectively, as compared with PHBV scaffolds. Additionally, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay indicated MG63 cells had a higher proliferation rate on PHBV/CS composite scaffolds than that on PHBV. Alkaline phosphatase (ALP) staining assay demonstrated the incorporation of CS significantly promoted osteogenic differentiation of MG63 cells on the scaffolds. These results suggest that the PHBV/CS composite scaffolds have the potential in serving as a substitute in bone tissue engineering.

Abstract:Calcium silicate (CS), the recent developed biodegradable ceramic, shows potential in stimulating bone regeneration. However, the rapid degradation restricts its further application, that is, the cytotoxicity is highly suspicious. In this study, the attempt with the addition of strontium by partially replacing the site of calcium shows appropriate, not only enhancing the bioactivity of CS, but also promoting its biocompatibility. Thus, the modification of biomaterial by strontium seems to be an effective approach for biological application.

Abstract:The biomaterials in system CaO-P2O5-SiO2 were synthesized via sol-gel method. The biomaterials can be applied to bone reparation and bone tissue engineering scaffolds The nano-pore structure, degradability, bioactivity and bio-mineralization characteristic of the biomaterials were investigated in details using XRD, SEM/EDX, FTIR, BET and DSC/TG techniques. It was indicated that the sol-gel derived biomaterials have a higher bioactivity than that of the melt derived bioactive glasses or glass-ceramics. It just takes 4-8 hours for HCA to form on the surface of the sol-gel samples in SBF solution at 37°C. The spherical HCA crystal clusters formed on the surface of the sol-gel derived samples immersed in SBF for 8 hours have a low crystallinity. Owing to their interconnected nano-sized pores, the sol-gel samples possess much higher surface areas and the hydrous porous SiO2 gel layer containing a great amount of ºSi-OH groups can be rapidly formed on the biomterials’ surface through a quick ion exchange between H3O+ in the solution and Ca2+ in the surface of the materials. ºSi-OH groups can play a very important role in inducing formation of HCA. They make the material surfaces electronegative, which resulted in a double electrode layer formed between the samples surface and SBF solution. The double electrode layer is in favor of formation of HCA on the surface of the materials.
Calcium Silicate and Calcium Hydroxide Materials for Pulp Capping: Biointeractivity, Porosity, Solubility and Bioactivity of Current Formulations

Abstract:Aim The chemical-physical properties of novel and long-standing calcium silicate cements versus conventional pulp capping calcium hydroxide biomaterials were compared. Methods Calcium hydroxide–based (Calxyl, Dycal, Life, Lime-Lite) and calcium silicate–based (ProRoot MTA, MTA Angelus, MTA Plus, Biodentine, Tech Biosealer capping, TheraCal) biomaterials were examined. Calcium and hydroxyl ion release, water sorption, interconnected open pores, apparent porosity, solubility and apatite-forming ability in simulated body fluid were evaluated. Results All calcium silicate materials released more calcium. Tech Biosealer capping, MTA Plus gel and Biodentine showed the highest values of calcium release, while Lime-Lite the lowest. All the materials showed alkalizing activity except for Life and Lime-Lite. Calcium silicate materials showed high porosity values: Tech Biosealer capping, MTA Plus gel and MTA Angelus showed the highest values of porosity, water sorption and solubility, while TheraCal the lowest. The solubility of water-containing materials was higher and correlated with the liquid-to-powder ratio. Calcium phosphate (CaP) deposits were noted on materials surfaces after short aging times. Scant deposits were detected on Lime-Lite. A CaP coating composed of spherulites was detected on all calcium silicate materials and Dycal after 28 days. The thickness, continuity and Ca/P ratio differed markedly among the materials. MTA Plus showed the thickest coating, ProRoot MTA showed large spherulitic deposits, while TheraCal presented very small dense spherulites. Conclusions calcium silicate-based cements are biointeractive (ion-releasing) bioactive (apatite-forming) functional biomaterials. The high rate of calcium release and the fast formation of apatite may well explain the role of calcium silicate biomaterials as scaffold to induce new dentin bridge formation and clinical healing.

Synthesis and in Vitro Bioactivity of Calcium-Phosphate Bioglass-Ceramic with 10mol％ Silica

Abstract:The bioactive glass-ceramics based on a SiO2-CaO-P2O5 system were synthesized using the polyacrylamide-gel method. The prepared glass-ceramic material is attractive for its low content of silica and high calcium-phosphorus molar ratio of 1.57, which is closer to the composition of hard tissues in human body. The bioactivity of the material was assessed by simply immersing it in the simulated body fluid (SBF) for different time durations. The prepared powders and the samples surface after the immersion were characterized by using TG/DTA, XRD, FTIR and SEM techniques. The concentration of Ca, P and Si ions in the SBF were measured by ICP-AES. The results show that the addition of silica contributes to the formation of the apatite on the surface of the glass-ceramics. With the extension of immersion time, the carbonated hydroxyl apatite (CHA) layer deposited is changed from spherical bumps to blades, and the concentration of calcium and phosphorus in the SBF increases while that of silica decreases. The above results indicate that the prepared glass-ceramic material has good bioactivity and may be used as bone and teeth repair material or filler.

Abstract:Gypsum is a promising material for bone defect repair due to its osteoconductivity, whereas it is still limited in orthopedic and dental surgeries due to its low bioactivity and too rapid resorption so that one major concern is the significant loss in microstructural stability in vivo. In the present strategy some key features were significantly improved by introducing rapidly biodegradable but highly bioactive calcium silicate (CS) for regulating the physicochemical properties and biological performances of the gypsum-based cements at the same liquid/solid ratio. We demonstrated that introduction of 23% CS into beta-calcium sulfate hemihydrate (CSH) could improve the physicochemical properties but would not compromise the mechanical strength of the composite. The surface bioactivity was significantly enhanced by introducing 23% CS, and these biphasic composites were favorable for decelerating the biodegradation rate by nearly 18.5% in 28 days in vitro. A mild bioresorption rate, with 39.6% of composite residual 4 weeks after operation, was determined when implanted in subcutaneous tissue of rats. 8 weeks after implantation, the composite cement containing 23% CS significantly enhanced new bone tissue regeneration with a much higher relative bone content (similar to 68.6%) than pure gypsum in critical size femoral defects in rabbits. The novel CSH-CS biocements represent promising candidates for rapid bone resconstruction and repair in trauma and pathological conditions.
Calcium silicate biocomposites: effects of selenium oxide on the physico-mechanical features and their in-vitro biological assessments

Abstract:Bone tissue regenerative material serves as a prospective recovery candidate with self-adaptable biological properties of bio-activation, degradability, compatibility, and antimicrobial efficacy instead of metallic implants. Such materials are highly expensive due to chemical reagents and complex synthesis procedures, making them unaffordable for patients with financial constraints. This research produced an efficient bone tissue regenerative material using inexpensive naturally occurring source materials, including silica sand and limestone. The extracted SiO2and CaO particles (75:25 wt%) were subjected to hydrothermal synthesis (water treatment instead of chemical solvents) to produce the CaSiO3biomaterial (code: S). Selenium oxide was doped with calcium silicate at 3, 5, and 10 wt.% to enhance its properties, yielding biocomposite materials (i.e. S3, S5, and S10). The physico-mechanical properties of these materials were investigated with x-ray diffraction, Fourier transform infrared, FESEM-EDS, and micro-universal testing machine. The results revealed that the synthesized biocomposites have a crystalline wollastonite phase with a porously fused rough surface. From structural parametric calculations, we found that the biocomposites have reduced particle size and enhanced surface area due to the influence of selenium oxide. The biocomposite S10, having high SeO2content, attained the maximum compressive strength of 75.2 MPa.In-vitrostudies of bioactivity, biodegradability, biocompatibility, and antibacterial activity were performed. At 7 and 14 d of bioactivity, the synthesized biocomposites are capable of dissolving their ions into simulated body fluid (SBF) solution to precipitate hydroxyapatite and a required Ca/P ratio of 1.69 was achieved by S3. A comparative analysis has been performed on the degradation activity in Tris-HCl and the consequent pH changes during SBF treatment. The bio-analysis revealed that the biocomposite S3 shows enhanced bioactivity through a controlled degradation rate and secured cell viability of 88% at a concentration of 100 μg ml-1. It also offers significant bacterial inhibition potency againstE.coliandS.aureusbacteria.
Calcium Phosphate Glass-Ceramics for Biomedical Applications

Abstract:Calcium phosphate glass-ceramics are widely applied in dental and bone repairing,substituting and bone tissue engineering because the glass-ceramics have good bioactivity and biocompatibility. In the present paper, processing of Ca phosphate glass-ceramics, the relations between compositions and properties, in vitro and in vivo behaviors of the glassceramics, and clinic applications were described in detail.

Abstract:Bioactive tricalcium silicate ceramics (Ca(3)SiO(5)) were fabricated by spark plasma sintering (SPS), and their sinterability and mechanical properties were examined. The bioactivity and in vitro biocompatibility of Ca(3)SiO(5) ceramics were evaluated. Ca(3)SiO(5) ceramics show higher density and superior mechanical properties compared with those prepared by conventional pressureless sintering. In addition, hydroxyapatite was induced to form on the surface of Ca(3)SiO(5) ceramics when soaked in simulated body fluid and bone marrow mesenchymal stem cells were attached and spread well on the ceramics. Ca(3)SiO(5) ceramics fabricated by SPS possess excellent mechanical properties, bioactivity, and biocompatibility and are promising bone repaired materials.
