Optimisation process of walnut protein hydrolysed as an antioxidant candidate

Abstract:Walnut protein was extracted from defatted walnut meal, the preparation process of walnut protein antioxidant peptides was determined by response surface optimization. The amino acid composition, simulated digestion, chemical antioxidant activity and HepG-2 cell antioxidant activity were investigated. The results showed that the optimal enzymatic hydrolysis conditions of walnut protein antioxidant peptides were tryptic hydrolysis for 3.5 h, adding enzyme dosage 0.1%, pH 8.0, substrate concentration 5.8%, temperature 39.4 °C. Under these conditions, the DPPH free radical scavenging rate of the walnut protein antioxidant peptides was 89.59%. Hydrophobic amino acids accounted for 32.46% of the hydrolysate of walnut protein antioxidant peptides. Walnut protein antioxidant peptides still demonstrated strong antioxidant activity after simulated gastric digestion, but the activity diminished moderately after simulated intestinal digestion. Walnut protein antioxidant peptides had significant chemical antioxidant activity, the IC 50 value of DPPH and ABTS radical scavenging rate were 0.098 mg/mL and 0.051 mg/mL, respectively, and the ORAC value was 465.568 μmol TE/g; the EC 50 value of CAA was 6.104 μg/mL. This research provides theoretical reference for the development and use of walnut protein peptides.
