Features of Variation and Genetic Polymorphisms in Mtdna of Seventeen Chinese Cattle Breeds

Abstract:China has a great variety of the genetic resources in bovine species reflected by more than 28 indigenous cattle breeds in different areas.To further understand the origin and the genetic background of these breeds,we carried out phylogenetic analysis of the cattles from different geographical locations based on mtDNA polymorphism.The complete mtDNA D-loop was sequenced from a total of 84 indigenous Chinese cattles belong to 17 different breeds.The analyzed sequences were classified into 53 haplotypes defined by polymorphisms at 102 polymorphic sites.These haplotypes were clustered into two distinct lineages:taurine Bos taurus and zebu Bos indicus.The major characteristic of the variations in the D-loop sequences was the observed high transition/transversion ratio;a second characteristic was the coexistence of deletions and substitutions,primarily the high deletion frequency in D-loop region.The overall nucleotide diversity and haplotype diversity of the D-loop sequences collected from Chinese cattle samples were determined to be 0.026 78± 0.000 50 and 0.919±0.027,respectively,which were significantly higher than the reports from other studies.The taurine mtDNA appeared to be dominant in the northern breeds(80%—100%),whereas the zebu mtDNA was dominant in the southern breeds(42.9%—100%).Meanwhile,the frequency and geographical distribution of taurine and zebu mtDNA among the breeds from the central regions were also observed to be divergent.The geographical distribution of the mtDNA frequencies revealed a declining south-to-north trend of female zebu introgression in China,which were in correlation with and could be explained by the factors such as historic events,geographical segregation and weather conditions.
