Latest review of methylation status of colorectal cancer related genes in carcinogenesis

Abstract:OBJECTIVE: To review the research on abnormal methylation changes of colorectal cancer related genes in carcinogenesis. METHODS: The articles about abnormal methylation of colorectal cancer related genes in carcinogenesis were searched for by using colorectal cancer,methylation and colorectal cancer related genes(such as K-ras, p53, c-myc, APC, p16, RASSF1A, hMLH1 and ERβ) as keywords in Medline and CNKI data-base. RESULTS: K-ras and p53 demonstrate DNA point mutation in colorectal cancer. Localized hypomethylation of c-myc is an original case in carcinogenesis correlating with cell proliferation.Not only the loss of heterozygosity(LOH) but also hypermethylation results in APC inactivation. p16 distributes a different methylation densities in colorectal cancer tissues. RASSF1A is suppressed mainly by promoter hypermethylation. Suppression of hMLH1 by hypermethylation induces MSI(+) phenotype involving in tumor stages in ascending colorectal cancer. Analysis of ERβ methylation mode plays a role on estimating tumor prognosis. CONCLUSIONS: The abnormal changes of colorectal cancer related genes include DNA point mutation, deletion, DNA rearrangements as well as methylation. Emphasis on methylation will benefit prevention and therapy of colorectal cancer.
