Abstract:Lightness is the apparent reflectance of a surface, and it depends not only on the actual luminance of the surface but also on the context in which the surface is viewed 1, 2, 3, 4, 5, 6, 7, 8, 9, 10. The cortical mechanisms of lightness processing are largely unknown, and the role of early cortical areas is still a matter of debate 11, 12, 13, 14, 15, 16, 17. We studied the cortical responses to lightness variations in early stages of the human visual system with functional magnetic resonance imaging (fMRI) while observers were performing a demanding fixation task. The set of dynamically presented visual stimuli included the rectangular version of the classic Craik-O'Brien stimulus 3, 18, 19 and a variant that led to a weaker lightness effect, as well as a pattern with actual luminance variations. We found that the cortical activity in retinotopic areas, including the primary visual cortex (V1), is correlated with context-dependent lightness variations.
Contrast Adaptation and Representation in Human Early Visual Cortex.

Abstract:The human visual system can distinguish variations in image contrast over a much larger range than measurements of the static relationship between contrast and response in visual cortex would suggest. This discrepancy may be explained if adaptation serves to re-center contrast response functions around the ambient contrast, yet experiments on humans have yet to report such an effect. By using event-related fMRI and a data-driven analysis approach, we found that contrast response functions in V1, V2, and V3 shift to approximately center on the adapting contrast. Furthermore, we discovered that, unlike earlier areas, human V4 (hV4) responds positively to contrast changes, whether increments or decrements, suggesting that hV4 does not faithfully represent contrast, but instead responds to salient changes. These findings suggest that the visual system discounts slow uninformative changes in contrast with adaptation, yet remains exquisitely sensitive to changes that may signal important events in the environment.
Optical imaging of contrast response in functional domains in V1 and V2 of macaque visual cortex

Abstract:Our recent optical imaging and single unit studies in visual cortex suggested a preferential involvement of V1 blobs and V2 thin stripes in real and illusory brightness response (Hung et al. Soc Neurosci 2002; cf. Hung et al Vision Res 2001). To further explore this possibility and to better relate full field brightness response to standard contrast sensitivity responses, we have measured contrast response of different functional domains in V1 and V2 with optical imaging and electrophysiological recording. Visual stimuli consisted of drifting horizontal and vertical luminance sine wave gratings with different spatial frequencies, ranging from 0.84 — 7.6 cyc/deg. Luminance contrast of the gratings was varied over four different levels (10%, 20%, 40%, 80%). Intrinsic signals were imaged from area V1 and V2 of anesthetized macaque monkeys. Average reflectance values from functional domains (orientation domains in V1 and V2, blobs in V1, and thin and thick/pale stripes in V2) were obtained and were also compared to single cell responses sampled from superficial layers of imaged domains. Results showed that, as expected, cortical reflectance values changed systematically with the contrast level of the stimulus: the greater the contrast, the greater the response. As measured by reflectance values, we find that, at low spatial frequencies and low contrast levels, V1 blobs have greater response than V1 interblobs. In comparison to V1 domains, V2 domains exhibit higher contrast gain and saturate at a low-mid contrast levels. V2 thin stripes also showed higher contrast gain than thick/pale stripes. Retinotopic distribution of chromatic responses in human primary visual cortex

Abstract:In non-human primates at least three anatomically and functionally distinct channels convey signals from the retina to the primary visual cortex (V1). Two of these channels, the parvocellular and the koniocellular, are sensitive to chromatic contrasts and form the basis of color vision. In humans, common phylogenetic history with other primates and psychophysical experiments suggest identical retinocortical mechanisms but separate evaluation of the distinct anatomical channels has been difficult because signals are already combined in V1. We studied the spatial distribution of activation to chromatic stimuli along the two opponent chromatic axes in human V1 with multifocal functional magnetic resonance imaging. The signal strength was quantified from three experiments with stimuli up to 20 degrees eccentricity. The hypothesis was that, although the parvo- and koniocellular signals are mixed in V1, distinct distributions of signal strength would be evident. We found that whereas different conditions activated the same areas of cortex, indicating that they have identical magnification factors, the responses to red/green stimulation were stronger close to the fovea whereas the blue/yellow responses were much less diminished with increasing eccentricity. Both chromatic axes showed saturating contrast response functions. Our measure directly from human V1 is in line with earlier psychophysical studies suggesting relatively stronger parvocellular channel representation close to the fovea, and more uniform distribution of the koniocellular and achromatic channels. In addition, our study presents a way to rapidly quantify retinotopic signal transmission in distinct retinocortical pathways of individual subjects.
Optical Imaging of Contrast Response in Macaque Monkey V1 and V2.

Abstract:Our studies on brightness information processing in Macaque monkey visual cortex suggest that the thin stripes in the secondary visual area (V2) are preferentially activated by brightness stimuli (such as full field luminance modulation and illusory edge-induced brightness modulation). To further examine this possibility, we used intrinsic signal optical imaging to examine contrast response of different functional domains in primary and secondary visual areas (V1 and V2). Color and orientation stimuli were used to map functional domains in V1 (color domains, orientation domains) and V2 (thin stripes, thick/pale stripes). To examine contrast response, sinusoidal gratings at different contrasts and spatial frequencies were presented. We find that, consistent with previous studies, the optical signal increased systematically with contrast level. Unlike single-unit responses, optical signals for both color domains and orientation domains in V1 exhibit linear contrast response functions, thereby providing a large dynamic range for V1 contrast response. In contrast to domains in V1, domains in V2 exhibit nonlinear responses, characterized by high gain at low contrasts, saturating at a mid-high contrast levels. At high contrasts, thin stripes exhibit increasing response, whereas thick/pale stripes saturate, consistent with a strong parvocellular input to thin stripes. These findings suggest that, with respect to contrast encoding, thin stripes have a larger dynamic range than thick/pale stripes and further support a role for thin stripes in processing of brightness information.
Model-Based Approach Shows ON Pathway Afferents Elicit a Transient Decrease of V1 Responses

Abstract:Neurons in the primary visual cortex (V1) receive excitation and inhibition from distinct parallel pathways processing lightness (ON) and darkness (OFF). V1 neurons overall respond more strongly to dark than light stimuli, consistent with a preponderance of darker regions in natural images, as well as human psychophysics. However, it has been unclear whether this "dark-dominance" is because of more excitation from the OFF pathway or more inhibition from the ON pathway. To understand the mechanisms behind dark-dominance, we record electrophysiological responses of individual simple-type V1 neurons to natural image stimuli and then train biologically inspired convolutional neural networks to predict the neurons' responses. Analyzing a sample of 71 neurons (in anesthetized, paralyzed cats of either sex) has revealed their responses to be more driven by dark than light stimuli, consistent with previous investigations. We show that this asymmetry is predominantly because of slower inhibition to dark stimuli rather than to stronger excitation from the thalamocortical OFF pathway. Consistent with dark-dominant neurons having faster responses than light-dominant neurons, we find dark-dominance to solely occur in the early latencies of neurons' responses. Neurons that are strongly dark-dominated also tend to be less orientation-selective. This novel approach gives us new insight into the dark-dominance phenomenon and provides an avenue to address new questions about excitatory and inhibitory integration in cortical neurons. SIGNIFICANCE STATEMENT Neurons in the early visual cortex respond on average more strongly to dark than to light stimuli, but the mechanisms behind this bias have been unclear. Here we address this issue by combining single-unit electrophysiology with a novel machine learning model to analyze neurons' responses to natural image stimuli in primary visual cortex. Using these techniques, we find slower inhibition to light than to dark stimuli to be the leading mechanism behind stronger dark responses. This slower inhibition to light might help explain other empirical findings, such as why orientation selectivity is weaker at earlier response latencies. These results demonstrate how imbalances in excitation versus inhibition can give rise to response asymmetries in cortical neuron responses.

Abstract:The lightness of a surface depends not only on the amount of light reflected off, it but also on the context in which it is embedded. Despite a long history of research, neural correlates of context-dependent lightness perception remain a topic of ongoing debate. Here, we seek to expand on the existing literature by measuring functional magnetic resonance imaging (fMRI) responses to lightness variations induced by the context. During the fMRI experiment, we presented 10 participants with a dynamic stimulus in which either the luminance of a disk or its surround is modulated at four different frequencies ranging from 1 to 8 Hz. Behaviorally, when the surround luminance is modulated at low frequencies, participants perceive an illusory change in the lightness of the disk (lightness induction). In contrast, they perceive little or no induction at higher frequencies. Using this frequency dependence and controlling for long-range responses to border contrast and luminance changes, we found that activity in the primary visual cortex (V1) correlates with lightness induction, providing further evidence for the involvement of V1 in the processing of context-dependent lightness.

Abstract:A fundamental task of the visual system is to respond to both increases and decreases of luminance with action potentials (ON and OFF responses1-4). OFF responses are stronger, faster, and more salient than ON responses in primary visual cortex (V1) of both cats5,6 and primates,7,8 but in ferrets9 and mice,10 ON responses can be stronger, weaker,11 or balanced12 in comparison to OFF responses. These discrepancies could arise from differences in species, experimental techniques, or stimulus properties, particularly retinotopic location in the visual field, as has been speculated;9 however, the role of retinotopy for ON/OFF dominance has not been systematically tested across multiple scales of neural activity within species. Here, we measured OFF versus ON responses across large portions of visual space with silicon probe and whole-cell patch-clamp recordings in mouse V1 and lateral geniculate nucleus (LGN). We found that OFF responses dominated in the central visual field, whereas ON and OFF responses were more balanced in the periphery. These findings were consistent across local field potential (LFP), spikes, and subthreshold membrane potential in V1, and were aligned with spatial biases in ON and OFF responses in LGN. Our findings reveal that retinotopy may provide a common organizing principle for spatial modulation of OFF versus ON processing in mammalian visual systems.

Abstract:Human patients rendered cortically blind by lesions to V1 can nevertheless discriminate between visual stimuli presented to their blind fields. Experimental evidence suggests that two response modes are involved. Patients are either unaware or aware of the visual stimuli, which they are able to discriminate. However, under both conditions patients insist that they do not see. We investigate the fundamental difference between percepts derived for the normal and affected hemifield in a human hemianope with visual stimuli of which he was aware. The psychophysical experiments we employed required the patient, GY, to make comparisons between stimuli presented in his affected and normal hemifields. The subject discriminated between, and was allowed to match, the stimuli. Our study reveals that the stimulus parameters of colour and motion can be discriminated and matched between the normal and blind hemifields, whereas brightness cannot. We provide evidence for associations between the percepts of colour and motion, but a dissociation between the percepts of brightness, derived from the normal and hemianopic fields. Our results are consistent with the proposal that the perception of different stimulus attributes is expressed in activity of functionally segregated visual areas of the brain. We also believe our results explain the patient's insistence that he does not see stimuli, but can discriminate between them with awareness.

Asymmetric distribution of color-opponent response types across mouse visual cortex supports superior color vision in the sky

Abstract:Color is an important visual feature that informs behavior, and the retinal basis for color vision has been studied across various vertebrate species. While many studies have investigated how color information is processed in visual brain areas of primate species, we have limited understanding of how it is organized beyond the retina in other species, including most dichromatic mammals. In this study, we systematically characterized how color is represented in the primary visual cortex (V1) of mice. Using large-scale neuronal recordings and a luminance and color noise stimulus, we found that more than a third of neurons in mouse V1 are color-opponent in their receptive field center, while the receptive field surround predominantly captures luminance contrast. Furthermore, we found that color-opponency is especially pronounced in posterior V1 that encodes the sky, matching the statistics of natural scenes experienced by mice. Using unsupervised clustering, we demonstrate that the asymmetry in color representations across cortex can be explained by an uneven distribution of green-On/UV-Off color-opponent response types that are represented in the upper visual field. Finally, a simple model with natural scene-inspired parametric stimuli shows that green-On/UV-Off color-opponent response types may enhance the detection of "predatory"-like dark UV-objects in noisy daylight scenes. The results from this study highlight the relevance of color processing in the mouse visual system and contribute to our understanding of how color information is organized in the visual hierarchy across species.

Abstract:Magnetic resonance imaging sensitized to activity-related changes in cerebral blood oxygenation was performed to map responses to selective stimulation of the parvo- and magnocellular visual pathways in calcarine and adjacent ventral occipital cortex of human subjects. In a repetitive stimulation protocol isoluminant chromatic or isochromatic luminance modulation was alternated with steady light of the same mean chromaticity and luminance as a reference condition. While no significant effects were observed for diffuse luminance modulation, two consistent cortical foci responded to isoluminant chromatic stimulation. A strong response was obtained in calcarine cortex at both 2 and 10 Hz, and even for selective S-cone stimulation. A second weaker colorsensitive response was seen bilaterally in the collateral sulcus. Thus, the data not only confirm color-sensitive activation in the collateral sulcus elicited in previous studies by selective cognitive tasks, but additionally demonstrate color-sensitive activation in primary visual cortex. With stimuli defined according to electrophysiological response properties of early visual processing stages, this study complements phenomenological or cognitive approaches in functional mapping of the human visual system.

Cortical Processing. of A Brightness Illusion

Abstract:Several brightness illusions indicate that borders can affect the perception of surfaces dramatically. In the Cornsweet illusion, two equiluminant surfaces appear to be different in brightness because of the contrast border between them. Here, we report the existence of cells in monkey visual cortex that respond to such an "illusory" brightness. We find that luminance responsive cells are located in color-activated regions (cytochrome oxidase blobs and bridges) of primary visual cortex (V1), whereas Cornsweet responsive cells are found preferentially in the color-activated regions (thin stripes) of second visual area (V2). This colocalization of brightness and color processing within V1 and V2 suggests a segregation of contour and surface processing in early visual pathways and a hierarchy of brightness information processing from V1 to V2 in monkeys.
Joint representations of color and form in mouse visual cortex described by random pooling from rods and cones

Abstract:Abstract Spatial transitions in color can aid any visual perception task, and its neural representation – the “integration of color and form” – is thought to begin at primary visual cortex (V1). An integration of color and form is untested in mouse V1, yet studies show that the ventral retina provides the necessary substrate from green-sensitive rods and UV-sensitive cones. Here, we used two-photon imaging in V1 to measure spatial frequency (SF) tuning along four axes of rod and cone contrast space, including luminance and color. We first reveal that V1’s sensitivity to color is similar to luminance, yet average SF tuning is significantly shifted lowpass for color. Next, guided by linear models, we used SF tuning along all four color axes to estimate the proportion of neurons that fall into classic models of color opponency – “single-”, “double-”, and “non-opponent”. Few neurons (~6%) fit criteria for double-opponency, which are uniquely tuned for chromatic borders. Most of the population can be described as a unimodal distribution ranging from strongly single-opponent to non-opponent. Consistent with recent studies of the rodent and primate retina, our V1 data is well-described by a simple model in which ON and OFF channels to V1 sample the photoreceptor mosaic randomly.
Characterization of human lightness discrimination thresholds for independent spectral variations

Abstract:The lightness of an object is an intrinsic property that depends on its surface reflectance spectrum. The visual system estimates an object's lightness from the light reflected off its surface. However, the reflected light also depends on object extrinsic properties of the scene, such as the light source. For stable perception, the visual system needs to discount the variations due to the object extrinsic properties. We characterize this perceptual stability for variation in two spectral properties of the scene: the reflectance spectra of background objects and the intensity of light sources. We measure human observers’ thresholds of discriminating computer-generated images of 3D scenes based on the lightness of a spherical target object in the scene. We measured change in discrimination thresholds as we varied the reflectance spectra of the objects and the intensity of the light sources in the scene, both individually and simultaneously. For small amounts of extrinsic variations, the discrimination thresholds remained nearly constant indicating that the thresholds were dominated by observers’ intrinsic representation of lightness. As extrinsic variation increased, it started affecting observers’ lightness judgment and the thresholds increased. We estimated that the effects of extrinsic variations were comparable to observers’ intrinsic variation in the representation of object lightness. Moreover, for simultaneous variation of these spectral properties, the increase in threshold squared compared to the no-variation condition was a linear sum of the corresponding increase in threshold squared for the individual properties, indicating that the variations from these independent sources combine linearly.

Abstract:The retinal ganglion cells (RGCs) receive different combinations of L, M, and S cone inputs and give rise to one achromatic and two chromatic post-receptoral channels. Beyond the retina, RGC outputs are subject to filtering and normalization along the geniculo-striate pathway, ultimately producing the properties of human vision. The goal of the current study was to determine temporal sensitivity across the three post-receptoral channels in subcortical and cortical regions involved in vision. We measured functional magnetic resonance imaging (MRI) responses at 7 Tesla from three participants (two males, one female) viewing a high-contrast, flickering, spatially-uniform wide field (∼140°). Stimulus flicker frequency varied logarithmically between 2 and 64 Hz and targeted the L+M+S, L–M, and S–[L+M] cone combinations. These measurements were used to create temporal sensitivity functions of primary visual cortex (V1) across eccentricity, and spatially averaged responses from lateral geniculate nucleus (LGN), V2/V3, hV4, and V3A/B. Functional MRI responses reflected known properties of the visual system, including higher peak temporal sensitivity to achromatic vs. chromatic stimuli, and low-pass filtering between the LGN and V1. Peak temporal sensitivity increased across levels of the cortical visual hierarchy. Unexpectedly, peak temporal sensitivity varied little across eccentricity within area V1. Measures of adaptation and distributed pattern activity revealed a subtle influence of 64 Hz achromatic flicker in area V1, despite this stimulus evoking only a minimal overall response. Comparison of measured cortical responses to a model of integrated retinal output to our stimuli demonstrates that extensive filtering and amplification is applied to post-retinal signals. Significance Statement We report the temporal sensitivity of human visual cortex across the three canonical post-receptoral channels from central vision to the far periphery. Functional MRI measurements of responses from the LGN, V1, and higher visual cortical areas demonstrate modification of temporal sensitivity across the visual hierarchy. This includes amplification of chromatic signals between the LGN and V1, and an increase in peak temporal sensitivity in visual areas beyond V1. Within V1, we find a surprising stability of peak temporal sensitivity in the periphery for all three post-receptoral directions. Comparison of our results to a model of retinal output demonstrates the presence of substantial post-retinal filtering, yielding greater uniformity of responses across area V1 than would be predicted from unmodified retinal signals.

Large-scale color biases in the retinotopic functional architecture are region specific and shared across human brains

Abstract:Despite the functional specialization in visual cortex, there is growing evidence that the processing of chromatic and spatial visual features is intertwined. While past studies focused on visual field biases in retina and behaviour, large-scale dependencies between coding of color and retinotopic space are largely unexplored in the cortex. Here we asked whether spatial color biases are shared across different human observers, and whether they are idiosyncratic for distinct areas. We tested this by predicting the color a person was seeing using a linear classifier that has never been trained on chromatic responses from that same brain, solely by taking into account: (1) the chromatic responses in other individuals’ brains and (2) commonalities between the spatial coding in brains used for training and the test brain. We were able to predict the color (and luminance) of stimuli seen by an observer based on other subjects’ activity patterns in areas V1-V3, hV4 and LO1. In addition, we found that different colors elicited systematic, large-scale retinotopic biases that were idiosyncratic for distinct areas and common across brains. The area-specific spatial color codes and their conservation across individuals suggest functional or evolutionary organization pressures that remain to be elucidated.

Abstract:<p>Cells in the primary visual cortex (V1) generally respond weakly to large uniform luminance stimuli. Only a subset of V1 cells is thought to encode uniform luminance information. In natural scenes, local luminance is an important feature for defining an object that varies and coexists with local spatial contrast. However, the strategies used by V1 cells to encode local mean luminance for spatial contrast stimuli remain largely unclear. Here, using extracellular recordings in anaesthetized cats, we investigated the responses of V1 cells by comparing with those of retinal ganglion (RG) cells and lateral geniculate nucleus (LGN) cells to simultaneous and rapid changes in luminance and spatial contrast. Almost all V1 cells exhibited a strong monotonic increasing luminance tuning when they were exposed to high spatial contrast. Thus, V1 cells encode the luminance carried by spatial contrast stimuli with the monotonically increasing response function. Moreover, high contrast decreased luminance tuning of OFF cells but increased that of in ON cells in RG and LGN. The luminance and contrast tunings of LGN ON cells were highly separable as V1 cells, whereas those of LGN OFF cells were lowly separable. These asymmetrical effects of spatial contrast on ON/OFF channels might underlie the robust ability of V1 cells to perform luminance tuning when exposed to spatial contrast stimuli.</p>

Abstract:Abstract Some patients, who are rendered perimetrically blind in one hemifield by cortical lesions, nevertheless exhibit residual visual capacities within their field defects. The neural mechanism that mediates the residual visual responses has remained the topic of considerable debate. One explanation posits the subcortical visual pathways that bypass the primary visual cortex and innervate the extrastriate visual areas as the substrate that underlies the residual vision. The other explanation is that small islands of the primary visual cortex remain intact and provide the signals for residual vision. We have performed behavioral and functional magnetic resonance imaging experiments to investigate the validity of the two explanations of residual vision. Our behavioral experiments indicated that of the seven hemianopes tested, two had the ability to discriminate the direction of a drifting grating. This residual visual response was shown with fMRI to be the result of spared islands of calcarine cortical activity in one of the hemianopes, whereas only lateral occipital activity was documented in the other patient. These results indicate that the underlying neural correlates of residual vision can vary between patients. Moreover, our study emphasizes the necessity of ruling out the presence of islands of preserved function and primary visual cortex before assigning residual visual capacities to the properties of visual pathways that bypass the primary visual cortex.
