Hypoxia-inducible factor 1alpha expression in colorectal cancer and its correlation with N-myc downstream regulated gene

Abstract:Objective To investigate the correlation between the expression levels of N-myc downstream regulated gene 1(NDRG1)and hypoxia inducible factor-1α(HIF-1α)in tumor tissues of human colorectal carcinoma.Methods Plasmid expressing small interfering RNA(siRNA)against HIF- 1α(pSilence-2.1-U6-siRNA)was constructed and transfected into LS174T cells by Lipofectamine meth- ods.After transfection,cells were incubated at Hypoxia condition for 24 hours.The inhibitory effects of siR- NA against HIF-1αon expression of HIF-1αand NDRG1 genes were measured by semi-quantitative re- verse transcription PCR.In addition,mRNA levels of HIF-1αwere also analyzed by using in situ hybridiza- tion,whereas,NDRG1 protein levels in colorectal adenomas and primary colorectal adenocarcinomas were determined by immunohistochemistry.Results The successful construction of pSilenee-2.1-U6-siRNA plasmid was confirmed by sequencing analysis.After expression of siRNA against HIF-1αin LS174T cells, expressions of HIF-1αand NDRG1 gene were inhibited significantly.HIF-1αtranscripts were positive in 67.7%(42/62)and 44.4%(8/18)of colorectal adenocarcinoma and adenoma,respectively.The mean percentage of cells with positive hybridization of HIF-1αmRNA increases with the development from Duke stage A to stage C+D(P<0.05).The frequencies of HIF-1αmRNA positive cells in pathologic stages of the specimens were as follow:adenoma 8%,Dukes A 14%,Dukes B 23%,and Dukes C+D 35%.The positive staining rate of NDRG1 protein was significant higher in eolorectal adenocarcinoma group than that in colorectal adenoma group(P<0.05).The expression of NDRG1 protein correlates with Dukes stages very well.The level of HIF-1αwas positively correlated with the level of NDRG1 protein(P<0.05)dur- ing eolorectal tumor progression.Conclusion HIF-1αmay play an important role in tumor progression by regulating the expression of NDRG1 in human colorectal carcinomas.
