Abstract:P1068 Aims: To investigate the long-term effect on human embryonic kidney cell line HEK-293 after co-culture with porcine endogenous retrovirus (PERV). Methods: Pig kidney cell line PK15 supernatant through a 0.45-μm low protein-binding filter was used to infect HEK-293. PERV particles, reverse transcriptase and PERV RNA were detected by Electron microscopy, ELISA and RT-PCR respectively. Then PERV-gag, pol gene in the two-month infected HEK-293 cells were examined by PCR and RT-PCR. PERV-gag protein was further confirmed by immunofluorescence analysis. The morphology, total DNA/cell and cell doubling time were detected to estimate the long-term effect of PERV infected HEK-293 cells. Results: All retroviral particles in the harvested PK15 supernatant were round and different in size (70-240μm) under electron microscopy. The concentration of reverse transcriptase was 1.3pg / ml, which was nearly one thousand times lower than that of HIV-1 control (1034pg/ml). Viral RNA persisted in PK15 supernatant. PERV-gag, pol gene and gag protein expressed in the infected HEK-293 cells (Fig. 1, 2). While the morphology between the uninfected and infected HEK-293 was similar under light microscopy, HEK-293 cells are polygonal or spindle. The total DNA/cell and the cell doubling time had no significant difference between the uninfected and infected HEK-293, only PERV infected HEK-293 cells doubled little earlier than the control (Fig. 3, 4).Figure 1. The cell doubling time was detected by cell counting.Conclusions: PERV could infect HEK-293 in vitro and PERV gene could functionally express by the infected HEK-293 after two-month infection. While the morphology, total DNA/cell and cell doubling time of uninfected and infected HEK-293 had no significant difference. The further study of the infected cell genome, specific protein product and the variety PERV sequence are under conducting.
