Abstract:The process of preparing 4-hydroxy-3-methoxyphenylacetonitrile,as one of the important intermediates for synthesis was studied.Beginning from the raw material—vanillin,the title compound was prepared,via protection of hydroxyl,reduction,chloridization and nitrilation.The influences of various reaction conditions on the yield were investigated,and the optimal reaction conditions(reagent,temperature,time and yield) were as follows:acetic anhydride as acetylation reagent,reacting at room temperature for 3 h and yield was 99%;Raney Ni as catalyst,reaction time was 8 h at room temperature,and yield was 96.8%;dropping thionyldichloride at 0℃,then reacting at room temperature for 2 h,and the yield was 78%;refluxing with sodium cyanide for 9 h,and the yield was 78.5%.The products were characterized with MS,1H NMR and elemental analysis.The results showed that the process proposed overcame the defects of traditional synthesis route.

Abstract:A new route for the synthesis of 6-bromo-4,4-dimethylthiochroman was proposed and the optimized conditions for each step were obtained.Bromobenzene was selected as starting material and 6-bromo-4,4-dimethylthiochroman was prepared via chlorosulfonation,reduction,thioetherification and cyclization with the yield of 88.5%.Except the reduction,which had to be performed at a higher temperature,the other three steps proceeded at temperatures lower than 50oC.Thus both the energy cost and byproducts were decreased.Compared to previous routes,the proposed one in this work is advanced for its lower cost and less contamination,so it is suitable to be industrially amplified.

Abstract:A parallel process for the new synthesis of 2,7-dimethyl-2,4,6-octatriene-1,8-dial was investigated.3-(5,5-Dimethyl-1,3-dioxan-2-yl)-2-butenal was obtained by a process involving aldehyde protection,ester exchange and catalytic oxidation from 4-acetoxy-2-methyl-2-butenal.A new catalytic oxidation process was proposed to get 3-(5,5-dimethyl-1,3-dioxan-2-yl)-2-butenal by oxidating 3-(5,5-dimethyl-1,3-dioxan-2-yl)-2-buten-1-ol with copper(Ⅰ) chloride、2,2,6,6-tetramethyl-1-piperidinyloxy and oxygen.3-(5,5-Dimethyl-1,3-dioxan-2-yl)-2-buten-1-ol was chlorizated with SOCl_2 in DMF to get 2-(3-chloro-1-methyl-1-propenyl)-5,5-dimethyl-1,3-dioxane.2,7-Dimethyl-2,4,6-octatriene-1,8-dial was acquired in an overall yield of 48% by one-pot synthesis method from 3-(5,5-dimethyl-1,3-dioxan-2-yl)-2-butenal,2-(3-chloro-1-methyl-1-propenyl)-5,5-dimethyl-1,3-dioxane and triethylphosphite.

Abstract:The title compound, C20H47NO6Si4, was synthesized by per-O-trimethylsilylation of N-acetyl-d-glucosamine using chlorotrimethylsilane in the presence of hexamethyldisiloxane. The trimethylsilyl group and acetamido group are located on the same side of the pyran ring, showing an α-configuration glycoside. One of the trimethylsilyl groups is disordered over two orientations, with site-occupancy factors of 0.625 (9) and 0.375 (9). In the crystal, N—H⋯O hydrogen bonds link the molecules into supramolecular chains along the a-axis direction.

Abstract:In the presence of BF3. OEt2, 1-O-trimethylsilyl-2,3,4,6-tetra-O-benzyl-alpha -D-mannopyranose (1) reacted stereoselectively with a series of carboxylic acids to give the corresponding a-mannosyl carboxylates in good yields. The configuration of products was assigned mainly by H-1-NMR spectra (delta (H1) and (3)J(1,2)).A few glycosyl carboxylates have been found in nature and they show a series of important biological activities(1)). There have been numerous attempts to prepare them(2)).In the previous paper(3)), we have reported that 1-O-trimethylsilyl-2,3,4,6-tetra-O-benzyl-alpha -D-glucopyranose reacted with carboxylic acids to give the 1-O-alpha -acyl-D-glucopyranose with high stereoselectivity in good yield (58.8-84.0%). Now. we used the same method for the synthesis of mannosyl carboxylates. It is a highly stereoselective approach for the preparation of 1-O-acyl-D-mannopyranose with the intermediate 1.

Abstract:3, 3-Dimethoxypropanenitrile was synthesized from 2-chloro-3-phenylsulfonylpropanenitrile by elimination and addition reactions. 3-phenylsulfonylpropanenitrile was obtained by eliminating HCl from 2-chloro-3-phenylsulfonylpropanenitrile at 0-20℃ with a yield of 88%. 3-Phenylsulfonylpropanenitrile was reacted with sodium methoxide in methanol, giving 3, 3-dimethoxypropanenitrile with a yield of 98%. Elimination reaction was innovated by using THF as a solvent instead of ethyl ether and addition reaction was innovated by using an excess of sodium methoxide in methanol, so as to have the reaction condition optimized.

Abstract:An improved process was reported to solve the problems such as low yield and high costs in the synthesis of 3-n-propyl-2, 4-dihydroxyacetophenone, which is a key intermediate of Leukotriene receptor antagonist. The target compound was synthesized starting from m-dihydroxybenzene, via acylation, allylation, Claisen rearrangement and hydrogenation reduction. The optimal reaction conditions were as follows. The acylation reaction which used acetic acid as acylation agent and ZnCl 2 as catalyst was carried out under reflux for 5 h, and 77.9% yield was obtained. The allylation reaction using allyl chloride as allylation reagent and NaBr, KI as catalyst was carried out at 40°C for 2 h and continued at 70°C for 14 h, to give 98.6% yield. In Claisen rearrangement carried out at 205°C for 4.5 h, diphenyl ether was used as reaction solvent and NaCl was used as catalyst, and the yield was improved to 67.4%. In the final step, the hydrogenated reduction with Pd/C (w(Pd) is 5%) as catalyst was carried out at 45°C for 6 h, and 88.6% yield was obtained. The process is much of industrial value because of high yield, cheap and available materials, and convenient operations. The structure of the main compounds was confirmed by 1H-NMR, GC-MS and IR.

Abstract:Two new compounds of cluster mannosides were synthesized by a convenient method. Ally-D-mannopyranoside (2), a key intermediate, was obtained in good yield by Fischer's glycosidation method. Protection of 2 with acetyl group and the oxidization of double C C bond of allyl group with NaIO4/RuCl3 afforded compound 4. The coupling of 1,6-hexanediamine or tris(2-aminoethyl)amine with 4 was carried out by using NHS/DCC to give 5 and 7, respectively. After deprotection, target molecules 6 and 8 were obtained. Their structures were confirmed by IR, H-1 NMR, C-13 NMR and HRMS Spectra.

Abstract:A concise synthesis of 3-deoxy-3-nucleobase-2,5-anhydro-d-mannitol 6(a–d) has been achieved, using the deamination of 2-amino-2-deoxy-d-glucose to construct in one step the sugar skeleton with the desired sense of chirality at each asymmetric center. The selective dibenzoylation of 2,5-anhydro-d-mannitol 9 was investigated, and the key epoxide intermediate 13 was obtained in good yield via an intramolecular Mitsunobu reaction. The process of opening of epoxide 13 by nucleobases appeared to be regioselective.

Abstract:Some synthetic methods of 1-bromo-3,4,5-trimethoxybenzene BTMOB are compared. One can obtain good yield by Sandmeyer reaction after 3,4,5-trimethoxyaniline is diazotized. The optimum reaction conditions are determined: n (ArNH2):n(NaNO2):n(NaBr) = 1.0:1.2:1.5, temperature of diazotization below 5°C and the complex of diazonium salt refluxed for 1 h during decomposition. Yield of BTMOB is about 87.5%.

Abstract:2,5:3,4-Dianhydro-L-talofuranose dimethylacetal 7 was synthesized from D-glucose in 7 steps. A series of 4-deoxy-4-nucleobase-2,5-anhydro-L-mannitols 13-16 were synthesized regioselectively from 7 in good yields. 6-O-p-Tolylsulfonyl-2,5:3,4-dianhydro-L-talofuranose dimethylacetal 8 reacted with uracil or thymine to give the corresponding isonucleosides 17 and 19, but in the case of reaction of 8 with adenine, reformation of tetrahydrofuran ring took place, giving 4-(S)-adenyl-5-(R)-[1'-(R)-hydroxy-2',2'-dimethoxyl] ethyl-2,3-dihydrofuran 21 and 4-deoxy-4-adenyl-2,5:3,6-dianhydro-L-mannofuranose dimethylacetal 22. Abstract:The thiomannoside is a key intermediate in the synthesis of polymannosaccharide. But its crystallographic study has not been reported. X-ray crystal structure analysis shows that the crystal of phenyl 2,3,4,6-tetra-O-acetyl-1-thio-α-d-mannopyranoside is in orthorhombic system, has P212121 space group. Its unit cell dimensions are a=9.1731(11), b=11.574(2), and c=21.199(3)Å, α=β=γ=90.00°, Z=4, V=2250.6(6)Å3, Dc=1.300g/cm3. The crystallographic study reveals that the thiomannoside is an α-anomer. Most interestingly, the structure provides an evidence of existing C–H⋯O and C–H⋯S intramolecular and intermolecular hydrogen bonds.

Abstract:An efficient and reproducible process for the synthesis of methyl 2,3,4,5-tetradeoxy-7,8-O-isopropylidene-d-arabino-nanonate (2), a key intermediate in the total synthesis of (+)-biotin (1), starting from readily available d-mannose is described. The crucial part of this synthesis was the development of a practical route to a novel O-benzyl protected unsaturated ester methyl (benzyl 5,6,7,8-tetradeoxy-2,3-O-isopropylidene-α-d-lyxo-nona-5,7-dienofuranosid) uronate (7), allowing the one-step preparation of hydroxy ester methyl 5,6,7,8-tetradeoxy-2,3-O-isopropylidene-α-d-lyxo-nanofuranuronate (8) by the catalytic debenzylation and hydrogenation over palladium on carbon catalyst. This procedure requires no chromatographic purification, which makes it ideal for synthetic preparation on an industrial scale.

Abstract:An expeditious three-component, one-pot sequential glycosylation protocol has been developed for the preparation of 3,6-branched unsymmetrical mannopentaose (Man5), employing a mannose trisaccharide donor, a mannose monosaccharide donor and a mannose monosaccharide acceptor.
