Abstract:In this study, in vitro induction of tetraploid Lychnis senno Siebold et Zucc. and its cytological and morphological characterization were conducted. For polyploid induction, nodal segments with axillary buds from in vitro grown plants were kept for 3 days in MS (Murashige and Skoog, 1962) liquid or solid media added with a series of concentrations of colchicine. Out of total 588 recovered plants, 15 tetraploids and 6 mixoploids determined by flow cytometry analysis were obtained. The tetraploid contained 48 chromosomes, twice the normal diploid number of 24, as observed under light microscope. The tetraploid plants exhibited much larger but less stomata than diploid plants. Moreover, significant differences in stem height and leaf size between the diploid and tetraploid plants were noted. The tetraploid plants were more compact than diploids.

Abstract:The results demonstrated that the same species of different geographical populations have the same amount of chromosome and karyotypic formulae. Rana limmocharis boie and Rana j. Japonica have 2n=26, NF=52 and 5+8 karyotypic formulae. Microhyla ornata has 2n=24, NF=48 and 6+6 karyotypic formulae. But some dissimilarities were found among them. First, the number and sequence of submetacentric chromosome were different among them, and then the secondary contriction (SC) or satellite (Sat) were also different. It was showed that the chromosomes of the same species of different geographical population have diversities.
Analysis of cytogenetics for the aneuploids in pentaploid progenies from intergeneric cross between Brassica napus and Orychophragmus violaceus

Abstract:Cytological observation on progenies of intergeneric pentaploids between Brassica napus and Orychophragmus violaceus was conducted and the results indicated that frequency of cells with 47, 44 and 41 chromosomes in somatic cells or pollen mother cells (PMCs) was higher than that with other chromosome number in aneuploids among the aneuploid progenies. Except for the plants with 38 chromosomes, they were mixoploid with extra chromosomes derived most likely from O. violaceus. Chromosome elimination, due to exclusion from telophase nuclei and sebsequent digestion of the extra chromosomes, occurred in all PMCs (2n>38). Some modes of segregation that were apt to cause symmetrical distribution of chromosome in PMCs at the metaphase I (MI) stage. Moreover, several special segregation modes were discovered in PMCs which led to aggregation of 18 or 17 chromosomes at this stage. In each of three parts (anaphase I (AI) stage of PMCs, female gametes and male gametes), chromosome number was lowered in plants with 44 or 41 chromosomes according to gamete constitutes and frequency. Among these three parts, the ratio of male gametes with decreased chromosome number was larger than both of others. Gametes with lower chromosome numbers (n=22-19) were more easily transferred to offsprings. Somatic chromosome diminution was observed in root tips of seedling according to their somatic constituent.

Abstract:Objective:For better prenatal diagnosis,2397cases of karyotypes of amniotic fluid cell were analysed.Methods:Amniotic fluid cells were cultured in silu and their karyotypes were analyzed by G band,followed them after delivery.Results :65 abnormal karyotypes were found from 2397 cases among them.Conclusion:It is necessary to analysis karyotypes of amniotic fluid cell for high risk gravida,combined with molecule diagnosis can be better to prenatal diagnosis.

Abstract:The lotus (Nelumbo Adans.) is a perennial aquatic plant with important value in horticulture, medicine, food, religion, and culture. It is rich in germplasm and more than 2000 cultivars have been cultivated through hybridization and natural selection. Microsporogenesis and male gametogenesis in the anther are important for hybridization in flowering plants. However, little is known about the cytological events, especially related to the stamen, during the reproduction of the lotus. To better understand the mechanism controlling the male reproductive development of the lotus, we investigated the flower structure of the Asian lotus (N. nucifera). The cytological analysis of anther morphogenesis showed both the common and specialized cytological events as well as the formation of mature pollen grains via meiosis and mitosis during lotus anther development. Intriguingly, an anatomical difference in anther appendage structures was observed between the Asian lotus and the American lotus (N. lutea). To facilitate future study on lotus male reproduction, we categorized pollen development into 11 stages according to the characterized cytological events. This discovery expands our knowledge on the pollen and appendage development of the lotus as well as improving the understanding of the species differentiation of N. nucifera and N. lutea.

Abstract:The lotus (Nelumbo Adans.) is an important aquatic plant with ornamental, medicinal and edible values and cultural connotations. It has single-, semi-double-, double- and thousand-petalled types of flower shape and is an ideal material for developmental research of flower doubling. The lotus is a basal eudicot species without a morphological difference between the sepals and petals and occupies a critical phylogenetic position in flowering plants. In order to investigate the genetic relationship between the sepals and petals in the lotus, the class E genes which affect sepal formation were focused on and analyzed. Here, SEPALLATA 1(NnSEP1) and its homologous genes AGAMOUS-LIKE MADS-BOXAGL9 (NnAGL9) and MADS-BOX TRANSCRIPTION FACTOR 6-like (NnMADS6-like) of the class E gene family were isolated from the flower buds of the Asian lotus (Nelumbo nucifera Gaertn.). The protein structure, subcellular localization and expression patterns of these three genes were investigated. All three genes were verified to locate in the nucleus and had typical MADS-box characteristics. NnSEP1 and NnMADS6-like were specifically expressed in the sepals, while NnAGL9 was highly expressed in the petals, suggesting that different developmental mechanisms exist in the formation of the sepals and petals in the lotus. The significant functional differences between NnSEP1, NnMADS6-like and NnAGL9 were also confirmed by a yeast two-hybrid assay. These results expand our knowledge on the class E gene family in sepal formation and will benefit fundamental research on the development of floral organs in Nelumbo.

Characterization of flower-bud transcriptome and development of genic SSR markers in Asian lotus (Nelumbo nucifera Gaertn.)

Abstract:Background: Asian lotus (Nelumbo nucifera Gaertn.) is the national flower of India, Vietnam, and one of the top ten traditional Chinese flowers. Although lotus is highly valued for its ornamental, economic and cultural uses, genomic information, particularly the expressed sequence based (genic) markers is limited. High-throughput transcriptome sequencing provides large amounts of transcriptome data for promoting gene discovery and development of molecular markers. Results: In this study, 68,593 unigenes were assembled from 1.34 million 454 GS-FLX sequence reads of a mixed flower-bud cDNA pool derived from three accessions of N. nucifera. A total of 5,226 SSR loci were identified, and 3,059 primer pairs were designed for marker development. Di-nucleotide repeat motifs were the most abundant type identified with a frequency of 65.2%, followed by tri- (31.7%), tetra- (2.1%), penta- (0.5%) and hexa-nucleotide repeats (0.5%). A total of 575 primer pairs were synthesized, of which 514 (89.4%) yielded PCR amplification products. In eight Nelumbo accessions, 109 markers were polymorphic. They were used to genotype a sample of 44 accessions representing diverse wild and cultivated genotypes of Nelumbo. The number of alleles per locus varied from 2 to 9 alleles and the polymorphism information content values ranged from 0.6 to 0.9. We performed genetic diversity analysis using 109 polymorphic markers. A UPGMA dendrogram was constructed based on Jaccard's similarity coefficients revealing distinct clusters among the 44 accessions. Conclusions: Deep transcriptome sequencing of lotus flower buds developed 3,059 genic SSRs, making a significant addition to the existing SSR markers in lotus. Among them, 109 polymorphic markers were successfully validated in 44 accessions of Nelumbo. This comprehensive set of genic SSR markers developed in our study will facilitate analyses of genetic diversity, construction of linkage maps, gene mapping, and marker-assisted selection breeding for lotus.

Abstract:Abstract Background Lotus ( Nelumbo nucifera ) is an aquatic plant with important agronomic, horticulture, art and religion values. It was the basal eudicot species occupying a critical phylogenetic position in flowering plants. After the domestication for thousands of years, lotus has differentiated into three cultivated types -flower lotus, seed lotus and rhizome lotus. Although the phenotypic and genetic differentiations based on molecular markers have been reported, the variation on whole-genome level among the different lotus types is still ambiguous. Results In order to reveal the evolution and domestication characteristics of lotus, a total of 69 lotus accessions were selected, including 45 cultivated accessions, 22 wild sacred lotus accessions, and 2 wild American lotus accessions. With Illumina technology, the genomes of these lotus accessions were resequenced to > 13× raw data coverage. On the basis of these genomic data, 25 million single-nucleotide polymorphisms (SNPs) were identified in lotus. Population analysis showed that the rhizome and seed lotus were monophyletic and genetically homogeneous, whereas the flower lotus was biphyletic and genetically heterogeneous. Using population SNP data, we identified 1214 selected regions in seed lotus, 95 in rhizome lotus, and 37 in flower lotus. Some of the genes in these regions contributed to the essential domestication traits of lotus. The selected genes of seed lotus mainly affected lotus seed weight, size and nutritional quality. While the selected genes were responsible for insect resistance, antibacterial immunity and freezing and heat stress resistance in flower lotus, and improved the size of rhizome in rhizome lotus, respectively. Conclusions The genome differentiation and a set of domestication genes were identified from three types of cultivated lotus- flower lotus, seed lotus and rhizome lotus, respectively. Among cultivated lotus, flower lotus showed the greatest variation. The domestication genes may show agronomic importance via enhancing insect resistance, improving seed weight and size, or regulating lotus rhizome size. The domestication history of lotus enhances our knowledge of perennial aquatic crop evolution, and the obtained dataset provides a basis for future genomics-enabled breeding.

Abstract:This comprehensive review systematically examines the multifarious aspects of Nelumbo nucifera , elucidating its ecological, nutritional, medicinal, and biomimetic significance. Renowned both culturally and scientifically, Nelumbo nucifera manifests remarkable adaptability, characterized by its extensive distribution across varied climatic regions, underpinned by its robust rhizome system and prolific reproductive strategies. Ecologically, this species plays a crucial role in aquatic ecosystems, primarily through biofiltration, thereby enhancing habitat biodiversity. The rhizomes and seeds of Nelumbo nucifera are nutritionally significant, being rich sources of dietary fiber, essential vitamins, and minerals, and have found extensive culinary applications. From a medicinal perspective, diverse constituents of Nelumbo nucifera exhibit therapeutic potential, including anti-inflammatory, antioxidant, and anti-cancer properties. Abstract:MYB superfamily is one of the most abundant transcription factors in plants, which are involved in many physiological and biochemical processes. Among them, R2R3 MYB might be the most widely studied subfamily. Here, we characterized the gene structures, expressional patterns, and potential functions of 116 R2R3 MYB genes in a genome-wide manner in lotus (Nelumbo nucifera). Compared with those in Arabidopsis and grape, the lotus R2R3 MYB genes are conserved in exon’s number and length. Most of them constitute three exons and two introns with the first two exons being highly conserved in their length. According to the structure of exon and DNA-binding domain-imperfect repeat (R), these R2R3 MYB genes were classified into four groups. The expression of 13 candidate MYB genes that are related to flavonoid biosynthesis was analyzed in lotus different tissues. One gene expresses highly in all tissues and might positively regulate common flavonoid biosynthesis. Two genes might positively regulate anthocyanin and proanthocyanidin biosynthesis, respectively, resulting in the pigmentation of flower and seed. While five members that are similar to Zm38 are expressed lowly in different tissues. This genome-wide analysis of R2R3 MYB genes in lotus may provide more comprehensive insights on the structure and function of this gene family, and hence contribute a lot to plant’s genetic engineering in improving the colorization of flowers or other tissues.

Abstract:NAC (NAM, ATAF, and CUC) is a ubiquitously expressed plant-specific transcription factor (TF) family which is involved in the regulation of various biological processes. However, a systematic characterization of NAC gene family is yet to be reported in lotus. Here, 82 NnNAC genes which included five predicted membrane-bound NAC proteins were identified in the lotus genome. Phylogenetic analysis revealed seven-subfamily clusters (I–VII) of NnNAC proteins, with homologous gene pairs displaying similar conserved motifs and gene structure characteristics. Transactivation assay of NnNAC proteins revealed an extensive transcriptional activation capacity which is mediated by the highly divergent C-terminal activation domain (AD). Expression analysis of NnNAC genes in lotus tissues showed high transcript levels in root, stamen, petal and seed coat. In addition, 30 and 29 differentially expressed NnNAC candidate genes putatively involved in lotus seed development and response to complete submergence stress, respectively, were identified. Overall, our study provides potentially useful candidate gene resources for future molecular breeding of lotus varieties with novel agronomic traits.

Abstract:Lotus is a good source of medical herbs. Analyzing lotus components will enable us to make better use of lotus resources. Compounds of nelumbo lotus have been analyzed by using GC-MS and GC, and 48 volatile compounds have been identified from nelumbo lotus which counted up to 92.5% of the total peak area, and 4 alkaloids of nelumbo lotus have been identified by GC-MS.

Abstract:Abstract Background: Lotus ( Nelumbo nucifera ) is a famous flower with high ornamental value. Flower color and flower morphology are two main factors for flower lotus breeding. Petaloid is a universal phenomenon in lotus flowers. However, the genetic regulation of floral organ petaloid in lotus still remains unclear. Result: In this study, transcriptomic analysis was performed among five organs, including petal, stamen petaloid, stamen, carpel petaloid, and carpel in lotus. Using WGCNA analysis, 37 candidate genes were found to be related to carpel petaloid. Additionally, one floral homeotic gene encoded MADS box transcription factor, AGAMOUS (AG) , was identified as candidate gene for petaloid in lotus. Conclusion: The above results explored the candidate genes related to petaloid, setting a theoretical basis for the molecular regulation of petaloid.

Abstract:Genetic and physical maps are powerful tools to anchor fragmented draft genome assemblies generated from next-generation sequencing. Currently, two draft assemblies of Nelumbo nucifera, the genomes of 'China Antique' and 'Chinese Tai-zi', have been released. However, there is presently no information on how the sequences are assembled into chromosomes in N. nucifera. The lack of physical maps and inadequate resolution of available genetic maps hindered the assembly of N. nucifera chromosomes. Here, a linkage map of N. nucifera containing 2371 bin markers [217 577 single nucleotide polymorphisms (SNPs)] was constructed using restriction-site associated DNA sequencing data of 181 F2 individuals and validated by adding 197 simple sequence repeat (SSR) markers. Additionally, a BioNano optical map covering 86.20% of the 'Chinese Tai-zi' genome was constructed. The draft assembly of 'Chinese Tai-zi' was improved based on the BioNano optical map, showing an increase of the scaffold N50 from 0.989 to 1.48 Mb. Using a combination of multiple maps, 97.9% of the scaffolds in the 'Chinese Tai-zi' draft assembly and 97.6% of the scaffolds in the 'China Antique' draft assembly were anchored into pseudo-chromosomes, and the centromere regions along the pseudo-chromosomes were identified. An evolutionary scenario was proposed to reach the modern N. nucifera karyotype from the seven ancestral eudicot chromosomes. The present study provides the highest-resolution linkage map, the optical map and chromosome level genome assemblies for N. nucifera, which are valuable for the breeding and cultivation of N. nucifera and future studies of comparative and evolutionary genomics in angiosperms.

Abstract:BackgroundFlower morphology, a phenomenon regulated by a complex network, is one of the vital ornamental features in Nelumbo nucifera. Stamen petaloid is very prevalent in lotus flowers. However, the mechanism underlying this phenomenon is still obscure.ResultsHere, the comparative transcriptomic analysis was performed among petal, stamen petaloid and stamen through RNA-seq. Using pairwise comparison analysis, a large number of genes involved in hormonal signal transduction pathways and transcription factors, especially the MADS-box genes, were identified as candidate genes for stamen petaloid in lotus.ConclusionsTaken together, these results provide an insight into the molecular networks underlying lotus floral organ development and stamen petaloid.

Abstract:Leucanthemum paludosum is a small, popular, perennial herbaceous flower that is widely used as a potted plant and in flower beds. It is highly adaptable and fast-growing, blooms from early to late spring, and can be maintained for two-three months, thus making it a potential model plant for cytological studies. However, it lacks applicable markers for identifying its chromosome numbers, and no karyotyping or cytological study about L. paludosum has been reported. In the present study, we aimed to analyze the morphology and chromosomal status of L. paludosum to provide a reference for its systematic utilization. The results showed that L. paludosum is diploid (2n = 2x = 18). The bivalent number was substantially higher than the monovalent bodies. L. paludosum also conducted a loosely patterned karyotype, with no fluorescence in situ hybridization (FISH) signal when using a conventional oligonucleotide (oligo) probe library. To address this problem, we developed a novel repetitive oligo probe ((ATC)10) using transcriptome sequences. Furthermore, we explored its feasibility in different plants and provided effective repetitive oligoprobes for species such as Avena sativa, Vicia faba, Allium sativum, Lilium davidii var. unicolor, and Antirrhinum majus. Our study provides new cytological data for L. paludosum and developed a novel method for designing oligo probes for karyotype detection.
Molecular cytogenetic study on the plants of Elymus nutans with varying fertility on the Qinghai Plateau

Abstract:A molecular cytogenetic investigation was conducted on plants of the allohexaploid species Elymus nutans with varying fertility on the Qinghai-Tibet Plateau. Molecular karyotyping revealed that chromosome variants were distributed unevenly among genomes and among different homologue chromosomes in each genome. The plants with varying fertility exhibited significantly higher numbers of chromosome variants than did the normal fertility samples, although both kinds of plants showed the same pattern of high-to-low polymorphism from the Y to St and H genomes. Heterozygosis and karyotype heterozygosity in the plants with varying fertility were 3- and 13-fold higher than those in normal samples, respectively. Significant negative correlations were found not only between seed setting rates and total genome heterozygosity but also between seed setting rates and heterozygosity of each genome in the plants of varying fertility. Chromosome pairing analysis was performed using genomic in situ hybridization in selected plants of different fertility levels. The pairing of chromosomes at meiotic metaphase I was mostly bivalent, although univalent, trivalent, quadrivalent, and other polyvalents also occurred; in addition, chromosome configuration forms and frequencies varied among the studied samples. ANOVA results showed that the average number of ring bivalents in the Y genome was significantly higher than those in the St and H genomes. Significant positive correlations between pollen grain fertility and ring bivalent number were found in the St and H genomes but not in the Y genome. Furthermore, chromosome configuration parameters (total bivalents, numbers of ring and rod bivalents) were found to be significantly correlated with heterozygosity and seed setting rates in the St and H genomes, respectively, but not in the Y genome. It was inferred that the seed setting rate and pollen grain fertility in E. nutans are strongly influenced by the heterozygosity of each genome, but the Y genome differs from the St and H genomes due to chromosome pair alterations. The St and H genomes may contain more chromosome structural variations than the Y genome in E. nutans.

Abstract:Under pot culture condition, growth characters and some physiological indexes of above-silt part ( leaf and flower ) and under-silt part ( rhizome ) of different cultivars ( rhizome lotus cultivar'Dazihong' , flower lotus cultivar'Zhongguohongshanghai' and dual purpose of flower and rhizome lotus cultivar 'Yixianlian ' ) of Nelumbo nucifera Gaertn. were compared and researched, and correlation analysis on growth characters and some physiological indexes of above-silt part and rhizome of three cultivars of N. nucifera were conducted. The results show that'Dazihong' does not flower throughout the growth and development period. 'Zhongguohongshanghai' has a relatively early flowering time ( the first ten days of June) , with double flower and bright color. 'Yixianlian' has a relatively late flowering time ( the first and middle ten days of July) , with single flower and white color. Among three cultivars of N.nucifera , number of standing leaves of'Dazihong' is the least, standing leaf area is the biggest, rhizome expands obviously, and contents of soluble sugar and soluble protein in rhizome are the highest;number of standing leaves of 'Zhongguohongshanghai' is the most, chlorophyll content in standing leaf is the highest, internode length of rhizome is the longest, but rhizome does not expand obviously; number of standing leaves of 'Yixianlian' is in middle, petiole length is the longest, rhizome expands obviously, and contents of soluble protein, soluble sugar, starch and vitamin C in rhizome are between those of other two cultivars of N. nucifera. The correlation analysis result shows that correlation of standing leaf area of N. nucifera with internode length of rhizome is significantly negative, while that with contents of soluble sugar, soluble protein and vitamin C in rhizome is obviously significantly or significantly positive;correlation of petiole length with diameter and internode mass of rhizome is significantly positive;correlation of chlorophyll content in standing leaf with internode length of rhizome and starch content in rhizome is significantly positive, while that with soluble sugar content in rhizome is significantly negative. It is suggested that growth, development and nutritional quality of rhizome can be indirectly judged by standing leaf area, petiole length and chlorophyll content in standing leaf of three cultivars tested of N. nucifera, which can be further applied to rapid selection of dual purpose of flower and rhizome lotus cultivar.
