Abstract:Two undescribed protostane triterpenoids, 11‐deoxy‐13(17),15‐dehydro‐alisol B 23‐acetate (2) and alisol S (3), together with 21 known ones (1, 4‐23), were isolated from the dried rhizome of Alisma plantago‐aquatica. Of these compounds, 13(17),15‐Dehydro‐alisol B 23‐acetate (1) and 11‐deoxy‐alisol Q 23‐acetate (2) are two protostane trierpenoids containing conjugated double bonds in the five‐membered ring D that are rarely found from nature resource, while alisol S (3) is a protostane triterpenoid with undescribed tetrahydrofuran moiety linked via C20‐O‐C24 at the side chain. Additionally, compound 18 is a new natural product, and cycloartenol triterpenoid 23 is a non protostane triterpenoid firstly isolated from genus Alisma. Their structures were elucidated by extensive spectral analysis of the UV, IR, MS, 1D and 2D NMR, and comparison of the experimental and calculated CD curves.

Abstract:AIM: To study the chemical constituents of the rhizome of Alisma orientale. METHODS: Silica gel column chromatography was used to isolate the compounds, and spectroscopic techniques were employed to elucidate the structures. RESULTS: One new protostane triterpenoid, 24 (R)-hydroxypmtosta-11, 13-diene 16 (S), 23 (S)-epoxide, named 24-deacetyl alisol O (1), together with 3 known compounds, alisol O (2), alisol F 24-acetate (3) and alisol A 24-acetate (4), was obtained from the rhizome of Alisma oriental. CONCLUSION: Compound 1 is a new protostane triterpenoid.

Abstract:Sixteen triterpenoids and nine sesquiterpenoids were isolated from the rhizome of Alisma orientate. Structures of 16-oxo-11-anhydroalisol A 24-acetate, 13 beta,17 beta-epoxy-24,25,26,27-tetranor-alisol A 23-oic acid, 1 alpha H,5 alpha H-guaia-6-ene-4 beta,10 beta-diol, and alisguaiaone were elucidated by comprehensive spectroscopic data analysis. The cytotoxic, antibacterial, antifungal, anti-inflammatory, and alpha-glucosidase inhibitory activities of isolated terpenoids were evaluated. Triterpenoids alisol A, alisol A 24-acetate, 25-O-ethylalisol A, 11-deoxyalisol A, alisol E 24-acetate, alisol G, alisol B 23-acetate and sesquiterpenoids 10-hydroxy-7,10-epoxysalvialane exhibited cytotoxicities against the three tested human cancer cell lines with IC50 values ranging from 11.5 +/- 1.7 mu M to 76.7 +/- 1.4 mu M. Triterpenoids alisol A, 25-O-ethylalisol A, 11-deoxyalisol A, alisol E 24-acetate, alisol G, and 25-anhydroalisol F showed antibacterial activities against the Gram-positive strains Bacillus subtilis and Staphylococcus aureus with MIC values of 12.5-100 mu g/mL. Sesquiterpenoid 4 beta,10 beta-dihydroxy-1 alpha H,5 beta H-guaia-6-ene exhibited antibacterial activity against B. subtilis with an MIC value of 50 mu g/mL, and 10-hydroxy-7,10-epoxysalvialane exhibited activity against S. aureus with an MIC value of 100 Compounds 16-oxo-il-anhydroalisol A 24-acetate, alisol F, 25-anhydroalisol F, and alisguaiaone exhibited inhibitory effects on lipopolysaccharide-induced NO production in RAW 264.7 macrophage cells. Abstract:Twenty‐eight protostane triterpenoids, including a new degraded one (1), nine new ones (2 – 10), and two new natural ones (11 and 12), have been isolated from the dried rhizomes of Alisma orientale. Alisol R (1) was the first 20,21,22,23,24,25,26,27‐octanorprotostane triterpenoid. The absolute configurations of 25‐methoxyalisol F (2) and 16β‐hydroperoxyalisol B 23‐acetate (3) were determined by X‐ray diffraction analysis. In addition, alismaketone‐B 23‐acetate (28) showed potent vasorelaxant activity on endothelium‐intact thoracic aorta rings precontracted with KCl.

Abstract:A nor-protostane, alisolide (1), a rearranged protostane, alisol O (2), and a 2,3-seco-protostane triterpene, alisol P (3), were isolated from the rhizomes of Alisma orientale, along with eight known protostane triterpenes. The structures were elucidated to be (17S)-3,11-dioxo-23-nor-protost-12-en-23(17)-olide, 3-oxo-11beta,23-dihydroxy-24,24-dimethyl-26,27-dinorprotost-13(17)-en-25-oic acid, and (20R,23S,24R)-23,24,25-trihydroxy-2,3-seco-protost-13(17)-en-3-oic acid 2,11beta-lactone, respectively, by interpretation of spectroscopic data.

Abstract:An abietane diterpenoid, triptobenzene Y and six sesquiterpene polyol esters, wilforsinines C–H, together with 14 known compounds, have been isolated from the roots of Tripterygium wilfordii. The structures of the compounds were elucidated on the basis of spectroscopic analyses. The quinone reductase (QR) induction assay indicated that two compounds showed moderate QR-inducing activities at concentrations of 25μM and 50μM, respectively.

Abstract:A new triterpenoid named alisol O ( 1) was isolated from the rhizomes of Alisma orientalis, together with six known compounds: alisol A 24-acetate ( 2), 25-anhydroalisol A ( 3), 13 beta,17 beta-epoxyalisol A ( 4), alisol B 23-acetate ( 5), alisol F ( 6), and alisol F 24-acetate ( 7). Based on 1D and 2D-NMR data (HMQC, HMBC, COSY, ROESY), the structure of the new compound was deduced to be 11-dehydroxy-12-dehydroalisol F-24-acetate ( 1). Compounds 2 - 7 exhibited inhibitory activity in vitro on hepatitis B virus (HBV) surface antigen (HBsAg) secretion of the Hep G2.2.15 cell line with IC (50) values of 2.3, 11.0, 15.4, 14.3, 0.6 and 7.7 microM, and on HBV e antigen (HBeAg) secretion with IC (50) values of 498.1, 17.6, 41.0, 19.9, 8.5 and 5.1 microM, respectively.

Abstract:Abstract A new protostane-type triterpenoid, 5β,29-dihydroxy alisol A (1) was isolated from Alisma plantago-aquatica subsp. orientale (Sam.) Sam. as well as 12-deoxyphorbol-13α-pentadecanoate (2). We first report the presence of compound 2 in the genus Alisma. Their structures were established on the basis of 1D and 2D NMR, and HRESIMS spectroscopic analyses. All the isolated compounds were assayed for their inhibitory effects against human carboxylesterase 2 (HCE-2). Compounds 1 and 2 displayed inhibitory activities against HCE-2 with IC50 values of 29.2 and 4.6 μM, respectively. The interaction mechanisms of HCE-2 with compounds 1 and 2 were investigated by molecular docking, respectively.

Abstract:Two new oleanane-type triterpenoids, (3E, 23E) -3-caffeoyl-23-coumaroylhederagenin (1) and (3E, 23E) -dicoumaroylhederagenin (2), together with two known analogues were isolated from the stems of traditional Chinese medicine Alangium chinense.The structures of two new compounds were determined by the spectroscopic method, and the known compounds were defined as (23E) -coumaroyl-hederagenin (3) and (23Z) -coumaroylhederagenin (4) based on the observed and reported NMR da-ta.All compounds were isolated from the genus Alangium for the first time.

Abstract:Four pairs of undescribed enantiomeric guaiane sesquiterpenoids, (±)-alismaenols A-D ( 1a/1b, 3a/3b-5a/5b ), together with a pair of known ones ( 2a/2b ) were isolated from the rhizomes of Alisma plantago-aquatica . The structures and relative configurations of the isolates were established by analysis of their 1D, 2D-NMR and HRESIMS data. Their absolute configurations were determined by comparison of their experimental CD spectra and calculated electronic circular dichroism (ECD) spectra or by single-crystal X-ray diffraction analysis. All compounds ( 1a / 1b - 5a / 5b ) were evaluated for their inhibitory effects on nitric oxide (NO) production in LPS-induced RAW 264.7 cells, and compound 1a exhibited stronger activity (IC 50 = 12.89 μ M) than indomethacin (IC 50 = 14.03 μ M).

Abstract:Twenty-one protostane-type triterpenoids with diverse structures, including nine new compounds (1-9), were isolated from the of Alisma plantago-aquatica Linn. Structurally, alisolides A-F (1-6), composed of an oxole group coupled to a five-membered ring, represent unusual C-17 spirost protostane-type triterpenoids. Alisolide H (8) is a novel triterpenoid with an unreported endoperoxide bridge. Alisolide I (9) represents the first example of 23,24-acetal triterpenoid. Their structures were elucidated based on spectroscopic analysis, wherein the absolute configurations of 4-6, 8 were further confirmed by the Mo-2(OAc)(4)-induced ECD method. Furthermore, all isolates were evaluated for their inhibitory effects on LPS-induced NO production in Caco-2 cells, and all the compounds showed remarkable inhibitory activities, with IC50 values in the range of 0.76-38.20 mu mol/L. Abstract:From the roots of Helicteres angustifolia, collected from Anhui province, two triterpenoids of oleanolic acid type were obtained and were identified as new compounds, methyl 3beta-hydroxyolean-12-en-27-benzoyloxy-28-oate(1) and 3beta-O-(p-hydroxy-(E)-cinnamoyl)-12 oleanen-28-oic acid(2). The variety of spectroscopic methods such as IR, HRMS, EIMS, H-H COSY, H-C COSY NMR have been applied in the structural elucidation of these two compounds.

Abstract:Two new compounds (1) and (2) were obtained from the roots and rhizomes of Trillium tschonoskii, and their structures were identified as (2,3-S-trans,10R,6E)-7,11-dimethyl-3-methylene-1,6-dodecadien-10,11-diol 10-O-β-d-glucopyranosyl-(1→4)-O-β-d-glucopyranosyl-(1→4)-O-β-d-glucopyranoside (1) and (23S,24S,25S)-spirost-5-en-1β,3β,21,23,24-pentaol-1-O-β-d-xylopyranosyl-(1→3)-[O-α-l-rhamnopyranosyl-(1→2)]-O-α-l-arabinopyranoside (2) on the basis of physico-chemical properties and spectral analysis. In addition, cytotoxic activity toward HepG2 cell line was tested by the MTT method, and compound 2 showed strong cytotoxicity to the HepG2 cell (IC50 0.499μmolL−1).

Abstract:A new phenylpropanoid allopyranoside (1) and a new indolinone alkaloid (2) were isolated from the rhizomes of Actaea dahurica (syn. Cimicifuga dahurica). The structures of those two compounds were deduced as cimicifugaside F (1) and 3E,11E-(3-methyl-2-butenylidene acid)-2-indolinone-1-O-β-d-glucopyranoside (2) by detailed analysis of their MS, 1D and 2D NMR data and comparison with literatures. Additionally, the isolates were evaluated for their inhibitory effects on the production of NO by LPS-stimulated RAW 264.7 macrophages.

Abstract:Eight pentacyclic triterpenoids were isolated from the methanol extract of the roots of Rhodomyrtus tomentosa. By application of spectroscopic methods (especially 1D-, 2D-NMR, MS), the structures were established as 23-cis-p-coumaroyloxy-2 alpha,3 beta-dihydroxyolean-12-en-28-oic acid (1), 23-trans-p-coumaroyloxy-2 alpha,3 beta-dihydroxyolean-12-en-28-oic acid (2), 3 beta-O-trans-ferulyl-2 alpha,23-dihydroxyolean-12-en-28-oic acid (3), 3 beta-O-trans-p-coumaroyl-2 alpha,23-dihydroxyolean-12-en-28-oic acid (4), 3 beta-O-cis-p-coumaroyl-2 alpha,23-dihydroxyolean-12-en-28-oic acid (5), maslinic acid (6), arjunolic acid (7), and 2 alpha,3 beta-dihydroxytaraxer-20-en-28-oic acid (8). Among them, compounds 1 and 3 were new oleanane-type triterpenoids, while compounds 2, 4 similar to 6 and 8 were isolated from this plant for the first time. This is the first report on the chemical constituents from the roots of R. tomentosa.

Abstract:The title compound, alisol B monoacetate or 11-hydroxy-24,25-epoxy-3-oxoprotost-13(17)-en-23-yI acetate, C32H50O5, is a protostane-type triterpenoid which was isolated from the Chinese herbal medicine Alismatis Rhizoma (the rhizome of Alisma orientalis Juzep.). The molecule contains three six-membered rings, two of which adopt slightly distorted half-chair conformations while the third exhibits a chair conformation, and one five-membered ring, which adopts an envelope conformation. There is an intermolecular hydrogen bond between the hydroxy and epoxy groups, forming zigzag molecular chains along the b axis.
