Abstract:OBJECTIVE To establish an on-line sweeping proconcentration method in capillary electrophoresis for determination of the trace strychnine in Chinese patent medicine.METHODS On-line sweeping proconcentration method in capillary electrophoresis was used with running buffer of 50 mmol·L-1 NaH2PO4-90 mmol·L-1 SDS-acetonitrile(83:83:34,pH2,injection time of 3.7 min at 0.5 psi and an applied voltage of 25 kV at 25 ℃.The detection wavelength was 205 nm.RESULTS The linearity of strychnine was obtained over the range of 0.5-8.0 μg·mL-1(r=0.9998) and the detection limit was 0.020 μg·mL-1.The mean recovery of this method ranged form 97.6% to 103.4%.The intra and inter day precision of strychnine was no more than 1.8%.CONCLUSION This method is rapid,accurate and effectively improved the sensitivity of capillary electrophoresis.
