Animal model of sudden-onset sensorineural hearing loss accompanied by vertigo induced by mitochondrial toxin

Abstract:Objective To establish an animal model for sudden-onset sensorineural hearing loss(SSNHL) for elucidating the mitochondrial mechanism and exploring the treatment. Methods A gel foam saturated with 3-nitropropionic acid(3-NP) was placed on the round window membrane and kept for 30 min in 49 male guinea pigs either at low dose(0.3 mmol/L) or at high dose(0.5 mmol/L).Ten control guinea pigs were tested by placing gel foam immersed in phosphate buffered saline(PBS) on the round window with respected time.In histological evaluation JB4 embedding was used for light microscopy. Results All animals demonstrated from profound hearing loss to total ABRs loss after 3-NP exposure.Spontaneous horizontal nystagmus with the fast phase beating away from the treated ear was developed in all animals treated with high dose 3-NP and 20%(1/5) of those exposed to low dose of 3-NP.Various degrees of gait disturbances typical for unilateral vestibular dysfunction were seen in all animals exposed to high dose of 3-NP and in 40%(2/5) of those exposed to low dose of 3-NP.The vestibular symptom resolved within 2 days following 3-NP exposure.Histologically,temporary edema was found throughout the organ of Corti 3 days after 3-NP administration.Cytoplasm and nucleus vacuolation in the spiral ganglion cells started within 12 hours post-3-NP exposure and signs of degeneration were visible at day 7. Conclusion Administration of 3-NP into the inner ear through the round window membrane can reproduce the clinical manifestation of SSNHL,and spiral ganglion damage is the most prominent pathological change in this model.
