Enzyme-Enzyme Interaction in the Chloroplast: Physical Evidence for Association Between Phosphoglycerate Kinase and Glyceraldehyde-3-Phosphate Dehydrogenase in Vitro

Abstract:Protein-protein hybridization, fluorescence anisotropy, ultrafiltration, and isoelectric focusing experiments provide evidence for physical interaction between the pea (Pisum sativum L.) chloroplast reductive pentose phosphate pathway enzymes P-glycerate kinase (EC 2.7.2.3) and glyceraldehyde-3-P dehydrogenase (EC 1.2.1.13) in vitro. Association between these two enzymes, suggested by previous kinetic experiments (J. Macioszek, J.B. Anderson and L.E. Anderson (1990) Plant Physiol., 94, 291–296), would presumably make CO2 fixation possible despite enzyme and substrate levels within the chloroplast which appear to be too low to support operation of the Calvin cycle (I.M. Marques, D.M. Ford, G. Muschinek and L.E. Anderson (1987) Arch. Biochem. Biophys., 252, 458–466).
