Abstract:Aromatic aldehydes are very important organic intermediates,which are widely used in pharmaceuticals,pesticides and other areas.In this paper,an improved synthetic method of aromatic aldehydes containing electrowithdrawing groups in ortho or para(or both) position of the aromatic ring was introduced.4-choloro-2-nitrotoluene,4-nitrotoluene and 4-cyanotoluene were used as raw materials,respectively,via enamination with N,N-dimethylformamide dimethyl acetal(DMFDMA) in the present of N,N-dimethylformamide(DMF) as solvent and under reflux,the corresponding styrylamine compounds were produced;then the intermediates were further oxidized by H2O2 under different reaction conditions.The effects of solvent species,reaction temperature and the used amount of H2O2 on the product yield were studied,and the optimal reaction conditions were determined.Under those optimal conditions,the best yields of different aldehydes are 75.6%(4-chloro-2-nitrobenzaldehyde),58.3%(4-nitrobenzaldehyde) and 68.2%(4-cyanobenzaldehyde),respectively with the advantages of simple process,low cost and being environment-friendly..The main intermediates and objective products were confirmed by HPLC and 1H-NMR.

Abstract:An efficient protocol for chemoselective synthesis of 2-substituted benzimidazoles from a variety of aliphatic/aromatic/heteroaryl aldehydes and o-phenylenediamine derivatives promoted by NaHSO3 in water had been developed. The amount of NaHSO3 had a great effect on the reaction selectivity of 2-substituted benzimidazole and 1,2-disubstituted benzimidazole when the reaction was carried out in water. When the amount of the NaHSO3 was more than 11 equivalents, the 2-substituted benzimidazole could be highly selectively formed as the sole product. NaHSO3 was firstly reacted with aldehyde to form the aldehyde sodium bisulfite, which reacted with o-phenylenediamine to form the 2-substituted benzimidazole and inhibited the formation of 1,2-disubstituted benzimidazole. This protocol solved the poor selectivity problem appearing in traditional method when cyclocondensation between o-phenylenediamine and aldehydes. The method also had advantage of simple work up by filtrating the single 2-substituted benzimidazole precipitates from reaction mixture at the end of the reaction without further purification. In addition, the method was applicable to both electron-rich and electron-poor starting materials, which was successfully used for synthesizing nine novel 2-substituted benzimidazole derivatives containing a 1,2,3-triazole moiety. They were characterized by NMR, IR and HRMS spectrum. Moreover, this method had been applied to a large scale synthesis of 2-substituted benzimidazole derivatives.

Abstract:A general and transition-metal-free method for the construction of the carbazol-4-amine motif via a vinylogous Michael addition/cyclization/isomerization/elimination reaction of 3-nitroindoles with alkylidene malononitriles has been developed. This novel methodology allows the facile synthesis of a series of di- and trisubstituted carbazol-4-amine derivatives in moderate to good yields. A gram-scale experiment was successfully performed, highlighting the practicability of this method. Moreover, this strategy is also applicable to 3-nitrobenzothiophene, affording the corresponding dibenzo[ b, d]thiophen-1-amine derivatives in moderate yields.

Abstract:A strategy for synthesis of β‐aryl‐α‐naphthols has been established through an iron‐promoted domino C(sp3)–H/C(sp)–H and C(sp2)–H/C(sp)–H dehydrogenative coupling of deoxybenzoins and alkynes. The synthesis uses inexpensive materials with a broad substrate scope and features simple operations with excellent regioselectivity. This study provides an alternative access to various β‐aryl‐α‐naphthols and the synthetic protocol should have potential applications in medicinal, materials science, and related areas.

Abstract:A family of optically pure 7,7'-disubsituted-2,2'-dihydroxy-1,1'-dinaphthyls have been conveniently prepared from easily accessible 7-alloxyl-2-naphthol by reaction sequences beginning with the asymmetric oxidative coupling and followed by key synthetic steps including deprotection, alkylation, and Suzuki coupling. Application of these binaphthols in catalytic phenylacetylene addition to aldehydes resulted in excellent enantioselectivities.

Abstract:Organocatalytic dearomatization of β‐naphthols has been achieved via an 1,6‐conjugated addition of 2‐naphthols to alkynyl 7‐methylene‐7H‐indoles in situ generated from α‐(7‐indolyl)methanols. In the presence of racemic phosphoric acid, a series of tetrasubstituted allenes was obtained in high yields. Particularly, with the aid of chiral phosphoric acid, asymmetric dearomatization of β‐naphthols was investigated, affording axially chiral tetrasubstituted allenes with moderate enantioselectivity.

Abstract:A direct regioselective benzoyloxylative dearomatization of both α- and β-naphthols by benzoyl peroxide under an air atmosphere, and radical inhibitor- and catalyst-free conditions at room temperature is described. The methodology provides a new efficient strategy for the construction of α-ketol derivatives bearing an oxo-quaternary carbon center from naphthols with good to excellent yields.

Abstract:A catalytic, regioselective Friedel-Crafts alkylation of beta-naphthol with allylic alcohols has been developed. This procedure allows for selective α-alkylation of β-naphthol with a p-toluenesulfonic acid catalyst. This transformation demonstrated functionalized naphthol synthesis under mild reaction conditions with high product yields – 20 examples with up to 96% yields. The synthetic utility further proved the versatility of the allyl naphthol products

Abstract:A novel route for dinitrification of N-phenylbenzenesulfonamide to synthesize N-(2,4-dinitrophenyl)benzenesul- fonamide has been developed. The method features convenient operation and good functional group tolerance. This reaction may go through a nitrogen dioxide radical (NO2 center dot) intermediate, which is generated by the thermal reaction process of sodium nitrate and ammonium persulfate. In addition, it provides direct approach for the preparation of 2,4-dinitroaniline derivatives which are crucial intermediate for the synthesis of benzimidazoles and quinoxaline derivatives.

Abstract:A catalyst‐free intermolecular dearomatization reaction of β‐naphthols with hypervalent‐iodine‐based nitrooxylating reagent is reported. Various nitrooxylated β‐naphthalenones bearing a quaternary carbon stereogenic center were obtained smoothly in good to excellent yields (up to 87%) under mild conditions. Abstract:A one-pot [3+3] aldol-SNAr-dehydration annulation sequence was utilized to fuse hindered phenols onto aromatic substrates. The transformation joins doubly activated 1,3-disubstituted acetone derivatives (dinucleophiles) with C5-activated 2-fluorobenzaldehyde SNAr acceptors (dielectrophiles) in the presence of K2CO3 in DMF at 65–70 °C to form polysubstituted 2-naphthols and 7-hydroxyquinolines. The reaction is regioselective in adding the most stable anionic center to the aldehyde followed by SNAr closure of the less stabilized anion to the electron-deficient aromatic ring. Twenty-seven examples are reported, and a probable mechanism is presented. In two cases where SNAr activation on the acceptor ring was lower (a C5 trifluoromethyl group on the aromatic ring or a 2-fluoropyridine), diethyl 1,3-acetonedicarboxylate initiated an interesting Grob-type fragmentation to give cinnamate esters as the products.

Abstract:A novel proline derivative, (4R)-4-(beta-Naphthalenyl)methoxy-(S)-proline, was conveniently prepared from the naturally occurring (4S)-hydroxy-(S)-proline; 5 mol % of this compound efficiently catalyzes the asymmetric aldol reactions of various benzaldehydes with acetone in excess of acetone as the solvent, giving the aldol adducts in good yields with ee up to 89.8%.

Abstract:An efficient synthesis for polysubstituted benzenes was successfully developed by the reaction of ninhydrin (=2,2-dihydroxyindane-1,3-dione), malononitrile (=propanedinitrile), and alkylidenemalononitrile. The method involves vinylogous Michael addition of alkylidenemalononitrile to 2-(1,3-dioxo-1H-inden-2(3H)-ylidene) malononitrile, which formed by condensation of malononitrile and ninhydrin in the presence of Et3N, and the alcoholic solvent has participated in the reaction as a reagent. The method has the advantages of good yields and of not requiring a metal catalyst. The structures were confirmed spectroscopically (IR, H-1- and C-13-NMR, and EI-MS) and by elemental analyses, and, in the case of 2c, by X-ray crystallography. A plausible mechanism for this reaction is proposed (Scheme).

Abstract:Described here is a modular strategy for the rapid synthesis of beta-functionalized electron-rich naphthalenes, a family of valuable molecules lacking general access previously. Our approach employs an intermolecular benzannulation ofin situgenerated isobenzopyrylium ions with various electron-rich alkynes, which were not well utilized for this type of reaction before. These reactions not only feature a broad scope, complete regioselectivity, and mild conditions, but also exhibit unusual product divergence depending on the substrate substitution pattern. This divergence allows further expansion of the product diversity. Control experiments provided preliminary insights into the reaction mechanism.

Abstract:[GRAPHICS]A novel multicomponent reaction of arynes, beta-keto sulfones, and Michael-type acceptors is presented, providing an efficient method for the synthesis of polysubstituted naphthols and polysubstituted naphthalenes. Further investigation suggests that the tandem reaction may proceed via a sequential nucleophilic attack to arynes, intramolecular nucleophilic substitution followed by a Michael addition, and a ring closure-elimination process.
