Abstract: Objective To develop a radioimmunoassay(RIA) for human serum liver F protein, and investigate its clinical significance in liver diseases. Methods The homogenate of liver tissue were centrifuged at 4℃, 105,000g.The supernatants were isolated with chromatography on Sepharose 6B and subsequently subjected to DEAE-cellulose column. The purified F protein was obtained with isoelectric focusing method.1 A polyclonal antiserum was raised in NZ rabbits immunized with purified human liver F protein. The purified F protein was labeled to 125I with Iodogen method. Human liver F protein RIA kits were made from it. The serum F protein concentrations were examined in 116 healthy adults and 167 patients with liver diseases or gastrointestinal carcinomas. Results Antiserum affinity constant(Ka) was 1.33×1010L/mol. Iodination percentage of F protein was 34.8%,the specific activity of the product was 3548GBq/g,the radiochemical purity 92.0%,ED50(22.39±4.15)μg/L, NSB<5.0%. The sensitivity was 1.26μg/L, and the assay covered range 0～80μg/L. The intra-as31say and inter-assay coefficient of variation(CV) were 4.1%and 8.5%, respectively. The assay recovery was (98.77±2.87)%. The normal value was (14.52±4.15)μg/L in 116 cases of healthy adults. The serum F protein concentration was obviously elevated(P<0.01)when liver cells were damaged. The F protein was found in 89.2% patients with acute hepatitis, 85.4% hepatic carcinoma, 79.6% alcoholic hepatitis, and 64.5% liver cirrhosis. Conclusion RIA of F protein is a sensitive、specific, and reliable means for studying the role of F protein in various liver diseases. F protein might be a new marker that could be used to show the severity of liver damages.
